Suppression of Ciliogenesis Alleviates Cellular Senescence via AKT Signaling in Gingival Aging.

Aging, as an intrinsic risk factor, accelerates gingival inflammation and periodontal diseases. However, the cellular and molecular mechanisms underlying gingival aging remain unclear, hindering the development of targeted therapies. In this study, we performed the first single-cell transcriptomic analysis of aging human gingiva, identifying primary cilia as potential regulators of gingival fibroblast senescence. We demonstrated that aged gingival tissues exhibit increased fibroblast senescence and enhanced ciliogenesis compared to young tissues. Suppression of ciliogenesis significantly reduc

Cellular senescence in Crohn's disease: a double-edged sword in intestinal fibrosis.

Cellular senescence represents a pivotal yet paradoxical determinant in the pathogenesis of intestinal fibrosis associated with Crohn's disease (CD). While acute senescence may facilitate tissue repair and limit fibrogenesis, the chronic accumulation of senescent cells drives persistent inflammation and excessive extracellular matrix (ECM) deposition, ultimately leading to irreversible fibrotic strictures. This review systematically delineates the triggers of cellular senescence within the CD microenvironment-including oxidative stress, telomere dysfunction, endoplasmic reticulum stress, and g

CRISPR screening identifies GNPTAB as a noncanonical STING activator driving cellular senescence.

Cellular senescence is accompanied by profound lysosomal alterations, yet whether lysosome-associated factors actively drive aging remains unclear. Through a focused CRISPR/Cas9 screen in human mesenchymal progenitor cells (hMPCs), we identified N-acetylglucosamine-1-phosphotransferase subunits alpha and beta (GNPTAB), an enzyme responsible for lysosomal hydrolase targeting, as a potent regulator of cellular senescence. Genetic ablation of GNPTAB attenuated senescence, whereas its overexpression accelerated senescence. This pro-senescent function occurred independently of GNPTAB's canonical en

Renal IGFBP6 Interacts With THBS1 to Drive Renal Cellular Senescence and Fibrosis.

Epithelial dedifferentiation and myofibroblast activation are critical drivers of chronic kidney disease (CKD) progression. Elevated levels of IGFBP6 have been linked to decreased renal function in CKD patients, but its precise role and underlying mechanisms remain unclear. In this study, we observed significantly increased IGFBP6 expression in the kidney tissues of both renal fibrosis patients and animal models. Global or tubule-specific IGFBP6 knockout attenuated renal cellular senescence and fibrosis development in mice. In vitro, IGFBP6 deficiency preserved epithelial cell phenotype and in

A covalent resveratrol-nitroxide conjugate protects against oxidative stress-induced cellular senescence.

Cellular senescence, frequently driven by excess reactive oxygen species (ROS), is a major contributor to organismal aging. Anti-aging strategies that reduce ROS often show limited efficacy due to poor bioavailability of therapeutics, and the development of more effective anti-senescent agents remains an important research objective. In this study, we sought to integrate the activity of resveratrol (RSV), a polyphenolic antioxidant with documented anti-senescence properties, with the (2,2,6,6-tetramethylpiperidin-1-yl)oxyl radical (TEMPO), a superoxide dismutase mimetic. While RSV and TEMPO di

Energy-replenishing, mitochondria-targeted hydrogel microspheres mitigate sarcopenia via cellular senescence amelioration.

Sarcopenia is a degenerative skeletal muscle disorder closely associated with aging, characterized by the gradual loss of muscle mass and function, which severely impacts the quality of life in the elderly. In general, the severity of sarcopenia varies significantly among different patients and across different muscle groups, which increases the complexity and challenge of sarcopenia treatment. Based on the core pathological mechanism of sarcopenia, namely the deficiency of Nicotinamide adenine dinucleotide (NAD(+)) induced mitochondrial dysfunction, this study has developed a novel Energy-rep

Cellular senescence-related gene variants as risk factors for recurrent pregnancy loss.

BACKGROUND: Recurrent pregnancy loss (RPL) is characterized by two or more pregnancy losses. Despite its etiology encompassing known risk factors, over half of the cases remain idiopathic. In this sense, the search for molecular and cellular mechanisms that could be related to this condition is essential for explaining, at least in part, these cases. Cellular senescence (CS) is a state of cell cycle arrest that is present during reproduction and development; however, its impact on pregnancy remains unclear. Thus, our purpose was to evaluate the involvement of CS-related genes in the RPL contex

Targeting cellular senescence with Traditional Chinese Medicine: advances in neurodegenerative disorder treatment.

Neurodegenerative diseases (NDDs), including Alzheimer's disease (AD), Parkinson's disease (PD), and Huntington's disease (HD), represent formidable health challenges, particularly within aging populations. Cellular senescence, marked by irreversible cell cycle arrest and the secretion of pro-inflammatory cytokines, has emerged as a crucial factor in the pathogenesis of these disorders. The accumulation of senescent cells in the nervous system exacerbates neuroinflammation, impairs neuronal function, and accelerates tissue degeneration, all of which are key features of NDDs. In this context, b

DHCR24 promotes endometrial carcinoma progression and is associated with cellular senescence regulation.

Endometrial carcinoma (EC) is a common gynecological malignancy with an increasing incidence, particularly among younger and obese women. Although early-stage EC generally has favorable outcomes, advanced and recurrent disease remains difficult to treat, highlighting the need for reliable biomarkers and biologically relevant therapeutic targets. DHCR24, a terminal enzyme in cholesterol biosynthesis, has been implicated in tumor progression, but its clinical relevance and biological role in EC remain incompletely defined. In this study, we evaluated DHCR24 expression, prognostic significance, a

Fermented Porcine Placenta and Its Dipeptides Modulate Cellular Senescence in Human Keratinocytes.

