Unit activity in human thalamic reticularis neurons. II. Activity evoked by significant and non-significant verbal or sensory stimuli.

In the nucleus reticularis thalami (n.Rt.) of 46 dyskinetic patients the responses of 340 single units to significant and non-significant verbal and sound stimuli and ordered voluntary movements were studied. The spontaneous activity of the same neuronal populations previously examined allowed the classification of these neurons into 3 groups, named A, B and C types. Only A and B cells were found to be activated during the verbal command to perform a movement and its realization. The patterns of the responses of these units were studied by means of principal component analysis (PCA) and of cor

Drug response analysis for scaffold-free cardiac constructs fabricated using bio-3D printer.

Cardiac constructs fabricated using human induced pluripotent stem cells-derived cardiomyocytes (iPSCs-CMs) are useful for evaluating the cardiotoxicity of and cardiac response to new drugs. Previously, we fabricated scaffold-free three-dimensional (3D) tubular cardiac constructs using a bio-3D printer, which can load cardiac spheroids onto a needle array. In this study, we developed a method to measure the contractile force and to evaluate the drug response in cardiac constructs. Specifically, we measured the movement of the needle tip upon contraction of the cardiac constructs on the needle

Persistent parkinsonism with dopaminergic denervation following axicabtagene ciloleucel therapy for diffuse large B cell lymphoma: a case report.

While immune effector cell-associated neurotoxicity syndrome (ICANS) remains one of the major adverse events in CD19-targeted CAR-T cell therapy, neurodegenerative-like movement disorders are characteristically associated with BCMA-targeted therapies, not CD19. We report a distinct case of persistent parkinsonism that became evident during recurrent neurological deterioration accompanied by a decline in ICE score in a patient treated with axicabtagene ciloleucel for relapsed diffuse large B cell lymphoma. Notably, 123I-Ioflupane SPECT revealed reduced striatal dopamine transporter uptake, cons

Purkinje Cell spike patterns do not correlate with nuclei cell spike patterns in mouse models for cerebellar disease.

Cerebellar dysfunction causes various movement disorders, including ataxia, dystonia, and tremor. Previous work demonstrated that spike patterns in cerebellar nuclei neurons were distinct between different movement disorder mouse models. However, often these changes arise from neural dysfunction in the cerebellar cortex, through misfiring, miswiring, or degenerating Purkinje cells. Even though Purkinje cells form the sole output from the cerebellar cortex, their information is relayed to other regions of the motor network via cerebellar nuclei cells. Purkinje cells make GABAergic synapses onto

'Real-motion' cells in area V3A of macaque visual cortex.

The stability of visual perception despite eye movements suggests the existence, in the visual system, of neural elements able to recognize whether a movement of an image occurring in a particular part of the retina is the consequence of an actual movement that occurred in the visual field, or self-induced by an ocular movement while the object was still in the field of view. Recordings from single neurons in area V3A of awake macaque monkeys were made to check the existence of such a type of neurons (called 'real-motion' cells; see Galletti et al. 1984, 1988) in this prestriate area of the vi

Plant Nuclei Move to Escape Ultraviolet-Induced DNA Damage and Cell Death.

A striking feature of plant nuclei is their light-dependent movement. In Arabidopsis (Arabidopsis thaliana) leaf mesophyll cells, the nuclei move to the side walls of cells within 1 to 3 h after blue-light reception, although the reason is unknown. Here, we show that the nuclear movement is a rapid and effective strategy to avoid ultraviolet B (UVB)-induced damages. Mesophyll nuclei were positioned on the cell bottom in the dark, but sudden exposure of these cells to UVB caused severe DNA damage and cell death. The damage was remarkably reduced in both blue-light-treated leaves and mutant leav

Orthodontics. Part 11: orthodontic tooth movement.

Orthodontic tooth movement is dependent on efficient remodelling of bone. The cell-cell interactions are now more fully understood and the links between osteoblasts and osteoclasts appear to be governed by the production and responses of osteoprotegerin ligand. The theories of orthodontic tooth movement remain speculative but the histological documentation is unequivocal. A periodontal ligament placed under pressure will result in bone resorption whereas a periodontal ligament under tension results in bone formation. This phenomenon may be applicable to the generation of new bone in relation t

Paraneoplastic movement disorders.

Paraneoplastic movement disorders are rare, autoimmune-mediated, nonmetastatic complications of malignant neoplasms. Common paraneoplastic movement disorders include paraneoplastic chorea, dystonia, cerebellar degeneration, different types of encephalitis, opsoclonus-myoclonus syndrome, stiff person syndrome, and neuromyotonia. Syndromes usually develop before tumor diagnosis, have subacute onset, and are associated with serum or cerebrospinal fluid antibodies. Two types of antibodies can be distinguished: antibodies against nuclear and cytoplasmic neuronal antigens (anti-Hu, anti-Ri, anti-Yo,

Tomato Brown Rugose Fruit Virus Evades Tm-22-Mediated Resistance by Avoiding the Induction of Tm-22 Self-Association.

Tomato brown rugose fruit virus (ToBRFV) overcomes Tm-22-mediated resistance, posing a serious threat to global tomato production. We previously identified that key residues in the movement protein (MP)-H67, N125, K129, A134, I147 and I168 are essential for ToBRFV evasion of Tm-22 resistance. However, the underlying mechanisms remain unclear. In Tm-22-transgenic Nicotiana benthamiana plants at later infection stages, MP mutations H67C, N125A, K129Q and A134N abolished ToBRFV-GFP cell-to-cell and systemic movement, whereas I168N abolished ToBRFV-GFP systemic but not cell-to-cell movement, and I

bZIP60 and Bax inhibitor 1 contribute IRE1-dependent and independent roles to potexvirus infection.

