Calmodulin and calbindin localization in retina from six vertebrate species.

Calmodulin is abundant in the central nervous system, including the retina. However, the localization of calmodulin in the retina has not been described in detail. We therefore decided to investigate calmodulin localization in retinae from six vertebrate species, by using immunohistochemical labeling with four different rabbit polyclonal antibodies against calmodulin. The localization of calbindin-D28k, another calcium-binding protein already well described in retina, was compared. We found that calmodulin distribution is more highly conserved among species, contrasting with calbindin variabil

Ca2+ and calmodulin-sensitive inositol trisphosphate kinase from bovine parathyroid.

A Ca2+ and calmodulin-activated inositol 1,4,5 trisphosphate kinase activity was detected in both soluble and membrane fractions from bovine parathyroid glands. Ca2+ activated the soluble enzyme in the concentration range 100 nM to 1 microM, which corresponds to the Ca2+ concentration range observed in the intact cell following maximal variation in extracellular Ca2+, the principal regulator of parathyroid hormone release. The Ca2+ sensitivity of the enzyme was absolutely dependent upon calmodulin. A similar activity was detected in the membranes but could be progressively removed by repeated

Calmodulin antagonists inhibit the phagocytic activity of cultured Kupffer cells.

The mechanism of phagocytosis by Kupffer cells is still unknown. In this study we found that trifluoperazine, chlorpromazine, and W-7, drugs which bind to Ca2+-calmodulin and inhibit its interaction with other proteins, inhibit phagocytosis by cultured Kupffer cells using polystyrene beads, time-lapse VTR systems, and fluorescent staining techniques. Inhibitory effects of these drugs on phagocytosis suggests that the Ca2+-calmodulin system may be involved in this complex cellular function and the integrity of the cytoskeletal system of Kupffer cells is essential to this phenomenon.

Calmodulin inhibitors protect against cadmium-induced testicular damage in mice.

Recent reports showing that cadmium can interact with calmodulin and activate calmodulin-sensitive enzymes have lead us to examine the effects of calmodulin inhibitors on cadmium-induced testicular toxicity in mice. Male CF-1 mice were pretreated with the various calmodulin inhibitors or inactive analogs and then, one hour later, given CdCl2 X 2 1/2 H2O (32 mumoles/kg). After 24 hours, the mice were killed and the testes were removed and weighed. The extent of hemorrhaging was quantified by determining the absorbance of hemoglobin at 414 nm in the soluble fraction of testicular homogenates. Ex

Calmodulin antagonists stimulate LDL receptor synthesis in human skin fibroblasts.

The LDL receptor synthesis of human skin fibroblasts in the presence of the specific calmodulin antagonists trifluoperazine, condensation product of N-methyl-p-methoxyphenethylamine with formaldehyde (compound 48/80) and N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide) (W-7) was studied. Labelling of cells with [35S]methionine followed by immunoprecipitation of radioactive LDL receptor protein with monospecific antibodies revealed that calmodulin antagonists caused a 3-fold increase in the radioactivity of the LDL receptor protein as compared with values found in control cells. Calmodulin activation and calcium regulation of parotid gland adenylate cyclase.

The effect of Ca2+ on the adenylate cyclase activity associated with membranes prepared from mouse parotid gland has been examined. Ca2+ stimulated then inhibited adenylate cyclase activity, with values for half-maximal stimulation and inhibition of 0.6 and 10 microM, respectively. Maximal activation (1.4-fold) was observed at 2 microM free Ca2+. These membranes contained 1.2 microgram calmodulin/mg protein. Exogenous calmodulin (0.2-1.2 microgram) activated, in a concentration-dependent manner, adenylate cyclase activity, with maximal activation being 2.5-fold at 12 micrograms calmodulin. Pre

Characterization of a calmodulin-dependent protein phosphatase from human platelets.

