Promutagen activation in parasite-infected organisms: preliminary observations with Fasciola hepatica-infected mice and aflatoxin B1.

Mutagenic activity of aflatoxin B1 (AFB1) was followed in the Ames Salmonella/microsome test using liver S-9 from uninfected and Fasciola hepatica infected mice. The S-9 preparations from parasite-infected animals were capable of inducing significantly greater AFB1 mutagenic activity to strain TA100 than S-9 preparations from either control, uninfected mice or Aroclor 1254-induced mice. The differences in activity between infected and uninfected mice were consistent between sex and strains of mice studied. The observed increase in biological activity was apparently due to an induction phenomen

Toxic effects of aflatoxin B1 in male Holstein calves with prior infection by flukes (Fasciola hepatica).

This experiment was designed to compare 3 dose levels of aflatoxin B1 (0.0, 0.5, and 1.0 mg/kg of body weight) and 2 infection levels of Fasciola hepatica metacercariae (0 and 220) to determine whether an additive effect from aflatoxin B1 occurs when fascioliasis is present in dairy calves. Twenty-four male, Holstein calves, 4 weeks old, and averaging 45.8 kg each, were assigned at random to 6 treatment groups, 4 calves per group: group 1--negative control; group 2--0.5 mg of aflatoxin B1/kg; group 3--1.0 mg of aflatoxin B1/kg group 4--220 metacercariae; group 5--220 metacercariae plus 0.5 mg

The tea specific pretreatment methods for rapid detection of aflatoxin B1 and ochratoxin A on dark tea samples.

Rapid test strips have been widely used for mycotoxins detection in foods; however, their application in tea is strictly limited by pronounced matrix interference. In this study, tea-specific cleaning methods were developed to enable rapid detection of aflatoxin B1 (AFB1) and ochratoxin A (OTA) in dark tea and tea infusions. The proposed methods achieved recoveries ranging from 81.5% to 128.07% for AFB1 and ranging from 80.05% to 131.48% for OTA. Method validation demonstrated robust accuracy and precision, with AFB1 recoveries of 83.82%-110.00% (RSDs: 0.46%-12.74%) and OTA recoveries of 82.83

Effect of Aflatoxin B1 on the Nervous System: A Systematic Review and Network Analysis Highlighting Alzheimer's Disease.

Exposure to aflatoxin (AF) triggers the production of inflammatory molecules and free radicals, leading to chronic inflammation, cancer, and neurodegenerative diseases. This systematic review evaluated the effects of AFB1 on the nervous system, particularly focusing on Alzheimer's disease (AD). A comprehensive search was conducted in Scopus, Cochrane Library, PubMed, and Web of Science databases up to 1 June 2024, without restrictions. From 993 records retrieved, 16 articles were included in the systematic review. AFB1 participates in various biochemical processes and pathological conditions.

Effect of Curcumin as Feed Supplement on Immune Response and Pathological Changes of Broilers Exposed to Aflatoxin B1.

In this study, we examined the protective effects of curcumin against the AFB1-induced immune response of and pathological changes in broilers. Histopathology examinations showed that at day 28, AFB1 (5 mg/kg) exposure leads to severe histological changes in the spleen, thymus and bursa of Fabricius with a decrease in the number and karyoplasmic area ratio of plasma cells. Curcumin alleviated the AFB1-induced immune organs' damage as well as the changes in plasma cells in a dose-dependent manner. RT-PCR data showed that AFB1 significantly downregulated the IL-2 and IFN-γ mRNA expression levels

Evaluation of berberine efficacy in reducing the effects of aflatoxin B1 and ochratoxin A added to male broiler rations.

Many types of mycotoxins are found in food sources contaminated with fungi, and if these are ingested in large quantities or over a long period, they can affect the health of humans and domestic animals. Berberine (BBR) is a plant alkaloid with multiple pharmacological functions. This study aimed to investigate the effect of different levels of the plant alkaloid BBR on reducing toxic effects of aflatoxin B1 (AFB) and ochratoxin A (OTA) in broilers by examining performance characteristics, blood biochemistry, antioxidant systems, ileum morphology, and histopathology of the liver. The isolation and culture of DHBV-infected embryo and duckling hepatocytes and the effect of aflatoxin B1 or irradiation on these cells.

The preparation of primary cultures of control and DHBV-infected duck hepatocytes from embryos and young ducklings is described. Cultures of both embryo and duckling hepatocytes secreted duck serum proteins. Cultures of hepatocytes established from ducklings maintained initial morphology for up to 3 weeks in culture and also exhibited high levels of metabolism of aflatoxin B1. Embryonic cell cultures rapidly lost ability to metabolise AFB1 and became overgrown by spindle-shaped cells. Both embryo and duckling cell cultures secreted infective DHBV, and had intracellular replicative forms of the

[Various aspects of the effect of nutritional factors on the metabolism and biological action of aflatoxin B1 in the rat liver].

Interrelationships between the quality of nutrition, oxidative hydroxylation of aflatoxin B1 and lipid peroxidation were studied in biomembranes of hepatocyte endoplasmic reticulum of growing male rats of the WAG strain. Activity of the liver microsomal monooxygenase, lipid peroxidation as well as metabolism of aflatoxin B1 were shown to depend on presence of three essential amino acids (lysine, methionine and threonine) and vitamins A, E and C. Deficiency of these compounds altered the aflatoxin metabolism causing formation of highly reactive metabolites, which might accelerate the carcinogen

The metabolism of aflatoxin B1 by hepatocytes isolated from rats following the in vivo administration of some xenobiotics.

