Subclinical markers of cardiovascular toxicity of benzene inhalation in mice.

Benzene is a ubiquitous environmental pollutant. Recent population-based studies suggest that benzene exposure is associated with an increased risk for cardiovascular disease. However, it is unclear whether benzene exposure by itself is sufficient to induce cardiovascular toxicity. We examined the effects of benzene inhalation (50 ppm, 6 h/day, 5 days/week, 6 weeks) or HEPA-filtered air exposure on the biomarkers of cardiovascular toxicity in male C57BL/6J mice. Benzene inhalation significantly increased the biomarkers of endothelial activation and injury including endothelial microparticles,

Effects of ethanol and phenobarbital administration on the metabolism and toxicity of benzene.

Effects of ethanol- and phenobarbital(PB)-treatment on the metabolism of benzene in vitro and in vivo, and on the benzene-induced hemotoxicity, were investigated. Ethanol consumption markedly enhanced in vitro metabolism of both benzene and phenol in rat liver, whereas PB-treatment, which enhanced the metabolism of phenol to some degree (about one-third of ethanol-induced enhancement), did not affect the metabolism of benzene. In a single exposure experiment with rats, ethanol increased benzene metabolism in vivo as evidenced by accelerated disappearance of benzene from the blood as well as by

The joint toxic action of perchloroethylene with benzene or toluene in rats.

The joint toxic action of mixtures of perchloroethylene with benzene or toluene was tested at five dose levels for each of six binary combinations with 20 rats per dose. LD50 data with 95% fiducial limits are reported for each mixture and tests for additive joint toxicity were made. The results indicate that the mixtures of perchloroethylene and benzene gave values for the LD50 which were slightly less than additive while perchloroethylene and toluene mixtures had LD50 values which could not be predicted by an additive model.

An advanced bath model to simulate association followed by ensuing dissociation dynamics of benzene + benzene system: a comparative study of gas and condensed phase results.

The role of the environment (N2 molecules) on the association followed by the ensuing dissociation reaction of benzene + benzene system is studied here with the help of a new code setup. Chemical dynamics simulations are performed to investigate this reaction in vacuum as well as in a bath of 1000 N2 molecules, equilibrated at 300 K. Bath densities of 20 and 324 kg m-3 are considered with a few results from the latter density. The simulations are performed at three different excitation temperatures of benzene, namely, 1000, 1500, and 2000 K, with an impact parameter range of 0-12 Å for both va

Monitoring of benzene-exposed workers for genotoxic effects of benzene: improved-working-condition-related decrease in the frequencies of chromosomal aberrations in peripheral blood lymphocytes.

The genotoxic effects of benzene were assessed in peripheral blood lymphocytes of 49 workers occupationally exposed to benzene (3-68.7 mg/m3 in the work environment) for 0-2, 2-10 and more than 10 years (10, 22 and 17 workers, respectively). Chromosomal aberrations, SCEs and UV-induced DNA synthesis were used as indicators of genotoxic effects. Most of the workers were followed up in 1991 and 1992, while the benzene concentrations were reduced to 1-18.4 mg/m3 air. Considered overall, in the "exposed" groups, the frequencies of chromosomal aberrations were significantly higher than in controls

Benzene exposures during tank washing activities on crude oil tankers.

Personal benzene exposure data during tank washing aboard crude oil tankers are presented. The data are from 4 monitoring surveys conducted by a tanker ship company in the 1990s. Among 59 partial-shift samples for personnel on deck during tank washing, the sample mean was 0.49 ppm and 44% of the corresponding minimum 8-h time-weighted average (TWA) benzene exposure values exceeded 0.2 ppm (the European Union 8-h TWA benzene limit). Among 13 full-shift samples for personnel on deck, the equivalent 8-h TWA benzene exposure values had a sample mean of 0.44 ppm, and a parametrically estimated 85%

Simulation of nonequilibrium heat conduction in benzene single-molecule junctions.

Problems of heat transport are ubiquitous to various technologies such as power generation, cooling, and thermoelectrics. In particular, the increased heat generated due to the miniaturization of electronic devices entails the degradation of its semiconductor components. This problem has lead to the emergence of single-molecule electronics, where long-chain molecules attached to tiny electrodes are used to transport and switch electrons. Thus the related problem of thermal transport through molecular junctions (MJs) has received considerable theoretical and experimental attention. In this work

RAD51 m6A and PARylation modification orchestrate benzene-induced haematological toxicity.

It is well-established that occupational benzene exposure can cause leukemia. DNA damage repair has great potential in assessing the health risk of benzene, but the role of RAD51, an executor of DNA damage repair, remains unclear in benzene-induced hematotoxicity. Here, we demonstrate that exposure to hydroquinone (HQ), a key metabolite of benzene, upregulates RAD51 expression in human lymphoblast (TK6), consequently suppressing DNA damage. Mechanistically, we demonstrate that METTL3, an m6A methyltransferase, catalyzes m6A modification on RAD51 mRNA, and that the m6A reader protein YTHDC1 sub

Benzene-Linked Polymeric Carbon Nitride for Enhanced Photocatalytic Hydrogen Production.

Cross-linking with functional molecular species in polymeric carbon nitride (PCN) could offer a positive strategy that tunes its molecular structure with excellent conductivity to improve photocatalytic activity. Herein, the benzene ring-cross-linked photocatalyst is obtained via the polymerization of urea, melamine, and trimesic acid. Benzene ring-cross-linked PCN narrows the band gap and augments the push-pull effect of carriers, thus enhancing visible light harvesting and transfer easiness of photogenerated electron/hole pairs. Notably, the amount of trimesic acid was optimized during the b

Dearomative Intramolecular meta-Thermocycloadditions of Benzene Rings via Wheland Intermediates.

