Postmortem biochemistry in deaths from ischemic heart disease.

Ischemic heart disease (IHD) is one of the leading causes of morbidity and sudden cardiac death worldwide and is an important public health problem. The presence of ischemia in clinical applications can be detected by ECG, biochemical markers, and radiological methods. Myocardial infarction is also frequently encountered in forensic autopsies. Postmortem diagnosis is determined as a result of histopathological examinations and additional exclusionary examinations (toxicology, microbiology, etc.). However, routine histopathological examinations are insufficient, especially when death occurs in

Lysosomal multienzyme complex: biochemistry, genetics, and molecular pathophysiology.

Lysosomal enzymes sialidase (alpha-neuraminidase), beta-galactosidase, and N-acetylaminogalacto-6-sulfate sulfatase are involved in the catabolism of glycolipids, glycoproteins, and oligosaccharides. Their functional activity in the cell depends on their association in a multienzyme complex with lysosomal carboxypeptidase, cathepsin A. We review the data suggesting that the integrity of the complex plays a crucial role at different stages of biogenesis of lysosomal enzymes, including intracellular sorting and proteolytic processing of their precursors. The complex plays a protective role for a

Snake venom constituents: biochemistry and toxicology (Part 1).

Complicated and mystifying as the snake envenomation process may appear, the toxic principles of snake venoms are biochemical entities that could be isolated, purified and characterized. In this review, the toxicological process of envenomation and the biochemical basis of venom toxicity are presented for those not directly involved in venom research but who nevertheless have an interest in snake venom poisoning.

The Biochemistry Behind Cognitive Decline: Biomarkers of Alzheimer's Disease.

Alzheimer's disease (AD) is the most prevalent type of dementia. Pathologically, the disease is marked by neurofibrillary tangles (NFT), which are aberrant accumulations of the tau protein that develop inside neurons, and extracellular plaque deposits of the amyloid β peptide (Aβ). These pathological lesions are present in the brain before the beginning of clinical manifestations. However, despite advancements in the comprehension of AD pathophysiology, timely and accurate clinical diagnosis remains challenging. Therefore, developing biomarkers capable of detecting AD during the preclinical ph

Muscular dystrophy meets protein biochemistry, the mother of invention.

Muscular dystrophies result from a defect in the linkage between the muscle fiber cytoskeleton and the basement membrane (BM). Congenital muscular dystrophy type MDC1A is caused by mutations in laminin α2 that either reduce its expression or impair its ability to polymerize within the muscle fiber BM. Defects in this BM lead to muscle fiber damage from the force of contraction. In this issue of the JCI, McKee and colleagues use a laminin polymerization-competent, designer chimeric BM protein in vivo to restore function of a polymerization-defective laminin, leading to normalized muscle structu

Comparative biochemistry of Giardia, Hexamita and Spironucleus: Enigmatic diplomonads.

The diplomonad genera are here represented by three highly diverse species, both free-living (Hexamita inflata), and parasitic (Spironucleus vortens and Giardia intestinalis). All three are moderately aerotolerant flagellates, inhabiting environments where O2 tensions are low and fluctuating. Many diplomonads are opportunistic pathogens of avian, terrestrial and aquatic animals. Hexamitids inhabit deep waters and sediments of lakes and marine basins, S. vortens commonly infects the intestinal tract of ornamental fish, particularly of cichlids and cyprinids, and G. intestinalis, the upper intes

A focused assignment encouraging deep reading in undergraduate biochemistry.

Encouraging undergraduate students to access, read, and analyze current primary literature can positively impact learning, especially in advanced courses. The incorporation of literature into coursework typically involves reading and responding to full research reports. Such exercises have clear value as students make connections between experiments and are able to probe and critique scientific logic. The exclusive use of full papers, though, may reinforce certain students' tendencies to rely on textual clues rather than a critical analysis of the actual data presented. I propose that structur

A pharmacy practice laboratory exercise to apply biochemistry concepts.

OBJECTIVES: To develop exercises that allow pharmacy students to apply foundational knowledge discussed in a first-professional year (P1) biochemistry course to specific disease states and patient scenarios. DESIGN: A pharmacy practice laboratory exercise was developed to accompany a lecture sequence pertaining to purine biosynthesis and degradation. The assignment required students to fill a prescription, provide patient counseling tips, and answer questions pertaining to the disease state, the underlying biochemical problem, and the prescribed medication. ASSESSMENT: Students were graded o

Variant late infantile neuronal ceroid-lipofuscinosis: pathology and biochemistry.