Skin aging is primarily driven by oxidative stress, mitochondrial dysfunction, and cell cycle dysregulation. This study investigated the anti-senescence effects of fermented porcine placenta (FPP) and its dipeptides, leucine-glycine (LG) and proline-hydroxyproline (PH), in human epidermal keratinocytes (HEKs), using nicotinamide mononucleotide (NMN) as a reference for nicotinamide adenine dinucleotide (NAD+)-related pathways. FPP suppressed senescence-associated β-galactosidase (SA-β-gal) activity and Cyclin-dependent kinase inhibitor 2A (p16) expression while enhancing adenosine triphosphate

Entwined processes in Alzheimer's disease: Brain cellular senescence and Alzheimer's disease pathology.

Alzheimer's disease (AD) is one of the most common age-related neurodegenerative diseases with severe progressive dementia. Amyloid beta (Aβ) deposition and phosphorylated tau accumulation are two typical AD pathologies, which could drive senescence in various types of brain cells. These factors interact to form an entwined vicious cycle, in which each cell could play an irreplaceable role. Thus, it is essential to stress the cause-and-effect relationship in this process and to identify the key factor leading to the vicious cycle. In this narrative review, we discuss (1) how AD pathology induc

Alterations in mitochondria and cellular senescence in aged sEH null female kidneys.

Age-related structural and functional deterioration of the kidneys is common among elderly individuals and contributes to increased mortality and morbidity. Mitochondrial dysfunction and cellular senescence are two hallmarks of aging that drive a progressional renal decline; however, the underlying molecular mechanisms and endogenous regulators behind these processes remain incompletely understood. The metabolism of polyunsaturated fatty acids by CYP450 enzymes produces numerous bioactive lipid mediators that can be further metabolized by soluble epoxide hydrolase (sEH) and microsomal epoxide

Update of cellular senescence in kidney fibrosis: from mechanism to potential interventions.

Kidney fibrosis is the final common pathway of virtually all chronic kidney disease (CKD). However, despite great progress in recent years, no targeted antifibrotic therapies have been approved. Epidemiologic, clinical, and molecular evidence suggest that aging is a major contributor to the increasing incidence of CKD. Senescent renal tubular cells, fibroblasts, endothelial cells, and podocytes have been detected in the kidneys of patients with CKD and animal models. Nonetheless, although accumulated evidence supports the essential role of cellular senescence in CKD, the mechanisms that promot

Leucokinin VIII acetate mitigates asthma severity by targeting ADCK5-induced cellular senescence.

Severe asthma poses a significant challenge in clinical management due to its poor responsiveness to conventional therapies and worse overall prognosis. Identifying differentially expressed genes (DEGs) between severe asthma and non-severe asthma, exploring potential therapeutic targets, and intervening in these targets could offer new strategies to overcome the treatment bottleneck in severe asthma and improve clinical outcomes. Bioinformatics analysis first revealed that DEGs between severe and non-severe asthma were predominantly enriched in the longevity-regulating pathway, which is closel

Cellular Senescence Mediates Doxorubicin Chemotherapy-Induced Aortic Stiffening: Role of Glycation Stress.

BACKGROUND: Mechanisms underlying doxorubicin chemotherapy-induced aortic stiffening are incompletely understood. To determine the role of cellular senescence and the senescence-associated secretory phenotype (SASP) in mediating doxorubicin-induced aortic stiffening and the influence of senolytic therapy. METHODS: Aortic stiffness (aortic pulse wave velocity) and associated mechanisms were assessed in young adult p16-3MR mice, a model that allows for genetic-based clearance of senescent cells with ganciclovir. Young (4-6 months) mice were injected with doxorubicin and subsequently treated wit

Pharmacologic activation of Δ133p53α reduces cellular senescence in progeria patients-derived cells.

BACKGROUND: Patients with Hutchinson-Gilford progeria syndrome (HGPS) show accelerated aging phenotypes and have shortened lifespan, with implications in physiological aging processes as well. While therapeutic approaches targeting the disease-causing abnormal protein, progerin, have been developed, further efforts to explore mechanistically distinct and complementary strategies are still critical to better treatment regimens. We previously showed that lentiviral vector-driven expression of Δ133p53α, a natural inhibitory isoform of p53, rescued HGPS patients-derived fibroblasts from early entr

Uncovering cellular senescence as a therapeutic target in NF2-related vestibular schwannoma.

BACKGROUND: Vestibular schwannomas (VS) are complex and heterogeneous human tumors arising from the Schwann cell compartment of the vestibulocochlear nerve. VS cause significant neurological deficit such as hearing loss and vestibular impairment, and in some cases death due to brainstem compression. There is an urgent need to find pharmacotherapies for VS since surgical removal and stereotactic radiosurgery are the only effective treatments. Cancer therapy based in the combination of drug-induced senescence and senolytics may provide an innovative pharmacological alternative for VS management.

Targeted delivery of liposomal senolytics to alleviate cellular senescence-induced bone loss.

The senescence of bone marrow-derived mesenchymal stem cells is involved in osteoporosis. The combination of dasatinib and quercetin has been explored to alleviate bone loss by efficiently reducing senescent cell populations. However, senolytic therapy by dasatinib and quercetin requires a precise ratio for better therapeutic effects, which is hard to achieve by oral administration. Meanwhile, the poor water solubility of these compounds limits their bioavailability, and their non-specific action could hamper effective penetration and targeting within relevant tissues. Herein, we developed ale
