Research progress on Sertoli cell secretion during spermatogenesis.

Sertoli cells (SCs), as the somatic cells in the testis of male mammals, play a crucial role in the close association with germ cells. The blood-testicular barrier (BTB), established by their tight junctions, provides immune protection to germ cells, leading to their characterization as "sentinel" cells. Moreover, the physiological process of testicular development and spermatogenesis in male animals is intricately tied to the secretory activities of SCs. These cells secrete a diverse array of proteins and cytokines that interact with various targets, working in concert with mechanisms in the

Possible physiological bases for contraceptive techniques in the male.

Aspects of the physiology of the testis and epididymis are discussed which might be relevant to future methods of contraception in the male. These include the blood-testis barriers, both at the tubular and vascular level, testicular and scrotal thermoregulation and maturation of spermatozoa in the epididymis. Possible adverse consequences of interfering with spermatogenesis are also considered, such as reduced endocrine responsiveness of the testis, increased incidence of genetic abnormalities and abnormal development of embryos produced by sub-fertile males.

Tissue microcircumstances for leukocytic infiltration into the testis and epididymis in mice.

Spermatozoa do not appear in the seminiferous epithelium until puberty, when immune tolerance has already been established. Therefore, they contain various autoimmunogenic materials which are recognized as foreign by the self immune system. However, the testis and epididymis are known as immunologically privileged organs. In particular, the blood-testis barrier (BTB) formed by Sertoli cells and the blood-epididymal barrier formed by epididymal epithelial cells protect autoimmunogeneic spermatozoa from attack by the self immune system. The immune privileged circumstances in the testis and epidi

Revisiting cadmium-induced toxicity in the male reproductive system: an update.

Heavy metals like cadmium (Cd) are one of the main environmental pollutants, with no biological role in the human body. Cd has been well-documented to have disastrous effects on both plants and animals. It is known to accumulate in kidneys, lungs, liver, and testes and is thought to affect these organs' function over time, which is linked to a very long biological half-life and a very poor rate of elimination. According to recent researches, the testes are extremely vulnerable to cadmium. The disruption of the blood-testis barrier, seminiferous tubules, Sertoli cells, and Leydig cells caused b

Immunological mechanisms and precision stratification in male infertility: from testicular immune privilege and danger signal amplification to seminal immune biomarkers and mechanism-tailored intervention.

Male infertility presents significant heterogeneity, yet traditional assessments primarily rely on semen parameters and endocrine, imaging, and genetic screening. Although these methods can identify some definitive etiologies, a mechanistic explanatory gap persists in large populations with mild to moderate oligoasthenoteratozoospermia, elevated sperm DNA fragmentation, or recurrent adverse reproductive outcomes. The testis maintains a dynamic immune-privileged balance by balancing infection defense against tolerance to post-pubertal germ cell antigens. In this context, infections, varicocele-

Testicular immunity.

The testis is a unique environment where immune responses are suppressed to allow the development of sperm that possess autoimmunogenic antigens. There are several contributors responsible for testicular immune privilege, including the blood-testis barrier, testicular immune cells, immunomodulation by Sertoli cells, and high levels of steroid hormones. Despite multiple mechanisms in place to regulate the testicular immune environment, pathogens that disrupt testicular immunity can lead to long-term effects such as infertility. If testicular immunity is disturbed, autoimmune reactions can also

Determinants and mechanisms of inorganic nanoparticle translocation across mammalian biological barriers.

Biological barriers protect delicate internal tissues from exposures to and interactions with hazardous materials. Primary anatomical barriers prevent external agents from reaching systemic circulation and include the pulmonary, gastrointestinal, and dermal barriers. Secondary barriers include the blood-brain, blood-testis, and placental barriers. The tissues protected by secondary barriers are particularly sensitive to agents in systemic circulation. Neurons of the brain cannot regenerate and therefore must have limited interaction with cytotoxic agents. In the testis, the delicate process of

Exercise improves testicular morphology and oxidative stress parameters in rats with testicular damage induced by a high-fat diet.