A calmodulin-dependent protein phosphatase has been identified in human platelets by its cross-reactivity with an antibody developed against a bovine brain calmodulin-dependent protein phosphatase and by its calmodulin-stimulated dephosphorylation of 32P-labeled substrates. The platelet enzyme was partially purified to separate it from calmodulin and calmodulin-independent phosphatases. The partially purified enzyme was stimulated by calmodulin, requiring 15 nM calmodulin for half-maximal activation. Calmodulin increased the Vmax of the phosphatase, with no significant effect on its Km. The en

Circular dichroism and 1H NMR studies on the structures of peptides derived from the calmodulin-binding domains of inducible and endothelial nitric-oxide synthase in solution and in complex with calmodulin. Nascent alpha-helical structures are stabilized by calmodulin both in the presence and absence of Ca2+.

There exist two types of nitric-oxide synthase (NOS); constitutive isozymes that are activated by binding calmodulin in response to elevated Ca2+ and an inducible isozyme that binds calmodulin regardless of Ca2+. To study the structural basis of the difference in Ca2+ sensitivity, we have designed synthetic peptides of minimal lengths derived from the calmodulin-binding domain of endothelial NOS (eNOS) and that of macrophage NOS (iNOS). A peptide, KRREIPLKVLVKAVLFACMLMRK, derived from human iNOS sequence, retained the ability to bind to calmodulin both in the presence and absence of Ca2+, whil

Proteomic Snapshots of Structural Cross-Linking Rearrangements in Ca2+/Calmodulin-Dependent Kinase-1-Delta Associated with Its Regulation by ATP, Ca2+/Calmodulin, and Reduction Potential.

Ca2+/calmodulin-dependent kinase 1 delta (CaMK1δ) plays a central role in regulatory pathways associated with ATP, reduction potential, and Ca2+/calmodulin (CaM). Mass spectrometry (MS)-based structural proteomics incorporating FragPipe and pLink cross-link analysis was used to reveal conformation selection induced by dialysis with ATP, reducing agents, and CaM. The structural changes were mediated via cysteine and phosphate cross-linking and loop-linking of the activation loop within the C-terminal. Phosphate loop-linking was validated by β-elimination and Michael addition (BEMAD) reactions,

Increased intracellular calcium activates serum and glucocorticoid-inducible kinase 1 (SGK1) through a calmodulin-calcium calmodulin dependent kinase kinase pathway in Chinese hamster ovary cells.

SGK1 is one of the protein-serine/threonine kinases that is activated by insulin in a PI3K-dependent manner. Although SGK1 mediates a variety of biological activities, the mechanisms regulating its activity remain unclear. In this study, we examined the potential roles of calcium signaling in the activation of SGK1. Treatment of CHO-IR cells with a cell-permeable calcium chelator, BAPTA-AM, abolished the insulin-induced activation of SGK1. Increasing intracellular calcium concentration by treating cells with thapsigargin or ionomycin induced a 6-8 fold increase in SGK1 activation. This was not

Inhibition of agonist-mediated calcium entry by calmodulin antagonists and by the Ca2+/calmodulin kinase II inhibitor KN-62. Studies with thyroid FRTL-5 cells.

Calmodulin and calmodulin-dependent mechanisms are probably important in regulating thyroid cell function. However, calmodulin antagonists may directly modify calcium fluxes in cells. In the present investigation the effects of several calmodulin inhibitors and of KN-62, a specific calcium/calmodulin kinase II inhibitor, on the ATP- and thapsigargin-evoked changes in intracellular free calcium ([Ca2+]i) were investigated in Fura-2 loaded thyroid FRTL-5 cells. All of the inhibitors tested attenuated agonist-evoked calcium entry. The inhibitor calmidazolium per se potently released sequestered c

Let-7a Is an Antihypertrophic Regulator in the Heart via Targeting Calmodulin.

Background: MicroRNAs (miRNAs) have been emerged as important regulator in a multiple of cardiovascular disease, including arrhythmia, cardiac hypertrophy and fibrosis, and myocardial infarction. The aim of this study was to investigate whether miRNA let-7a has antihypertrophic effects in angiotensin II (AngII)-induced cardiac hypertrophy. Methods: Neonatal rat ventricular myocytes (NRVMs) were exposed to AngII for 36 h as a cellular model of hypertrophy; subcutaneous injection of AngII for 2 weeks was used to establish a mouse model of cardiac hypertrophy in vivo study. Genotyping species of the Sporothrix schenckii complex by PCR-RFLP of calmodulin.

