CD19 CAR T cells for B cell malignancies: a systematic review and meta-analysis focused on clinical impacts of CAR structural domains, manufacturing conditions, cellular product, doses, patient's age, and tumor types.

CD19-targeted chimeric antigen receptors (CAR) T cells are one of the most remarkable cellular therapies for managing B cell malignancies. However, long-term disease-free survival is still a challenge to overcome. Here, we evaluated the influence of different hinge, transmembrane (TM), and costimulatory CAR domains, as well as manufacturing conditions, cellular product type, doses, patient's age, and tumor types on the clinical outcomes of patients with B cell cancers treated with CD19 CAR T cells. The primary outcome was defined as the best complete response (BCR), and the secondary outcomes

Calmodulin Directly Interacts with the Cx43 Carboxyl-Terminus and Cytoplasmic Loop Containing Three ODDD-Linked Mutants (M147T, R148Q, and T154A) that Retain α-Helical Structure, but Exhibit Loss-of-Function and Cellular Trafficking Defects.

The autosomal-dominant pleiotropic disorder called oculodentodigital dysplasia (ODDD) is caused by mutations in the gap junction protein Cx43. Of the 73 mutations identified to date, over one-third are localized in the cytoplasmic loop (Cx43CL) domain. Here, we determined the mechanism by which three ODDD mutations (M147T, R148Q, and T154A), all of which localize within the predicted 1-5-10 calmodulin-binding motif of the Cx43CL, manifest the disease. Nuclear magnetic resonance (NMR) and circular dichroism revealed that the three ODDD mutations had little-to-no effect on the ability of the Cx4

Structure-Based Discovery Targeting GSK-3α Reveals Potent Nanomolar Selective 4-Phenyl-1H-benzofuro[3,2-b]pyrazolo[4,3-e]pyridine Inhibitor with Promising Glioblastoma and CNS-Active Potential in Cellular Models.

Glycogen synthase kinase-3 (GSK-3) is linked with multiple CNS conditions, including glioblastoma (GBM). Compared to the GSK-3β isoform, structure-based inhibitor design targeting GSK-3α is limited. Virtual screening was employed to identify GSK-3α inhibitors with CNS-active potential. Using a GSK-3α homology model, an optimized protocol with three-dimensional (3D)-pharmacophore filtering and Glide-SP docking was used to screen the ZINC20 biogenic subset. From 14 compounds selected for binding assay validation, three novel hit compounds were identified, with 1 (4-phenyl-1H-benzofuro[3,2-b]pyra

Postnatal development of the rabbit pineal gland. A light- and electron-microscopic study.

The development of the rabbit pineal gland has been studied by light and electron microscopy from the 1st to the 120th postnatal day. After 24 h of postnatal life, the pineal parenchyma is highly cellular, showing two identifiable cell types: pinealocytes I and II. Immature type II pinealocytes arrange either in cellular cords or clusters or forme rosette-like structures. At the 5th postnatal day, corticomedullar differentiation is established. Rosette-like structures and cellular cords are absent from the cortex. Along the postnatal period, nuclei of pinealocytes are set apart due to cytoplas

A Plesiohedral Cellular Network of Graphene Bubbles for Ultralight, Strong, and Superelastic Materials.

Advanced materials with low density and high strength impose transformative impacts in the construction, aerospace, and automobile industries. These materials can be realized by assembling well-designed modular building units (BUs) into interconnected structures. This study uses a hierarchical design strategy to demonstrate a new class of carbon-based, ultralight, strong, and even superelastic closed-cellular network structures. Here, the BUs are prepared by a multiscale design approach starting from the controlled synthesis of functionalized graphene oxide nanosheets at the molecular- and nan

Goyangamins A-C, Three Pairs of Enantiomeric 2(1H)-Quinolinone Alkaloid Dimers with Antisenescent Activity from Tetradium daniellii.

