BACKGROUND: The postpartum administration of an adequate amount of anti-D immunoglobulin to the mother in cases of Rhesus incompatibility requires the exact quantification of the amount of Rh-positive fetal cells that may be present in the Rh-negative maternal circulation. The classical methods to detect an intrapartum fetomaternal hemorrhage are either time-intensive (such as the Kleihauer-Betke test), of low specificity (such as the indirect Coombs test), or technically cumbersome (such as flow cytometry). The goals of our study were to develop a simple screening test that may be used routin

The card agglutination test for trypanosomiasis (CATT) was evaluated and compared to the classical immunofluorescent antibody test (IFAT) in the immunological diagnosis of Gambian trypanosomiasis. Tests were performed on serum and whole blood. Cross-reactions were found in the CATT with sera from patients suffering from parasitic infections other than sleeping sickness, but could be largely overcome by selecting 1/10 as the specific threshold dilution. At 1/40 dilution no false positive result was observed in the IFAT. At the specific threshold dilution, the sensitivity of IFAT was 94.7%, comp

A passive haemagglutination test, using sheep red blood cells sensitised with Salmonella typhi lipopolysaccharide, was compared with the Widal test for the serological diagnosis of typhoid fever in an endemic area. The results obtained on sera from 152 patients with bacteriologically confirmed typhoid and 183 patients who did not have typhoid were analysed in terms of sensitivity, specificity, simplicity, and rapidity of the respective tests. The passive haemagglutination test was found to be more sensitive (80%) than the S typhi O antigen (71%) but marginally less sensitive than the H antigen

We evaluated the field use of two serologic tests for visceral leishmaniasis (VL), the direct agglutination test (DAT) and rK39 dipstick test, in the context of a case-control study. Most VL cases in Nepal are currently diagnosed on clinical grounds and with relatively non-specific tests such as the formol-gel test. Among 14 newly diagnosed VL patients with bone-marrow slides confirmed positive in two independent laboratories, the sensitivity of both tests was 100%. Among 113 controls with no personal or household history of VL, the specificity of the rK39 was 100% while that of the DAT was 93

In our nursery, we identified neonates with positive urine latex particle agglutination (LPA) tests for group B streptococcus (GBS) antigen who did not have corroborating cultural evidence of infection. To investigate the mechanisms underlying these apparent false-positive reactions, we examined the urine LPA test in an unselected population of neonates suspected of sepsis. Urine specimens for LPA testing and culture and simultaneous perirectal cultures were obtained from 134 neonates with suspected sepsis. Six infants had positive blood cultures for GBS; four of the six were positive by LPA t

Fifty-one calves were divided into six trial groups of seven to eleven animals and vaccinated with a commercial leptospiral vaccine containing serovars pomona and hardjo. Vaccinations were given at 6,7,14 or 21 months of age and animals in various groups were vaccinated on one to four occasions. Antibody responses were determined by the microscopic agglutination test (MAT) and the complement fixation test (CFT) at one to four weeks intervals until 66 weeks after the start of the trial. Fifty percent or greater agglutination in serum diluted 1:100 or more and 50% or greater fixation of compleme

Brucellosis is a zoonotic disease causing significant economic losses, and necessitating standardized diagnostic kits for effective surveillance. The objectives of this study are to investigate brucellosis serosurveillance in ruminants, and to evaluate three serological tests-Rose Bengal Plate Tests (RBPTs), Serum Tube Agglutination Tests (STATs), and Enzyme-Linked Immunosorbent Assays (ELISAs)-each from three manufacturers. A total of 5797 serum samples (5260 sheep, 83 bovines, and 454 spotted deer) were initially screened with the RBPT, and 54 RBPT-positive selected samples were subsequently

