Rab27a dependent exosome releasing participated in albumin handling as a coordinated approach to lysosome in kidney disease.

Exosomes are increasingly recognized as vehicles of intercellular communication. However, the role of exosome in maintaining cellular homeostasis under stress conditions remained unclear. Here we show that Rab27a expression was upregulated exclusively in tubular epithelial cells (TECs) during proteinuria nephropathy established by adriamycin (ADR) injection and 5/6 nephrectomy as well as in chronic kidney disease patients, leading to the increased secretion of exosomes carrying albumin. The active exosome production promoted tubule injury and inflammation in neighboring and the producing cells

CSF1 inhibits ferroptosis in colorectal cancer cells by autocrine activation of the JAK/STAT3 signaling pathway.

Colorectal cancer (CRC) remains a major cause of cancer-related mortality, particularly in patients with advanced disease. Although colony stimulating factor 1 (CSF1) has been implicated in tumor progression, its role in CRC remains unclear. In this study, we showed that CSF1 was significantly upregulated in CRC tissues and cell lines, and that high CSF1 expression was associated with adverse clinicopathological features and poorer patient outcomes. Functional assays demonstrated that CSF1 promoted CRC cell proliferation, migration, invasion, and xenograft tumor growth. Importantly, CSF1 also

CGRP derived from cardiac fibroblasts is an endogenous suppressor of cardiac fibrosis.

AIMS: Aberrant activation of cardiac fibroblasts leads to cardiac fibrosis, and evolving evidences suggest that endogenous bioactive substances derived from cardiac fibroblasts regulate cardiac fibroblasts activation in an autocrine/paracrine manner. Here we first presented evidence that cardiac fibroblasts can synthesize and secrete calcitonin gene-related peptide (CGRP), therefore, this study aimed to investigate the role of cardiac fibroblasts-derived CGRP in cardiac fibroblasts activation and its regulative mechanism. METHODS AND RESULTS: The abundantly expression of CGRP in rat, mouse, a

Regulation of neutrophil apoptosis differs after in vivo transmigration to skin chambers and synovial fluid: a role for inflammasome-dependent interleukin-1β release.

Short-lived neutrophils are major players in inflammation, arriving early to infected and/or injured tissues. After performed duty, neutrophils are programmed to die by apoptosis and are thereafter rapidly cleared by other phagocytes. In vitro, modulation of the apoptotic process has been thoroughly investigated in neutrophils isolated from peripheral blood, but less is known about the regulation of this process in neutrophils derived from extravascular tissues. We recently demonstrated that neutrophils having transmigrated in vivo, obtained from experimental skin chambers of healthy human sub

Physiology of Mechanotransduction: How Do Muscle and Bone "Talk" to One Another?

The complexity of cell interactions with their microenvironment and their ability to communicate at the autocrine, paracrine, and endocrine levels has gradually but significantly evolved in the last three decades. The musculoskeletal system has been historically recognized to be governed by a relationship of proximity and function, chiefly dictated by mechanical forces and the work of gravity itself. In this review article, we first provide a historical overview of the biomechanical theory of bone- muscle interactions. Next, we expand to detail the significant evolution in our understanding of

miR-146a-5p targets IRAK1/TRAF6 to promote bacillus Calmette-Guérin survival by exosome-mediated autocrine actions.

BACKGROUND: Exosomes carry signaling molecules between cells and play important roles in the interaction between macrophages and Mycobacterium tuberculosis (Mtb). This study aimed to examine the function and content of exosomes secreted by macrophages infected with Bacillus Calmette-Guérin (BCG). METHODS: THP-1 monocytes and HEK293T cells were used. Macrophages were infected with BCG. A Transwell system was used to evaluate the effect of the exosomes secreted by macrophages. Cells were transfected with the miR-146a-5p plasmid or inhibitor to examine the effects of miR-146a-5p overexpression o

Autocrine IL-10 activation of the STAT3 pathway is required for pathological macrophage differentiation in polycystic kidney disease.

