Tropolone derivative hinokitiol ameliorates cerulein-induced acute pancreatitis in mice.

Hinokitiol is a natural bio-active tropolone derivative with promising antioxidant and anti-inflammatory properties. This study was conducted to evaluate the ameliorative effects of hinokitiol against acute pancreatitis induced by cerulein. Mice were pre-treated with hinokitiol intraperitoneally for 7 days (50 and 100 mg/kg), and on the final day of study, cerulein (6 × 50 μg/kg) was injected every hour for six times. Six hours after the last dose of cerulein, blood was collected from the mice through retro-orbital plexus for biochemical analysis. After blood collection, mice were euthanized a

Effects of pantoprazole in experimental acute pancreatitis.

AIMS: Oxidative stress with free radicals plays a crucial role in acute pancreatitis (AP). Pantoprazole (PPZ), widely used as a proton pump inhibitor, possesses reactivity towards hydroxyl radicals. The aim of the study was to examine the effect of PPZ on the course of experimental AP. MAIN METHODS: Mild AP was induced in rats by caerulein (n=12). Severe AP was induced by infusion of glycodeoxycholic acid (10mM) into the pancreatic duct combined with caerulein (n=12). Both AP models were randomized to PPZ treatment (20mg/kg at baseline and after 12h) or placebo. Control animals received Ringe

The influence of cholecystokinin on gastric myoelectrical activity in duodenal ulcer following Helicobacter pylori eradication--an electrogastrographic study.

Cholecystokinin (CCK) plays an important role in the regulation of postprandial gastric motor activity which was found to be abnormal in duodenal ulcer patients. This study was designed to compare the influence of CCK on gastric myoelectrical function in duodenal ulcer patients and healthy controls. Fifteen patients with active duodenal ulcer and Helicobacterpylori (H. pylori) infection and 15 healthy controls were included into this study. Electrogastrography (EGG) was performed before and 4 weeks after the eradication of H. pylori in ulcer patients and in healthy controls. We compared EGG pa

[Pancreatic duct antibodies and subclinical insufficiency of the exocrine pancreas in Sjögren's syndrome].

In previous studies we reported evidence of subclinical exocrine pancreatic insufficiency in primary or secondary Sjögren's syndrome (SSI, SSII) and rheumatoid arthritis (RA). In present study we evaluated the occurrence of pancreatic duct antibodies (PDA), and their relationship to exocrine pancreatic function in 36 women. Of these patients, 12 were classified as SSI, 12 as SSII, and 12 as RA. Exocrine pancreatic function was evaluated using the Secretin-Caerulein test (S.Cae test). The indirect immunofluorescent technique was used to evaluate patient sera for PDA. S.Cae test results, compare

Influence of chronic intracerebroventricular infusion of vasoactive intestinal peptide (VIP) on memory processes in Morris water pool test in the rat.

In our previous study, bolus injection of VIP into the lateral cerebral ventricle, at nonphysiological high doses, has been shown to produce amnesia. Accordingly, in the present study, VIP was infused chronically into the cerebral ventricle of the rat at a rate of 10 ng per day for 2 weeks. During the infusion period, the animals were subjected to the Morris water pool test. The VIP infusion caused an apparent impairment of memory, particularly in the acquisition of new information; VIP(1-12) also caused similar impairment, but to a lesser extent. The VIP antagonists did not affect the perform

beta-Endorphin involvement in the antidopaminergic effect of caerulein.

Caerulein has been shown to possess a long-lasting antagonistic effect on amphetamine hyperactivity in rats when given in combination with haloperidol. We found that this effect of caerulein involved beta-endorphin. Naloxone pretreatment and hypophysectomy abolished the caerulein effect, while intracerebroventricular or intra-nucleus accumbens injection of beta-endorphin together with haloperidol administration produced an effect similar to that of caerulein. The results suggest that the long-term antagonism of the amphetamine effect of caerulein is mediated by the endogenous opioid beta-endor

The effect of gastrin and CCK-like peptides on epithelial cell proliferation in the stomach.

