Demystifying a buzzword: Use of the term "human-animal-interface" in One Health oriented research based on a literature review and expert interviews.

As of today, 75% of infectious human diseases are caused by zoonotic pathogens, which use the interface between humans and animal species to cross. Due to this ability, zoonoses affect more than just one health sector and the effective control is a matter of the One Health concept. One defining feature of this concept is the "human-animal-interface". However, even though the term is ubiquitously used in the field of infectious disease research, a clear definition of the term is lacking, leading to a rather nebulous understanding of what this interface really encompasses. Based on this observat

Vaccines for emerging pathogens: from research to the clinic.

In this two-part series of reviews, we have invited experts in their fields to contribute articles on the status of vaccine research and development for emerging pathogens. This topic has been brought into sharp focus in recent years following significant outbreaks of viral diseases such as those causing severe acute respiratory syndrome and Middle East respiratory syndrome, as well as devastating outbreaks of diseases caused by the Ebola, Marburg, Zika and Lassa fever viruses, to name only a few examples. Additionally, bacterial infections leading to bubonic and pneumonic plague, most notably

Host-Pathogen-Vector Continuum in a Changing Landscape: Potential Transmission Pathways for Bartonella in a Small Mammal Community.

Bacterial infections account for a large proportion of zoonoses. Our current understanding of zoonotic spillover, however, is largely based on studies from viral systems. Small mammals such as rodents and their ectoparasites present a unique system for studying several bacterial pathogens and mapping their spillover pathways. Using Bartonella spp. (a Gram-negative bacteria) as a model system within a rainforest human-use landscape, we investigated (1) ecological correlates of Bartonella prevalence in small mammal hosts and (2) evolutionary relationships between Bartonella spp. and various host

Intracellular trafficking study of a RB51 B. abortus vaccinal strain isolated from cow milk.

Brucella is responsible for one of the major worldwide zoonoses. Over the last century, several vaccines have been used against brucellosis. Among these, the rough vaccine Brucella abortus RB51 was introduced with the idea that it would not interfere with the diagnosis of brucellosis. Recently, RB51 has been isolated from milk and vaginal exudates from vaccinated cows, thus raising the possibility of extensive bacterial replication in these animals. We hypothesized that shedding of RB51 might be related to a change in its intracellular cell cycle. Therefore, we have compared the intracellular

Brucellosis: a worldwide zoonosis.

Brucella is one of the world's major zoonotic pathogens, and is responsible for enormous economic losses as well as considerable human morbidity in endemic areas. Control of brucellosis requires practical solutions that can be easily applied to the field. Rapid DNA-based diagnostic tests for both humans and livestock have now proved themselves on an experimental level. Data on the virulence of Brucella suggest common mechanisms shared with plant pathogens and endosymbionts of the alpha-proteobacteria. Understanding virulence will have practical repercussions in the realms of vaccine developmen

Understanding leptospirosis eco-epidemiology by environmental DNA metabarcoding of irrigation water from two agro-ecological regions of Sri Lanka.

BACKGROUND: Leptospirosis is one of the most significant zoonoses across the world not only because of its impact on human and animal health but also because of the economic and social impact on agrarian communities. Leptospirosis is endemic in Sri Lanka where paddy farming activities, the use of draught animals in agriculture, and peridomestic animals in urban and rural areas play important roles in maintaining the infection cycle of pathogenic Leptospira, especially concerning animals as a potential reservoir. In this study, an environmental DNA (eDNA) metabarcoding methodology was applied i

High prevalence of carbapenem-resistant Pseudomonas spp. in the pork production chain: a potential reservoir for zoonotic transmission.

Multidrug-resistant Pseudomonas aeruginosa is currently one of the primary threats to human health, accounting for a significant proportion of difficult-to-treat infections globally. Although pork meat is frequently contaminated with Pseudomonas spp., well-known spoilage organisms, their potential role as reservoirs of antibiotic-resistant strains and associated risks to human health remains unclear. This study investigated the prevalence of carbapenem-resistant Pseudomonas spp. (CRP) throughout the same pork processing chain, and assessed their pathogenicity and genetic relatedness. We sample

Patchy promiscuity: machine learning applied to predict the host specificity of Salmonella enterica and Escherichia coli.

Salmonella enterica and Escherichia coli are bacterial species that colonize different animal hosts with sub-types that can cause life-threatening infections in humans. Source attribution of zoonoses is an important goal for infection control as is identification of isolates in reservoir hosts that represent a threat to human health. In this study, host specificity and zoonotic potential were predicted using machine learning in which Support Vector Machine (SVM) classifiers were built based on predicted proteins from whole genome sequences. Analysis of over 1000 S.enterica genomes allowed the

Molecular Epidemiology of Brucella abortus in Northern Ireland-1991 to 2012.