Obesity and male infertility are problems that affect population. Exercise is a nonpharmacological way to reduce the negative health effects of obesity. The purpose of this study was to examine the effects of exercise on hormone levels, blood-testis barrier, and inflammatory and oxidative biomarkers in rats that became obese due to a high-fat diet (HFD). Male rats received a standard diet (STD group) or a HFD (HFD group) for 18 weeks. During the final 6 weeks of the experiment, swimming exercises (1 h/5 days/week) were given to half of these animals (STD + EXC and HFD + EXC groups). Finally, b

Sperm proteins and cancer-testis antigens are released by the seminiferous tubules in mice and men.

Sperm develop from puberty in the seminiferous tubules, inside the blood-testis barrier to prevent their recognition as "non-self" by the immune system, and it is widely assumed that human sperm-specific proteins cannot access the circulatory or immune systems. Sperm-specific proteins aberrantly expressed in cancer, known as cancer-testis antigens (CTAs), are often pursued as cancer biomarkers and therapeutic targets based on the assumption they are neoantigens absent from the circulation in healthy men. Here, we identify a wide range of germ cell-derived and sperm-specific proteins, including

Toxicological Effects of Cadmium on Mammalian Testis.

Cadmium is a heavy metal, and people are exposed to it through contaminated foods and smoking. In humans and other mammals, cadmium causes damage to male testis. In this review, we summarize the effects of cadmium on the development and function of the testis. Cadmium causes severe structural damage to the seminiferous tubules, Sertoli cells, and blood-testis barrier, thus leading to the loss of sperm. Cadmium hinders Leydig cell development, inhibits Leydig cell function, and induces Leydig cell tumors. Cadmium also disrupts the vascular system of the testis. Cadmium is a reactive oxygen spec

Cantharidin-induced toxic injury, oxidative stress, and autophagy attenuated by Astragalus polysaccharides in mouse testis.

Cantharidin (CTD) is a chemical constituent derived from Mylabris and has good antitumor effects, but its clinical use is restricted by its inherent toxicity. However, few researches have reported its reproductive toxicity and mechanisms. This study aims to assess CTD's toxicity on mouse testes and the protective effect of Astragalus polysaccharides (APS). Briefly, biochemical analysis, histopathology, transmission electron microscopy, immunohistochemistry, and Western blotting were used to evaluate the oxidative damage of mouse testicular tissue after exposure to CTD and treatment by APS. Expression of occludin in testis and epididymis of wild rabbits, Lepus sinensis coreanus.

Tight junctions (TJs) in inter-Sertoli junctional areas and epididymal epithelia build up the blood-testis barrier (BTB) and the blood-epididymal barrier (BEB), respectively. In this study, the expression of occludin, an integral member of the TJs, was examined in testis and different regions of epididymis of Lepus sinensis coreanus, an Korean wild rabbit species. In testis, intense occludin immunoreactivity was found in the basally located inter-Sertoli junctional area together with diffused immunoreactivity of occludin in the cytoplasm of Sertoli cells. It can be suggested that occludin is o

Androgen rapidly activates phosphorylation signaling pathways in Sertoli cells.

Androgen-activated phosphorylation signaling pathways are of critical importance during spermatogenesis. The present study comprehensively analyzed androgen-activated phosphorylation signaling pathways in testicular Sertoli cells. Primary cultured mouse Sertoli cells were treated with testosterone for varying durations. Using tandem mass tag-labeled phosphoproteomics technology combined with Mfuzz clustering and functional enrichment analysis, key phosphorylation signaling pathways were identified. Mass spectrometry revealed 13,015 quantifiable phosphorylation sites, and Mfuzz clustering of ex

Gut-testis axis: how microbiota influence male reproductive health.

