NFKB1-miR-612-FAIM2 pathway regulates tumorigenesis in neurofibromatosis type 1.

Neurofibromatosis type I (NF1) is a carcinoma mainly featured by malignant peripheral nerve sheath tumor (MPNST). Dysregulated microRNAs (miRNAs) play decisive roles in tumor initiation and development. Our study sought for the possible roles of miR-612 in NF1. RT-qPCR estimated the expression of nuclear factor kappa B subunit 1 (NFKB1), miR-612, and Fas apoptotic inhibitory molecule 2 (FAIM2) in NF1, separately. Cell proliferation and migration were detected by CCK-8 and transwell experiments. Cell apoptosis was measured via flow cytometry and detection of the expression and activity of caspa

Solamargine inhibits the migration and invasion of HepG2 cells by blocking epithelial-to-mesenchymal transition.

Solamargine (SM), a steroidal alkaloid glycoside purified from the Chinese traditional herb Solanum incanum, is known to possess various biological activities. However, only a few previous studies have reported the anti-metastatic activity of SM. In the present study, the inhibitory effects of SM on metastatic action were investigated in human HepG2 cells. The proliferation effects of SM on the HepG2 cells was evaluated by MTT and colony formation assays. Wound-healing and Transwell assays were performed to examine the migration and invasion effects on SM-treated HepG2 cells. The epithelial-to

Protein arginine methyltransferase 1 stimulates basal cell proliferation and migration to maintain corneal epithelial homeostasis.

The corneal epithelium is a constantly self-renewing, stratified squamous tissue that protects the inner eye from external stimuli. The organization of the corneal epithelium involves multiple biological activities, including basal cell proliferation and centripetal migration. However, the underlying molecular mechanisms remain unclear. Herein, we identify protein arginine methyltransferase 1 (PRMT1) as a key regulator of corneal epithelial homeostasis. We exploited an inducible Prmt1 knockout mouse model and observed apparent disruption in the corneal epithelial homeostasis. PRMT1-deficient m

MiR-29b suppresses proliferation and mobility by targeting SOX12 and DNMT3b in pancreatic cancer.

Pancreatic cancer is one of the leading causes of solid carcinoma with the worst survival rate. The reasons for the worst survival rate include the lack of biomarkers for early detection, diagnosis at a late stage, and the limitation of the current therapy. Further study to investigate the underlying molecular mechanism in pancreatic cancer patients is necessary. A previous study showed that the miR-29b expression level is dysregulated, suggesting that it may serve an important function in pancreatic cancer. The CCK8 assay and the colony formation assay were used to detect the proliferation ab

AIM: Ovarian cancer (OC) is a fatal female malignant tumor that severely impacts the health of women worldwide. Due to the lack of diagnostic biomarkers, 70% of OC patients are considered in the advanced stage at the first diagnosis. Exploring novel biomarkers for OC diagnosis has become an urgent clinical need to address. TRIB1 is a newly discovered oncogene in several malignant tumors, including acute myeloid leukemia, prostate cancer, and breast cancer. However, the biological function of TRIB1 in OC remains uncertain and, therefore, was explored in the present study. METHODS: Levels of TR

Long noncoding RNA MINCR regulates cellular proliferation, migration, and invasion in hepatocellular carcinoma.

PURPOSE: Accumulating evidence indicates that long noncoding RNAs (lncRNAs) are aberrantly expressed in many cancer types, including hepatocellular carcinoma (HCC). lncRNA MYC-induced long non-coding RNA (MINCR) were revealed to be markedly up-regulated in gallbladder cancer and Burkitt lymphoma cells. However, the biological role and function of MINCR in HCC progression are still unknown. METHODS: The expression of MINCR in HCC tissues and cell lines was determined using quantitative real-time polymerase chain reaction assays. The effects of MINCR in HCC cell proliferation, migration, and in

CNN3 promotes vascular smooth muscle cells proliferation and migration to deteriorate atherosclerosis by β-catenin signaling pathway.

Atherosclerosis (AS) is a complex pathophysiological process. Evidence has demonstrated that abnormal proliferation and migration of vascular smooth muscle cells (VSMCs) play an essential role in AS. Calponin 3 (CNN3) is implicated in the pathogenesis of various diseases by modulating cell proliferation and migration. However, the roles of CNN3 in VSMCs and AS remains elusive. Experiments were performed in tissues and in vitro to investigate the role of CNN3 in the pathophysiology of AS. The expression of CNN3 in aortic plaques was determined using immunohistochemistry, and the serum CNN3 leve

Biophysical and Morphological Cell Features Retrieved by Digital Holographic Microscopy Correlate with Drug-Induced Changes in Cell Migration Behavior.

Quantitative analysis of cancer cell migration is critical for developing effective therapies to curb cancer metastasis. However, traditional methods are time-consuming and labor-intensive and lack quantitative capabilities. Cell volume change, a key physiological indicator of cell migration, is directly linked to phase change. In this work, we have developed a model that connects phase features from digital holographic microscopy (DHM) with cell healrate values from the wound healing assay. This approach aims to provide a rapid and quantitative evaluation of the breast cancer cell migration c

Macrophage Enhancement of Tumor Cell Extravasation and Intravasation In Vitro.

Macrophages orchestrate multiple functions in the tumor microenvironment leading to metastatic progression including promotion of tumor cell invasion, intravasation, and extravasation at metastatic sites. In vitro assays have been developed to explore these processes in high resolution. Here we describe an in vitro protocol used to examine macrophage enhanced tumor cell extravasation and intravasation in 3D using a stratified co-culture transendothelial migration (TEM) transwell system. These assays may be generally applied in order to study different aspect of metastatic dissemination, and we
