The effect of atropine on calculated sinoatrial conduction time in man.

This study investigates the effect of atropine on the length of the return cycles after premature atrial stimulation in man. On the assumption that sinus node automaticity is not changed by premature depolarization of the sinus node, sinoatrial conduction time (SACT) was calculated from the differences between the length of the return cycles and the spontaneous cycle length. 11 patients were studied before and after the injection of atropine. In all cases atropine caused an increase in heart rate. In 8 patients the return cycles exhibited a typical behavior. In 6 patients without signs of sinu

Bradycardia in preterm infants: indications and hazards of atropine therapy.

Eight preterm infants are presented to demonstrate the indications and hazards of using atropine for treatment of bradycardia in the high-risk premature nursery. Three infants developed bradycardia following initiation of nipple feedings, one following gavage feedings, three following surgical manipulation of visceral structures, and one associated with presence of chronic pulmonary disease. It is suggested that the first seven cases represent "reflexic bradycardia," probably vagally mediated and thus amenable to atropine therapy, whereas the eighth is an example of "hypoxic bradycardia" where

Stimulation of intestinal smooth muscle by atropine, procaine, and tetrodotoxin.

In order to determine whether or not atropine, procaine, and tetrodotoxin (TTX) can stimulate intestinal smooth muscle directly, we examined the effects of these drugs on the mechanical and electrical activities of several types of cat intestinal smooth muscle preparations. The preparations consisted of isolated rings of 1) intact intestinal wall, 2) intact longitudinal and circular muscle, 3) ganglion-free circular muscle, and 4) ganglion-free circular muscle devoid of its dense layer and plexus muscularis profundus. Atropine and procaine (greater than 10(-4) M) stimulated all four types of p

A NOTE ON THE ACTION OF CURARE, ATROPINE, AND NICOTINE ON THE INVERTEBRATE HEART.

1. The alkaloids (curare, atropine, and nicotine) in molluscs and arthropods stimulate and paralyze the central nervous system and peripheral (visceral) ganglia, but do not paralyze the motor nerve endings to skeletal or visceral muscle. 2. They stimulate and paralyze the denervated heart. 3. They paralyze or block the cardioinhibitory nerves, but not the cardioaccelerator nerves. 4. In the Limulus heart these drugs act primarily on the heart ganglion, not on the heart muscle or the intrinsic motor nerve fibers.

Translational changes: evidence for low-dose atropine and myopia control.

Myopia control, as opposed to myopia correction, is emerging as the new standard of care for clinical management of paediatric myopia by optometrists. The prevalence of myopia, and high myopia, is increasing worldwide, while the prevalence of pathologies associated with myopia increases with increasing axial length. With increasing global prevalence, preventing the axial elongation and pathologies associated with higher levels of myopia is therefore of paramount importance. Pharmacological management with atropine is one of the key myopia control options available to practitioners. Management

0.01% Atropine for Myopia Prevention in Pre-myopia (AMPP study).

PURPOSE: This study aimed to evaluate the efficacy and safety of 0.01% atropine eye drops in preventing myopia in children with pre-myopia. METHODS: In this randomised controlled trial (ChiCTR2200055482), children aged 5-12 years with cycloplegic spherical equivalent (SE) between +0.75D and -0.50D were randomised 1:1 to receive either 0.01% atropine (once nightly) or placebo (once nightly). The primary endpoint was the change in SE after 12 months. RESULTS: Sixty-four subjects (48.4% female) completed the 12-month follow-up (32 per group, mean age, 7.47 ± 1.72 vs. 7.84 ± 1.32 years). Juvenile toxicity of atropine sulfate eye drops in young rats.

OBJECTIVES: This study was to investigate the effects of atropine sulphate eye drops (ASED)on the development of partial systems in young rats and their toxic reactions following repeated eye-drop administration over a period of 40 days. METHODS: SD rats of 20 days old were randomly assigned to control group, 0.01, 0.02, and 0.04% ASED groups, with 60 females and 25 males per group. ASED was given by eye drops from PND21 onwards and normal saline was given in the control group at 10 μL/eye once a day for 40 days, in both right and left eyes. Rats of ASED groups were instilled with eye drops a

Functional atropine sensitive purinergic responses in the healthy rat bladder.

While acetylcholine is regarded to be the main directly contractile transmitter substance in the urinary bladder, interactions with other transmitters likely occur. Presently, the interplay between purinergic and cholinergic signalling was investigated to unravel the involvement of the urothelium and efferent neurons in the functionally important purinergically evoked release of acetylcholine in vitro. Functional characterization of receptor subtypes involved in this interplay was also performed. In vitro organ bath experiments with electrical field stimulation (EFS) or administration of agoni

Adenosine A1 antagonism attenuates atropine-resistant hypoxic bradycardia in rats.

OBJECTIVES: To test the following hypotheses: Hypoxia induces bradycardia and hemodynamic compromise that are resistant to atropine but responsive to selective antagonism of the adenosine A1 receptor (A1AdoR). The mechanism for such attenuation is independent of the vagus nerve. METHODS: Ten minutes after sham or actual bilateral cervical vagotomy, paralyzed ventilated rats were made hypoxic (5% fractional inspired oxygen, continued until death). Five minutes after beginning hypoxia, intravenous treatment with BG-9719, a selective A1AdoR antagonist (0.1 mg/kg); atropine (0.1 mg/kg); BG-9719 v

Subacute atropine toxicity in a pygmy sperm whale, Kogia breviceps.

Atropine, an anticholinergic agent commonly used in human and veterinary medicine, is reported to cause toxicity associated with its antimuscarinic action. A juvenile pygmy sperm whale, Kogia breviceps, was treated with atropine in an attempt to relieve symptoms similar to pyloric stenosis, as has been used in humans. Two doses of 0.01 mg/kg were given i.m., 12 hr apart, followed by three doses of 0.005 mg/kg i.m. s.i.d. over the next 3 days. Symptoms associated with atropine toxicity developed gradually and included hyperexcitability, a generalized ascending paralysis of body musculature, sha

Dose-dependent inhibition of cold air-induced bronchoconstriction by atropine.

We undertook a study to demonstrate whether inhalation of atropine could inhibit cold air-induced bronchoconstriction in a dose-dependent fashion. In seven subjects with asthma we assessed the effects of placebo and of various doses of inhaled atropine (0.13-2.08 mg) on a base-line specific airway resistance (sRaw) and on the increase in sRaw produced by 5 min of voluntary eucapnic hyperventilation with subfreezing air at -17 degrees C. We also assessed the effect of the lowest doses of atropine on the increase in sRaw produced by five breaths of 1.0% metacholine. Atropine in doses of 0.13 or

Atropine suppresses gastrin release by food intact and vagotomized dogs.

We have demonstrated that at doses lower than those used by others in dogs, atropine consistently inhibited food-stimulated gastrin release irrespective of vagal innervation of the stomach. Gastrin release induced by food placed directly into the stomach was studied in four gastric fistula dogs with intact vagi and in three other similar dogs with fundic vagotomy. The studies were repeated in dogs after conversion to truncal vagotomy. Fasting serum gastrin was lower in the intact dogs (33 +/- 1.7 pg/ml) than after fundic vagotomy (61 +/- 13 pg/ml) or truncal vagotomy (97 +/- 20 pg/ml). The sam
