Apelin-induced cardioprotection against ischaemia/reperfusion injury: roles of epidermal growth factor and Src.

AIM: Apelin, the ligand of the G-protein-coupled receptor (GPCR) APJ, exerts a post-conditioning-like protection against ischaemia/reperfusion injury through activation of PI3K-Akt-NO signalling. The pathway connecting APJ to PI3K is still unknown. As other GPCR ligands act through transactivation of epidermal growth factor receptor (EGFR) via a matrix metalloproteinase (MMP) or Src kinase, we investigated whether EGFR transactivation is involved in the following three features of apelin-induced cardioprotection: limitation of infarct size, suppression of contracture and improvement of post-is

Serum apelin levels in established pulmonary arterial hypertension: A cross-sectional case-control study.

BACKGROUND: Pulmonary arterial hypertension (PAH) is a progressive condition involving pulmonary vasoconstriction, vascular remodeling, abnormal angiogenesis, and right ventricular dysfunction. Apelin, an endogenous peptide acting via the APJ receptor, modulates vascular tone, enhances nitric oxide-mediated vasodilation, and counteracts the renin-angiotensin system, suggesting relevance to PAH. AIMS: To compare serum apelin levels between patients with established PAH and healthy controls and to examine their relationship with cross-sectional measures of disease severity. METHODS: In this pr

Apelin Inhibits Cyst Growth and Improves Kidney Function in Mice with Polycystic Kidney Disease.

BACKGROUND: Autosomal dominant polycystic kidney disease (ADPKD) is a common inherited disorder marked by numerous renal cysts that impair kidney function, with about half of affected individuals progressing to kidney failure by midlife. Patients exhibit reduced circulating apelin, a ligand of the apelin receptor known to regulate cardiovascular function including hypertension. We tested whether diminished apelin signaling contributes to cystogenesis and if exogenous apelin receptor activation can improve disease outcomes. METHODS: Plasma samples from children and young adults with ADPKD and

The Apelin-APJ Axis in Heart Failure: Mechanistic Insights, Clinical Evidence, and Translational Challenges.

Heart failure remains a major cause of morbidity and mortality despite contemporary guideline-directed therapy, highlighting the need for additional therapeutic strategies, particularly across heterogeneous heart failure phenotypes. The apelin-Elabela-APJ axis has emerged as a biologically relevant pathway because it influences cardiac contractility, vascular tone, endothelial signalling, metabolism, and remodelling through mechanisms distinct from conventional neurohormonal targets. In this review, we examine the molecular biology of apelin, Elabela, and APJ, and critically appraise the precl

Apelin signaling acts as a molecular switch between endothelial and hematopoietic stem cell fates.

Hematopoietic stem and progenitor cells (HSPCs) emerge from arterial endothelial cells (ECs) through a process termed endothelial-to-hematopoietic-transition (EHT), a process induced by paracrine signals and driven by a transcriptional cascade. Despite inductive signals being broadly received by ECs in the dorsal aorta (DA), only a subset of ECs undergoes EHT, while others maintain their vascular identity. The molecular mechanisms that determine this selective fate decision remain poorly understood. Here, we discover Apelin signaling as a critical regulator of cell fates in the DA, acting as a

Apelin regulates skeletal muscle adaptation to exercise in a high-intensity interval training model.

Plasma apelin levels are reduced in aging and muscle wasting conditions. We aimed to investigate the significance of apelin signaling in cardiac and skeletal muscle responses to physiological stress. Apelin knockout (KO) and wild-type (WT) mice were subjected to high-intensity interval training (HIIT) by treadmill running. The effects of apelin on energy metabolism were studied in primary mouse skeletal muscle myotubes and cardiomyocytes. Apelin increased mitochondrial ATP production and mitochondrial coupling efficiency in myotubes and promoted the expression of mitochondrial genes both in pr

Activation of the apelin/APJ system by vitamin D attenuates age-related muscle atrophy.

AIMS: Age-related frailty and reduced physical activity contribute to a degenerative loss of muscle mass, function, and strength, which is known as sarcopenia. Increasing evidence has shown that vitamin D has beneficial effects on the muscle health. However, the molecular mechanisms of vitamin D have not been fully elucidated. In this study, we aimed to demonstrate whether vitamin D can overcome muscle atrophy due to aging, especially with respect to the regulation of myokines. MAIN METHODS: Young (3-month-old) and aged (18-month-old) C57BL/6 mice were assigned to the following 3 groups: norm

Impaired heart contractility in Apelin gene-deficient mice associated with aging and pressure overload.

Apelin constitutes a novel endogenous peptide system suggested to be involved in a broad range of physiological functions, including cardiovascular function, heart development, control of fluid homeostasis, and obesity. Apelin is also a catalytic substrate for angiotensin-converting enzyme 2, the key severe acute respiratory syndrome receptor. The in vivo physiological role of Apelin is still elusive. Here we report the generation of Apelin gene-targeted mice. Apelin mutant mice are viable and fertile, appear healthy, and exhibit normal body weight, water and food intake, heart rates, and hear

Cardioprotective Effects of Apelin in Myocardial Ischemia/Reperfusion Injury: A Systematic Review and Meta-Analysis.

Myocardial ischemia/reperfusion (I/R) injury remains a major clinical challenge, because blood flow restoration can exacerbate tissue damage. Apelin, an endogenous peptide acting through the APJ receptor, has demonstrated cardioprotective effects in experimental models. The APJ receptor, a G-protein-coupled receptor widely expressed in cardiovascular tissues, mediates vasodilation, cardiac contractility, and angiogenesis. This systematic review and meta-analysis evaluates its efficacy in myocardial I/R injury. A systematic search in Medline (PubMed), Embase, Scopus, and Web of Science was cond
