Mechanistic insights into the regulation of circular dorsal ruffle formation.

Growth factor stimulations induce dynamic changes in the cytoskeleton beneath the plasma membrane. Among them is the formation of membrane ruffles organized in a circular array, called 'circular dorsal ruffles' (CDRs). Physiological functions of CDRs include downregulation of cell growth by desensitizing the signalling from growth factor receptors as well as rearrangement of adhesion sites at the onset of cell migration. For the formation of CDRs, not only the activators of actin polymerization, such as N-WASP and the Arp2/3-complex, but also membrane deforming proteins with BAR/F-BAR domains

A Mussel-Inspired Bioadhesive Patch to Selectively Kill Glioblastoma Cells.

Glioblastoma, the most prevalent and aggressive brain tumor, presents significant challenge due to its rapid proliferation, invasive nature, and resistance to conventional therapies. Current treatments, including surgery, radiation, and chemotherapy, frequently lead to recurrence, underscoring the urgent need for innovative solutions. This work develops and evaluates bioinspired adhesive membranes designed as novel strategy to address glioblastoma recurrence post-surgery. Inspired by mussel adhesion, these membranes exhibit strong bioadhesion in wet environments and incorporate various phenoli

Parietal cell antibodies in pernicious anemia inhibit H+, K+-adenosine triphosphatase, the proton pump of the stomach.

Antibodies to a membrane-bound antigen, localized to the canalicular structures of the parietal cell, are found in most sera of patients with chronic atrophic gastritis and pernicious anemia. In the present study immunoglobulins containing parietal cell antibodies were found to inhibit the activity of H+,K+-adenosine triphosphatase (EC 3.6.1.36) in a tubulovesicular membrane preparation from porcine gastric mucosa. The degree of inhibition correlated to the titer of parietal cell antibodies as assessed by an enzyme-linked immunosorbent assay. The specificity of the enzymatic inhibition was con

Transient acantholytic dermatosis with involvement of oral mucosa.

Histologic, immunologic and electron microscopic studies were performed in a patient with transient acantholytic dermatosis which involved oral mucosa. Hitologic and electron microscopic findings were almost identical in both cutaneous and mucous membrane lesions, and these were similar to pemphigus vulgaris; suprabasilar separation with acantholytic cells. Desmosome-desmosome complexes were separated without disruption of cell membranes and cytoplasmic structures were well-preserved without dyskeratosis. Lamina lucida, however, was often separated in mucous membrane lesions, in contrast to th

Protein 4.1N is required for the formation of the lateral membrane domain in human bronchial epithelial cells.

The membrane skeleton forms a scaffold on the cytoplasmic side of the plasma membrane. The erythrocyte membrane represents an archetype of such structural organization. It has been documented that a similar membrane skeleton also exits in the Golgi complex. It has been previously shown that βII spectrin and ankyrin G are localized at the lateral membrane of human bronchial epithelial cells. Here we show that protein 4.1N is also located at the lateral membrane where it associates E-cadherin, β-catenin and βII spectrin. Importantly, depletion of 4.1N by RNAi in human bronchial epithelial cells

Structural insights into the elevator-like mechanism of the sodium/citrate symporter CitS.

The sodium-dependent citrate transporter of Klebsiella pneumoniae (KpCitS) belongs to the 2-hydroxycarboxylate transporter (2-HCT) family and allows the cell to use citrate as sole carbon and energy source in anaerobic conditions. Here we present crystal structures of KpCitS in citrate-bound outward-facing, citrate-bound asymmetric, and citrate-free inward-facing state. The structures reveal that the KpCitS dimerization domain remains stationary throughout the transport cycle due to a hydrogen bond network as well as extensive hydrophobic interactions. In contrast, its transport domain undergo

Cortactin (CTTN), N-WASP (WASL), and clathrin (CLTC) are present at podosome-like tubulobulbar complexes in the rat testis.

Tubulobulbar complexes are actin filament-rich plasma membrane protrusions that form at intercellular junctions in the seminiferous epithelium of the mammalian testis. They are proposed to internalize intact junctions during sperm release and during the translocation of spermatocytes through basal junction complexes between neighboring Sertoli cells. Tubulobulbar complexes morphologically resemble podosomes found at cell/substrate attachments in other systems. In this study we probe apical tubulobulbar complexes in fixed epithelial fragments and fixed frozen sections of rat testis for two key

Role of membrane organization and membrane domains in endocytic lipid trafficking.

Lipid compositions vary greatly among organelles, and specific sorting mechanisms are required to establish and maintain these distinct compositions. In this review, we discuss how the biophysical properties of the membrane bilayer and the chemistry of individual lipid molecules play a role in the intracellular trafficking of the lipids themselves, as well as influencing the trafficking of transmembrane proteins. The large diversity of lipid head groups and acyl chains lead to a variety of weak interactions, such as ionic and hydrogen bonding at the lipid/water interfacial region, hydrophobic

Lipopolysaccharide biogenesis and transport at the outer membrane of Gram-negative bacteria.

The outer membrane (OM) of Gram-negative bacteria is an asymmetric lipid bilayer containing a unique glycolipid, lipopolysaccharide (LPS) in its outer leaflet. LPS molecules confer to the OM peculiar permeability barrier properties enabling Gram-negative bacteria to exclude many toxic compounds, including clinically useful antibiotics, and to survive harsh environments. Transport of LPS poses several problems to the cells due to the amphipatic nature of this molecule. In this review we summarize the current knowledge on the LPS transport machinery, discuss the challenges associated with this p

Fluorescence as a method to reveal structures and membrane-interactions of amyloidogenic proteins.

