Mandible asymmetry and genetic diversity in island populations of the common shrew, Sorex araneus.

Mandibles from 13 island and six mainland populations of common shrews from the west coast of Scotland were subjected to geometric morphometric analysis in order to investigate the relationship between genetic diversity and fluctuating asymmetry. Although population mean shape fluctuating asymmetry (FA) and size FA were significantly inversely correlated with population genetic diversity this result was substantially due to one island. Sanda, the smallest island with by far the lowest genetic diversity, also had the highest FA. When Sanda was removed from the analysis, the relationship was not

Platelet alpha 2-adrenoceptor binding and function during the menstrual cycle.

Blood platelet receptors are widely used as peripheral models of central nervous system receptors, particularly in attempts to understand the biological basis of a number of neurological and psychiatric disorders. It is important to differentiate factors other than the primary disease process which may influence platelet receptors. One such potential factor is the menstrual cycle. In this study we have determined platelet high-affinity alpha 2-adrenoceptor binding, using the agonist ligand 3H-UK-14,304 and platelet aggregatory responses to adrenaline in 14 healthy young women, sampled on four

High baseline expression of IL-6 and IL-10 decreased CCR7 B cells in individuals with previous SARS-CoV-2 infection during BNT162b2 vaccination.

The current pandemic generated by SARS-CoV-2 has led to mass vaccination with different biologics that have shown wide variations among human populations according to the origin and formulation of the vaccine. Studies evaluating the response in individuals with a natural infection before vaccination have been limited to antibody titer analysis and evaluating a few humoral and cellular response markers, showing a more rapid and intense humoral response than individuals without prior infection. However, the basis of these differences has not been explored in depth. In the present work, we analyz

Why don't all birds lay early? Causes and consequences of breeding timing for a long-lived migratory seabird.

The relationship between earlier laying and higher breeding success is well-established in avian species. However, a wide distribution of breeding timings is often maintained within a population, and the causes and consequences of this variation require further exploration, particularly as many avian species advance breeding phenology in response to climate change. Using a 16-year dataset combining colony observations and biologging of Manx shearwaters (Puffinus puffinus), we show that earlier laying does not always result in higher breeding success, with later laying becoming less costly in r

Population pharmacokinetics of efavirenz in HIV and TB/HIV coinfected children: the significance of genotype-guided dosing.

OBJECTIVES: The current WHO weight-based dosing recommendations for efavirenz result in a wide variability of drug exposure in children. Our objectives are to characterize the effects of rifampicin- and isoniazid-containing anti-TB therapy and CYP2B6 activity on efavirenz concentrations in children, using non-linear mixed-effects modelling. METHODS: This is a pharmacokinetic (PK) substudy of a prospective study that examined the interactions between anti-TB therapy and efavirenz in HIV-infected children with and without TB. PK samples were obtained 4 weeks after starting efavirenz (PK1) and r

Clinical Outcomes of Atlantoaxial Dislocation Combined with High-Riding Vertebral Artery Using C2 Translaminar Screws.

OBJECTIVE: Atlantoaxial stabilization procedures in high-riding vertebral artery (HRVA) cases are challenging. C2 translaminar screws are rigid and pose no risk to the vertebral artery. The aim of this study was to present clinical outcomes of atlantoaxial dislocation combined with HRVA using C2 translaminar screws. METHODS: Cases of atlantoaxial dislocation combined with HRVA surgically treated in our institution from 2007 to 2015 were retrospectively reviewed. Atlantodental interval and clivus-axial angle were measured. The Japanese Orthopaedic Association scale was used to evaluate neurolo

Germline variants are associated with increased primary melanoma tumor thickness at diagnosis.

Germline genetic variants have been identified, which predispose individuals and families to develop melanoma. Tumor thickness is the strongest predictor of outcome for clinically localized primary melanoma patients. We sought to determine whether there is a heritable genetic contribution to variation in tumor thickness. If confirmed, this will justify the search for specific genetic variants influencing tumor thickness. To address this, we estimated the proportion of variation in tumor thickness attributable to genome-wide genetic variation (variant-based heritability) using unrelated patient

The influence of short-term temporal variability on the efficacy of dragonfly larvae as mercury biosentinels.

