Treatment of drug-induced exogenous psychosis in parkinsonism with clozapine and fluperlapine.

A total of 13 patients with drug-induced psychosis in Parkinson's disease were treated with two non-classical neuroleptics-clozapine and fluperlapine. Patients mainly complained about severe hallucinatory symptoms and different degrees of paranoid delusions. Complete relief was observed in 8 patients, moderate improvement in 3 and no effects in 2. Parkinsonian disability did not increase under neuroleptic medication with clozapine and fluperlapine, but could be ameliorated by additional L-dopa or bromocriptine medication. The non-classical neuroleptics employed are dopamine D2 blocking agents

Alterations in cortical and basal ganglia levels of opioid receptor binding in a rat model of l-DOPA-induced dyskinesia.

Opioid receptor-binding autoradiography was used as a way to map sites of altered opioid transmission in a rat model of l-DOPA-induced dyskinesia. Rats with unilateral 6-hydroxydopamine lesions of the nigrostriatal pathways sustained a 3-week treatment with l-DOPA (6 mg/kg/day, combined with 12 mg/kg/day benserazide), causing about half of them to develop dyskinetic-like movements on the side of the body contralateral to the lesion. Autoradiographic analysis of mu-, delta-, and kappa-opioid binding sites was carried out in the caudate-putamen (CPu), the globus pallidus (GP), the substantia nig

Noradrenoceptor antagonism with idazoxan improves L-dopa-induced dyskinesias in MPTP monkeys.

Treatment of Parkinson's disease with L-dopa is plagued in a majority of patients by dyskinesias. Noradrenaline/dopamine interactions are proposed on behavioral, biochemical, physiological and anatomical grounds. The aim of the study was to test the potential antidyskinetic effect of the alpha2-adrenoceptor antagonist, idazoxan, in a primate model of Parkinson's disease. Six female cynomolgus monkeys previously rendered parkinsonian by the toxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and presenting an unchanged syndrome for several months were used. All responded readily to L-dopa

3,4-dihydroxyphenylalanine reverses the motor deficits in Pitx3-deficient aphakia mice: behavioral characterization of a novel genetic model of Parkinson's disease.

Parkinson's disease (PD) is a neurodegenerative disease characterized by a loss of dopaminergic neurons in the substantia nigra. There is a need for genetic animal models of PD for screening and in vivo testing of novel restorative therapeutic agents. Although current genetic models of PD produce behavioral impairment and nigrostriatal dysfunction, they do not reproduce the loss of midbrain dopaminergic neurons and 3,4-dihydroxyphenylalanine (L-DOPA) reversible behavioral deficits. Here, we demonstrate that Pitx3-deficient aphakia (ak) mice, which have been shown previously to exhibit a major

Therapeutic value of Madopar HBS: judgment after 2 years experience.

The study included 19 parkinsonian patients presenting with fluctuations in motor performance after L-dopa therapy (9.9 years on average, range 6 months to 17 years) and 2 other patients not pretreated with L-dopa. In all patients of the first group, after 3 months on stable 'optimal' dosage schedule, the previous L-dopa treatment was abruptly replaced, dose for dose, from one day to another by Madopar HBS, a new controlled-release form of Madopar. The first clinical assessment was performed just before the beginning of the HBS treatment. Then, 4-6 weeks were allowed to optimize the dosage sch

Central depression of carotid baroreceptor pressor response, arterial pressure and heart rate by 5-hydroxytryptophan: influence of supracollicular areas of the brain.

5-Hydroxytryptophan (5-htp) administered into the fourth cerebral ventricle of cats decreased resting arterial blood pressure, preganglionic sympathetic nerve activity and heart rate in both intact brain and midcollicular decerebrate preparations. The increases in sympathetic nerve discharge and blood pressure in response to bilateral carotid occlusion were reduced after 5-HTP administration when the brain was intact but not in midcollicular decerebrate preparations. Resting arterial pressure, heart rate and bilateral carotid occlusion response were not affected when the distribution of 5-HTP

Metabolic interactions of aldehyde dehydrogenase with therapeutic and toxic agents.

The specific physiological roles of ALDH have not been determined. In this overview, we have identified malondialdehyde as one potential endogenous toxic aldehyde that is oxidized by ALDH. Undoubtedly, there are other cytotoxic aldehydes which occur in several different tissues that are detoxified by this same system of enzymes. Intuitively, the role of aldehyde dehydrogenases in the detoxification of xenobiotic aldehydes should also receive additional consideration and attention. An interesting feature of specific studies described in this overview is that a single aldehydic substrate, like m

Nardosinone improves levodopa-induced dyskinesia in Parkinsonian rats through the microbiota-gut-brain axis.

Levodopa-induced dyskinesia (LID) is a side effect of Parkinson's disease (PD) treated with L-3,4-dihydroxyphenylalanine (L-DOPA). Nardosinone is the major active component of the dried root and rhizome of Nardostachys jatamansi DC, which is effective for treating PD. The purpose of this study was to explore the effects of nardosinone on LID and the underlying mechanism involving the microbiota-gut-brain axis. A PD rat model was established with rotenone. Behavioral and immunohistochemical staining experiments were used to investigate the synergistic effect of nardosinone on L-DOPA in PD rats.

Efficacy of Madopar and trihexyphenidyl combination therapy for dystonia in children with cerebral palsy.

