Investigation of antibacterial mode of action for traditional and amphiphilic aminoglycosides.

Aminoglycoside represents a class of versatile and broad spectrum antibacterial agents. In an effort to revive the antibacterial activity against aminoglycoside resistant bacteria, our laboratory has developed two new classes of aminoglycoside, pyranmycin and amphiphilic neomycin (NEOF004). The former resembles the traditional aminoglycoside, neomycin. The latter, albeit derived from neomycin, appears to exert antibacterial action via a different mode of action. In order to discern that these aminoglycoside derivatives have distinct antibacterial mode of action, RNA-binding affinity and fluoro

Evaluation of extended interval dosing aminoglycosides in the morbidly obese population.

Aminoglycoside dosing has been studied in the obese population, typically recommending an adjusted weight utilizing a 40% dosing weight correction factor (IBW + 0.4 × (TBW-IBW)). These studies included limited numbers of morbidly obese patients and were not done in the era of extended interval aminoglycoside dosing. Here, we report a retrospective evaluation of morbidly obese patients receiving gentamicin or tobramycin at our hospital. The objective of this study was to evaluate the accuracy of the commonly recommended adjusted weight for weight-based dosing. There were 31 morbidly obese patie

Engineering an allosteric binding site for aminoglycosides into TEM1-β-Lactamase.

Allosteric regulation of enzyme activity is a remarkable property of many biological catalysts. Up till now, engineering an allosteric regulation into native, unregulated enzymes has been achieved by the creation of hybrid proteins in which a natural receptor, whose conformation is controlled by ligand binding, is inserted into an enzyme structure. Here, we describe a monomeric enzyme, TEM1-β-lactamase, that features an allosteric aminoglycoside binding site created de novo by directed-evolution methods. β-Lactamases are highly efficient enzymes involved in the resistance of bacteria against β

TRPA1-mediated accumulation of aminoglycosides in mouse cochlear outer hair cells.

Aminoglycoside ototoxicity involves the accumulation of antibiotic molecules in the inner ear hair cells and the subsequent degeneration of these cells. The exact route of entry of aminoglycosides into the hair cells in vivo is still unknown. Similar to other small organic cations, aminoglycosides could be brought into the cell by endocytosis or permeate through large non-selective cation channels, such as mechanotransduction channels or ATP-gated P2X channels. Here, we show that the aminoglycoside antibiotic gentamicin can enter mouse outer hair cells (OHCs) via TRPA1, non-selective cation ch

[Interaction of aminoglycosides and the other antibiotic on selected bacterial strains].

The aim of this study was to analyse the activity interaction of aminoglycosides (gentamicin, kanamycin, streptomycin and dihydrostreptomycin) when combined with other antibiotics (lincomycin, benzylpenicillin, amoxicillin, cephalexin, spectinomycin and erythromycin), on selected clinical bacterial strains. The checkerboard method has been selected from the traditional assays for the measurement of antibiotic interaction. Checkerboard results for all strains demonstrated synergism for nine cases (9/112--8%). Additive effects were predominant--73/112--65.2%. In 12.5% neutral effects were shown,

Inhibition of the proteolytic activity of anthrax lethal factor by aminoglycosides.

The anthrax lethal factor (LF), a Zn-dependent endopeptidase, is considered the dominant virulence factor of anthrax. Because pharmacological inhibition of the catalytic activity of LF is considered a plausible mechanism for preventing the lethality of anthrax, a high-throughput screening experiment based on LF-catalyzed cleavage of a fluorescent substrate was performed to identify novel inhibitors of LF. The RNA-targeting antibiotics, neomycin B and some synthetic dimeric aminoglycosides, were found to be nanomolar active-site inhibitors of LF.

Bacterial resistance to aminoglycosides and beta-lactams: the Tn1331 transposon paradigm.

Aminoglycosides (Ags) are a group of antibiotics that exert their bactericidal activity primarily by inhibition of protein synthesis. Aminoglycoside (Ag) molecules bind to the bacterial 30S ribosomal subunit rendering the ribosomes unavailable for translation, which results in cell death. Although these antibiotics are and have been very useful to treat a variety of bacterial infections, in recent years the number of Ag resistant and multiresistant isolates has seriously increased. Mechanisms of resistance to Ag include enzymatic inactivation by acetyltransferases, nucleotidyltransferases (ade

In vitro synergism of ceftriaxone combined with aminoglycosides against Pseudomonas aeruginosa.

The antipseudomonal activities of ceftriaxone (CEF) or ceftazidime (CAZ), each combined with tobramycin (TOB) or netilmicin (NET), against 90 clinically significant Pseudomonas aeruginosa isolates were examined both by checkerboard and time-kill assays. As expected, susceptibility testing of single antibiotics by agar dilution demonstrated good activity for CAZ (89% susceptible), TOB (94%), and NET (58%), but poor activity for CEF (15%). Checkerboard studies revealed striking synergy (FIC indices < or = 0.5) for CEF, however, in combination with either TOB (72%) or NET (81%), compared with CAZ

In vitro activities of aminoglycosides, lincosamides, and rifamycins against Mycobacterium leprae.

The in vitro activities of a variety of aminoglycosides, lincosamides, and rifamycins against Mycobacterium leprae were evaluated with the BACTEC 460 system. At 20 micrograms/ml, gentamicin, kanamycin, tobramycin, streptomycin, and amikacin were inactive. Lincomycin was active at 20 micrograms/ml, and clindamycin was active at 0.31 micrograms/ml. Rifamycin SV, rifabutin, and rifampin were active at 3.1, 3.1 to 12.5, and 200 ng/ml, respectively. The in vitro assay correlates well with the in vivo response of M. leprae to antimicrobial agents, with the exception of the aminoglycosides.

