An In Vitro test battery using human whole blood for immunotoxicity hazard identification: Proof of concept studies with dexamethasone and benzo(a)pyrene.

Immunotoxicity assessment is nearing a crossroads predicated on mounting pressure for reduction/replacement of animals in toxicology. This has fueled the development of alternative New Approach Methodologies (NAMs) for hazard identification. This work details a comprehensive human whole blood NAM battery for immunotoxicity testing. The test system is a closed tube culture containing whole blood diluted 1:3 in culture media with or without an immune stimulant, anti-CD3/CD28 or viral peptide pool. Model immunotoxicants dexamethasone (DEX; 0 - 1.94 μM) and benzo(a)pyrene [B(a)P; 0-6.3 μM], were a

Development and validation of an all-in-one rabies virus Bat-Clade genomic sequencing and host identification protocol.

Rabies virus (RABV), remains a significant public health concern, with bat-maintained lineages accounting for all currently documented cases in Brazil. Despite the availability of pharmacological prophylaxis for humans and animals, the high genetic diversity of RABV in diverse natural bat hosts and continued circulation in multiple animals pose challenges for effective surveillance. Here, we developed and validated a novel, rapidly deployable amplicon-based sequencing approach for RABV genomic surveillance. This "all-in-one" protocol integrates whole RABV genome sequencing with host species id

Hybrid 99mTc-magnetite tracer for dual modality sentinel lymph node mapping.

Accuracy of sentinel lymph node identification using radioactive tracers in non-superficial cancers can be limited by radiation shine through and low spatial resolution of detection systems such as intraoperative gamma probes. By utilising a dual radioactive/magnetic tracer, sensitive lymphoscintigraphy can be paired with high spatial resolution intraoperative magnetometer probes to improve the accuracy of sentinel node detection in cancers with complex multidirectional lymphatic drainage. Dextran-coated magnetite nanoparticles (33 nm mean hydrodynamic diameter) were labelled with 99mTc and ap

Sperm Recovery and Evaluation in Domestic Species: Challenges and Current Practices.

Semen collection is a key step in the development of assisted reproductive technologies (ARTs) in domestic mammals, as the collection method determines the functionality and fertility of the sperm. The artificial vagina or manual collection yields ejaculates similar to those obtained during natural mating, although they require prior training to enable semen collection. When semen collection cannot be achieved using the aforementioned techniques, alternative methods such as electroejaculation may be employed. This method has been associated with increased stress and pain in animals; therefore,

Inhibition of glycogen synthase kinase 3 by lithium, a mechanism in search of specificity.

Inhibition of Glycogen synthase kinase 3 (GSK3) is a popular explanation for the effects of lithium ions on mood regulation in bipolar disorder and other mental illnesses, including major depression, cyclothymia, and schizophrenia. Contribution of GSK3 is supported by evidence obtained from animal and patient derived model systems. However, the two GSK3 enzymes, GSK3α and GSK3β, have more than 100 validated substrates. They are thus central hubs for major biological functions, such as dopamine-glutamate neurotransmission, synaptic plasticity (Hebbian and homeostatic), inflammation, circadian r

Iron and erythropoiesis: a dual relationship.

Iron is essential for cell life and especially for erythropoiesis which is the major body consumer of iron for red cell production. The study of genetic disorders of iron metabolism, the identification of iron transporters and of the role of hepcidin as the key regulator of systemic iron homeostasis have greatly contributed to our understanding of iron handling by the erythroid marrow. Spontaneous and engineered animal models of iron disorders have help to add further insights to the issue. A still incompletely understood aspect remains the regulation that erythropoiesis exerts on iron.

Dendritic cells as regulators of the immune response to inhaled allergen: recent findings in animal models of asthma.

Antigen-presenting dendritic cells are essential for the recognition and presentation of allergens to the cells of the immune system. Airway dendritic cells capture allergen in the mucosa and present it to naive T cells after migration into the draining lymph nodes. In this review article, we discuss the most recent findings from animal models of asthma, which highlight an essential role for these cells in the induction and maintenance of eosinophilic airway inflammation. This increasing knowledge might lead to the identification of new targets for the prevention and therapy of asthma.

Diagnostic evaluation of a recombinant chimeric Sporothrix brasiliensis protein-based ELISA for serological detection of feline sporotrichosis.

Sporotrichosis is a zoonotic infection caused by fungi of the Sporothrix schenkii complex, with S. brasiliensis being the most prevalent etiological agent in animals, particularly cats. Felines are highly susceptible to infection and may develop severe clinical forms characterized by extensive cutaneous lesions and systemic dissemination. In addition, infected cats play a major role in zoonotic transmission, as Sporothrix spp. can be transmitted to humans through bites and scratches. The gold standard for sporotrichosis diagnosis remains the isolation and identification of Sporothrix species f

Avian Hepatitis E Virus: With the Trend of Genotypes and Host Expansion.

UNLABELLED: Avian hepatitis E virus (HEV) is a single-stranded, positive-sense RNA virus with a complete genome of approximately 6.6 kb in size. To date, four major genotypes of avian HEV have been identified and classified into the Orthohepevirus B genus of the family Hepeviridae. The avian HEV associated with hepatitis-splenomegaly syndrome, big liver and spleen disease or hepatic rupture hemorrhage syndrome in chickens is genetically and antigenically related to mammalian HEV. With the increased genotypes of avian HEV identified, a broader host tropism is also notable in the epidemiological

From Control to Eradication: The Role of Point-of-Care Testing in Modernising Australia's Bovine Viral Diarrhoea (BVD) Disease Management.

