Glial responses to neonatal hypoxic-ischemic injury in the rat cerebral cortex.

Neurogenesis is nearly completed after birth, whereas gliogenic activities remain intense during the postnatal period in the developing rat cortex. These include involution of radial glia, proliferation of astrocytes and oligodendrocytes and myelin formation. Little is known about the effects of hypoxic-ischemic (HI) injury on these critical postnatal processes. Here we explored the glial reactions to mild HI injury of the neonatal rat cerebral cortex at P3. We show that the HI lesion results in disruption of the normal radial glia architecture, which was paralleled by an increase in GFAP immu

Structure of internal interneuronal connections in field 17 of the cat cerebral cortex.

The spatial distribution of horizontal internal connections in field 17 of the cat cortex was studied after microiontophoretic application of horseradish peroxidase to individual cortical columns. Cluster analysis of the distribution of labeled cells in the superficial layers in the tangential plane of the cortex was performed. Field 17 included 7 +/- 1 clusters of up to five cells. Clusters were distributed into two layers, separated by 1.2 +/- 0.3 mm. The distance between the centers of the clusters forming rows was 0.8 +/- 0.3 mm. The spatial characteristics of the grouping of cells sending

Reorganization of human cerebral cortex: the range of changes following use and injury.

Animal and human research over the past decades have increasingly detailed the brain's capacity for reorganization of neural network architecture to adapt to environmental needs. In this article, the authors outline the range of reorganization of human representational cortex, encompassing 1) reconstruction in concurrence with enhanced behaviorally relevant afferent activity (examples include skilled musicians and blind Braille readers); 2) injury-related response dynamics as, for instance, driven by loss of input (examples include stroke, amputation, or in blind individuals); and 3) maladapti

Isoflurane and pentobarbital reduce AMPA toxicity in vivo in the rat cerebral cortex.

BACKGROUND: Isoflurane and pentobarbital can reduce alpha-amino-d-hydroxy-5-methyl-4-isoxazole-propionate (AMPA) receptor-mediated toxicity in vitro. However, their effect on AMPA toxicity in vivo is not known. The present study was undertaken to evaluate the effects of isoflurane and pentobarbital on the in vivo neurotoxicity produced by AMPA. METHODS: Wistar-Kyoto rats were allocated to one of seven groups (n = 8 per group): isoflurane 1 minimum alveolar concentration, isoflurane electroencephalogram burst suppression (EEG-BS), low-dose pentobarbital, pentobarbital EEG-BS, NBQX, conscious,

[Acetaldehyde adducts in the cerebral cortex of long-term ethanol-fed mice].

It is assumed that the acetaldehyde binds to the proteins after the consumption of ethanol to form an adduct. Such acetaldehyde adducts are related to organ diseases. We examined 8-week-old female BALB/cAJcl mice which had been on a liquid diet for six months. The alcohol-fed mice's liquid diet comprised vitamins, minerals, amino acid and a ethanol solution. The five groups of mice examined were 1.5% ethanol-fed, 2% ethanol-fed, 2.5% ethanol-fed, 3% ethanol-fed, and 3.5% ethanol-fed mice respectively. All mice fed by 2.5% or more ethanol died within a month. Tissue samples were taken from the

[The modulation of cerebral cortex and subcortical nuclei on NRM and their role in acupuncture analgesia].

The vast research have demonstrated that the acupuncture analgesia is effected through a physiological mechanism brought about by the nervous system, particularly the central nervous system. We combined the acupuncture effects and theory of channels and collaterals with the new advance of pain neurophysiology, and centred attention on nucleus raphe magnus (NRM), that is one of the origins of the important descending inhibitory pathways of the intrinsic analgesic systems in brain. The unit discharges of NRM neurons and their nociceptors/ph responses were recorded extracellularly with glass micr

Microsphere embolism-induced changes in presynaptic function of the cerebral cortex in rats.

The present study was undertaken to elucidate pathophysiological changes in the cortical presynaptic function, K(+)-stimulated calcium influx, noradrenaline release and noradrenaline uptake, on the 1st and 3rd days after microsphere embolism in rats. Voltage-dependent calcium channels were characterized pharmacologically using three types of calcium channel blockers, L-type (nifedipine and diltiazem), N-type (omega-conotoxin GVIA), and P-type channel (omega-agatoxin IVA) blockers. K(+)-stimulated calcium influx of the normal rat synaptosome was inhibited by 100 nM omega-agatoxin IVA, but not b

Regional difference in cerebral blood flow and oxidative metabolism in human cortex.

UNLABELLED: We sought to determine if there are regional differences in cerebral blood flow (CBF) and cerebral metabolic ratio for oxygen (CMRO2) in normal subjects during the resting state. METHODS: Regional CBF, CMRO2 and oxygen extraction fraction (OEF) in 15 normal volunteers (mean age 58.8 +/- 8.2 yr) were measured during rest using PET and a 15O-gas steady-state technique. RESULTS: CBF and CMRO2 in the visual cortex were significantly higher than those in other cortices. Additionally, OEF in the sensorimotor cortex was significantly lower than that in other cortical regions. CONCLUSIO

A case of primary progressive aphasia with abnormally ubiquitinated neurites in the cerebral cortex.

We report the histopathological and immunohistochemical findings in a patient with primary progressive aphasia and abnormally ubiquitinated neurites in the cerebral cortex. Neuropathological examination showed severe neuronal loss and astrocytosis with a spongy change in the frontal cortex and neostriatum. Immunohistochemistry for ubiquitin antibody showed many immunoreactive dystrophic neurites in the superficial layer of the affected cortices and putamen. Those neurites were neither argentophilic nor stained with other antibodies against neurofilament, tau, or microtubule-associated protein-

Histamine-and isoprenaline-evoked stimulation of cAMP formation in chick cerebral cortex.