Sporotrichosis is one of the most common subcutaneous mycosis in Latin America and is caused by 4 pathogenic thermodimorphic fungi in the genus Sporothrix. From both therapeutic and epidemiological perspectives, it is essential to identify the causative agents down to the species level. Traditional parameters may overlap among closely related species, and we propose polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) as an alternative approach. In the present study, the calmodulin gene was amplified and digested with HhaI to yield 5 different electrophoretic patterns

Identification and inhibitory properties of a novel Ca(2+)/calmodulin antagonist.

We developed a high-throughput yeast-based assay to screen for chemical inhibitors of Ca(2+)/calmodulin-dependent kinase pathways. After screening two small libraries, we identified the novel antagonist 125-C9, a substituted ethyleneamine. In vitro kinase assays confirmed that 125-C9 inhibited several calmodulin-dependent kinases (CaMKs) competitively with Ca(2+)/calmodulin (Ca(2+)/CaM). This suggested that 125-C9 acted as an antagonist for Ca(2+)/CaM rather than for CaMKs. We confirmed this hypothesis by showing that 125-C9 binds directly to Ca(2+)/CaM using isothermal titration calorimetry.

Calmodulin is a functional regulator of Cav1.4 L-type Ca2+ channels.

Cav1.4 channels are unique among the high voltage-activated Ca2+ channel family because they completely lack Ca2+-dependent inactivation and display very slow voltage-dependent inactivation. Both properties are of crucial importance in ribbon synapses of retinal photoreceptors and bipolar cells, where sustained Ca2+ influx through Cav1.4 channels is required to couple slow graded changes of the membrane potential with tonic glutamate release. Loss of Cav1.4 function causes severe impairment of retinal circuitry function and has been linked to night blindness in humans and mice. Recently, an in

The effect of calmodulin antagonists on experimental scoliosis: a pinealectomized chicken model.

STUDY DESIGN: Randomized controlled. OBJECTIVE: To evaluate the effects of Tamoxifen (TMX) and trifluoperozine (TFP) on pinealectomized chicken scoliosis. SUMMARY OF BACKGROUND DATA: Pinealectomized chicken develops scoliosis probably due to the lack of melatonin. In addition to other functions, melatonin also acts as a calmodulin antagonist. We postulate that loss of this antagonistic effect may be the cause of scoliosis in this model. TMX and TFP are known calmodulin antagonists, which may alter the incidence and severity of scoliosis. METHODS: Seventy-two newly hatched chicken that under

The transcriptional activity of estrogen receptor-alpha is dependent on Ca2+/calmodulin.

Estrogen binds to estrogen receptors in cells, thereby activating the receptors and eliciting biological effects. One of the best characterized effects of estrogen receptor-alpha (ERalpha) is transcriptional activation that regulates selected target genes in the nucleus. Work from several laboratories has documented a Ca2+-dependent interaction between ERalpha and calmodulin. In addition, we previously showed that antagonism of calmodulin function in cells prevented estradiol from inducing ERalpha transcriptional activity, suggesting that association of ERalpha with calmodulin participates in

Structural basis for the activation of anthrax adenylyl cyclase exotoxin by calmodulin.

Oedema factor, a calmodulin-activated adenylyl cyclase, is important in the pathogenesis of anthrax. Here we report the X-ray structures of oedema factor with and without bound calmodulin. Oedema factor shares no significant structural homology with mammalian adenylyl cyclases or other proteins. In the active site, 3'-deoxy-ATP and a single metal ion are well positioned for catalysis with histidine 351 as the catalytic base. This mechanism differs from the mechanism of two-metal-ion catalysis proposed for mammalian adenylyl cyclases. Four discrete regions of oedema factor form a surface that r
