Virtual screening of potential inhibitors against catechol-o-methyltransferase (COMT) for temporomandibular joint disorders.

BACKGROUND: Temporomandibular joint pain dysfunction syndrome (TMJPDS) is a musculoligamentous condition causing dull, aching pain in the masticatory muscles, influenced by jaw movement. Computer-aided drug design (CADD) uses computational methods to support modern drug discovery. Molecular docking, a core CADD technique, predicts drug-target interactions, aiding in stable, specific, and effective drug design. AIM: The aim of this study was to use the docking technique for COMT inhibitors. MATERIALS AND METHODS: Four COMT inhibitors were selected from the literature, and their compound struc

Versatile phosphonium-catechol dispersant for inorganic nanoparticle stability across diverse media and manufacturing processes.

Inorganic nanoparticles (NPs) have significant potential for various applications; however, achieving stable dispersion in organic media and polymers remains a key challenge for their industrial use. We developed a novel dispersant, tributyl(3-((3,4-dihydroxyphenylamino)-3-oxopropyl)phosphonium chloride) (TPC), which contains a catechol group for strong NPs binding and a phosphonium chloride moiety for effective dispersion. The versatility of TPC was demonstrated by stabilizing TiO₂, Fe₂O₃, and ZnO NPs in methanol using bead milling, as confirmed by TGA, DLS, zeta potential, and TEM analyses.

Study on Phosphorus Compound/Catechol-Catalyzed Dehydrative Amidation and Its Database Development for Machine Learning.

Herein, we report that the catalytic activity of phosphorus compounds with a P─H bond in dehydrative amidation reactions is substantially enhanced when using catechol derivatives as additives. A systematic investigation into the catalytic activity of four phosphorus compounds and 17 catechol derivatives allowed identifying several effective combinations, for example, dimethyl phosphite and 2,3-dihydroxynaphthalene, for the catalytic amidation of carboxylic acids with amines. The developed catalytic system enables the synthesis of pharmaceuticals and facilitates the amidation of anilines, which

Periplasmic Binding Protein YiuA Enables Yersinia pestis to Scavenge Fe(III)-Catechol Siderophores.

Yersinia pestis, the pathogen causing plague, requires iron to grow. Y. pestis employs several uptake pathways for iron, including the siderophore yersiniabactin, as well as hemin and inorganic iron. The Y. pestis iron assimilation repertoire further harbors the uncharacterized YiuRABC pathway, presumed to transport an unidentified Fe(III)-siderophore(s). Through intrinsic fluorescence quenching of the periplasmic binding protein YiuA, we discovered that YiuA displays high affinity toward Fe(III) complexes of the hydrolysis products of enterobactin, Fe(III)-[di(DHB-LSer)] and Fe(III)-[DHB-LSer

Catechol-induced covalent modifications modulate the aggregation tendency of α-synuclein: An in-solution and in-silico study.

Parkinson's disease (PD) stands as a challenging neurodegenerative condition characterized by the emergence of Lewy Bodies (LBs), intracellular inclusions within dopaminergic neurons. These LBs harbor various proteins, prominently including α-Synuclein (Syn) aggregates, implicated in disease pathology. A promising avenue in PD treatment involves targeting Syn aggregation. Recent findings from our research have shown that 3,4-dihydroxyphenylacetic acid (DOPAC) and 3,4-dihydroxyphenylethanol (DOPET) possess the ability to impede the formation of Syn fibrils by disrupting the aggregation process.

Mucoadhesive, antioxidant, and lubricant catechol-functionalized poly(phosphobetaine) as biomaterial nanotherapeutics for treating ocular dryness.

Dry eye disease (DED) is associated with ocular hyperosmolarity and inflammation. The marketed topical eye drops for DED treatment often lack bioavailability and precorneal residence time. In this study, we investigated catechol-functionalized polyzwitterion p(MPC-co-DMA), composed of 2-methacryloyloxyethyl phosphorylcholine (MPC) and dopamine methacrylamide (DMA) monomers, as potential topical nanotherapeutics for DED. The copolymers were synthesized via random free-radical copolymerization, producing different proportions of catecholic functionalization. All as-prepared polymer compositions

Catechol-Modified and MnO2-Nanozyme-Reinforced Hydrogel with Improved Antioxidant and Antibacterial Capacity for Periodontitis Treatment.

Periodontitis is an inflammatory disease characterized by tooth loss and alveolar bone resorption. Bacteria are the original cause of periodontitis, and excess reactive oxygen species (ROS) encourage and intensify inflammation. In this study, a mussel-inspired and MnO2 NPs-reinforced adhesive hydrogel capable of alleviating periodontitis with improved antibacterial and antioxidant abilities was developed. The hydrogel was created by combining polyvinyl alcohol (PVA), 3,4-dihydroxy-d-phenylalanine (DOPA), and MnO2 nanoparticles (NPs) (named PDMO hydrogel). The hydrogel was demonstrated to be ab

Catechol-O-methyltransferase gene (COMT) is associated with neurocognitive functioning in patients with sickle cell disease.