IRE1, BI-1, and bZIP60 monitor compatible plant-potexvirus interactions though recognition of the viral TGB3 protein. This study was undertaken to elucidate the roles of three IRE1 isoforms, the bZIP60U and bZIP60S, and BI-1 roles in genetic reprogramming of cells during potexvirus infection. Experiments were performed using Arabidopsis thaliana knockout lines and Plantago asiatica mosaic virus infectious clone tagged with the green fluorescent protein gene (PlAMV-GFP). There were more PlAMV-GFP infection foci in ire1a/b, ire1c, bzip60, and bi-1 knockout than wild-type (WT) plants. Cell-to-cel

A common cause for nystagmus in different congenital stationary night blindness mouse models.

In Nyxnob mice, a model for congenital nystagmus associated with congenital stationary night blindness (CSNB), synchronous oscillating retinal ganglion cells (RGCs) lead to oscillatory eye movements, i.e. nystagmus. Given the specific expression of mGluR6 and Cav 1.4 in the photoreceptor to bipolar cell synapses, as well as their clinical association with CSNB, we hypothesize that Grm6nob3 and Cav 1.4-KO mutants show, like the Nyxnob mouse, oscillations in both their RGC activity and eye movements. Using multi-electrode array recordings of RGCs and measurements of the eye movements, we demonst

Thermosensitive TRPV4 channels mediate temperature-dependent microglia movement.

Microglia maintain central nervous system homeostasis by monitoring changes in their environment (resting state) and by taking protective actions to equilibrate such changes (activated state). These surveillance and protective roles both require constant movement of microglia. Interestingly, induced hypothermia can reduce microglia migration caused by ischemia, suggesting that microglia movement can be modulated by temperature. Although several ion channels and transporters are known to support microglia movement, the precise molecular mechanism that regulates temperature-dependent movement of

Endoderm convergence controls subduction of the myocardial precursors during heart-tube formation.

Coordination between the endoderm and adjacent cardiac mesoderm is crucial for heart development. We previously showed that myocardial migration is promoted by convergent movement of the endoderm, which itself is controlled by the S1pr2/Gα13 signaling pathway, but it remains unclear how the movements of the two tissues is coordinated. Here, we image live and fixed embryos to follow these movements, revealing previously unappreciated details of strikingly complex and dynamic associations between the endoderm and myocardial precursors. We found that during segmentation the endoderm underwent thr

Ecological paradigms to understand the dynamics of metastasis.

The process by which prostate cancer cells non-randomly disseminate to the bone to form lethal metastases remains unknown. Metastasis is the ultimate consequence of the long-range dispersal of a cancer cell from the primary tumor to a distant secondary site. In order to metastasize, the actively emigrating cell must move. Movement ecology describes an individual's migration between habitats without the requirement of conscious decision-making. Specifically, this paradigm describes four interacting components that influence the dynamic process of metastasis: (1) the microenvironmental pressures

Cell division and targeted cell cycle arrest opens and stabilizes basement membrane gaps.

Large gaps in basement membrane (BM) occur during organ remodelling and cancer cell invasion. Whether dividing cells, which temporarily reduce their attachment to BM, influence these breaches is unknown. Here we analyse uterine-vulval attachment during development across 21 species of rhabditid nematodes and find that the BM gap that forms between these organs is always bounded by a non-dividing vulval cell. Through cell cycle manipulation and live cell imaging in Caenorhabditis elegans, we show that actively dividing vulval cells facilitate enlargement of this breach by promoting BM movement.

Treatment of Movement Disorder Emergencies in Autoimmune Encephalitis in the Neurosciences ICU.

Immune response against neuronal and glial cell surface and cytosolic antigens is an important cause of encephalitis. It may be triggered by activation of the immune system in response to an infection (para-infectious), cancer (paraneoplastic), or due to a patient's tendency toward autoimmunity. Antibodies directed toward neuronal cell surface antigens are directly pathogenic, whereas antibodies with intracellular targets may become pathogenic if the antigen is transiently exposed to the cell surface or via activation of cytotoxic T cells. Immune-mediated encephalitis is well recognized and ma

Investigation of a two-patch within-host model of hepatitis B viral infection.

Chronic infection with hepatitis B virus (HBV) can lead to formation of abnormal nodular structures within the liver. To address how changes in liver anatomy affect overall virus-host dynamics, we developed within-host ordinary differential equation models of two-patch hepatitis B infection, one that assumes irreversible and one that assumes reversible movement between nodular structures. We investigated the models analytically and numerically, and determined the contribution of patch susceptibility, immune responses, and virus movement on within-patch and whole-liver virus dynamics. Mechanoresponsive Properties of the Periodontal Ligament.

The periodontal ligament (PDL) functions as an enthesis, a connective tissue attachment that dissipates strains created by mechanical loading. Entheses are mechanoresponsive structures that rapidly adapt to changes in their mechanical loading; here we asked which features of the PDL are sensitive to such in vivo loading. We evaluated the PDL in 4 physiologically relevant mechanical environments, focusing on mitotic activity, cell density, collagen content, osteogenic protein expression, and organization of the tissue. In addition to examining PDLs that supported teeth under masticatory loading

Ultrastructure of Merkel cells in the hard palate of the squirrel monkey (Saimiri sciureus).

The distribution and ultrastructure of Merkel cells in the hard palate was investigated in the squirrel monkey (Saimiri sciureus) after fixation by vascular perfusion. Merkel cells were clustered at the base of the epithelial rete pegs of the hard palate. They were characterized by concentrations of dense-cored granules and closely associated intraepithelial nerve endings. Numerous spiny processes extending from the Merkel cell were intercalated with adjacent keratinocytes, probably serving to detect and amplify movement of adjacent cells. These spiny processes contained a rigid core of parall