The growth promoting effect of gastrin on the glandular stem cells and parietal cells of the oxyntic mucosa has been confirmed by several observations. Gastrin has no effect on the antral mucosa. Cholecystokinin, another trophic hormone, stimulates cell proliferation in the exocrine pancreas, duodenum and gallbladder but not in the oxyntic mucosa. More recently, we have observed that caerulein stimulates epithelial cell proliferation in the antrum. The following is seen after acute administration of this cholecystokinin analog: a, after 18 h, there is an increased nuclear 3H-thymidine uptake i

Prostaglandin E2 derived from phosphatidylinositol breakdown in the exocrine pancreas facilitates secretion by an action on the ducts.

The observation that stimulation of a variety of endocrine and exocrine glands, synaptic tissue and certain other structures with appropriate agonists is associated with a breakdown of phosphatidylinositol (PI) and a synthesis of phosphatidic acid (accompanied by increased turnover) has long been known. Our laboratory recently showed that arachidonate released from PI breakdown forms prostaglandin (PG) E2 and the latter is involved in secretagogue-stimulated secretion from the exocrine pancreas. The data presented here are consistent with the view that there is a "pool" of enzymes in the ducts

MiR-324-5p mitigates experimental acute pancreatitis by suppressing Rock2 expression.

Acute pancreatitis (AP) is a lethal pancreatic disease with limited therapeutic options. MicroRNA-324-5p (miR-324-5p) has shown dual roles across diseases, but its specific role and molecular targets in AP remain unclear. Plasma miR-324-5p levels were measured in 80 AP patients and 40 healthy controls using RT-qPCR. An in vivo AP mouse model was induced by caerulein, and agomir miR-324-5p was administered to assess its therapeutic effect. In vitro, MPC-83 pancreatic acinar cells were treated with caerulein and transfected with miR-324-5p mimics or co-transfected with Rock2 overexpression plasm

Arachidonate 15-lipoxygenase: A promising therapeutic target for alleviating inflammation in acute pancreatitis.

This article discusses the significant findings from the study on the transfection of arachidonate 15-lipoxygenase (ALOX15) and its therapeutic potential in managing acute pancreatitis (AP). The research highlights the role of ALOX15 in attenuating inflammatory responses, apoptosis, and autophagy in a cerulein-induced AP murine model. By using a recombinant lentiviral vector for efficient gene delivery, the study provides compelling evidence for the protective effects of ALOX15 transfection on pancreatic tissue. The authors demonstrate that ALOX15 reduces the expression of key inflammatory mar

High-mobility group box 1 fragment ameliorates chronic pancreatitis induced by caerulein in mice.

BACKGROUND: Chronic pancreatitis (CP) is a progressive disease characterized by pancreatic fibrosis for which effective treatment options are lacking. Mesenchymal stem cells (MSCs) have shown potential for fibrosis treatment but face limitations in clinical application. The high-mobility group box 1 (HMGB1) fragment mobilizes MSCs from bone marrow into the blood and has emerged as a promising therapeutic agent for tissue regeneration in various pathological conditions. The aim of this study was to investigate the potential therapeutic effects of systemic administration of the HMGB1 fragment in

A Single-Cell Atlas of the Murine Pancreatic Ductal Tree Identifies Novel Cell Populations With Potential Implications in Pancreas Regeneration and Exocrine Pathogenesis.

BACKGROUND & AIMS: Pancreatic ducts form an intricate network of tubules that secrete bicarbonate and drive acinar secretions into the duodenum. This network is formed by centroacinar cells, terminal, intercalated, intracalated ducts, and the main pancreatic duct. Ductal heterogeneity at the single-cell level has been poorly characterized; therefore, our understanding of the role of ductal cells in pancreas regeneration and exocrine pathogenesis has been hampered by the limited knowledge and unexplained diversity within the ductal network. METHODS: We used single cell RNA sequencing to compre

Vagus Nerve Stimulation Decreases Pancreatitis Severity in Mice.

