Fatty acid amide hydrolase (FAAH) is a critical metabolic enzyme in the endocannabinoid system. Through regulating the metabolism of lipid signaling molecules, FAAH is involved in a variety of physiological and pathological processes, including cell proliferation, inflammatory responses, and metabolic homeostasis. Polycystic ovary syndrome (PCOS), one of the most common endocrine and metabolic disorders affecting women of reproductive age, is closely associated with abnormal follicular development, dysregulated hormone secretion, insulin resistance, dyslipidemia, and inflammatory imbalance. Mycobacterium tuberculosis (MTB), the pathogen responsible for tuberculosis (TB), remains a significant global health concern, especially with the growing prevalence of multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains. This study focuses on understanding the molecular basis of pyrazinamide (PZA) resistance, particularly mutations in the pyrazinamidase (Pzase) enzyme, including D8G, H71R, K96T, and S104R. We used computational methods to explore the effects of bioactive compounds on these PZA-resistant mutations. The structures of wild-type (WT) Pzase and its mutant varian

A promising yet clinically unexploited antibiotic target in difficult-to-treat Gram-negative bacteria is LpxC, the key enzyme in the biosynthesis of lipopolysaccharides, which are the major constituents of the outer membrane. Despite the development of dozens of chemically diverse LpxC inhibitor molecules, it is essentially unknown how bacteria counteract LpxC inhibition. Our study provides comprehensive insights into the response against five different LpxC inhibitors. All compounds bound to purified LpxC from Escherichia coli. Treatment of E. coli with these compounds changed the cell shape

MCH1 is a synthetic macamide that has shown in vitro inhibitory activity on fatty acid amide hydrolase (FAAH), an enzyme responsible for endocannabinoid metabolism. This inhibition can modulate endocannabinoid and dopamine signaling in the nucleus accumbens (NAc), potentially having an antidepressant-like effect. The present study aimed to evaluate the effect of the in vivo administration of MCH1 (3, 10, and 30 mg/kg, ip) in 2-month-old BALB/c male mice (n=97) on forced swimming test (FST), light-dark box (LDB), and open field test (OFT) and on early gene expression changes 2 h after drug inje

Here, we describe a clinical case of pyrazinamide-resistant (PZA-R) tuberculosis (TB) reported as PZA-susceptible (PZA-S) by common molecular diagnostics. Phenotypic susceptibility testing (pDST) indicated PZA-R TB. Targeted Sanger sequencing reported wild-type PncA, indicating PZA-S TB. Whole Genome Sequencing (WGS) by PacBio and IonTorrent both detected deletion of a large portion of pncA, indicating PZA-R. Importantly, both WGS methods showed deletion of part of the primer region targeted by Sanger sequencing. Repeating Sanger sequencing from a culture in presence of PZA returned no result,

The endocannabinoid system is prominently implicated in the control of cocaine reinforcement due to its relevant role in synaptic plasticity and neurotransmitter modulation in the mesocorticolimbic system. The inhibition of fatty acid amide hydrolase (FAAH), and the resulting increase in anandamide and other N-acylethanolamines, represents a promising strategy for reducing drug seeking. In the present study, we aimed to assess the effects of the FAAH inhibitor URB597 (1 mg/kg) on crucial features of cocaine addictive-like behaviour in mice. Therefore, we tested the effects of URB597 on acquisi

A series of hexafluoroisopropyl carbamates with indolylalkyl- and azaindolylalkyl-substituents at the carbamate nitrogen was synthesized and evaluated for inhibition of the endocannabinoid degrading enzymes fatty acid amide hydrolase (FAAH) and monoacylglycerol lipase (MAGL). The synthesized derivatives with butyl to heptyl spacers between the heteroaryl and the carbamate moiety were inhibitors of both enzymes. For investigated compounds in which the alkyl chain was partially incorporated into a piperidine ring, different results were obtained. Compounds with a methylene spacer between the pip

Modulation of the endocannabinoid system (ECS) is a novel putative target for therapeutic intervention in depressive disorders. Altering concentrations of one of the principal endocannabinoids, N-arachidonoylethanolamine, also known as anandamide (AEA) can affect depressive-like behaviors through several mechanisms including anti-inflammatory, hormonal, and neural circuit alterations. Recently, isoflavonoids, a class of plant-derived compounds, have been of therapeutic interest given their ability to modulate the metabolism of the endogenous ligands of the ECS. To determine the therapeutic pot

Antisocial personality disorder (ASPD) imposes a high societal burden given the repetitive reactive aggression that affected individuals perpetrate. Since the brain endocannabinoid system (ECS) has been implicated in ASPD and aggressive behavior, we utilized [11C]CURB positron emission tomography to investigate fatty acid amide hydrolase (FAAH), an enzyme of the ECS that degrades anandamide, in 16 individuals with ASPD and 16 control participants. We hypothesized that FAAH density would be lower in the amygdala for several reasons. First, decreased FAAH expression is associated with increased