Histamine (HA) and isoprenaline (ISO) strongly stimulated cAMP formation in chick cerebral cortical slices. The effects of HA and ISO were selectively antagonized by HA H2-receptor blockers (aminopotentidine, ranitidine) and beta-adrenoceptor blocker propranolol, respectively. The combination of HA (0.1-100 microM) and ISO (0.1-10 microM) produced some additive effects when the drugs were used at low concentrations, i.e. 0.1-1 microM but not when HA was applied at 10 or 100 microM doses (which evoked nearly maximal effect). It is suggested that HA H2-like receptors and beta-adrenoceptors (whos

Alzheimer's disease-type neurofibrillary degeneration in verrucose dysplasias of the cerebral cortex.

Verrucose dysplasias, found at autopsy in the cerebral cortex of three elderly individuals (two without neurological disorders and one with motor neuron disease), are shown to present neurofibrillary degeneration of Alzheimer's disease type. This neurofibrillary degeneration immunoreacted with antibodies against abnormally phosphorylated tau (5E2 and AT8), disclosed acetyl- and butyrylcholinesterase activity, and was consistently stained with thioflavin-S. Cortical dysplasias, found either as isolated verrucose nodules or comprising multiple nodules, contained cell-sparse areas around which a

Preamyloid deposits in the cerebral cortex of patients with Alzheimer's disease and nondemented individuals.

A polyclonal antibody to a 28 residue synthetic peptide, homologous to the NH2 terminal region of amyloid beta-protein, was employed in a study of the frontal and temporal cortex of 8 Alzheimer patients and 13 non-demented individuals aimed to define the relationship of immunolabelled to argyrophilic, congophilic and thioflavine S-positive cortical lesions. In Alzheimer patients, this antiserum labelled not only senile plaques and congophilic angiopathy, but also cortical deposits that were neither argyrophilic, congophilic nor thioflavine S-positive and were unrelated to degenerating neurites

Adenosine receptors in post-mortem human cerebral cortex and the effect of carbamazepine.

Representative A1 site- and A2 site-selective ligands for adenosine receptors were found to bind to a single class of non-co-operative sites in synaptic membranes prepared from pieces of normal human temporal lobe cerebral cortex obtained at autopsies performed within 24 h post-mortem. The known adenosine antagonists theophylline and 1-isobutyl-3-methylxanthine also bound to these sites and could completely displace either ligand. Computer analysis combining data from 21 separate experiments which used membranes prepared from seven different autopsy cases gave the following kinetic parameters

Free amino acid changes in the cerebral cortex of experimental uremic rat.

The levels of free amino acids in the cerebral cortex of acute and chronic uremic rats were examined. Amino acids significantly elevated were aspartate, glutamine, glycine, histidine, ornithine, phenylalanine, phosphoethanolamine and taurine, whereas 1-methyl histidine and 3-methyl histidine were specifically detected in uremic rats. Glutamate, arginine and carnosine disclosed a significant reduction. There was no change in the concentrations of gamma-aminobutyrate and alanine. The above findings were essentially identical in both acute and chronic uremia. It was conjectured that these changes

Affinity labeling of muscarinic receptors in rat cerebral cortex with a photolabile antagonist.

Highly potent photoaffinity probes for muscarinic binding sites were prepared by the incorporation of an azido group into the benzilic acid moiety in two compound, 3-quinuclidinyl benzilate (3QNB) and N-methyl-4-piperidyl benzilate (4NMPB). Inactivation of muscarinic sites in rat cortex depends on the formation of a reversible complex with the azides prior to their photolytic conversion to the highly reactive nitrenes. During photolysis, radiolabeled azido-4NMPB interacted specifically and with high affinity (Kd = 1.06 nM) with the muscarinic receptors, and the ligand could be covalently incor

[Penetration of amoebae into ganglion cells of the cerebral cortex in primary amoebic meningoencephalitis].

The intracellular penetrating activity of amoebas of Limax-type (vs. Naegleria fowleri) was observed in a previously published case of the PAME with fulminant lethal course in man. This invasion concerned some ganglion cells of the cerebral cortex and - exceptionally - also the Purkinje-cells in the cerebellum. As to the formal genesis, adjacent and penetrating forms were identified in relation to the ganglion cells, and finally invaded neurons containing solitary or - exceptionally - two parasites were noted. This phenomenon of the intraneuronal lesion due to amoebae in PAME is said to be in

The toxicity to rat cerebral cortex or topical applications of acetaldehyde, ammonia or bilirubin.

The relative toxicity of acetaldehyde, ammonia and bilirubin to cerebral cortex of rats has been tested by superfusion of the intact dura for 1 h with each toxin separately. The cortex was examined histologically after 6 days survival using a silver strain which selectively impregnates degenerating axons and their synaptic terminals. A tissue concentration of 30 mM acetaldehyde was found to cause axonal and terminal degeneration, whereas 11 mM acetaldehyde was not toxic, and did not produce any structural changes detectable by electron microscopy. In terms of tissue concentrations, acetaldehyd

Representation pattern in the second somatic sensory area of the monkey cerebral cortex.

The body representation in the second somatic sensory area of macaques has been studied by tracing with anatomical techniques the projections from defined parts of the body representation in the first somatic sensory area (SI) to their terminal regions within the lateral sulcus. The second somatic sensory area (SH), as identified in terms of cytoarchitecture and its connection with the thalamic ventrobasal complex, is the only region of the lateral sulcus to receive a projection from SI. The nearby retroinsular area and area 7b receive a projection from area 5. Within SII the face and head rep