BACKGROUND: Vagus nerve stimulation (VNS) is effective in reducing inflammation in various diseases, such as rheumatoid arthritis, colitis and acute kidney injury. The anti-inflammatory effect of vagus nerve in these diseases necessitates the interactions of neural activation and α7 nicotinic acetylcholine receptors (α7nAChRs) on splenic macrophages. In this study, we aimed to investigate the effect of VNS on severity in experimental acute pancreatitis (AP). METHODS: Two independent AP models were used, which induced in ICR mice with caerulein or pancreatic duct ligation (PDL). Thirty minutes

Macrophages-induced IL-18-mediated eosinophilia promotes characteristics of pancreatic malignancy.

Reports indicate that accumulated macrophages in the pancreas are responsible for promoting the pathogenesis of chronic pancreatitis (CP). Recently, macrophage-secreted cytokines have been implicated in promoting pancreatic acinar-to-ductal metaplasia (ADM). This study aims to establish the role of accumulated macrophage-activated NLRP3-IL-18-eosinophil mechanistic pathway in promoting several characteristics of pancreatic malignancy in CP. We report that in a murine model of pancreatic cancer (PC), accumulated macrophages are the source of NLRP3-regulated IL-18, which promotes eosinophilic in

Exocrine pancreatic function after alcoholic or biliary acute pancreatitis.

OBJECTIVES: There have been various studies of exocrine pancreatic function after acute pancreatitis, but few have examined the relationship between this function and the etiology of the pancreatitis. The aim of this work was to study pancreatic function in patients who had had acute alcoholic or acute biliary pancreatitis. METHODS: Seventy-five patients who had had a single attack of acute pancreatitis were studied. The etiology was alcohol in 36 and cholelithiasis in 39. Pancreatic function was studied between 4 and 18 months after pancreatitis by duodenal intubation in 18 patients (8 alcoh

Effects of alpha 1-acid glycoprotein on acute pancreatitis and acute lung injury in rats.

alpha 1-Acid glycoprotein (AAG), a highly negatively charged glycoprotein, well known for its capillary stabilizing effect, was tested in rat models of acute edematous pancreatitis, acute hemorrhagic-necrotizing pancreatitis, and acute respiratory distress syndrome (ARDS). In cerulein-elicited edematous pancreatitis AAG improved histological alterations at 200 mg/kg i.v. and plasma amylase activity at 1800 or 4200 mg/kg i.v. All other parameters (edema, plasma lipase) were not affected in a biologically relevant manner. In glycodeoxycholic acid-induced hemorrhagic-necrotizing pancreatitis AAG

The role of intracellular calcium signaling in premature protease activation and the onset of pancreatitis.

The exocrine pancreas synthesizes and secretes large amounts of digestive proteases as inactive precursor zymogens. Under physiological conditions a variety of cellular defense mechanisms protect the pancreatic acinar cell against a premature and intracellular activation of these zymogens. When these defenses fail, pancreatic autodigestion is initiated and acute pancreatitis can develop. A number of experimental observations suggest that extra- as well as intracellular calcium concentrations play an important part in the initiation of pancreatic protease activation, but the intracellular signa

Neutrophil behavior in pancreas and liver and the role of nitric oxide in rat acute pancreatitis.

The behavior of neutrophils in a rat acute pancreatitis model was observed in the pancreas and liver using fluorescence microscopy with an image analyzing system after labeling with a specific fluorescent reagent. Nonviable cells of both organs were also labeled and quantified. The role of nitric oxide in neutrophil accumulation and organ damage was estimated by administering a relatively selective inhibitor of constitutive nitric oxide synthase, N-nitro-L-arginine (L-NNA). The animal model of acute pancreatitis was induced by cerulein injection (80 mg/kg). Two groups were created, one given a