Isolated rat hepatocytes, an intact cellular system capable of performing phase I and phase II metabolism, have been used to investigate metabolism of aflatoxin B1. These cells were found to metabolise [14C]aflatoxin B1 to aflatoxins M1 and Q1, and to radiolabelled polar material, presumably conjugates, as analysed by h.p.l.c., t.l.c. and radioactive determination. In vivo administration of the mixed function oxidase inducers, phenobarbitone and 3-methylcholanthrene, resulted in enhanced hepatocyte phase I (microsomal) metabolism of aflatoxin B1. In contrast to metabolism of AFB1 by in vitro s

A ratiometric fluorescent probe with distinct response to Aflatoxin B1 and B2a: probe design, bioimaging, and quantification.

Aflatoxin B1 (AFB1) and its photo-degraded metabolite Aflatoxin B2a (AFB2a) are both highly toxic, yet most analytical methods focus only on AFB1, neglecting AFB2a. We developed a human serum albumin (HSA)-based fluorescent probe that exhibits distinct ratiometric responses to AFB1 and AFB2a, which can be owing to the different binding preference of two analytes that facilitates the distinguished sensing mechanisms: indicator-displacement assay for AFB2a and indicator-analyte co-existence for AFB1. This probe enables quantitative, on-site detection of both toxins in corn-based food extracts vi

A target-responsive and signal-amplified DNA hydrogel biosensor for visual and quantitative detection of aflatoxin B1.

Aflatoxin B1 (AFB1) poses a severe threat to food safety due to its potent carcinogenicity. To address the need for rapid and on-site detection, this study developed a colorimetric biosensor based on an AFB1-responsive DNA aptamercross-linked sodium alginate hydrogel. Gold nanoparticles (AuNPs) were encapsulated within the hydrogel as a visual signal reporter. The sensing mechanism relies on target-induced dissociation of aptamer crosslinkers, leading to hydrogel disintegration and release of AuNPs. The hydrogel formulation was optimized regarding sodium alginate concentration and crosslinking

Preparation and utilization of an immunoadsorption column for aflatoxin B1 composed of nanobodies and pre-crosslinked agarose microspheres.

Recent studies on immunoaffinity chromatography (IAC) columns for aflatoxin B1 (AFB1) have not provided a comprehensive preparation procedure or explored their capacity, reusability, and stability. In this study, we outline a preparation process for an AFB1 immune affinity column filler (AFB1-IAC) for extracting AFB1 from food sources like rice, peanuts, and soybeans. Using commercially available IAC is often limited due to high costs, low capacity, slow flow rates, and poor reusability. To overcome these issues, we developed succinic anhydride (SA)-modified agarose pre-crosslinked microsphere

The Ameliorative Role of Lico A on Aflatoxin B1-Triggered Hepatotoxicity Partially by Activating Nrf2 Signal Pathway.

Aflatoxin B1 (AFB1) is one of the most harmful and toxic mycotoxins in foods and feeds, posing a serious health risk to both humans and animals, especially its hepatotoxicity. Nuclear factor-erythroid 2-related factor 2 (Nrf2), an important nuclear transcription factor, is generally recognized as a potential target for phytochemicals to ameliorate liver injury. The current study sought to elucidate the molecular processes by which licochalcone A (Lico A), a compound derived from Xinjiang licorice Glycyrrhiza inflate, protects against AFB1 toxicity. In vivo, male wild-type (WT) and Nrf2 knockou

A semiconductor quantum dot-based ratiometric electrochemical aptasensor for the selective and reliable determination of aflatoxin B1.

In recent years, a ratiometric electrochemical method has been investigated due to its ability to effectively reduce the background electrical signals via the introduction of an internal calibration mechanism, which has great practical significance in the detection of mycotoxins in foods. Herein, we report a ratiometric electrochemical aptasensor based on two semiconductor quantum dots (i.e. CdTe and PbS QDs) for the detection of aflatoxin B1 (AFB1). The aptasensor was fabricated by immobilizing PbS QD-coated silica hybrid spheres (SiO2@PbS) onto CdTe QD-modified Fe3O4@SiO2 (Fe3O4@SiO2/CdTe) s

Acute oral toxicity test and assessment of combined toxicity of cadmium and aflatoxin B1 in kunming mice.

Cadmium and aflatoxin B1 (AFB1) are both common and widespread pollutants in food and feed. There are several reports on toxicity induced by Cadmium or AFB1 alone, but few address the toxicity caused by co-exposure to the two substances. In this study, 42 female and 42 male Kunming (KM) mice were divided into seven groups to test the acute oral toxicity of CdCl2 and AFB1, using Karber's method. The combined toxicity was assessed using the Keplinger evaluation system. Acute toxicity symptoms, deaths, and body and organ weights were evaluated, and hematological, blood biochemical, and histopatho

Aflatoxin B1 levels, dietary exposure and cancer risk assessment in sesame and nut-based foods in Türkiye.

Aflatoxin B1 (AFB1), a highly toxic mycotoxin commonly found in oilseeds and nuts, poses serious public health and economic risks. Türkiye's climatic conditions are conducive to AFB1 contamination, particularly in sesame- and nut-based foods. This study investigated the presence of AFB1 in 100 widely consumed samples of tahini, tahini halva, hazelnut paste, and peanut paste. AFB1 was detected in 67% of the samples, with exceedances of the European Commission (EC) limit (2 µg/kg) notably high in tahini halva (80%) and peanut paste (60%); in contrast none of the hazelnut paste samples surpassed