Background/aim: It was aimed to evaluate the results of Rose Bengal (RB), ELISA total tests (IgM and IgG), and the Brucella Coombs gel test (BCGT), which are used as screening tests, with the combined results of a tube agglutination test (standard Wright test: SWT) and a tube agglutination test with antihuman globulin (AHG TAT). Materials and methods: Samples from 97 patients prediagnosed with brucellosis (age ≥18 years) were screened with RB, ELISA, and BCGT. SWT < 160 samples and RB-negative but ELISA- or BCGT-positive samples were tested by AHG TAT. SWT ≥ 160 or AHG TAT ≥ 160 was taken as r

In this study, 241 serum samples from individuals exposed to brucellosis were subjected to the Rose Bengal plate test (RBPT); the titre was estimated by standard tube agglutination test (STAT), with positive > or = 80 IU/ml. Randomly selected sera (n = 81) were analysed by complement fixation test (CFT): titre > or = 1:4 was considered positive. Of 241 sera subjected to RBPT and STAT, 177 were negative in both tests; 5 samples tested negative by RBPT but positive by STAT. None was positive by RBPT and negative by STAT. Of 81 sera subjected to CFT, 23 (28.4%) were positive. Both RBPT and CFT fo

Paracoccidioidomycosis is diagnosed from the direct observation of the causative agent, but serology can facilitate and decrease the time required for diagnosis. The objective of this study was to determine the influence of serum sample inactivation on the performance of the latex agglutination test (LAT) for detecting antibodies against Paracoccidioides brasiliensis. The sensitivity of LAT from inactivated or non-inactivated samples was 73% and 83%, respectively and the LAT selectivity was 79% and 90%, respectively. The LAT evaluated here was no more specific than the double-immunodiffusion a

Paracoccidioidomycosis (PCM) is a fungal disease caused by Paracoccidioides brasiliensis, and Brazil is one of the principal countries where it is endemic. Diagnosis is based on the observation of budding P. brasiliensis yeast in clinical specimens from patients; however, the sensitivity of the visualization of fungi is low, indicating that serological tests are used for early diagnosis. The double-immunodiffusion test (ID) is the "gold standard" test for serology in PCM, although the execution of this test requires the availability of laboratorial infrastructure. We report the improved perfor

Serum and urine samples from 30 cows (15 pregnant and 15 nonpregnant) from each of 10 Georgia dairy herds (total cows = 300) were examined by microscopic agglutination testing (MAT) and direct fluorescent antibody testing (FAT), respectively. Seven of the 10 herds had at least 1 cow with a positive FAT, and all of the herds had at least 1 cow with a reciprocal MAT titer > or =100 for 1 or more serovars. Serological testing was not helpful in identifying the infecting serovar for cows with a positive FAT result. The MAT titers for all 7 of the serovars evaluated were significantly correlated wi

We previously standardized the direct agglutination test (DAT) to detect anti-Leishmania chagasi promastigote antibodies (DAT-LPC) with good sensitivity and specificity for diagnosing visceral leishmaniasis (VL). In this paper, we present a technical upgrade by introducing some modifications into the antigen preparation. This antigen was evaluated in DAT (DAT-Mod) using 61 sera samples from VL patients and 96 samples from patients with other diseases. The DAT-Mod presented a cut-off of 1:100, satisfactory reproducibility (VC <5.8), sensitivity of 93.4%, specificity of 96.9%, and diagnostic eff

A prospective study in Thailand identified 106 patients with culture-proven leptospirosis. The accuracy of the microscopic agglutination test (MAT) in predicting the infecting serovar was evaluated in 78/106 (74%) patients with a diagnostic titer. MAT correctly determined the infecting serovar in 26 cases (33%), indicating that this assay is a poor predictor of infecting serovar in our setting.

INTRODUCTION: Visceral leishmaniasis is the most severe clinical form of leishmaniasis and is often fatal without proper treatment. Therefore, early and accurate diagnosis is important, but often difficult in endemic areas.
OBJECTIVE: The aim was to evaluate a direct agglutination test as a potential visceral leishmaniasis diagnostic method in endemic areas of Venezuela.
MATERIALS AND METHODS: The performance of the direct agglutination test, based on freeze-dried Leishmania donovani antigen was evaluated under laboratory conditions using serum samples of humans and dogs from several Venezue