Goyangamins A-C (1-3), three 2(1H)-quinolinone alkaloid dimers, were isolated from the roots of Tetradium daniellii. The structures of these dimers were elucidated by HRESIMS, 1D and 2D NMR spectroscopy, and ECD analysis. Compounds 1 and 2 feature an sp3-sp2 chiral axis, whereas compound 3 contains an sp2-sp2 chiral axis, giving rise to a pair of interconverting atropisomers. Variable-temperature NMR (VT NMR), in conjunction with computational potential energy surface (PES) scans, indicated that compounds 1-3 belong to class I atropisomers that can readily interconvert at room temperature. Pleiotropic Roles of Cholesteryl Sulfate during Entamoeba Encystation: Involvement in Cell Rounding and Development of Membrane Impermeability.

Entamoeba histolytica, a protozoan parasite, causes amoebiasis, which is a global public health problem. The major route of infection is oral ingestion of cysts, the only form that is able to transmit to a new host. Cysts are produced by cell differentiation from proliferative trophozoites in a process termed "encystation." During encystation, cell morphology is markedly changed; motile amoeboid cells become rounded, nonmotile cells. Concomitantly, cell components change and significant fluctuations of metabolites occur. Cholesteryl sulfate (CS) is a crucial metabolite for encystation. However

Molecular and spatial analysis of tertiary lymphoid structures in Sjogren's syndrome.

Tertiary lymphoid structures play important roles in autoimmune and non-autoimmune conditions. While many of the molecular mechanisms involved in tertiary lymphoid structure formation have been identified, the cellular sources and temporal and spatial relationship remain unknown. Here we use combine single-cell RNA-sequencing, spatial transcriptomics and proteomics of minor salivary glands of patients with Sjogren's disease and Sicca Syndrome, with ex-vivo functional studies to construct a cellular and spatial map of key components involved in the formation and function of tertiary lymphoid st

Cryo-EM structures of CENP-A nucleosomes isolated from human cells.

Centromeres ensure accurate chromosome segregation, and their identity is specified epigenetically by the histone H3 variant CENP-A. Although the structures of reconstituted CENP-A nucleosomes have provided important insights, how this epigenetic mark is structurally organized within native chromatin remains incompletely understood. Here, we combine cryogenic electron microscopy with chromatin immunoprecipitation (ChIP-CryoEM) to determine the structures of human CENP-A nucleosomes isolated directly from cells without crosslinking or stabilizing agents. We resolved two distinct cellular CENP-A

Impact response and energy absorption of human skull cellular bones.

A skull fracture, due to a composition of typical lightweight cellular structures, is the most common type of traumatic brain injury. This paper presents a systematic investigation on the failure mechanism and energy absorption of skull cellular bones under low- and medium-velocity impact loadings. Non-destructive three-dimensional micro-computed tomography (Micro-CT) is utilized to scan samples of human skull cellular bones, and relevant structural parameters are obtained to reconstruct a finite element (FE) model of these bones. Micro-structures, mechanical properties, and failure process an

Design and Additive Manufacturing of 3D Phononic Band Gap Structures Based on Gradient Based Optimization.

We present a novel approach for gradient based maximization of phononic band gaps. The approach is a geometry projection method combining parametric shape optimization with density based topology optimization. By this approach, we obtain, in a two dimension setting, cellular structures exhibiting relative and normalized band gaps of more than 8 and 1.6, respectively. The controlling parameter is the minimal strut size, which also corresponds with the obtained stiffness of the structure. The resulting design principle is manually interpreted into a three dimensional structure from which cellula

In vitro and in vivo efficacy of carvacrol against Echinococcus granulosus.

Currently, benzimidazoles are used as chemotherapeutic agents and as a complement to surgery and PAIR in the treatment of cystic echinococcosis (CE). They are generally applied at high doses causing side effects and, 50% of cases do not respond favorably to such chemotherapy. The use of essential oils obtained by distillation from aromatic plants would be an effective alternative or complementary to the synthetic compounds, because would not bring the appearance of side effects. Carvacrol and his isomer thymol are the main phenolic components from essential oils of Origanum vulgare (oregano) a

Mimicking Cellular Membranes With Micropatterned Lipid Bilayers.

Studying the role of membranes in cell biophysics requires experimental models which faithfully capture the compositional and topological complexity of cellular structures. This is particularly important for membrane structures that are just at the limit of, or even below optical resolution, such as bacterial cells. Micropatterning is one of the key enabling technologies for the precise manipulation of compositional heterogeneity, morphology, and topography in artificial membranes, bringing in vitro models closer to cellular reality. This perspective highlights recent works which implement mic

Carbohydrates in herpesvirus infections.

