Because cerebrospinal fluid (CSF) antibiotic levels fail to predict either clinical success or relapse in the treatment of bacterial meningitis, we examined simultaneous antibiotic concentrations in the blood, brain, and CSF of control rabbits and of animals with experimental pneumococcal meningitis. Cefamandole pharmacokinetics were analyzed in detail and compared with those of cephalothin, ampicillin, penicillin G, and tobramycin. After 4 h of continuous intravenous infusion, cefamandole reached concentrations in both brain and CSF in excess of the minimal bactericidal concentration for the

The interaction between postsynaptic density-95 (PSD-95) and neuronal nitric-oxide synthase (nNOS) forms a signaling hub that couples N-methyl-d-aspartate receptor (NMDAR) calcium influx to bursts of neurotoxic nitric oxide. Disrupting this protein-protein interaction (PPI) offers a strategy to suppress pathological NO production while sparing normal synaptic transmission-an advantage unattainable with channel blockers or active-site nNOS inhibitors. Over the past two decades, cell-penetrant peptides such as nerinetide (Tat-NR2B9c) have validated the target from rodent stroke models to phase-I

Studies have been made on gangliosides from various parts of the brain in 5 species of cartilaginous fishes, 2 species of teleosts, 2 amphibian species, 2 avian species and 3 species of mammals. Almost in all the species investigated, the highest content of gangliosides per weight unit of the brain were found in the forebrain (determinations were carried out by sialic acid, sometimes by sphingosine). Ganglioside content of brain structure in warm-blooded animals is higher that in corresponding structures of fishes and amphibia. Comparative studies on ganglioside composition in the brain of fis

Potential prodrugs for the TRH-like tripeptide pGlu-Glu-Pro-NH(2) were synthesized either by esterifying the Glu side-chain of the parent peptide in solution with alcohols in the presence of resin-bound dicyclohexylcarbodiimide or by solid-phase peptide chemistry. Affinities of these ester prodrugs to lipid membranes as predictors of the transport across the blood-brain barrier were compared by immobilized artificial membrane chromatography, and prodrug activation was tested in the brain tissue of experimental animals. Esters of pGlu-Glu-Pro-NH(2) with long-chain primary alcohols emerged as po

Transferrin-coated nanoparticles show promise in delivering chemotherapeutic drugs across biological barriers for treating glioblastoma multiforme (GBM). Employing a synthetic approach integrating advanced chemistries and bespoke in vitro biological assays, we developed versatile transferrin-conjugated polymeric nanoparticles designed for both blood-brain barrier (BBB) penetration and GBM targeting. First, tunable azido-functional polymeric nanoparticles were synthesized through reversible addition-fragmentation chain-transfer (RAFT)-mediated polymerization-induced self-assembly (PISA). Subseq

Monitoring chemistry in the central nervous system in vivo is an important route to better understand chemical signals and metabolism underlying brain function. Sampling from the brain extracellular space allows for in depth and multiplexed analysis of this compartment. Miniaturized sampling devices prepared by using microfabrication are emerging as tools that allow high spatial resolution measurements with low invasiveness. In this work, we describe a microfabrication method for Parylene-C-based push-pull sampling probes. Parylene-C is an attractive material because its biocompatibility and f

Cerebral hypoxia-ischemia remains a major cause of acute perinatal brain injury. Research in experimental animals over the past decade has greatly expanded our knowledge of those oxidative events which occur during a hypoxic-ischemic insult to the brain, as well as those metabolic alterations which evolve during the recovery period following resuscitation. The available evidence suggests that hypoxia alone does not lead to brain damage, but rather a combination of hypoxia-ischemia or isolated cerebral ischemia is a necessary prerequisite for tissue injury to occur. Furthermore, hypoxia-ischemi

