Collagen XVII/BP180: a collagenous transmembrane protein and component of the dermoepidermal anchoring complex.

Collagen XVII, or BP180, is a collagenous transmembrane protein and a structural component of the dermoepidermal anchoring complex. Molecular studies reveal that it has a globular cytosolic amino-terminal domain and flexible-rod extracellular carboxyterminal domain. The extracellular portion of collagen XVII is constitutively shed from the cell surface by ADAMs (proteinases that contain adhesive and metalloprotease domains). Cell biological analyses suggest that collagen XVII functions as a cell-matrix adhesion molecule through stabilization of the hemidesmosome complex. This concept is suppor

Airway Progenitor Clone Formation Is Enhanced by Y-27632-Dependent Changes in the Transcriptome.

The application of conditional reprogramming culture (CRC) methods to nasal airway epithelial cells would allow more wide-spread incorporation of primary airway epithelial culture models into complex lung disease research. In this study, we adapted the CRC method to nasal airway epithelial cells, investigated the growth advantages afforded by this technique over standard culture methods, and determined the cellular and molecular basis of CRC cell culture effects. We found that the CRC method allowed the production of 7.1 × 10(10) cells after 4 passages, approximately 379 times more cells than

Engineering Shape-Controlled Microtissues on Compliant Hydrogels with Tunable Rigidity and Extracellular Matrix Ligands.

In vitro models that recapitulate key aspects of native tissue architecture and the physical microenvironment are emerging systems for modeling development and disease. For example, the myocardium consists of layers of aligned and coupled cardiac myocytes that are interspersed with supporting cells and embedded in a compliant extracellular matrix (ECM). These cell-cell and cell-matrix interactions are known to be important regulators of tissue physiology and pathophysiology. In this protocol, we describe a method for mimicking the alignment, cell-cell interactions, and rigidity of the myocardi

Research on the construction of corneal endothelium transplantation with acellular amniotic membrane as a scaffold.

INTRODUCTION: This study aimed to develop a human acellular amniotic membrane (HAAM) scaffold suitable for corneal endothelial transplantation. The HAAM was engineered using sequential chemical treatments and physical agitation to remove cellular components while preserving the extracellular matrix structure. The study sought to evaluate the biocompatibility and functional properties of the HAAM when seeded with immortalized human corneal endothelial cells (HCECs), with the ultimate goal of providing a potential therapeutic option for corneal endothelial dysfunction. METHODS: The HAAM was fab

[Structure of the dermo-epidemic junction].

The basement membranes are extracellular proteic matrixes involved mainly in cell-substrate interactions. The dermo-epidermal junction (DEJ) is defined as the region formed by the dermal pole of the basal cell layer, the intercellular spaces, the lamina densa or basal lamina and the acellular fibrous components of the papillary dermis. The functions of the DEJ are to bind the epithelia to the connective tissue, and to preserve the viability of the epidermis by means of connecting it to the dermis. which is in charge of providing the necessary nutrients for its normal proliferation. The purpose

The cell adhesion molecule nectin-1 is critical for normal enamel formation in mice.

Nectin-1 is a member of a sub-family of immunoglobulin-like adhesion molecules and a component of adherens junctions. In the current study, we have shown that mice lacking nectin-1 exhibit defective enamel formation in their incisor teeth. Although the incisors of nectin-1-null mice were hypomineralized, the protein composition of the enamel matrix was unaltered. While strong immunostaining for nectin-1 was observed at the interface between the maturation-stage ameloblasts and the underlying cells of the stratum intermedium (SI), its absence in nectin-1-null mice correlated with separation of

Overcoming immunogenic gaps in malaria subunit vaccines by broadening CSP regions targeted.

The first approval of malaria vaccines RTS,S/AS01 and R21/Matrix M marked a major milestone. Both vaccines present a portion of the Plasmodium falciparum circumsporozoite protein (PfCSP) containing an immunodominant major repeat region, which may limit breadth to other protective epitopes, limiting efficacy and durability. Using B cell receptor (BCR) knock-in mice, we found that the R21-included PfCSP epitope elicited robust B cell responses to the immunodominant major repeats but not to other highly protective epitopes, the minor repeat and junction. We then defined a minimal peptide capable

Basal lamina of human seminiferous tubule- its role in material transport. I. In presence of tunica vaginal hydrocele.