Polycystic kidney disease (PKD) is characterized by slow expansion of fluid-filled cysts derived from tubules within the kidney. Cystic expansion results in injury to surrounding parenchyma and leads to inflammation, scarring and ultimately loss of renal function. Macrophages are a key element in this process, promoting cyst epithelial cell proliferation, cyst expansion and disease progression. Previously, we have shown that the microenvironment established by cystic epithelial cells can 'program' macrophages, inducing M2-like macrophage polarization that is characterized by expression of mark

Autocrine interferon-γ may affect malignant behavior and sensitivity to tamoxifen of MCF-7 via estrogen receptor β subtype.

Mitogenic actions of estrogens are mediated by two distinct estrogen receptors (ERs), which are critical in the progression and therapeutic response of breast cancer. ER expression is a dynamic phenomenon that is regulated by numerous factors, including cytokines, in the tumor microenvironment. Recently, studies have shown that autocrine production of IL-4 promotes cancer cell growth and there is negative correlation between tumor IL-4 and hormone receptor levels, suggesting that there is crosstalk between cytokine receptors and ER. Thus, we evaluated for interaction between the two ERs and th

Evidence for regulatory diversity and auto-regulation at the TAC1 locus in sensory neurones.

The neuropeptide substance-P (SP) is expressed from the TAC1 gene in sensory neurones where it acts as a key modulator of neurogenic inflammation. The promoter of TAC1 (TAC1prom) plays a central role in the regulation of the TAC1 gene but requires the presence of a second regulatory element; ECR2, to support TAC1 expression in sensory neurones and to respond appropriately to signalling pathways such as MAPkinases and noxious induction by capsaicin. We examined whether the effect of capsaicin on ECR2-TAC1prom activity in larger diameter neurones was cell autonomous or non- cell autonomous. We d

The dual vitamin D pathways: considerations for adequate supplementation.

Vitamin D insufficiency and deficiency have been identified as having a correlation with poor clinical outcomes in patients with chronic kidney disease (CKD). The availability of vitamin D for metabolism into 25(OH)D and the ability to further metabolize to 1,25(OH)D are known to have a significant impact on the endocrine system and the modulation of iPTH, calcium, and phosphorus imbalances in patients with CKD. Until recently, the focus of care for these patients has been to support the endocrine need for 1,25(OH)D because the loss of kidney function eliminates the ability to synthesize calci

Autocrine release of TGF-beta by portal fibroblasts regulates cell growth.

Portal fibroblasts (PF) are a newly isolated population of fibrogenic cells in the liver postulated to play a significant role in early biliary fibrosis. Because transforming growth factor-beta (TGF)-beta is a key growth factor in fibrosis, we characterized the response of PF to TGF-beta. We demonstrate that PF produce significant amounts of TGF-beta2 and, unlike activated hepatic stellate cells (HSC), express all three TGF-beta receptors and are growth inhibited by TGF-beta1 and TGF-beta2. Fibroblast growth factor (FGF)-2, but not platelet derived growth factor (PDGF), causes PF proliferation

Autocrine transforming growth factor beta stimulation of extracellular matrix production by fibroblasts from fibrotic human gingiva.

Hereditary gingival fibromatosis (HGF) is a fibrotic enlargement of the gingiva. HGF gingiva contains large amounts of collagen and other extracellular matrix (ECM) molecules. In vitro, HGF fibroblasts produce greater amounts of the ECM components fibronectin (FN) and type 1 collagen than normal human gingival (GN) fibroblasts. Transforming growth factor beta (TGF beta) is a cytokine important in regulating tissue repair and regeneration after injury, and stimulating fibroblast proliferation and the production of FN and collagens. The objective of this study was to determine whether HGF fibrob

Regional Heterogeneity of Perivascular Adipose Tissue: Morphology, Origin, and Secretome.

Perivascular adipose tissue (PVAT) is a unique fat depot with local and systemic impacts. PVATs are anatomically, developmentally, and functionally different from classical adipose tissues and they are also different from each other. PVAT adipocytes originate from different progenitors and precursors. They can produce and secrete a wide range of autocrine and paracrine factors, many of which are vasoactive modulators. In the context of obesity-associated low-grade inflammation, these phenotypic and functional differences become more evident. In this review, we focus on the recent findings of P

Cilia in the choroid plexus: their roles in hydrocephalus and beyond.