The neuronal ceroid-lipofuscinoses (NCL) are among the most common inherited neurodegenerative disorders of childhood. The genomic defect causing a variant late infantile neuronal ceroid-lipofuscinosis (vLINCL, also called CLN-5 or variant Jansky-Bielschowsky disease) has recently been localized to chromosome 13q22, thus delineating this disease as a separate entity. This particular form of NCL is clinically well defined, but lacks pathomorphological and biochemical description. The present analyses indicate that subunit c of the mitochondrial ATP synthase is the major protein in vLINCL brain

Biochemistry of thyroid regulation under normal and abnormal conditions.

Perhaps in an oversimplified view, abnormal thyroid growth can be classified into two main categories: a) those cases due to excess of thyroid stimulators extrinsic to the gland; b) situations in which an intrinsic alteration in the gland occurs: Extrinsic (excess thyroid stimulation) Iodide deficiency with elevated TSH Goitrogens Graves' immunoglobulins Thyroid stimulating factors produced by tumors Dishormonogenesis with hypothyroidism Intrinsic (normal TSH) Increased sensitivity to TSH (iodine depletion) Altered autoregulation (?) Abnormal TSH receptor Other biochemical abnormalities From t

[Biochemistry and prepared diagnosis of lung maturation (author's transl)].

The composition and structure of acellular alveolar material are described. Evidences as summarized that 1,2-dipalmityl-sn-glycero-phosphoryl-choline, depending of the presence of other components of the hypophasis, determines the surface tension of the air-alveolus interface. The foetal maturation of the lung is defined according to the Avery-Clements-Gluck theory, as a stabilisation of the alveolar lecthin production; and the dependence of the relevant enzyme activities on the stage of the pregnancy is discussed. Attention is drawn to the possibility of enzyme induction by medication; the bi

Alcohol consumption: from biochemistry to behavior, health, and beyond.

Alcohol metabolism is a multi-step biochemical process driven primarily by hepatic oxidation of ethanol to acetaldehyde and then to acetate. Although this pathway protects the body from prolonged ethanol exposure, transient acetaldehyde accumulation generates substantial toxicity, and chronic alcohol intake activates auxiliary pathways that amplify oxidative stress, inflammation, and tissue injury. Both metabolic capacity and physiological response to alcohol vary widely between individuals, reflecting genetic and epigenetic variation in the alcohol- and aldehyde-dehydrogenase enzymes, the cyt

Substrate for Thyroid Hormone Synthesis: Biochemistry, Evolution, and Physiology.

The use of iodotyrosines to control development and other biological processes predates the evolution of the thyroid gland. Endogenous thyroid hormone synthesis evolved with the ability to enzymatically iodinate protein secreted and directed for gastrointestinal proteolysis-and evolved further to specialize for iodination of protein entrapped extracellularly within closed follicles. The evolutionary appearance of the vertebrate thyroid gland with its follicular architecture correlates closely with the first appearance of thyroglobulin, which acts as the primary protein scaffold for thyroid hor

Morphology, physiology, and biochemistry of zinc-stressed caraway plants.

A greenhouse pot experiment was conducted to evaluate the impact of zinc supply (0, 1, and 2 mM Zn as ZnSO4) on morpho-physiological and biochemical parameters of caraway (Carum carvi L.). Exposure to different Zn concentrations for 12 weeks compromised severely all growth parameters (plant height, number of secondary branches, diameter of primary and secondary branches, fresh and dry weight of aerial parts and roots) yield and its components (number of umbels per primary branches and secondary branches; number of umbel per plant; number of seeds per plant; and the weight of 1000 seeds). Calcium citrate: from biochemistry and physiology to clinical applications.

Adequate daily calcium intake should normally be achieved by dietary sources. Since low calcium diets are quite common in subjects that do not reach the recommended intake and particularly those at risk of fractures, calcium supplements may become necessary. Different forms of calcium salts are available, but products containing calcium citrate and calcium carbonate complexes are the most frequently used. Although only limited evidence on the efficacy and long-term safety of calcium citrate is available, these supplements may represent a valuable product for the management of different chronic