BACKGROUND: Brucellosis is the most common bacterial zoonoses worldwide. Bovine brucellosis caused by Brucella abortus has far reaching animal health and economic impacts at both the local and national levels. Alongside traditional veterinary epidemiology, the use of molecular typing has recently been applied to inform on bacterial population structure and identify epidemiologically-linked cases of infection. Multi-locus variable number tandem repeat VNTR analysis (MLVA) was used to investigate the molecular epidemiology of a well-characterised Brucella abortus epidemic in Northern Ireland inv

[Brucella at the dawn of the third milenium: genomic organization and pathogenesis].

Bacteria of the genus Brucella, responsible for brucellosis, are pathogenic for animals and occasionally for humans. The cost of this widespread zoonotic infection is still very high for the community. Over the last few years, there have been advances in two main domains. First, the Brucella genome has been shown to be complex, with two circular chromosomes. Second, recent data on the virulence of Brucella suggest common mechanisms shared with plant pathogens and endosymbionts of the alpha-proteobacteria. Understanding virulence will have practical repercussions in the realms of vaccine develo

The impacts of animal agriculture on One Health-Bacterial zoonosis, antimicrobial resistance, and beyond.

The industrialization of animal agriculture has undoubtedly contributed to the improvement of human well-being by increasing the efficiency of food animal production. At the same time, it has also drastically impacted the natural environment and human society. The One Health initiative emphasizes the interdependency of the health of ecosystems, animals, and humans. In this paper, we discuss some of the most profound consequences of animal agriculture practices from a One Health perspective. More specifically, we focus on impacts to host-microbe interactions by elaborating on how modern animal

A Cellular Model of Infection with Brucella melitensis in Ovine Macrophages: Novel Insights for Intracellular Bacterial Detection.

Intracellular bacteria provoking zoonoses, such as those of the genus Brucella, present a host cell tropism mostly limited to the monocyte/macrophage lineage, leading to chronic inflammatory reactions, difficult-to-eradicate-infections, and widespread prevalence among ruminants. Eradication of brucellosis has been based on programs that translate into a substantial financial burden for both the authorities and stockbreeders, if not strictly followed. To this end, we sought to create an in vitro cell model that could be utilized as future reference for adequately measuring the number of engulfe

Zoonotic diseases associated with reptiles and amphibians: an update.

Reptiles and amphibians are popular as pets. There are increased concerns among public health officials because of the zoonotic potential associated with these animals. Encounters with reptiles and amphibians are also on the rise in the laboratory setting and with wild animals; in both of these practices, there is also an increased likelihood for exposure to zoonotic pathogens. It is important that veterinarians remain current with the literature as it relates to emerging and reemerging zoonotic diseases attributed to reptiles and amphibians so that they can protect themselves, their staff, an

Bacterial diseases of fish associated with human health.

As this country continues to increase its food fish output without increasing the water acreage, and environmental wastes continue to plague fish production, we can expect to encounter more zoonotic organisms, especially enteric-like organisms. Since 1972, the number of enteric genera has doubled and the species have tripled. The veterinarian engaged in aquatic animal medicine should be aware that aquatic pets or food fish can be associated with human disease and respond accordingly. In such cases, the veterinarian plays a major role in maintaining human health.

[Specific bacterial diversity in bats of different food guilds in Southern sierra Oaxaca, Mexico].

Specific bacterial diversity in bats of different food guilds in Southern sierra Oaxaca, Mexico. Bats have different ecologic roles in variable ecosystems that have been already described. They have been linked to several zoonoses, however little is known about the relationship between bat microbiota and their diet, and studies on the bacterial ecology of the microbiota in bats are limited. To contribute with the description of this important interaction between microbiota and host, the aim of this work was to characterize the composition and bacterial diversity in the oral and anal regions of

Clostridioides difficile as a Zoonotic and Foodborne Pathogen: Reviewing the Global Evidence and South African Data.

Clostridioides difficile infections (CDIs) are becoming increasingly problematic in developing countries, further burdening healthcare systems. In Southern Africa, limited information is available on the prevalence and diversity of C. difficile strains in animals, food, and the environment. This review aims to provide insight into global findings on strain diversity, antibiotic resistance, and the epidemiology of C. difficile associated with food-producing animals, wildlife, and food processing environments. The review further explores the zoonotic and foodborne risk potential of the pathogen