INTRODUCTION: Dystonia is a predominant and debilitating movement disorder associated with dyskinetic cerebral palsy (DCP). Although trihexyphenidyl (THP) is commonly used as a treatment, its efficacy often exhibits a plateau effect. The combination of dopaminergic and anticholinergic agents represents a rational therapeutic strategy; however, robust evidence for the combination of Madopar (levodopa/benserazide) and THP is lacking. METHODS: This retrospective cohort study compared THP monotherapy (n = 25) with combined Madopar + THP therapy (n = 24) in children with DCP and dystonia. Propensi

Kynu inhibition mitigates bile duct ischemic injury by rewiring tryptophan metabolism to restore tight junction integrity.

BACKGROUND: Disruption in bile duct barrier function contributes to hepatocyte toxicity in ischemia-reperfusion injury, often leading to surgical complications in liver resection, transplantation, and hemorrhagic shock. However, the underlying mechanisms remain incompletely understood. METHODS: Transcriptomic and proteomic analyses were conducted to examine tryptophan (Trp) metabolism in a Pringle maneuver-induced bile duct injury rat model; Hypoxia/Reoxygenation (H/R) was used to establish an in vitro cholangiocyte injury model. Cholangiocyte injury was assessed via hematoxylin and eosin (H&

Richardson Syndrome Variant of Progressive Supranuclear Palsy: A Case Report.

Progressive supranuclear palsy (PSP) is a chronic neurodegenerative disease, an atypical parkinsonism with four-repeat tau neuropathology. PSP-Richardson syndrome (PSP-RS) is the most prevalent clinical variant of PSP, originally described as the prototypical form of the disease. It is defined by vertical supranuclear gaze palsy (most prominently affecting downward gaze), early postural instability with frequent falls, and a subcortical-frontal pattern of cognitive impairment. The diagnosis of PSP is mainly based on clinical data and is only definitively confirmed at autopsy. Neuroimaging play

Cerebral differences between dopamine-resistant and dopamine-responsive Parkinson's tremor.

Rest tremor in Parkinson's disease is related to cerebral activity in both the basal ganglia and a cerebello-thalamo-cortical circuit. Clinically, there is strong interindividual variation in the therapeutic response of tremor to dopaminergic medication. This observation casts doubt on the idea that Parkinson's tremor has a dopaminergic basis. An interesting alternative explanation is that interindividual differences in the pathophysiology of tremor may underlie this clinical heterogeneity. Previous work showed that dopaminergic medication reduces Parkinson's tremor by inhibiting tremulous act

The effect of subchronic, intermittent L-DOPA treatment on neuronal nitric oxide synthase and soluble guanylyl cyclase expression and activity in the striatum and midbrain of normal and MPTP-treated mice.

We have investigated the effects of low (10 mg/kg) and high (100 mg/kg) doses of L-DOPA on the expression and activity of neuronal nitric oxide synthase (nNOS) and guanylyl cyclase (GC) in the striatum and midbrain of mice. L-DOPA was administered subchronically for 11 days (beginning 3 days after last MPTP/NaCl injection) or for 14 days (with dosing started immediately following the last MPTP/NaCl injection). Adult mice received three intraperitoneal (i.p.) injections of physiological saline or MPTP at 2h intervals (total dose of 40 mg/kg). Normal and MPTP-injected mice were treated twice a d

Differing effects of N-methyl-D-aspartate receptor subtype selective antagonists on dyskinesias in levodopa-treated 1-methyl-4-phenyl-tetrahydropyridine monkeys.

The antiparkinsonian and antidyskinetic profile of two N-methyl-D-aspartate (NMDA) receptor antagonists, a competitive antagonist, (R)-4-oxo-5-phosphononorvaline (MDL 100,453), and a novel noncompetitive allosteric site antagonist, 4-hydroxy-N-[2-(4-hydroxyphenoxy)ethyl]-4-(4-methylbenzyl)piper idi ne (Co 101244/PD 174494), was assessed in six levodopa-treated 1-methyl-4-phenyl-tetrahydropyridine-lesioned parkinsonian monkeys. The effects on motor function of these two drugs, alone and in combination with levodopa, were then correlated with NMDA subtype selectivity and apparent affinity for fo

Catechol-O-methyltransferase inhibition with tolcapone reduces the "wearing off" phenomenon and levodopa requirements in fluctuating parkinsonian patients.

BACKGROUND: More than 50% of patients with Parkinson's disease develop motor response fluctuations (the 'wearing off" phenomenon) after more than five years of levodopa therapy. Inhibition of catechol-O-methyltransferase by tolcapone has been shown to increase levodopa bioavailability and plasma elimination half life, thereby prolonging the efficacy of levodopa. OBJECTIVES: The primary objective was to evaluate the efficacy of tolcapone in reducing "wearing off" in levodopa treated, fluctuating parkinsonian patients. Secondary objectives included assessment of reduction in levodopa requiremen

Behavioral Effects of Renin-Angiotensin System Modulators in a Rotenone-Induced Model of Parkinson's Disease in Wistar Albino Rats.

INTRODUCTION: Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by motor and non-motor dysfunction resulting from dopaminergic neuronal loss. Emerging evidence suggests a role of the renin-angiotensin system (RAS) in neurodegeneration, making it a potential therapeutic target. The present study aimed to evaluate the behavioral effects of captopril, perindopril, and losartan on motor, behavioral, and anxiety parameters in a rotenone-induced model of Parkinson's disease in Wistar albino rats. MATERIALS AND METHODS: An experimental in vivo study was conducted usi
