Complex I drives glutamine-dependent TCA cycle to support viability of MYChigh breast cancer cells.

In many cancers, stably elevated MYC levels drive sustained activation of anabolic programs and the cell cycle, creating opportunities for the synthetic-lethal targeting of MYChigh tumors. Enhanced mitochondrial respiration is a hallmark of MYC overexpressing cancer cells. Mitochondrial respiration sustains the TCA cycle by regenerating NAD+ through complex I-mediated oxidation of NADH, supporting the anabolic demand of MYC-driven cells. Metabolic carbon tracing reveals that MYC shifts the TCA cycle carbon source from glucose to glutamine. Inhibition of the glutamine-fueled TCA cycle using NAD

Lengthening vegetation carbon turnover time across China's forests.

Vegetation carbon turnover time (τveg) dominates the uncertainty in terrestrial carbon cycle dynamics. Reports have shown that the τveg of mature or old-growth forests in North America and Europe has decreased because of faster carbon loss under global climate change. However, the temporal trend of τveg in widespread younger forests, which exhibit different growth patterns, remains inconclusive. Here, we consistently revealed a significant overall increase in τveg (0.025 ± 0.002 years per year) across China's forests that are characterized by a relatively young forest age structure, using mult

Seasonal variations of carbon fluxes and the drivers in typical Chinese forest ecosystems.

We integrated monthly eddy-covariance carbon flux data from 27 forest flux towers across China, compiled from ChinaFLUX during 2003 to 2023, literature, and our own observations. The study sites covered four forest biomes, namely boreal, temperate, subtropical, and tropical forests. Based on long-term mean monthly series, we quantified seasonal metrics of gross primary productivity (GPP), ecosystem respiration (Re), and net ecosystem productivity (NEP), including seasonal amplitude, seasonality index, and seasonal standard deviation, as well as flux magnitude metrics, including maximum monthly

Strategies to intensify CO2 capture by microalgae for the circular bioeconomy.

In this review, we explore pathways of coupling between microscale metabolic engineering with macroscale bioprocess design to transform microalgae into intelligent carbon management platforms, focusing on strategies that enhance CO2 fixation capacity, including synthetic enhancement of carbon-concentrating mechanisms (CCMs) and metabolic reprogramming. The integration of microalgae with microbial consortia further stabilizes carbon flow and supports system resilience under environmental fluctuations. Linking Freshwater Wetland Productivity and Methane Emissions: A Global Perspective.

Understanding the link between wetland gross primary productivity (GPP) and methane (CH4) emissions is crucial for global carbon cycle modeling, yet this coupling remains poorly constrained at a global scale due to data limitations. To address this critical gap, we compiled and analyzed the most comprehensive daily scale eddy covariance data set of GPP and CH4 fluxes. In this analysis, we define this coupling as the slope of the linear fit between carbon fixed by ecosystems and carbon emitted as CH4. Results indicate that the median lag time between CH4 emissions and GPP is 24.8 days. In terms

Direct and Legacy Effects of Varying Cool-Season Precipitation Totals on Ecosystem Carbon Flux in a Semi-Arid Mixed Grassland.

In the semi-arid grasslands of the southwest United States, annual precipitation is divided between warm-season (July-September) convective precipitation and cool-season (December-March) frontal storms. While evidence suggests shifts in precipitation seasonal distribution, there is a poor understanding of the ecosystem carbon flux responses to cool-season precipitation and the potential legacy effects on subsequent warm-season carbon fluxes. Results from a two-year experiment with three cool-season precipitation treatments (dry, received 5th percentile cool-season total precipitation; normal,

Predictive insights into CO2 capture carbonation conversion rates under different Ca-looping conditions in fluidized bed reactors.