Amyloidogenesis is a characteristic feature of the 40 or so known protein deposition diseases, and accumulating evidence strongly suggests that self-association of misfolded proteins into either fibrils, protofibrils, or soluble oligomeric species is cytotoxic. The most likely mechanism for toxicity is through perturbation of membrane structure, leading to increased membrane permeability and eventual cell death. There have been a rather limited number of investigations of the interactions of amyloidogenic polypeptides and their aggregated states with membranes; these are briefly reviewed here.

Tunneling Nanotubes in Myeloid Cells: Perspectives for Health and Infectious Diseases.

Tunneling nanotubes (TNTs) are cellular connections, which represent a novel route for cell-to-cell communication. Strong evidence points to a role for TNTs in the intercellular transfer of signals, molecules, organelles, and pathogens, involving them in many cellular functions. In myeloid cells (e.g., monocytes/macrophages, dendritic cells, and osteoclasts), intercellular communication via TNT contributes to their differentiation and immune functions, by favoring material and pathogen transfer, as well as cell fusion. This chapter addresses the complexity of the definition and characterizatio

Mitochondrial Transport from Mesenchymal Stromal Cells to Chondrocytes Increases DNA Content and Proteoglycan Deposition In Vitro in 3D Cultures.

OBJECTIVE: Allogeneic mesenchymal stromal cells (MSCs) are used in the 1-stage treatment of articular cartilage defects. The aim of this study is to investigate whether transport of mitochondria exists between chondrocytes and MSCs and to investigate whether the transfer of mitochondria to chondrocytes contributes to the mechanism of action of MSCs. DESIGN: Chondrocytes and MSCs were stained with MitoTracker, and CellTrace was used to distinguish between cell types. The uptake of fluorescent mitochondria was measured in cocultures using flow cytometry. Transport was visualized using fluoresce

Changes in the ultrastructure of human cells related to certain biological responses under hyperthermic culture conditions.

It has been reported that human cancer cells are more sensitive to high temperatures than normal human cells, and that cell proliferation and viability are affected by the temperature environment. In this study, we proceeded further, and turning our attention to the close relationship between cell morphology and temperature, used two human cancer cell lines and two normal cell strains to investigate how intracellular fine structure changes in a high temperature environment. The results showed that 1) both of the human cancer cell lines were more sensitive to high temperature than the normal hu

[Spindle-cell thymoma (a histological and electron microscopic study)].

The paper describes 12 cases of spindle cell thymoma without myasthenia or any other syndromes such as hypogammaglobulinemia, hypoplasia of the red sprout of the bone marrow. In most patients the tumours were detected roentgenologically. Microscopic diagnosis of spindle cell thymoma is based on the detection in it of epithelial cell structures. In those cases where the tumours were represented by spindle cells alone, verification of the tumor was possible only by electron microscopic studies (the presence in the cells of tonofilaments, desmosomes and basal membrane).

Pannexin1 channels contain a glycosylation site that targets the hexamer to the plasma membrane.

Pannexins are newly discovered channel proteins expressed in many different tissues and abundantly in the vertebrate central nervous system. Based on membrane topology, folding and secondary structure prediction, pannexins are proposed to form gap junction-like structures. We show here that Pannexin1 forms a hexameric channel and reaches the cell surface but, unlike connexins, is N-glycosylated. Using site-directed mutagenesis we analyzed three putative N-linked glycosylation sites and examined the effects of each mutation on channel expression. We show for the first time that Pannexin1 is gly

Mycobacterial surface-mediated mechanical priming initiates spacious to compact phagosome switching for egress.

Mycobacterium tuberculosis (Mtb) establishes intracellular niches by remodeling host membranes into either spacious or compact phagosomes, yet how direct bacterial contact within these distinct compartments facilitates bacterial egress remains unknown. Using fixed cell imaging, reconstituted phagosome-like vesicles, and numerical simulations, we uncover that mycobacterial load-mediated contact determines phagosome fate by driving membrane-bending, lipid-wrapping, and phase-separation. Low-to-moderate bacterial loads promote membrane vesiculation and formation of compact phagosome-like structur

Ultrastructure of Rickettsia sibirica during interaction with the host cell.

Rickettsia sibirica (strain Netsvetaev) was found within large translucent spaces in the cytoplasm of L-929 cell monolayer cultures on day 6 postinfection (p.i.). These rickettsiae were rod shaped 0.3 X 10 microns in size encircled by a halo of up to 200 nm wide corresponding to a capsule-like coat. Directly on the cell wall was a 12 nm thick microcapsule in which subunit structures with 12 nm thick spacing could be recognized. The cell wall membrane was 14 nm thick with a wider internal layer occasionally in a section found split into two electron-dense lamels; the internal layer corresponded

Ultrastructure of intra-epithelial nerve endings in the hard palate of the rat, Rattus norvegicus.

The ultrastructure is described of lanceolate and free nerve endings within the epithelium of the papillae of the intermolar palatal rugae, located either between the basement membranes and the bases of the epithelial cells, or suprabasally in the intercellular spaces of the epithelium. Cytoplasmic processes of epithelial cells invaginated the Schwann cells of the nerve endings; junctions between, or fusion of, the cell membranes of the epithelial cells and the Schwann cells were not found. The neurites of the nervous structures were characterized by numerous mitochondria, clear-cored vesicles
