Improvement of Antialveolar echinococcosis efficacy of novel Albendazole-Bile acids Derivatives with Enhanced Oral Bioavailability.

Alveolar echinococcosis (AE) is a chronic and fatal infectious parasitic disease, which has not been well-researched. Current recommended therapies for AE by the World Health Organization include complete removal of the infected tissue followed by two years of albendazole (ABZ), administered orally, which is the only effective first-line anti-AE drug. Unfortunately, in most cases, complete resection of AE lesions is impossible, requiring ABZ administration for even longer periods. Only one-third of patients experienced complete remission or cure with such treatments, primarily due to ABZ's low

Bioprocessing of Pea Protein can Enhance Fortified Fe But Reduce Zn In Vitro Bioaccessibility.

The bioaccessibility of minerals during food digestion is essential in facilitating absorption and hence mineral bioavailability. Bioprocessing approaches have shown promising effects on Fe and Zn bioaccessibility in plant food matrices. In this study, lactic acid bacteria fermentation or enzymatic hydrolysis was performed on pea protein concentrates (PPCs) to investigate their effects on the bioaccessibility of fortified Fe and Zn salts. Simulated digestion studies revealed that enzymatic hydrolysis was more effective than fermentation. Phytase treatment significantly (P < 0.05) improved Fe3+

Rapamycin nanoparticles improves drug bioavailability in PLAM treatment by interstitial injection.

BACKGROUND: Pulmonary lymphangiomyomatosis (PLAM) is a rare interstitial lung disease characterized by diffuse cystic changes caused by the destructive proliferation of smooth muscle-like cells or LAM cells. PLAM is more common in young women than other people, and a consensus is lacking regarding PLAM treatment. The clinical treatment of PLAM is currently dominated by rapamycin. By inhibiting the mTOR signaling pathway, rapamycin can inhibit and delay PLAM's occurrence and development. However, the application of rapamycin also has shortcomings, including the drug's low oral bioavailability a

Curcumin and Derivatives in Nanoformulations with Therapeutic Potential on Colorectal Cancer.

There is growing concern in the rise of colorectal cancer (CRC) cases globally, and with this rise is the presentation of drug resistance. Like other cancers, current treatment options are either invasive or manifest severe side effects. Thus, there is a move towards implementing safer treatment options. Curcumin (CUR), extracted from Curcuma longa, has received significant attention by scientists as possible alternative to chemotherapeutic agents. It is safe and effective against CRC and nontoxic in moderate concentrations. Crucially, it specifically modulates apoptotic effects on CRC. Howeve

Application of curcumin nanoformulations in Alzheimer's disease: prevention, diagnosis and treatment.

Objectives: Alzheimer's disease (AD) is a serious neurodegenerative disease. Although many therapeutic strategies have been studied, their clinical applications are immature. Moreover, these methods can only alleviate symptoms rather than cure it, posing a challenge to brain health in older adults worldwide. Curcumin (CUR) is a very promising natural compound for nerve protection and treatment. It can prevent and treat AD, and on the other hand, its fluorescence properties can be used in the diagnosis of AD. However, CUR is characterized by very low water solubility, fluid instability, rapid m

Insulin-loaded liposomes packaged in alginate hydrogels promote the oral bioavailability of insulin.

Compared to subcutaneous injections, oral administration of insulin would be a preferred route of drug administration for diabetic patients. For oral delivery, both liposomes and alginate hydrogels face many challenges, including early burst release of the encapsulated drug and poor intestinal drug absorption. Also, adhesion to the intestinal mucosa remains weak, which all result in a low bioavailability of the payload. This study reports on an alginate hydrogel loaded with liposomes for oral insulin administration. Liposomes (Lip) loaded with arginine-insulin complexes (AINS) were incorporate

Models to evaluate the barrier properties of mucus during drug diffusion.

Mucus is widely disseminated in the nasal cavity, oral cavity, respiratory tract, eyes, gastrointestinal tract, and reproductive tract to prevent the invasion of pathogenic bacteria and toxins. The mucus layer through its continuous secretion can prevent the passage of macromolecular substances such as pathogenic bacteria and toxins, thereby reducing the occurrence of inflammation. Without a doubt, mucus also hinders oral absorption. The physiological and biochemical properties of intestinal mucus and the different types of mucus barrier models need to be predominated. To find ways to increase

Extremely short bioavailability and fast pharmacodynamic effects of pMHC-based nanomedicines.

Nanoparticles (NPs) coated with autoimmune disease-relevant peptide-major histocompatibility complexes (pMHCs) can blunt autoimmune diseases by re-programming cognate effector T-lymphocytes into disease-suppressing regulatory T-cells, followed by massive expansion. Here, a method to quantify the absolute amounts of the active drug product is developed, to understand the relationship between bioavailability and pharmacodynamics. Incubation with plasma results in the formation of a protein corona that stabilizes the directional pMHC coat, shielding it from proteolysis or anti-drug antibody recog

Nicotinic acid supplementation at a supraphysiological dose increases the bioavailability of NAD+ precursors in mares.

