Light orchestrates cotyledon greening and growth by coordinating chloroplast development with cell division and expansion, yet the molecular integrator of these processes remains unknown. Here we identify RDE (REGULATOR OF DG1 EXPRESSION), a conserved DNA architectural factor that synchronizes thylakoid protein complex biogenesis during chloroplast development with the G1-S transition of cell cycle progression in Arabidopsis thaliana. RDE mediates promoter DNA bending to sequester DPa in stable nucleoprotein complexes, precluding DPa-E2Fa heterodimer formation. This repression mechanism contro

Dysregulated lipid metabolism plays an important role in prostate cancer, although the understanding of the essential regulatory processes in tumorigenesis is incomplete. We employ a CRISPR-Cas9 screen using a custom human lipid metabolism knockout library to identify essential genes for prostate cancer survival. Screening in three prostate cancer cell lines reveals 63 shared dependencies, with enrichment in terpenoid backbone synthesis and N-glycan biosynthesis. Independent knockout of key genes of the mevalonate pathway reduces cell proliferation. Further investigation focuses on NUS1, a sub

Due to the generally low growth rates of acetogens, conventional batch and continuously operated syngas fermentation processes in suspension cultures, e.g., stirred tank bioreactors, typically result in low cell concentrations and, thus, limited volumetric productivities (space-time yields). To increase cell concentrations in syngas fermentations, a continuously operated submerged microfiltration membrane bioreactor (MBR) was applied on a L-scale with C. autoethanogenum LAbrini for the production of alcohols. Cell retention was achieved by inserting a sintered metal microfiltration rod into a

Based on striking parallels between the cell kinetics in the epidermal lesion of psoriasis and the proliferation of hair matrix keratinocytes during the anagen phase of the hair growth cycle, the hypothesis is proposed that both phenomena may share the same "switch-on" mechanism. Particular emphasis is placed on a comparison between the Koebner phenomenon in psoriasis and wounding-induced anagen hair growth. In discussing alternative theoretical models for the proposed common "switch-on" mechanism, some useful experimental tools are suggested. Research into the mechanisms which control epithel

This study was designed to examine the role of opioids on cell migration, chemotaxis, invasion, and adhesion, with an emphasis on whether the opioid growth factor (OGF, [Met(5)]-enkephalin) or the opioid antagonist naltrexone (NTX) impacts any or all of these processes. Drug concentrations of OGF and NTX known to depress or stimulate, respectively, cell proliferation and growth were analyzed. Three different human cancers (pancreatic, colon, and squamous cell carcinoma of the head and neck), represented by seven different cancer cell lines (PANC-1, MIA PaCa-2, BxPC-3, CAL-27, SCC-1, HCT-116, a

Environmental stimuli are perceived and transduced inside the cell through the activation of signaling pathways. One common type of cell signaling transduction network is initiated by G-proteins. G-proteins are activated by G-protein-coupled receptors (GPCRs) and transmit signals from hormones, neurotransmitters, and other signaling factors, thus controlling a number of biological processes that include synaptic transmission, visual photoreception, hormone and growth factors release, regulation of cell contraction and migration, as well as cell growth and differentiation. G-proteins mainly act

Autophagy is a cellular process that is essential for maintaining homeostasis in mammalian cells by degrading and recycling intracellular components, particularly under stress conditions. To achieve high therapeutic protein production in recombinant Chinese hamster ovary (rCHO) cells, it is necessary to develop a fed-batch culture process using optimized culture media that maintains a rapid growth rate and extends the culture longevity of cell lines with high specific protein productivity. During the development of these culture processes, stressful conditions, such as rapid nutrient deprivati

Neurite outgrowth and motility are essential for neuronal development and migration, and they play crucial roles in nervous system formation and axonal regeneration. Cytosolic scaffold proteins are key regulators of these processes, and their dysregulation can lead to neurodevelopmental disorders. In flies, Bruchpilot (Brp), an ortholog of the mammalian active zone scaffold protein ELKS/CAST, is a key factor in synapse formation and maintenance and in neurotransmitter release at chemical synapses. Although mammalian ELKS has been implicated in the growth and motility of both neuronal and non-n

UNLABELLED: Bacteria in nature and disease frequently spend time in non-growing states, yet the genes that allow cells to survive without growing remain poorly understood. Using Pseudomonas aeruginosa strain PA14 as a model system to study non-growth powered by anaerobic phenazine cycling, we employed randomly barcoded transposon-insertion sequencing (RB-TnSeq) to identify the genes required for this maintenance state using a platform that sustains anaerobic survival via continuous phenazine reoxidation. We found 167 genes whose disruption altered survival, including genes involved in transcri

Both epithelialization and angiogenesis are indispensable processes in gastric ulcer healing. Coordination between these processes has not been well studied. In the present study, we have established a new primary culture system of human gastric epithelial cells and investigated the effect of epithelialization stimulants on a specific angiogenic factor, vascular endothelial growth characterized as epithelial cells. Both epithelialization stimulants, hepatocyte growth factor (HGF) and epidermal growth factor (EGF), significantly stimulated vascular EGF expression in gastric epithelial cells. HG