Resistance to aminoglycosides: the situation in the Federal Republic of Germany.

The susceptibility of important pathogens to aminoglycosides was analysed using data from five studies encompassing different patient groups. Enterobacteria were generally sensitive to all modern aminoglycosides without marked differences. The frequency of resistance of Pseudomonas aeruginosa and other non-fermenters to gentamicin was 5-15% and to amikacin was 0-3%, and similar values applied for staphylococci. A higher rate of resistance was observed in pathogens isolated from urological patients: for non-fermenters resistance to gentamicin, tobramycin and netilmicin was 24-28%, whereas resis

Activity of cefsulodin, other beta-lactams, and aminoglycosides against Pseudomonas aeruginosa.

Cefsulodin was the most active of the cephalosporins and exhibited 4-16 times more activity than carbenicillin or ticarcillin against 50 clinical isolates of Pseudomonas aeruginosa. Azlocillin and piperacillin showed good activity, while tobramycin was the most effective aminoglycoside. The activity of cefsulodin was unaltered by increases in inocula, but resistance was induced easily. When combined with gentamicin, no synergistic or antagonistic activity was observed against multiply resistant isolates.

Development of a predictive platform for target concentration intervention of aminoglycosides.

Aminoglycosides, including gentamicin, are potent bactericidal antibiotics that are widely utilized for the treatment of severe gram-negative bacterial infections. These drugs demonstrate a concentration-dependent bactericidal effect, whereby higher concentrations result in more rapid bacterial elimination. Despite their efficacy, determining optimal dosing regimens is challenging due to significant inter-individual pharmacokinetic (PK) variability and adverse drug reactions, such as nephrotoxicity and ototoxicity. This study developed a simplified, user-friendly PK-predictive system for targe

Biosynthesis and biotechnological engineering of aminoglycosides: Pathways, enzymes, and industrial prospects.

Aminoglycosides (AGs) are a broad class of antibiotics widely used in medicine since the 1950s. Challenges remain in creating highly stereoselective modifications with traditional chemical methods. Biosynthesis, which involves enzymatic formation of AGs via genetically encoded pathways in microbes, offers a more efficient, selective, and eco-friendly alternative. The biosynthetic routes of key AGs such as neomycin (NEO), kanamycin (KAN), gentamicin (GEN), tobramycin (TOB), apramycin (APR), and sisomicin (SIS) have been mapped out, involving enzymatic steps like aminocyclitol core formation, gl

Aminoglycosides, vancomycin, and metronidazole for people with cirrhosis and hepatic encephalopathy.

RATIONALE: Hepatic encephalopathy is a common complication of cirrhosis. Its development is associated with increased morbidity and mortality. Its exact pathogenesis is unknown, but ammonia, produced by bacterial action in the intestine, plays a key role. Antibiotics modulate the gut flora and may reduce intestinal ammonia production. Aminoglycosides such as neomycin, paromomycin, and ribostamycin have been used to treat hepatic encephalopathy, as have other antibiotics such as vancomycin and metronidazole. OBJECTIVES: To assess the beneficial and harmful effects of aminoglycosides, vancomyci

Sonoporation-enhanced the antibacterial efficacy of aminoglycosides against Gram-negative persisters.

Bacterial persisters pose a significant challenge to antibiotic therapy, often causing treatment failure and chronic infections. Improving antibiotic efficacy against persisters is thus clinically crucial. In this study, we developed a low-frequency and low-intensity ultrasound system (40 kHz, 0.2 W/cm2) to induce sonoporation in bacterial persisters. Combining ultrasound with aminoglycosides (AGs) reduced the persister ratio of Gram-negative bacteria (E. coli, P. aeruginosa, and A. baumannii) by approximately 5- to 15-fold, and notably, this approach achieves eradication below the detection t

Synergistic effect of regorafenib with aminoglycosides in ferroptosis-mediated liver injury.

BACKGROUND: Combination therapy of anticancer drugs with antibacterial agents has numerous advantages, but these may develop adverse drug events that were rarely studied. Specifically, regorafenib (REGO) mediates ferroptosis and causes liver injury when used in combination with aminoglycosides (AGs); however, this interaction remains unexplored. METHODOLOGY: The study was conducted using both in vivo and in vitro assays involving Sprague-Dawley (SD) rats and HepG2/Huh7 to investigate ferroptosis-associated liver injury. The drugs REGO, amikacin (AMK), and gentamicin (GNT) were administered in

Late-Stage Functionalization of Glucosides and Aminoglycosides to Combat Resistant Resistance.

The rapid emergence of resistant bacteria and the scarcity of the antibiotic pipeline have been a persistent threat to global health. To expand the antibiotic pipeline, the strategy has focused on revitalizing the current antibiotic via novel site-specific derivatization. Aminoglycosides (AGs), once considered effective therapeutics for treating clinical infections, are now used to a limited extent due to drug-induced toxicity and AG-resistant bacteria. Challenges in the synthesis and modification of AGs impede their potential for regain efficacy. Herein, a photoredox catalyzed late-stage modif

Site-Selective Palladium-catalyzed Oxidation of Unprotected Aminoglycosides and Sugar Phosphates.

The site-selective modification of complex biomolecules by transition metal-catalysis is highly warranted, but often thwarted by the presence of Lewis basic functional groups. This study demonstrates that protonation of amines and phosphates in carbohydrates circumvents catalyst inhibition in palladium-catalyzed site-selective oxidation. Both aminoglycosides and sugar phosphates, compound classes that up till now largely escaped direct modification, are oxidized with good efficiency. Site-selective oxidation of kanamycin and amikacin was used to prepare a set of 3'-modified aminoglycoside deri
