A Src family kinase inhibitor improves survival in experimental acute liver failure associated with elevated cerebral and circulating vascular endothelial growth factor levels.

BACKGROUND AND AIMS: Acute liver failure (ALF) is frequently complicated by cerebral oedema, systemic inflammation and multiorgan dysfunction. Vascular endothelial growth factor (VEGF) may stimulate liver regeneration but it can also be pro-inflammatory, activating endothelial cells and increasing permeability, actions mediated through Src kinase signalling. We therefore examined whether a Src inhibitor could have therapeutic potential in ALF. METHODS: Murine ALF was induced with azoxymethane. Liver pathology was graded by a blinded examiner and apoptosis quantified by immunohistochemistry. C

Protective effects of calcium from nonfat dried milk against colon carcinogenesis in rats.

A number of studies have demonstrated the protective effect of dietary calcium against risk for colon cancer. The objective of this experimental study was to test the efficacy of two sources of dietary calcium, elemental calcium in the form of CaCO3 and dairy calcium as nonfat dried milk (NFDM), in colon tumor inhibition. Male weanling F344 rats were fed six test diets containing low (LF, 5%) and high (HF, 20%) levels of corn oil and low (0.5%) and high (1.0%) levels of calcium supplemented as CaCO3 or NFDM in a 2 x 3 factorial design. Tumors were induced with two weekly injections of azoxymet

Protective mechanisms of dietary fibers in nutritional carcinogenesis.

Fibers in foods are complex carbohydrates. There are several types of fiber, but, for the purpose of mechanistic insight into their mode of protective action in carcinogenesis, classification into two broad types, soluble and insoluble fibers, is warranted. Soluble fibers are present in fruits, vegetables, and certain grains like oats. This type of fiber undergoes metabolism in the small intestine and especially in the large intestine through bacterial enzymes, converting it to products that increase stool size only moderately. But, they have appreciable effects in modifying the metabolism of

Cadmium exposure is associated with colorectal neoplasm risk: From epidemiological evidence to laboratory confirmation.

Cadmium (Cd) is a well-established carcinogenic heavy metal linked to the development of various cancers. However, its role in colorectal neoplasms remains unclear. Using a cross-sectional design involving 2332 Chinese participants, we examined the association between Cd exposure and colorectal neoplasms and further explored underlying mechanisms using murine models exposed to environmentally relevant Cd concentrations in drinking water. Epidemiological analysis showed that elevated whole-blood Cd levels were associated with an increased risk of both non-advanced (prevalence ratio [PR] per log

Regulation of Fatty Acid Metabolism and Inhibition of Colorectal Cancer Progression by Erchen Decoction.

Erchen decoction (ECD) is a traditional Chinese prescription widely used in the treatment of various diseases such as obesity, fatty liver, diabetes, and hypertension. In this study, we investigated the effect of ECD on fatty acid metabolism in a colorectal cancer (CRC) mouse model fed a high-fat (HF) diet. The HF-CRC mouse model was established by azoxymethane (AOM)/dextran sulphate sodium (DSS) combined with a high-fat diet. Mice were then gavaged with ECD. Change in the body weight was recorded every two weeks for 26 weeks. Changes in blood glucose (GLU), total cholesterol (TC), total trigl

Weight loss with exercise improves muscle architecture and progenitor cell populations compared with weight loss alone in mice with preneoplastic colorectal lesions.

Weight loss and exercise reduce colorectal cancer (CRC) risk in persons with obesity. Whether weight loss and exercise effect myofiber characteristics and muscle stem/progenitor cell populations in mice with preneoplastic colorectal lesions, a model of CRC risk, is unknown. To address this gap, male C57Bl/6J mice were fed a high-fat diet (HFD) to induce obesity or a control (CON) diet prior to azoxymethane injection to induce preneoplastic colorectal lesions. The HFD group was then randomized to weight loss conditions that included (1) switching to the CON diet only (HFD-SED) or switching to t

Behavioral and emotional abnormalities in mice with orthotopic colitis-associated colorectal cancer.

BACKGROUND: Colorectal cancer (CRC) is usually accompanied by emotional and behavioral changes in patients, yet the underlying neurobiological mechanisms remain unclear. There is a lack of robust experimental models to investigate how tumor progression may influence brain function and behavior. METHODS: The azoxymethane (AOM) and dextran sulfate sodium (DSS) were used to induce the orthotopic colitis-associated CRC (CAC) mouse models. Then we performed a range of behavioral tests including the sucrose preference test (SPT), elevated plus maze (EPM), tail suspension test (TST), novel object re

Zearalenone exposure is associated with increased tumor burden and coordinated host-microbiota-metabolic alterations in colitis-associated colorectal cancer.

Zearalenone (ZEN) is a common foodborne mycotoxin with intestinal toxicity, but its role in colitis-associated colorectal cancer (CAC) remains unclear. This study evaluated the association between ZEN exposure and CAC progression and explored the related molecular, microbial, and metabolic alterations. Network toxicology was used to predict potential ZEN-related targets and pathways associated with colorectal cancer, suggesting possible involvement of PI3K-Akt, MAPK, and apoptosis-related pathways. Subsequently, an azoxymethane/dextran sulfate sodium (AOM/DSS)-induced CAC model was established

The ketogenic diet modulates tumor-associated neutrophil polarization via the AMOT-YAP/TAZ axis to inhibit colorectal cancer progression.

Despite significant advances in the diagnosis and treatment of colorectal cancer (CRC), the prognosis for late-stage patients remains poor, highlighting the urgent need for new preventive and therapeutic strategies. Recent studies have focused on the ketogenic diet (KD) and its metabolite, β-hydroxybutyrate (BHB), for their tumor-suppressive effects and modulation of inflammatory responses. Using the azoxymethane (AOM) / dextran sulfate sodium (DSS)-induced mouse CRC model, we found that the ketogenic diet and BHB inhibit pro-tumor N2-type tumor-associated neutrophils (TANs) while promoting th

Identification of gene expression changes from colitis to CRC in the mouse CAC model.

A connection between colorectal carcinogenesis and inflammation is well known, but the underlying molecular mechanisms have not been elucidated. Chemically induced colitis-associated cancer (CAC) is an outstanding mouse model for studying the link between inflammation and cancer. Additionally, the CAC model is used for examining novel diagnostic, prognostic, and predictive markers for use in clinical practice. Here, a CAC model was established in less than 100 days using azoxymethane (AOM) with dextran sulfate sodium salt (DSS) in BALB/c mice. We examined the mRNA expression profiles of three