Carbonation cycles are vital for the efficiency, sustainability, and cost-effectiveness of the calcium looping process, making it a promising technology for carbon capture and storage. The research presented in this paper is based on experimental carbonation tests conducted on dual-interconnected fluidized bed reactors under realistic operational conditions in three different states including absence (NS, No Sulfur), 75 ppm (SP, Sulfur Poor), and 1500 ppm (SR, Sulfur Rich) SO2 concentration using high purity limestone as sorbent. Experimental results demonstrated that the carbonation conversio

Temporal Pattern and Metabolic Mechanisms of Ecosystem Carbon Fluxes Under Decadal-Long Nitrogen Enrichment in an Alpine Grassland.

Due to widespread nitrogen limitation, reactive nitrogen input is expected to stimulate vegetation carbon fixation, thus potentially offsetting the decomposition-induced soil carbon losses. However, this projection is largely based on short-term measurements. The crucial question is whether such responses persist over time. Combining a decade-long nitrogen-addition experiment in an alpine grassland with biochemical and plant metabolomics analyses, we showed that a decade of nitrogen addition elicited only a transient increase in net ecosystem productivity (NEP; balance between ecosystem carbon

Quantitative stable isotope probing (qSIP)-informed metagenomics identifies viruses infecting chemoautotrophs.

Aquatic environments absorb ~2.5 gigatonnes of atmospheric carbon each year1, more than the carbon stored in the atmosphere, soils, and all biomass combined. Primary producers transform this dissolved inorganic carbon into biomass that can subsequently flow into other trophic levels, or be released back into the environment through viral lysis. While there is substantial knowledge about the diversity and activity of viruses infecting photoautotrophic primary producers and the ecosystem impact, little is known about viruses infecting chemoautotrophs, representing a gap in our understanding of k

Resilience to Hurricanes Is High in Mangrove Blue Carbon Forests.

Mangrove forests are typically considered resilient to natural disturbances, likely caused by the evolutionary adaptation of species-specific traits. These ecosystems play a vital role in the global carbon cycle and are responsible for an outsized contribution to carbon burial and enhanced sedimentation rates. Using eddy covariance data from two coastal mangrove forests in the Florida Coastal Everglades, we evaluated the impact hurricanes have on mangrove forest structure and function by measuring recovery to pre-disturbance conditions following Hurricane Wilma in 2005 and Hurricane Irma in 20

CO2 drives ecological shifts through asymmetric selection in microalgae-bacteria granular for enhanced wastewater treatment and carbon fixation.

Microalgae-bacteria granular sludge (ABGS) systems show promising potential for simultaneous pollutant removal and CO2 fixation in wastewater treatment, aligning with carbon neutrality goals. However, the ecological mechanisms underlying CO2-introduced ABGS system performance adaptations remain poorly understood, hindering biosystem optimization. Here, we systematically investigated multi-level ecological adaptation mechanisms in ABGS systems under external CO2 in low-carbon wastewater. For the first time, our study reveals that CO2 acts as a selective force driving community evolution through

Strategies to Improve the Efficiency of Enzymatic Carbon Dioxide Conversion In Vitro.

With the rapid industrial development, massive fossil fuel use has caused excessive carbon dioxide (CO2) emissions, triggering global warming and environmental issues. Thus, CO2 recovery and reuse have become a research focus, among which artificially designed in vitro biocatalytic pathways for converting CO2 into high-value chemicals show promise, with advantages like shorter routes, higher efficiency and lower energy consumption compared to natural pathways. However, challenges remain due to natural enzymes' issues in specificity, affinity, efficiency, stability and oxygen sensitivity. To ta

The complexity of heatwaves impact on terrestrial ecosystem carbon fluxes: Factors, mechanisms and a multi-stage analytical approach.

Extreme heatwaves have become more frequent and severe in recent decades, and are expected to significantly influence carbon fluxes at regional scales across global terrestrial ecosystems. Nevertheless, accurate prediction of future heatwave impacts remains challenging due to a lack of a consistent comprehension of intrinsic and extrinsic mechanisms. We approached this knowledge gap by analyzing the complexity factors in heatwave studies, including the methodology for determining heatwave events, divergent responses of individual ecosystem components at multiple ecological and temporal scales,

Gross primary production responses to warming, elevated CO2 , and irrigation: quantifying the drivers of ecosystem physiology in a semiarid grassland.