Dearomative cycloadditions are powerful synthetic transformations utilizing aromatic compounds for cycloaddition reactions. They have been extensively applied to the synthesis of biologically relevant compounds not only because of the complexity generated from simplicity but also the atom- and step-economy. For the most studied yet challenging benzene ring systems, ortho- and para-cycloadditions have been realized both photochemically and thermally, while the meta-cycloadditions are still limited to the photochemical processes tracing back to the 1960s. Herein, we for the first time realized t

Novel benzene sulfonamides with acetylcholinesterase and carbonic anhydrase inhibitory actions.

A series of benzene sulfonamides 15-26 were synthesized and determined for their in vitro and in silico inhibitory profiles toward acetylcholinesterase (AChE) and carbonic anhydrases (CAs). Commercially available 3,4-dimethoxytoluene was reacted with chlorosulfonic acid to furnish benzene sulfonyl chloride derivatives. The reaction of substituted benzene sulfonyl chloride with some amines also including (±)-α-amino acid methyl esters afforded a series of novel benzene sulfonamides. In this study, the enzyme inhibition abilities of these compounds were evaluated against AChE and CAs. They exhib

Cancer incidence in Swedish oil refinery workers exposed to benzene.

BACKGROUND: Oil refinery workers are exposed to benzene, which is a well-known cause of leukaemia, but results on leukaemia in oil refinery workers have been mixed, and the data on workers' exposure is limited. Oil refinery workers are also exposed to asbestos and several studies have shown increased risk of mesothelioma. AIM: The objective was to investigate cancer incidence, especially leukaemia, at low to moderate exposure to benzene in an update of a previous study of employees at three Swedish oil refineries. METHODS: Cancer incidence was followed up in 2264 men (1548 refinery operators

Metagenomic and Genomic Sequences from a Methanogenic Benzene-Degrading Consortium.

Draft and complete metagenome assembled genomes (MAGs) were created from multiple metagenomic assemblies of DGG-B, a strictly anaerobic, stable mixed microbial consortium that degrades benzene completely to methane and CO2. Our objective was to obtain closed genome sequences of benzene-fermenting bacteria to enable the elucidation of their elusive anaerobic benzene degradation pathway.

Leukemia Mortality among Benzene-Exposed Workers in Brazil (2006-2011).

BACKGROUND: In this study, the annual leukemia mortality rate is estimated by occupational groups potentially exposed to benzene in Brazil and compared to non-exposed workers by sex. METHODS: Data were extracted from the Mortality Information System and the National Institute of Geography and Statistics from 2006 to 2011. Occupational groups exposed to benzene were defined by using the Finnish Job-Exposure Matrix, FINJEM. RESULTS: We found 21,049 leukemia deaths in 1917 in occupational groups potentially exposed to benzene, corresponding to an annual average mortality rate of 4.5/100,000, hi

Carbonic anhydrase inhibitory activity of phthalimide-capped benzene sulphonamide derivatives.

A series of phthalimide-capped benzene sulphonamides (1-22) reported by our group for dengue protease inhibitory activity have been evaluated for their carbonic anhydrase (hCA, EC 4.2.1.1) inhibitory activity against hCA I, hCA II. Compounds 1, 3, 10, and 15 showed hCA I inhibition, whereas 1, 4, and 10 showed hCA II inhibition at nanomolar concentrations. Among these compounds, 1 displayed potent inhibitory activity against the hCA I (Ki = 28.5 nM) and hCA II (Ki = 2.2 nM), being 10 and 6 times more potent than acetazolamide, a standard inhibitor (Ki = 250 nM and 12 nM), respectively. Non-coding RNAs: A new frontier in benzene-mediated toxicity.

One of the most frequent environmental contaminants, benzene is still widely used as an industrial solvent around the world, especially in developing nations, posing a serious occupational risk. While the processes behind the toxicity of benzene grounds are not fully understood, it is generally accepted that its metabolism, which involves one or more reactive metabolites, is crucial to its toxicity. In order to evaluate the many ways that benzene could influence gene regulation and thus have an impact on human health, new methodologies have been created. The pathophysiology of the disorder may

Synthesis and Antioxidant Properties of HeteroBisNitrones Derived from Benzene Dicarbaldehydes.

We report herein the synthesis and antioxidant profile of nine novel heterobisnitrones (hBNs) as new α-phenyl-tert-butylnitrone (PBN) analogues. The synthesized hBNs 1-9 were evaluated for their antioxidant activity using different in vitro techniques, while they were also tested as inhibitors of soybean LOX, as an indication of their anti-inflammatory effect. Nitrone hBN9 is the most potent antioxidant presenting higher anti-lipid peroxidation and hydroxyl radicals scavenging activities as well as higher lipoxygenase inhibition. In silico calculations reveal that hBN9 follows Lipinski's rule

Glycine/glycine N-methyltransferase/sarcosine axis mediates benzene-induced hematotoxicity.

Benzene, an important and widely used industrial chemical, is the cause of different types of blood disorders. However, the mechanisms of benzene-induced hematotoxicity are still unclear. This study aimed to explore the effects of benzene on metabolism, especially in amino acid metabolism, in human peripheral blood B lymphocyte cells (AHH-1 cells) treated with 1,4-benzoquinone (1,4-BQ) and in benzene-exposed population based on the un-targeted and targeted metabolomics platforms. The results showed that 1,4-BQ disturbed the metabolic activity, such as arginine biosynthesis, citrate cycle, glyc