Electron microscopy of testicular biopsies obtained from two adult males with tunica vaginal hydrocele revealed some protrusions from the basal lamina to the germinal epithelium in the seminiferous tubule. The protrusions were of three types: some between the spermatogonia and Sertoli cells, some directly within the Sertoli cell cytoplasm and others inside the spermatogonia. The protrusions inside the spermatogonia were only 0.5 mum deep whereas the other types were from 1-11 mum deep. Occasionally some cut off portions of these protrusions were seen inside the ground cytoplasm of the Sertoli

Cell-cell junction gene signatures as subtype-specific prognostic biomarkers in breast cancer.

Cell-cell junctions (CCJs) are essential for maintaining epithelial integrity, and adhesion-related molecules have long been implicated in breast cancer progression. However, the subtype-specific prognostic significance of CCJ-related gene expression patterns within individual intrinsic breast cancer subtypes has not been systematically characterized. We analyzed 179 genes annotated to the Gene Ontology term "cell-cell junction organization" (GO:0045216) across intrinsic breast cancer subtypes using the METABRIC and The Cancer Genome Atlas (TCGA) datasets. Subtype-specific prognostic CCJ genes

GOLPH3 promotes alveolar epithelial glycocalyx shedding and barrier disruption in lipopolysaccharide-induced acute lung injury via the ERK/MMP2/9 axis.

BACKGROUND: Acute lung injury (ALI) is a life-threatening syndrome with high mortality, characterized by alveolar epithelial barrier disruption and excessive inflammatory injury. The epithelial glycocalyx, a layer critical for barrier integrity, undergoes shedding during ALI. However, the underlying regulatory mechanisms remain poorly understood. This study investigates the role of Golgi phosphoprotein 3 (GOLPH3) in glycocalyx-mediated barrier dysfunction in ALI. METHODS: A Lipopolysaccharide (LPS)-induced ALI model was established in MLE-12 cells and C57BL/6 mice. Bioinformatics analysis of

Synaptic Homeostasis and Its Immunological Disturbance in Neuromuscular Junction Disorders.

In the neuromuscular junction, postsynaptic nicotinic acetylcholine receptor (nAChR) clustering, trans-synaptic communication and synaptic stabilization are modulated by the molecular mechanisms underlying synaptic plasticity. The synaptic functions are based presynaptically on the active zone architecture, synaptic vesicle proteins, Ca2+ channels and synaptic vesicle recycling. Postsynaptically, they are based on rapsyn-anchored nAChR clusters, localized sensitivity to ACh, and synaptic stabilization via linkage to the extracellular matrix so as to be precisely opposed to the nerve terminal.

[TOPICAL ANTI-INFLAMMATORY AGENTS FOR DRY EYE DISEASE].

Dry eye disease (dry eye) is a multifactorial disorder of the ocular surface. Dry eye is one of the most frequent ocular disorders, affecting 5% to 50% of the entire population at all ages. Evidence suggests that inflammation and hyperosmolarity are considered core mechanisms in the development of dry eye. Dry eye is accompanied by changes in tear composition including enhanced hyperosmolarity and secretion of pro-inflammatory mediators such as cytokines, chemokines, matrix metalloproteinases (MMPs) and adhesion molecules. All these factors may act as mediators of tissue damage leading to lysi

Insights into the solution structure of the actin-binding tail domain of metavinculin by small angle X-ray scattering and molecular dynamics simulations.

Vinculin is a ubiquitously expressed focal adhesion protein that plays an important role in cell-matrix and cell-to-cell junctions. Metavinculin, a muscle-specific splice variant of vinculin, contains a 68-amino acid disordered insert region in its actin binding tail domain (MVt). Mutations in this insert are linked to cardiomyopathies. This study investigates the solution structures and structural ensembles of wild-type (WT) and two mutant MVts, ΔLeu954 and R975W, which have been associated with cardiomyopathies, using small-angle X-ray scattering (SAXS) and molecular dynamics (MD) simulation

Galectin-3 organizes histological compartments associated with inflammatory reaction induced by gliadin in BALB/c mice.