The intestinal flora forms a complex ecosystem that interacts with the host, influencing health and fitness through mechanisms that connect with distant organs like the brain, liver, muscles, and testes. The gut microbiota plays a vital role in regulating androgen production and metabolism, and can cross the blood-testis barrier to influence spermatogenesis. This review highlights the significance of the gut-testis axis in male reproductive and sexual health, based on extensive studies exploring how gut microbes impact testicular function. Gaining this understanding deepens our knowledge of th

New perspectives on the renal slit diaphragm protein podocin.

Podocin is a critical component of the glomerular filtration barrier, its mutations causing recessive steroid-resistant nephrotic syndrome. A GenBank analysis of the human podocin (NPHS2) gene resulted in the possible existence of a new splice variant of podocin in the kidney, missing the in-frame of exon 5, encoding the prohibitin homology domain. Using RT-polymerase chain reaction and immunoblotting followed by sequence analysis, we are for the first time able to prove the expression of a novel podocin isoform (isoform 2), exclusively and constitutively expressed in human podocytes. Increased apoptosis in the contralateral testes of patients with testicular torsion as a factor for infertility.

PURPOSE: Unilateral testicular torsion is common and leads to bilateral testicular injury. Spermiography is abnormal in 70% of patients after testicular torsion. Histological changes in the contralateral testis at the time of torsion have been previously interpreted as the consequence of a predisposing testicular pathology or a noxious effect of the twisted testis. We hypothesized that increased apoptosis in the contralateral testis in unilateral testicular torsion is a consequence of a breakdown in the blood-testis barrier of the twisted testis, which may increase the risk of infertility. RAB3IL1: a key regulator in the autophagy-ferroptosis axis of male infertility†.

Ferroptosis, an iron-dependent form of regulated cell death characterized by phospholipid peroxidation, plays a pivotal role in various diseases. However, its involvement in male infertility, particularly in idiopathic non-obstructive azoospermia (iNOA), remains largely unexplored. Idiopathic non-obstructive azoospermia accounts for 10-15% of male infertility cases, presenting a significant clinical challenge due to its unknown origins. This study investigated the potential link between ferroptosis and iNOA, revealing excessive activation of both ferroptosis and autophagy in the testes of iNOA

The effect of temperature on the potential difference and input resistance of rat seminiferous tubules.

1. The p.d. of rat seminiferous tubules was 5.86 +/- 0.15 mV, lumen negative, at 33 degrees C and varied linearly with temperature between 24 and 37 degrees C, exhibiting an apparent Q(10) (25-35 degrees C) of 2.0 with a slope of 0.43 mV/ degrees C. Exposing testes to a temperature of 37-43 degrees C resulted in an initial hyperpolarization followed by depolarization of the tubules. These changes were more rapid in testes exposed to 41-43 degrees C than in testes exposed to 37-39 degrees C.2. The R(in) of seminiferous tubules was 198 +/- 7.8 kOmega at a testis temperature of 33 degrees C. The

Autoimmunity and Inflammaging in Male Infertility.

Male factors contribute, alone or in combination with female-related ones, to approximately 40-50% of infertility in couples. Within the spectrum of male infertility, growing evidence indicates that autoimmune mechanisms and chronic inflammation can impair spermatogenesis, sperm function, and reproductive outcomes. The disruption of the testicular immune privilege, blood-testis barrier dysfunction, local inflammatory pathways, and antisperm antibodies represent the best-characterized immune-mediated mechanisms, although their clinical relevance varies and several mechanistic observations remai

Microplastics and male reproductive toxicity: Research progress on injury pathways and regulatory mechanisms.

Microplastics and nanoplastics are ubiquitous across global environments and can bioaccumulate through food chains, arousing concerns over their links to rising rates of male infertility. Relevant literature was retrieved from PubMed, Web of Science Core Collection and Scopus, and eligible original human, animal and cell studies were screened and analyzed. Epidemiological data confirm microplastic accumulation in human reproductive tissues, which correlates with declining semen quality. Experimental evidence reveals that microplastics penetrate the blood-testis barrier to trigger testicular ox