PURPOSE: To evaluate the influence of atractylenolide (Atr) III on sepsis-induced lung damage.
METHODS: We constructed a mouse sepsis model through cecal ligation and puncture. These mice were allocated to the normal, sepsis, sepsis + Atr III-L (2 mg/kg), as well as Atr III-H (8 mg/kg) group. Lung injury and pulmonary fibrosis were accessed via hematoxylin-eosin (HE) and Masson's staining. We used terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) and flow cytometry for detecting sepsis-induced lung cell apoptosis. The contents of the inflammatory cytokines in lung tissue we

BACKGROUND: Bacillus cells faced with unfavorable environmental conditions undergo an asymmetric division process ultimately leading to the formation of the bacterial spore. In some instances the spore serves as an infectious agent; such is the case with the spore of Bacillus anthracis and the disease anthrax. Spores are resistant to a variety of environment conditions including traditional decontamination techniques due to the formation of specialized cellular structures. One such structure, the spore cortex, is a thick layer of modified peptidoglycan that contributes to spore dormancy throug

Pyrazinamide (PZA) is a unique antituberculosis (anti-TB) drug that plays a key role in shortening TB therapy. PZA kills nonreplicating persisters that other TB drugs fail to kill, which makes it an essential drug for inclusion in any drug combinations for treating drug-susceptible and drug-resistant TB such as multidrug-resistant TB. PZA acts differently from common antibiotics by inhibiting multiple targets such as energy production, trans-translation, and perhaps pantothenate/coenzyme A required for persister survival. Resistance to PZA is mostly caused by mutations in the pncA gene encodin

Synthetic cannabinoids, the psychoactive components of the Cannabis sativa (marijuana) plant and their endogenous counterparts, act through two G protein-coupled receptors, CB1 and CB2. The endocannabinoids are metabolized by fatty acid amide hydrolase (FAAH). Previous research has described the impact of cannabis consumption on pregnancy, potential roles of endocannabinoids and abnormalities of FAAH expression in recurrent miscarriage and pregnancy. However, the cellular localization of the CB1 cannabinoid receptor and FAAH in the human placenta has not been determined. We have examined CB1 r

The clinical features, causes and potential treatment of Canavan disease are reviewed. The course of the illness can show considerable variation, and can sometimes be protracted. It has an autosomal recessive mode of inheritance, and is caused by mutations in the gene for aspartoacetylase, localized to the short arm of chromosome 17. The changes in the central nervous system are related to the deficiency of aspartoacyclase which leads to an excess of N-acetylaspartate. Prenatal diagnosis of Canavan disease is possible by the measurement of N-acetylaspartate in the amniotic fluid, but the metho

Bacterial chemotactic F-met peptides have been identified in culture supernatants of intestinal bacteria and in human faecal dialysates. These potent inflammatory agents could play a role in intestinal inflammatory disorders should they cross the epithelial barrier of the gut. We have identified mucosal peptidases which degrade F-met-leu-phe (FMLP) in ileal and colonic mucosal biopsies obtained at colonoscopy. A carboxypeptidase, inhibited by D-L-benzyl succinate (BzS), accounted for more than 60% of total FMLP-ase activity, other uncharacterized peptidases contributing the rest of the activit

The effects of a disulfide reducing agent and sulfhydryl blocking agents on the biotinidase activity in human serum and on the purified biotinidase have been extensively studied by using a newly developed HPLC assay method. This HPLC method directly measures the product (p-aminobenzoate, PAB), and is not interfered with by sulfhydryl-reactive agents. Further, because the substrate solution of this HPLC assay method contains only substrate (biotin 4-amidobenzoate) and phosphate buffer, accurate studies on the effects of sulfhydryl blocking reagents on the purified enzyme could be performed. Because there are few persuasive data for selecting one semisynthetic penicillin or cephalosporin over another for treatment of serious staphylococcal infections, 118 recent clinical isolates of Staphylococcus aureus were studied to determine to what extent the presence of beta-lactamase affected the relative anti-staphylococcal activity of six penicillins and seven cephalosporins. In addition, the effect of inoculum was studied for its possible effect on the anti-staphylococcal activity of the 13 beta-lactam antibiotics. By all criteria, methicillin and nafcillin were clearly more resistant t

INTRODUCTION: Moyamoya (MM) is a progressive arteriopathy of the internal carotid artery and its branches that can lead to stroke and can be treated with surgical revascularization. The enzyme Nit2/ω-amidase (NIT2) is involved in glutaminase II and methionine salvage pathways. Recent studies have shown correlations between these metabolic pathways and MM arteriopathy, specifically increases in L-methionine, NO metabolites, and homocysteine. Here, we present data demonstrating that elevated levels of NIT2 correlate with the presence of MM, suggesting that it may have potential utility as a non-

BACKGROUND: Pyrazinamide is an important first-line antibiotic for treating tuberculosis, with resistance primarily driven by mutations in the pncA gene. Traditional machine learning models are able to predict pyrazinamide resistance with some success but are limited in their ability to incorporate 3-dimensional protein structural information. Graph neural networks offer the potential to integrate protein structure and residue-level features to better predict the impact of mutations on drug resistance. RESULTS: We trained a graph convolutional network on PncA variants containing missense mutat