The brain edema preventing effects of aldosterone and spirolactone were compared to the effects of dexamethasone in young rabbits, dehydrated by enteral application of Na2SO4-solution. The water-, sodium- and potassium content of total brains were measured after a four hour period of forced glucose infusion. The water content of the treated groups did not differ from the control group, whereas the electrolyte content differed significantly. Dexamethasone lowered the sodium content below normal value. Spirolactone treatment resulted in an elevated potassium content. It is assumed, that aldoster

Analysis of five brains from patients with Leigh's disease demonstrates an accumulation of thiamine pyrophosphate and a deficiency of thiamine triphosphate. The enzyme which converts thiamine pyrophosphate to thiamine triphosphate was normally active in two of these brains, suggesting that the inhibitor found in Leigh's disease is probably producing the observed neurochemical changes. Reasons for the histological similarity between Leigh's and Wernicke's diseases are suggested.

In 57% of 151 BSE positive brains of Swiss cattle, degenerating neurons were found in BSE predilection sites, either single scattered (34%) or involving large parts of brain stem nuclei (23%). The lesion consisted of central chromatolysis, shrinkage, karyolysis or nuclear pyknosis. In 43% of the BSE positive brains no neuronal changes besides vacuolation were present. Neuronal degeneration other than vacuolation alone is not pathognomonic for BSE but, according to our findings, seems to be of some importance together with the typical vacuolation of neuropil and neurons. There is no correlation

OBJECTIVE: This study investigated whether idiopathic generalized epilepsies (IGEs) are associated with neuroinflammation by measuring neurometabolite levels and brain temperature using whole-brain magnetic resonance spectroscopy imaging (MRSI). We compared patients with IGEs to healthy controls (HCs), and in IGEs examined relationships with seizure control, MRSI markers, epilepsy duration, and cognitive performance.
METHODS: Seventeen patients with IGEs and 15 HCs underwent 3T-MRI scans. IGEs were also administered the Montreal Cognitive Assessment (MoCA) and Flanker test. Whole-brain 1H-MRS

Current therapy for primary amoebic meningoencephalitis (PAM), a highly lethal brain infection in humans caused by Naegleria fowleri amoeba, is restricted to repurposed drugs with limited efficacy and success. Discovery of an antiamoebic benzylamine scaffold 2 precipitated a medicinal chemistry effort to improve potency, cytotoxicity profile, and drug-like properties. Thirty-four compounds were prepared, leading to compound 28 with significant gains in potency (EC50 = 0.92 μM), solubility, and microsomal stability and a demonstrated absence of cytotoxicity in SH-SY5Y human neuroblastoma cells

Pretargeted positron emission tomography (PET) is emerging as a transformative approach for imaging biologics in the brain, overcoming the limitations of direct radiolabeling. Conventional methods with long-lived radionuclides suffer from high radiation exposure, off-target accumulation, and suboptimal imaging properties. Pretargeting separates the targeting from the imaging step, enabling the use of short-lived radionuclides such as fluorine-18, the clinical gold standard, and thereby allowing high-contrast, low-dose imaging. Realizing this strategy in the brain remains challenging due to the

Before 1960, the brain was considered by scientists to be immutable, subject only to genetic control. In the early sixties, however, investigators were seriously speculating that environmental influences might be capable of altering brain structure. By 1964, two research laboratories proved that the morphology and chemistry or physiology of the brain could be experientially altered (Bennett et al. 1964, Hubel and Wiesel 1965). Since then, the capacity of the brain to respond to environmental input, specifically "enrichment," has become an accepted fact among neuroscientists, educators and othe

The blood-brain barrier (BBB) is a key physiological component of the brain, protecting the brain from peripheral processes and pathogens. The BBB is a dynamic structure that is heavily involved in cerebral blood flow, angiogenesis, and other neural functions. However, the BBB also creates a challenging barrier for the entry of therapeutics into the brain, blocking more than 98% of drugs from contact with the brain. Neurovascular comorbidities are common in several neurological diseases including Alzheimer's and Parkinson's Disease, suggesting that BBB dysfunction or break down likely has a ca