Carbohydrate structures constitute important factors in the pathogenesis of herpesvirus infections for three reasons: (i) Human herpesviruses specify several biologically active glycoproteins, where co-translational addition of carbohydrate units is essential for generation of functioning viral glycoproteins. (ii) Cellular carbohydrate structures are parts of the cellular receptor complex at least for some herpes-viruses. (iii) Peripheral carbohydrate structures are engaged in modulation of the immunological properties of herpesvirus glycoproteins. The present review will consider all these th

Visualizing viral protein structures in cells using genetic probes for correlated light and electron microscopy.

Structural studies of viral proteins most often use high-resolution techniques such as X-ray crystallography, nuclear magnetic resonance, single particle negative stain, or cryo-electron microscopy (EM) to reveal atomic interactions of soluble, homogeneous viral proteins or viral protein complexes. Once viral proteins or complexes are separated from their host's cellular environment, their natural in situ structure and details of how they interact with other cellular components may be lost. EM has been an invaluable tool in virology since its introduction in the late 1940's and subsequent appl

Ultrastructural features of the midgut epithelium of females Lutzomyia intermedia (Lutz & Neiva, 1912) (Diptera: Psychodidae: Phlebotominae).

A morphological study of the midgut of Lutzomyia intermedia, the primary vector of cutaneous leishmaniasis, in southeast Brazil, was conducted by light, scanning and transmission electron microscopy. The midgut is formed by a layer of epithelium of columnar cells on a non-cellular basal lamina, under which there is a musculature, which consists of circular and longitudinal muscular fibers. A tracheolar network is observed surrounding and penetrating in the musculature. Females were examined 12, 24, 48, 72 h and 5 days following a blood meal and were analyzed comparatively by transmission elect

Nucleolar reorganization on stress depends on physicochemical changes due to reduced nascent rRNA synthesis.

Successive maturation of ribosomal subunits occurs through multilayered phase-separated structures of the cell nucleolus. The spatio-functional relationship between transcription of rRNA and nucleolar substructures, and how this adapts to cellular stress remain incompletely understood. In this study, we resolve the sub nucleolar structures using expansion microscopy to reveal ordered structures of fibrillar center (FC) and dense fibrillar component (DFC) domains as nested shells, which are reorganized upon cellular stress like DNA damage or RNA polymerase I (RNAPI) inhibition. TRIP13 - a potential drug target in cancer pharmacotherapy.

ATPases Associated with Diverse Cellular Activity (AAA+ATPases) are important enzymatic functional proteins in human cells. Thyroid Hormone Receptor Interacting Protein-13 (TRIP13) is a member of this protein superfamily, that partly regulates DNA repair pathways and spindle assembly checkpoints during mitosis. TRIP13 is reported as an oncogene involving multiple pathways in many human malignancies, including multiple myeloma, brain tumors, etc. The structure of TRIP13 reveals the mechanisms for ATP binding and how TRIP13 recognizes the Mitotic Arrest Deficiency-2 (MAD2) protein, with p31comet

Cellular intermediate filament networks and their derangement in alcoholic hepatitis.

Intermediate filaments are major components of most eukaryotic cells that form from the polymerization of protein subunits that are expressed in tissue and development specific fashions. The interactions of intermediate filaments with a myriad of other cellular proteins and structures give rise to a complex overall cellular architecture that is likely responsible for cellular well-being. The mature 10-nm filaments are relatively stable cellular structures, but the intermediate filaments undergo major morphological and biochemical changes, especially during mitosis, differentiation, and in resp

Cutaneous Cellular Pseudoglandular Schwannoma: An Unusual Histopathologic Variant.

Cellular schwannoma and pseudoglandular schwannoma are both previously described rare variants of schwannoma. The authors present an unusual case of a cellular spindle cell neoplasm with prominent gland-like structures, having features of both variants. The nature of this lesion was confirmed by histology and immunohistochemistry, with diffuse and strong S100 and membranous collagen type IV staining. The gland-like structures were lined by S100 + cells and contained proteinaceous, mucicarmine-negative material, supporting a degenerative, not true glandular, phenomenon. This is the first case o