Mercury (Hg) exposure to fish, wildlife, and humans is widespread and of global concern, thus stimulating efforts to reduce emissions. Because the relationships between rates of inorganic Hg loading, methylmercury (MeHg) production, and bioaccumulation are extremely complex and challenging to predict, there is a need for reliable biosentinels to understand the distribution of Hg in the environment and monitor the effectiveness of reduction efforts. However, it is important to assess how temporal and spatial variation at multiple scales influences the efficacy of specific biosentinels. Seasonal

[Ultrasound-Guided PRP for Tendinopathy: a Practical, Multimodal Framework from Biology to Rehabilitation].

Tendinopathies are common and hete- rogeneous, and recovery depends on both the local biological milieu and appropriately progressed mechanical loading. Platelet-rich plasma (PRP) is a biologically plausible adjunct, yet reported clinical effects are inconsistent, likely due to variation in phenotype, PRP formulation and dosing, delivery technique, and rehabilitation protocols. Ultrasound guidance improves injection accuracy and safety, but its incremental clinical value may be context dependent. This review outlines tendon-healing biology, summarizes evidence for conservative management, and

Improving results in rat fracture models: enhancing the efficacy of biomechanical testing by a modification of the experimental setup.

BACKGROUND: Animal fracture models, primarily performed in rats, are crucial to investigate normal and pathological bone healing. However, results of biomechanical testing representing a major outcome measure show high standard deviations often precluding statistical significance. Therefore, the aim of our study was a systematical examination of biomechanical characteristics of rat femurs during three-point bending. Furthermore, we tried to reduce variation of results by individually adapting the span of bearing and loading areas to the bone's length. METHODS: We examined 40 paired femurs of

Radiobiological Implications of Fukushima Nuclear Accident for Personalized Medical Approach.

On March 11, 2011, a devastating earthquake and subsequent tsunami caused serious damage to areas of the Pacific coast in Fukushima prefecture and prompted fears among the residents about a possible meltdown of the Fukushima Daiichi Nuclear Power Plant reactors. As of 2017, over six years have passed since the Fukushima nuclear crisis and yet the full ramifications of the biological exposures to this accidental release of radioactive substances remain unclear. Furthermore, although several genetic studies have determined that the variation in radiation sensitivity among different individuals i

The Biobank Rare Variant consortium powers the discovery of rare genetic associations through global collaboration.

Rare coding variants can have large effects on disease risk and provide direct routes from human genetics to disease mechanisms and therapeutic targets, but their discovery is constrained by sample size, particularly for low-prevalence diseases. Here we establish the Biobank Rare Variant Analysis (BRaVa) consortium, a global rare variant association resource that integrates sequencing and linked health-record data from ten biobanks and cohorts comprising over 1.2 million individuals across diverse ancestries. We performed gene-based meta-analyses of rare coding variation across 33 clinical end

Comparison of library preparation protocols and bioinformatic pipelines in high-throughput 16S rRNA gene sequencing.

BACKGROUND: 16S rRNA gene sequencing is widely used for bacterial community profiling in both clinical and research contexts. The expanding availability of library preparation protocols and bioinformatic pipelines increases analytical flexibility but may also introduce method-dependent biases that affect inferred microbial composition and relative abundance estimates. The relative impact of library preparation protocol, amplicon region, and bioinformatic pipeline on species-level taxonomic inference and compositional agreement remains insufficiently characterised. We therefore compared the Ill

Bridging Gaps in Postpartum Depression Research: Pathways to Personalized Medicine in Women's Mental Health.

Postpartum depression (PPD) is a prevalent and serious mental health disorder that affects nearly 15% of women after childbirth. It disrupts maternal well-being, infant development, and family relationships, and often remains underdiagnosed due to its complex biological, psychological, and social determinants. Conventional treatment strategies largely follow generalized, symptom-based approaches that fail to address the wide individual variation in hormonal sensitivity, genetic background, and psychosocial context among affected women. This review emphasizes the importance of adopting a person

Organochlorines affect the major androgenic hormone, testosterone, in male polar bears (Ursus maritimus) at Svalbard.