PURPOSE: Neurocognitive impairment is a common and debilitating complication of sickle cell disease (SCD) resulting from a combination of biological and environmental factors. The catechol-O-methyltransferase (COMT) gene modulates levels of dopamine availability in the prefrontal cortex. COMT has repeatedly been implicated in the perception of pain stimuli and frequency of pain crises in patients with SCD and is known to be associated with neurocognitive functioning in the general population. The current study aimed to examine the associations of genetic variants in COMT and neurocognitive fun

Design and synthesis of functionalized 4-aryl-Catechol derivatives as new antiinflammtory agents with in vivo efficacy.

Oxidative stress and inflammation are two conditions that coexist in many multifactorial diseases and the discovery of antioxidants is an attractive approach that can simultaneously tackle two or more therapeutic targets of the arachidonic acid cascade. We report that the simple structural variations on the 4-aryl-benzene-1,2-diol side-arm of the scaffold significantly influence the selectivity against 5-LOX vs 12- and 15-LOX. Derivatives 4 a-l were evaluated for their antioxidant activity, using the DPPH, and ferric ion reducing antioxidant power (FRAP) methods. Docking simulations proposed c

A Catechol-Meter Based on Conventional Personal Glucose Meter for Portable Detection of Tyrosinase and Sodium Benzoate.

In this study, the personal glucose meter (PGM) was first used as a fast and user-friendly meter for analyzing catechol (CA) based on the reduction of the mediator K3[Fe(CN)6] to K4[Fe(CN)6] in the glucose test strip. Then, an easy, low-cost, and convenient PGM-based method for detecting tyrosinase (TYR) activity and sodium benzoate (SBA) was developed on the basis of the TYR-catalyzed reaction. In this method, CA is oxidized to form o-benzoquinone by TYR, thereby reducing the residual amount of CA and the PGM readout. On the other hand, SBA can inhibit the oxidation of CA catalyzed by TYR and

Genetic and epigenetic regulation of Catechol-O-methyltransferase in relation to inflammation in chronic fatigue syndrome and Fibromyalgia.

BACKGROUND: Catechol-O-methyltransferase (COMT) has been shown to influence clinical pain, descending modulation, and exercise-induced symptom worsening. COMT regulates nociceptive processing and inflammation, key pathophysiological features of Chronic Fatigue Syndrome and Fibromyalgia (CFS/FM). We aimed to determine the interactions between genetic and epigenetic mechanisms regulating COMT and its influence on inflammatory markers and symptoms in patients with CFS/FM. METHODS: A case-control study with repeated-measures design was used to reduce the chance of false positive and increase the

Colorimetric selective quantification of anthocyanins with catechol/pyrogallol moiety in edible plants upon zinc complexation.

Here is examined the colour development from common anthocyanins (i.e., cyanidin, delphinidin, malvidin, and pelargonidin glycosides) and from anthocyanins-rich extracts (i.e., bilberries, strawberries, and raspberries), using zinc-anthocyanin complexes as molecular probe. We have observed the absorbance increase in the blue region in presence of large excess of zinc ion at acidic pH for cyanidin and delphinidin derivatives, likely due to quinoidal base stabilization from catechol and pyrogallol moiety. The assay condition were studied and applied to natural extracts containing these compounds

Polyphosphazene and Non-Catechol-Based Antibacterial Injectable Hydrogel for Adhesion of Wet Tissues as Wound Dressing.

Wound dressings with excellent adhesiveness, antibacterial, self-healing, hemostasis properties, and therapeutic effects have great significance for the treatment of acute trauma. So far, numerous mussel-inspired catechol-based wet adhesives have been reported, opening a pathway for the treatment of acute trauma. However, catechol-based hydrogels are easily oxidized, which limits their applications. Here, the design of a polyphosphazene and non-catechol based antibacterial injectable hydrogel is reported as a multifunctional first aid bandage. Inspired by barnacle cement proteins, a series of

Catechol-O-methyltransferase gene Val158Met polymorphism and obsessive compulsive disorder susceptibility: a meta-analysis.

Obsessive-compulsive disorder (OCD) is a common psychiatric disorder that affects approximately 1-3% of the general population. It is characterized by disabling obsessions (intrusive unwanted thoughts) and/or compulsions (ritualized repetitive behaviors). Catechol-O-methyltransferase (COMT) enzyme has an important role in inactivation of dopamine and higher dopamine levels may be implicated in OCD, hence COMT gene is a suitable candidate for OCD. Several case-control studies have evaluated the role of COMT Val 158Met (rs4680;472G- > A) polymorphism as a risk factor for OCD but the results rema

Catechol and 1,2,4,5-tetrahydroxybenzene functionalized cyclodiphosphazane ligands: synthesis, structural studies, and transition metal complexes.

Syntheses of two novel cyclodiphosphazane derivatives appended on catechol and 1,2,4,5-tetrahydroxy benzene, [{(μ-N(tBu)P)2(C6H4O2)}] (1) and [{((μ-N(tBu)P)2)2(μ-C6H2O4)}] (2), are described. Reactions of 1 with copper(i) halides led to the isolation of one-dimensional (1-D) and two-dimensional (2-D) coordination polymers, depending on the reaction conditions, metal-to-ligand ratio and CuX (X = Cl, Br or I) employed. The 1 : 1 reaction between 1 and CuCl yielded a 1-D coordination polymer [{(μ-N(tBu)P)2(C6H4O2)}{Cu(μ2-Cl)(NCCH3)}2]n (3) containing [Cu(μ2-Cl)]2 rhombus units. Similar reactions