Determining whether the terrestrial biosphere will be a source or sink of carbon (C) under a future climate of elevated CO2 (eCO2 ) and warming requires accurate quantification of gross primary production (GPP), the largest flux of C in the global C cycle. We evaluated 6 years (2007-2012) of flux-derived GPP data from the Prairie Heating and CO2 Enrichment (PHACE) experiment, situated in a grassland in Wyoming, USA. The GPP data were used to calibrate a light response model whose basic formulation has been successfully used in a variety of ecosystems. The model was extended by modeling maximum

Time-resolved targeted metabolomics shows an abrupt switch from Calvin-Benson-Bassham cycle to tricarboxylic acid cycle when the light is turned off.

In leaves, major CO₂ fluxes alternate between fixation by the Calvin-Benson-Bassham (CBB) cycle during light and release by the tricarboxylic acid (TCA) cycle in darkness. The speed at which leaf metabolism transitions between these pathways likely influences plant tolerance to fluctuating light conditions. To investigate these rapid metabolic shifts, we exposed leaves to ¹³CO₂ for 20 min to establish a quasi-steady state before abruptly turning off the light while maintaining ¹³CO₂ feeding. Within 10 s of dark transition, 3-phosphoglycerate levels rose significantly while most other CBB cycle

Niche-specific regulation of central metabolic pathways in a fungal pathogen.

To establish an infection, the pathogen Candida albicans must assimilate carbon and grow in its mammalian host. This fungus assimilates six-carbon compounds via the glycolytic pathway, and two-carbon compounds via the glyoxylate cycle and gluconeogenesis. We address a paradox regarding the roles of these central metabolic pathways in C. albicans pathogenesis: the glyoxylate cycle is apparently required for virulence although glyoxylate cycle genes are repressed by glucose at concentrations present in the bloodstream. Using GFP fusions, we confirm that glyoxylate cycle and gluconeogenic genes i

Ecological, Functional, and Phylogenetic Determinants of Cyanobacterial Biomineralisation.

Cyanobacteria play a key role in the biomineralisation of carbon dioxide into solid carbonates, a critical process in the global carbon biogeochemical cycle that links atmospheric CO2 to lithospheric carbonate reservoirs. While photosynthetic carbon fixation by these microorganisms has been extensively studied and is relatively well understood, the biomineralisation pathway remains much less explored, likely leading to an underestimation of its global relevance. This review summarises current findings and highlights the ecological and cellular factors that contribute to cyanobacterial biominer

Plant diversity's positive effect on soil respiration diminishes with increasing productivity in global forests.

Plant diversity is essential for regulating ecosystem functions, yet its global-scale relationship with soil respiration, a critical component of the carbon cycle, remains unexplored. While plant productivity modulates ecosystem processes, understanding how plant species richness regulates soil respiration across net primary production gradients is key to predicting carbon-climate feedbacks. Here, we integrate two global plant species richness datasets (tree species richness and vascular plant species richness, including woody and non-woody plants) with a global soil respiration dataset estima

Decoding carbon, water, and energy exchange driven by agricultural and horticultural intensification in semi-arid ecosystem of India.

Terrestrial ecosystems play a vital role in sequestering carbon dioxide (CO2); however, growing both agri-horticultural intensifications can influence CO2 sequestration potential. This study was conducted in an agri-horticultural-intensive ecosystem of Central India to investigate the net ecosystem exchange (NEE) of CO2, water (H2O), and energy fluxes and assess the influence of meteorological factors using the eddy covariance datasets acquired from January to June 2024. The highest monthly mean diurnal CO2 flux was recorded in January (-7.98 µmol m-2 s-1), and the lowest was in June (-3.03 µm