Galectin-3 regulates cell-cell and cell-extracellular matrix interactions in distinct tissues, including intestinal epithelial cells interfering with inflammatory responses. In the gut, galectin-3 stabilizes epithelial junctions and intestinal permeability. Here, it was investigated whether gliadin (protein of gluten) can induce inflammatory reactions in the digestive system in the presence or absence of galectin-3. BALB/c wild-type (Lgals3+/+) and genetically mutated in LGALS3 (Lgals3-/-) mice were divided into controls (water) or orally supplemented with gliadin (100mg/day) for 30 days. Frag

Visualization of the Dynamics of Invasion and Intravasation of the Bacterium That Causes Lyme Disease in a Tissue Engineered Dermal Microvessel Model.

Lyme disease is a tick-borne disease prevalent in North America, Europe, and Asia. Despite the accumulated knowledge from epidemiological, in vitro, and in animal studies, the understanding of dissemination of vector-borne pathogens, such as Borrelia burgdorferi (Bb), remains incomplete with several important knowledge gaps, especially related to invasion and intravasation into circulation. To elucidate the mechanistic details of these processes a tissue-engineered human dermal microvessel model is developed. Fluorescently labeled Bb are injected into the extracellular matrix (ECM) to mimic ti

hTERT-Driven Immortalization of RDEB Fibroblast and Keratinocyte Cell Lines Followed by Cre-Mediated Transgene Elimination.

The recessive form of dystrophic epidermolysis bullosa (RDEB) is a crippling disease caused by impairments in the junctions of the dermis and the basement membrane of the epidermis. Using ectopic expression of hTERT/hTERT + BMI-1 in primary cells, we developed expansible cultures of RDEB fibroblasts and keratinocytes. We showed that they display the properties of their founders, including morphology, contraction ability and expression of the respective specific markers including reduced secretion of type VII collagen (C7). The immortalized keratinocytes retained normal stratification in 3D ski

Secretory component production by polarized epithelial cells from the human female reproductive tract.

At mucosal surfaces, the polymeric Ig receptor (pIgR) is responsible for transporting polymeric IgA across epithelial cells. The purpose of this study was to determine whether normal epithelial cells from the female reproductive tract form tight junctions and produce secretory component, the external domain of the pIgR. Uterine, cervical and vaginal tissues from women at different stages of the menstrual cycle and following menopause were used to prepare purified epithelial cell sheets, which were cultured in cell chambers. Transepithelial resistance was measured and the media from apical and

Melanoma's connections to the tumour microenvironment.

Melanoma cells interact with and depend on seemingly normal cells in their tumour microenvironment to allow the acquisition of the hallmarks of solid cancer. In general, there are three types of interaction of melanoma cells with their microenvironment. First, there is bilateral communication between melanoma cells and the stroma, which includes fibroblasts, endothelial cells, immune cells, soluble molecules, and the extracellular matrix. Second, while under normal conditions keratinocytes control localisation and proliferative behaviour of melanocytes in the epidermis, once this balance is di

Application of ECIS to Evaluate the Effects of Porcine Urinary Bladder Matrix Hydrogels on Caco-2 Cell Attachment, Migration, and Barrier Formation.

Recent studies have highlighted the potential of urinary bladder matrix (UBM) derived from decellularized porcine urinary bladder as a bioactive hydrogel. Despite its complex composition of over 100 proteins, Type I collagen is the primary constituent of UBM. Caco-2 cells are widely used as an in vitro model of the intestinal epithelium; however, to date, no published study has evaluated the effects of UBM on Caco-2 cells. In this study, Electric Cell-Substrate Impedance Sensing (ECIS) was used to measure Caco-2 cell attachment and wound-healing migration on UBM-coated microelectrodes. Our res