Cilia are whip-like projections that are widely conserved in eukaryotes and function as a motile propeller and/or sensory platform to detect various extracellular stimuli. In vertebrates, cilia are ubiquitously found in most cells, showing structural and functional diversities depending on the cell type. In this review, we focus on the structure and function of cilia in choroid plexus epithelial cells (CPECs). CPECs form one or two dozen non-motile 9+0 cilia, which display transient acquisition of motility during development. Genetic malfunction of cilia can lead to failure of multiple organs

Muscle in Endocrinology: From Skeletal Muscle Hormone Regulation to Myokine Secretion and Its Implications in Endocrine-Metabolic Diseases.

Skeletal muscle, traditionally recognized for its motor function, has emerged as a key endocrine organ involved in metabolic regulation and interorgan communication. This narrative review addresses the dual role of muscle as a target tissue for classical hormones-such as growth hormone (GH), insulin-like growth factor type 1 (IGF-1), thyroid hormones, and sex steroids-and as a source of myokines, bioactive peptides released in response to muscle contraction that exert autocrine, paracrine, and endocrine effects. Several relevant myokines are discussed, such as irisin and Metrnl-like myokines (

The "double-edged sword" effect of cytokines in cancer: coexisting opportunities and challenges.

As a key signaling molecule network in the tumor immune microenvironment, cytokines mediate intercellular communication through mechanisms such as autocrine and paracrine, exhibiting a significant "double-edged sword" effect during tumor initiation and progression. The dynamic regulation of this dual effect is influenced by the dependence on concentration, the variability within the tumor immune microenvironment, and the stages of tumor progression, ultimately representing the prolonged co-evolutionary result between tumors and the immune system. Cytokines, as a vital element of the immune mic

Glucose increases extracellular [Ca2+] in rat insulinoma (INS-1E) pseudoislets as measured with Ca2+-sensitive microelectrodes.

Secretory granules of pancreatic β-cells contain high concentrations of Ca2+ ions that are co-released with insulin in the extracellular milieu upon activation of exocytosis. As a consequence, an increase in the extracellular Ca2+ concentration ([Ca2+]ext) in the microenvironment immediately surrounding β-cells should be expected following the exocytotic event. Using Ca2+-selective microelectrodes we show here that both high glucose and non-nutrient insulinotropic agents elicit a reversible increase of [Ca2+]ext within rat insulinoma (INS-1E) β-cells pseudoislets. The glucose-induced increases

Cytokines of the placenta and extra-placental membranes: roles and regulation during human pregnancy and parturition.

Summary In an earlier, companion, review, we concluded that cytokines produced by the placenta and associated membranes are likely to be involved in control of the processes of implantation and placental development (Bowen et al., 2002). In this review, we discuss evidence that cytokines continue to be part of a paracrine/autocrine regulatory network in the placenta and membranes throughout the mid and late stages of gestation. Cytokines are involved in regulation of placental growth during these later stages of pregnancy and also function to protect the fetus from pathological organisms. The

Autocrine stimulation of interleukin 1 alpha in the growth of adult human T-cell leukemia cells.

In a previous study, we reported that adult T-cell leukemia (ATL) cells produce interleukin 1 (IL1)-like factors that stimulate murine thymocyte proliferation, the production of interleukin 2 (IL2), and the expression of IL2 receptors (IL2R) on normal human T-cells in the presence of concanavalin A. In this communication, we studied the effect of IL1 on the growth of ATL cells in vitro. When ATL cells freshly obtained from patients were cultured with recombinant (r) human IL1 alpha, IL1 beta, or IL1-like factors produced by ATL cell lines, the growth of ATL cells was stimulated in a concentrat

Exercise-induced myokines in metabolic regulation: mechanisms, mimetics, and translational potential.

Skeletal muscle, once regarded solely as a contractile tissue, is now recognized as a dynamic endocrine organ that secretes exercise-induced myokines-bioactive peptides with autocrine, paracrine, and endocrine functions. These myokines coordinate systemic energy homeostasis by regulating glucose and lipid metabolism, mitochondrial function, inflammation, and interorgan communication. Building on our previous review published in 2018, this review synthesizes major advances in exercise-induced myokines within an evidence-based framework considering mechanistic support and translational relevance