NAD+ deficiency has recently been linked with increased occurrences of congenital abnormalities and embryonic death in human and animal subjects. Early embryonic death is a major component of pregnancy loss in mares and very little is known regarding the requirement for NAD+ in horses. The aim of this study was to quantify NAD+ and its metabolites in the plasma and urine of mares after orally administering an acute dose of nicotinic acid and determine the absorption, metabolism and excretion of this essential precursor for NAD+ biosynthesis. Nicotinic acid (5 g per os) was administered to four

Evaluation of mercury bioavailability to vegetables in the vicinity of cinnabar mine.

Knowledge of the concentration of the bioavailable forms of mercury in the soil is necessary, especially, if these soils contain above-limit total mercury concentrations. The bioavailability of mercury in soil samples collected from the vicinity of abandoned cinnabar mines was evaluated using diffusive gradients in the thin films technique (DGT) and mercury phytoaccumulation by vegetables (lettuce, spinach, radish, beetroot, carrot, and green peas). Mercury was accumulated primarily in roots of vegetables. The phytoaccumulation of mercury into edible plant parts was site-specific as well as ve

Evolution of enzyme activity, heavy metals bioavailability and microbial community in different temperature stages of the co-bioevaporation process.

Laboratory investigations documented enzyme activity, heavy metals' bioavailability and the bacterial community during co-bioevaporation treatment of food waste and landfill leachate. The activities of dehydrogenase, protease, urease and phosphatase were sensitive to the changes in operating temperature inherent in co-bioevaporation. The maximum dehydrogenase activity was appeared at warming 30 °C. The maximum hydrolytic activity of the microorganisms on protein, urea and phosphorus-containing organic compounds appeared at warming 50 °C. The bacteria mainly gathered on the surface and in the p

Comparable effects of manure and its biochar on reducing soil Cr bioavailability and narrowing the rhizosphere extent of enzyme activities.

Chromium (Cr) contamination is especially hazardous to soil biota. Application of manure and biochar has been frequently proposed to remediate Cr-contaminated soil. However, the understanding of mechanisms behind manure and biochar impacts on soil enzyme activities requires advanced visualization technologies. For the first time, we compared manure and its biochar influence on the spatial distribution of β-glucosidase, N-acetyl-glucosaminidase and phosphomonoesterase activities in Cr-contaminated soil using direct zymography. Maize was planted for 45 days in (a) soil mixed with manure, (b) soi

Strategies to Improve Bioavailability and In Vivo Efficacy of the Endogenous Opioid Peptides Endomorphin-1 and Endomorphin-2.

Morphine and the other alkaloids found in the opium poppy plant still represent the preferred therapeutic tools to treat severe pain in first aid protocols, as well as chronic pain. The use of the opiate alkaloids is accompanied by several unwanted side effects; additionally, some forms of pain are resistant to standard treatments (e.g. neuropathic pain from cancer). For these reasons, there is currently renewed interest in the design and assay of modified versions of the potent endogenous opioid peptides endomorphin-1 and endomorphin-2. This review presents a selection of the strategies direc

Development of Comparative Toxicity Potentials of 14 cationic metals in freshwater.

Site-dependent and site-generic Comparative Toxicity Potentials (CTPs) (also known as Characterization Factors (CFs)) were calculated for 14 cationic metals (Al(III), Ba, Be, Cd, Co, Cr(III), Cs, Cu(II), Fe(II), Fe(III), Mn(II), Ni, Pb, Sr and Zn), to be applied in Life Cycle Impact Assessment. CTPs were calculated for 7 EU-archetypes, taking bioavailability and speciation pattern into account. The resulting site-dependent CTPs showed up to 2.4-6.5 orders of magnitude variation across archetypes for those metals that form stable hydroxyl compounds in slightly alkaline waters (Al(III), Be, Cr(I

Toxicity of sediment-associated unresolved complex mixture and its impact on bioavailability of polycyclic aromatic hydrocarbons.

Unresolved complex mixtures (UCMs) and polycyclic aromatic hydrocarbons (PAHs) are ubiquitous in sediment originating from oil leaks, shipping, and other human activities and thus it is necessary to understand the role of UCM on sediment toxicity and PAH bioaccumulation. In the current study, lethal and sublethal effects of sediment-associated UCM were examined in two benthic invertebrates (Chironomus dilutus and Lumbriculus variegatus) using two spiked sediments. Results showed that UCM alone was toxic to the organisms and its toxicity was species-dependent. Approximately 1% of UCM in sedimen

Inhibitory effect of encapsulated curcumin on ultraviolet-induced photoaging in mice.

Photoaging is the superposition of photodamage (ultraviolet [UV] radiation-induced) on the aging process. It is a major damaging consequence of free radical action. Curcumin (diferuloylmethane) is a phytochemical with diverse antioxidant and antiinflammatory properties. However, it shows a poor topical bioavailability. Therefore, we encapsulated curcumin in elastic vesicles (EVs) and investigated different doses (1, 3, 5 and 10 micromol) for its in vivo antiaging activity in mice. VICCO turmeric served as the marketed control, and free curcumin dispersed in an ointment base was another control