Pancreatic and duodenum homeobox 1 (PDX1) is considered as a pivotal transcription factor that acts as a "master regulator" in pancreatogenesis and maintenance of β-cells. Earlier study has reported that PDX1 also functions as a tumor suppressor in human gastric cancer cells by inhibiting cell growth. Here, we report the bioactivity of the purified human PDX1 fusion protein using various assays like cell migration, proliferation, cell cycle analysis, and gene expression. In cancer cells, recombinant PDX1 protein reduced cell migration and proliferation, and arrested cell growth by inducing apo

PURPOSE: The three-dimensional configurations of rod and cone bipolar cell (BC) dendrites and horizontal cell (HC) processes outside rod and cone synaptic terminals have not been fully elucidated. We reveal how these neurites are mutually arranged to coordinate formation and maintenance of the postsynaptic complex of ribbon synapses in mouse and monkey retinas.
METHODS: Serial section transmission electron microscopy was utilized to reconstruct BC and HC neurites in macaque monkey and mouse, including metabotropic glutamate receptor 6 (mGluR6)-knockout mice.
RESULTS: Starting from sporadical

The growing need for sustainable biotechnological solutions to address environmental challenges, such as climate change and resource depletion, has intensified interest in microbial-based production systems. Synthetic biofilms, which mimic natural microbial consortia, offer a promising platform for optimizing complex metabolic processes that can convert renewable feedstocks into valuable chemicals. In this context, understanding and harnessing the interactions between co-immobilized microorganisms are critical for advancing bioprocesses that contribute to circular bioeconomy goals. In this stu

Recent advances in AMP-activated protein kinase (AMPK) as a target in cancer waxed and waned over the past decade of cancer research. AMPK is a cellular energy sensor, present in almost all eukaryotic cells. An elevated AMP/ATP ratio activates the AMPK, which in turn inhibits energy-consuming processes and induces catabolic events that generate ATP to restore the energy homeostasis inside the cell. Several reports have indicated that AMPK regulates several metabolic pathways and may be a potential therapeutic target for the treatment of cancer. Cancer cells have specific metabolic changes that

Fed-batch processes using Chinese hamster ovary (CHO) cells are primarily employed to manufacture biologics. In these cultures, cells transition from exponential growth to the stationary phase, often resulting in a viability decline. This decline along, with similar reductions in cell growth and productivity is attributed to changes in cellular metabolism, the cell cycle, and apoptosis, potentially caused by nutrient imbalance and the accumulation of inhibitory byproducts. To enhance productivity in fed-batch processes, understanding the interactions among these factors is crucial for identify

Idiopathic pulmonary fibrosis (IPF) is characterized by the progressive and ultimately fatal accumulation of fibroblasts and extracellular matrix in the lung that distorts its architecture and compromises its function. IPF is now thought to result from wound-healing processes that, although initiated to protect the host from injurious environmental stimuli, lead to pathological fibrosis due to these processes becoming aberrant or over-exuberant. Although the environmental stimuli that trigger IPF remain to be identified, recent evidence suggests that they initially injure the alveolar epitheli

The fungal cell wall plays a large role in pathogenicity and is essential to the survival and virulence of fungi such as Candida albicans. Filamentous growth and its causes have been studied, however specific cell wall features that contribute to this morphology have yet to be investigated thoroughly. Understanding how the makeup of cell wall polymers changes during hyphal growth is therefore critical to developing novel targets for therapeutics. Bio-orthogonal chemistry has emerged as a powerful tool to profile critical cellular processes without interfering with canonical machinery. This cha

BACKGROUND: The rice blast fungus, Magnaporthe oryzae, poses a serious threat to global rice production. Cell cycle and polarized growth are two essential processes for host infection of M. oryzae; however, the association of them remains largely unknown.
RESULTS: MoHym1, a homolog of yeast RAM (regulation of Ace2 and morphogenesis) network component, was identified as a key regulator for fungal development and pathogenicity in M. oryzae. Deletion of MoHYM1 resulted in severe defects in vegetative growth, conidiation, polarized growth, cell wall integrity, surface hydrophobicity, and secretio

A family of IGF-binding proteins (IGFBP) exerts biological actions both dependent on and independent of IGF-I. A major effector of the insulin/IGF-I signaling pathway, the serine/threonine protein kinase Akt, mediates cellular processes such as glucose uptake, protein synthesis, cell survival, and growth. IGF-I is required for normal organismal growth, and in the pancreatic beta-cell, the insulin/IGF-I signaling pathway is critical for normal and adaptive maintenance of beta-cell mass. Expression of myrAkt1, an activated form of Akt, in the endocrine pancreas drives beta-cell expansion through

Restrictions in neuronal fate occur during the transition from a multipotential to a postmitotic cell. This and later steps in neuronal differentiation are determined by extracellular signals. We report that basic fibroblast growth factor is mitogenic for stem cells and is a differentiation factor for calbindin-expressing hippocampal neurons. The neurotrophin NT-3 is a differentiation factor for the same neurons but does not affect proliferation. NT-3 and brain-derived neurotrophic factor promote the maturation of neurons derived from stem cells that have been grown in vitro. These results def