Normal sexual development and subsequent reproductive function are dependent on appropriate testosterone production and action. The regulation of steroid hormones, including androgens, can be influenced by both biological and environmental factors, including environmental chemicals. Concentrations of organochlorines are considerably greater in Svalbard polar bears than in polar bears from other regions. Between 1995 and 1998, samples were collected from 121 male polar bears (Ursus maritimus) from the Svalbard area. In this study, testosterone concentration variations were described for male po

Epigenomics and Ayurgenomics: Molecular Analysis of Ayurvedic Prakriti in Personalized Healthcare.

Personalized healthcare increasingly requires models that explain individual variation in disease risk, prevention, and treatment response. Ayurvedic Prakriti offers a constitution-based framework for classifying human variability through Vata, Pitta, and Kapha phenotypes, while Ayurgenomics attempts to connect these traditional classifications with molecular biology. Despite growing interest, the biological relationship between Prakriti, epigenomic regulation, and precision healthcare remains insufficiently established, particularly because available evidence is limited by small cohorts, hete

Proteomic signatures of smoking and their associations with risk of incident diseases and mortality in diverse populations.

Smoking is the most important behavioural determinant of morbidity and mortality. Using machine learning on plasma levels of 2,917 proteins in the UK Biobank (n = 43,914), we develop a proteomic Smoking Index (pSIN) comprising 51 proteins that accurately distinguish current from never smokers (AUC = 0.95; 95% CI 0.94-0.95). Validation in the China Kadoorie Biobank (n = 3,977) shows similar accuracy (AUC = 0.91; 95% CI 0.89-0.92). pSIN is significantly associated with the risk of all-cause mortality and 18 major chronic diseases, including cardiovascular, renal, pulmonary, neurodegenerative, an

Cross-Country Individual Participant Analysis of 4.1 Million Singleton Births in 5 Countries with Very High Human Development Index Confirms Known Associations but Provides No Biologic Explanation for 2/3 of All Preterm Births.

BACKGROUND: Preterm birth is the most common single cause of perinatal and infant mortality, affecting 15 million infants worldwide each year with global rates increasing. Understanding of risk factors remains poor, and preventive interventions have only limited benefit. Large differences exist in preterm birth rates across high income countries. We hypothesized that understanding the basis for these wide variations could lead to interventions that reduce preterm birth incidence in countries with high rates. We thus sought to assess the contributions of known risk factors for both spontaneous

PDCD1 SNPs and alloimmune transplant outcomes: mechanistic basis and human evidence across solid organ transplantation and allogeneic hematopoietic stem cell transplantation.

BACKGROUND: The programmed cell death 1 gene (PDCD1) encodes programmed death-1 protein (PD-1), an inhibitory immune-checkpoint receptor that regulates peripheral tolerance and alloimmune activation. SCOPE: This narrative review synthesizes mechanistic and human evidence linking PDCD1 single-nucleotide polymorphisms (SNPs) to transplant-related alloimmune outcomes. In solid organ transplantation (SOT), outcomes are framed mainly as rejection-related endpoints, whereas in allogeneic hematopoietic stem cell transplantation (allo-HSCT), bone marrow transplantation (BMT), and cord blood transplan

Differences in Salmonella Typhimurium infection and excretion among laboratory and field strains of the German cockroach suggest a genomic basis for vector competence.

The German cockroach, Blattella germanica, can be a vector of human enteric bacterial pathogens, including Salmonella enterica serovar Typhimurium (S. Typhimurium). Transmission of such pathogens by cockroaches has largely been considered a passive mechanical process, but recent studies have argued against this dogma by demonstrating bacterial proliferation within the cockroach gut and the necessity of specific bacterial genes for successful transmission in the feces, revealing unappreciated biological complexity in the vector-pathogen relationship between cockroaches and S. Typhimurium. Howev