Bovine Viral Diarrhoea (BVD) is an infectious disease caused by the Bovine Viral Diarrhoea Virus (BVDV), a member of the genus Orthopestivirus. The disease remains endemic across Australian beef and dairy production systems, imposing a multi-million-dollar annual burden on animal health, welfare, and industry sustainability. BVDV can be transmitted both horizontally and vertically, with persistently infected (PI) animals serving as the primary source of infection. Rapid identification and subsequent culling of PI animals are fundamental requirements for any successful eradication program. Curr

Resilience in motion: emerging perspectives on stress, substance use and youth.

Adolescence and young adulthood are developmental stages characterized by heightened stress sensitivity and limited cognitive control. The identification of the risk factors of alcohol consumption in these stages is crucial for early interventions focused on reducing harmful alcohol use. This review examines how exercise can modulate stress responses, reduce cravings, and preserve cognitive and emotional functioning. In animal models, it has been well described that exercise is able to reduce craving and protect cognitive and affective domains. Translational studies in young people with risky

Identification of a Candidate Starch Utilizing Strain of Prevotella albensis from Bovine Rumen.

The inclusion of starch-rich feedstuffs, a common practice in intensive ruminant livestock production systems, can result in ruminal acidosis, a condition that can severely impact animal performance and health. One of the main causes of acidosis is the rapid accumulation of ruminal short chain fatty acids (SCFAs) resulting from the microbial digestion of starch. A greater understanding of ruminal bacterial amylolytic activities is therefore critical to improving mitigation of acidosis. To this end, our manuscript reports the identification of a candidate starch utilizer (OTU SD_Bt-00010) using

Epithelial plasma membrane transporters as drug targets.

Small-molecule discovery and drug development are increasingly being pursued in academic settings, expanding beyond their traditional confinement to the pharmaceutical industry. The initial steps in drug discovery typically include identification and validation of a target, screening of chemical libraries to identify modulators of target activity, and subsequent prioritization and optimization of lead compounds using in vitro systems and animal models, with emphasis on compound potency, selectivity, and pharmacological properties. This review focuses on early-stage discovery of small molecules

From Brain Organoids to Translational Neurology: Exploring Neuroprotective Targets and Molecular Approaches in Perinatal Brain Injury.

Perinatal brain injury (PBI) is a leading cause of long-term neurological deficits in newborns, yet effective therapies are limited. At the cellular level, PBI involves hypoxic-ischemic stress, neuroinflammation, oxidative damage, excitotoxicity, and disrupted neurovascular and glial development. Traditional animal models and 2D cultures cannot fully capture the spatiotemporal complexity of the developing human brain, highlighting the need for more physiologically relevant systems. Human brain organoids have emerged as advanced three-dimensional models that recapitulate region-specific cytoarc

High-altitude pulmonary hypertension: a comprehensive review of mechanisms and management.

High-altitude pulmonary hypertension (HAPH) is characterized by an increase in pulmonary artery pressure due to prolonged exposure to hypoxic environment at high altitudes. The development of HAPH involves various factors such as pressure changes, inflammation, oxidative stress, gene regulation, and signal transduction. The pathophysiological mechanisms of this condition operate at molecular, cellular, and genetic levels. Diagnosis of HAPH often relies on echocardiography, cardiac catheterization, and other methods to assess pulmonary artery pressure and its impact on cardiac function. Treatme

Archaeoparasitological and biomolecular analysis of the wells from the Black Sea city of Phanagoria during the Khazar period (8th-9th centuries A.D.).

Ancient water supply systems provide important insights into historical sanitation practices and disease transmission patterns. This study examined soils from 2 wells of the medieval city of Phanagoria (8th-9th centuries AD) to reconstruct parasite contamination patterns during the Khazar period. Soil samples from wells 33 and 46 were analysed using light microscopy for parasite egg identification and gas chromatography-mass spectrometry (GC-MS) for faecal biomarker detection. Microscopic analysis identified 6 types of parasite eggs: Trichuris trichiura/suis, Trichuris muris, Ascaris lumbricoi

Leishmania vaccine development: A comprehensive review.

Infectious diseases like leishmaniasis, malaria, HIV, tuberculosis, leprosy and filariasis are responsible for an immense burden on public health systems. Among these, leishmaniasis is under the category I diseases as it is selected by WHO (World Health Organization) on the ground of diversity and complexity. High cost, resistance and toxic effects of Leishmania traditional drugs entail identification and development of therapeutic alternative. Since the natural infection elicits robust immunity, consistence efforts are going on to develop a successful vaccine. Clinical trials have been conduc

Kocuria rosea Sepsis in an Immunocompromised Patient: A Case Report.

Kocuria species, classified within the phylum Actinomycetota, class Actinomycetes, order Micrococcales, family Micrococcaceae, are Gram-positive coccoid bacteria. They bear morphological resemblance to Staphylococci spp and Micrococci spp, which often leads to misidentification and oversight as contaminants, given their presence as normal flora on human and animal skin and mucous membranes. Accurate identification of these organisms typically relies on automated systems such as MALDI-TOF-MS, Vitek-2 System, and 16S rRNA studies. Once considered rare, there is increasing recognition of Kocuria
