Chalcones from Angelica keiskei (ashitaba) inhibit key Zika virus replication proteins.

Zika virus (ZIKV) is a dangerous human pathogen and no antiviral drugs have been approved to date. The chalcones are a group of small molecules that are found in a number of different plants, including Angelica keiskei Koidzumi, also known as ashitaba. To examine chalcone anti-ZIKV activity, three chalcones, 4-hydroxyderricin (4HD), xanthoangelol (XA), and xanthoangelol-E (XA-E), were purified from a methanol-ethyl acetate extract from A. keiskei. Molecular and ensemble docking predicted that these chalcones would establish multiple interactions with residues in the catalytic and allosteric si

Design, synthesis and antibacterial activity of chalcones against MSSA and MRSA planktonic cells and biofilms.

Staphylococcus aureus is the one of the most successful modern pathogens. The same bacterium that lives as a skin and mucosal commensal can be transmitted in health-care and community-settings and causes severe infections. Thus, there is a great challenge for a discovery of novel anti-Staphylococcus aureus compounds, which should act against resistant strains. Herein, we designed and synthesized a series of 17 chalcones, substituted by amino group on ring A, which were evaluated against methicillin-susceptible S. aureus (MSSA) and methicillin-resistant S. aureus MRSA planktonic cells. The anti

Chalcone Scaffolds as Anticancer Drugs: A Review on Molecular Insight in Action of Mechanisms and Anticancer Properties.

Cancer is the deadliest disease worldwide and the development of safer chemical entities to treat cancer is one of the major challenges of medicinal chemistry. The emergence of new cases every year and the development of multiple drug resistance against available molecular entities have turned the focus of researchers towards natural products. Chalcones are pharmacologically active compounds, present in plants, which have been derivatized and screened by many researchers for the treatment of cancer. Chalcones, consist of 1,3-diaryl-2- propen-1-one, is one such class exhibiting broad anticancer

Disruption of endolysosomal trafficking pathways in glioma cells by methuosis-inducing indole-based chalcones.

Methuosis is a form of non-apoptotic cell death involving massive vacuolization of macropinosome-derived endocytic compartments, followed by a decline in metabolic activity and loss of membrane integrity. To explore the induction of methuosis as a potential therapeutic strategy for killing cancer cells, we have developed small molecules (indole-based chalcones) that trigger this form of cell death in glioblastoma and other cancer cell lines. Here, we report that in addition to causing fusion and expansion of macropinosome compartments, the lead compound, 3-(5-methoxy-2-methyl-1H-indol-3-yl)-1-

Isoliquiritigenin, a chalcone compound, is a positive allosteric modulator of GABAA receptors and shows hypnotic effects.

Isoliquiritigenin (ILTG) is a chalcone compound and has valuable pharmacological properties such as antioxidant, anti-inflammatory, anticancer, and antiallergic activities. Recently, the anxiolytic effect of ILTG has been reported; however, its action mechanism and hypnotic activity have not yet been demonstrated. Therefore, we investigated the hypnotic effect and action mechanism of ILTG. ILTG significantly potentiated the pentobarbital-induced sleep in mice at doses of 25 and 50mg/kg. The hypnotic activity of ILTG was fully inhibited by flumazenil (FLU), a specific gamma-aminobutyric acid ty

Chalcone-based synthesis of a new Pyrazoline derivative: Molecular docking and anthelmintic evaluation against Schistosoma mansoni.

Chalcones and their derivatives have attracted considerable interest in drug discovery due to literature reports describing their diverse biological activities. In this study, we evaluated the in vitro schistosomicidal activity of a chalcone derivative and its corresponding pyrazoline derivative against adult Schistosoma mansoni worms. The chalcone (1a) and its newly synthesized pyrazoline derivative (1b), reported here for the first time, showed generally favorable predicted pharmacokinetic and ADMET profiles. Both compounds were predicted to exhibit high gastrointestinal absorption (GIA), su

New players from the old chalcone scaffold: Synthesis, identification, and characterization of new potent ABCG2 inhibitors.

The ABCG2 transporter is an efflux pump that can transport various anticancer drugs and is strongly associated with multidrug resistance (MDR) in cancer. A promising strategy to combat MDR mediated by this transporter is through functional inhibition. However, there are currently no potent selective ABCG2 inhibitors in clinical trials. Chalcone, a privileged scaffold, can be easily synthesized to produce a wide array of derivatives with diverse biological applications. Notably, chalcone derivatives have been identified as inhibitors of ABC transporters, including ABCG2. In this study, we synth

Fragment optimized chalcone derivatives targeting OmpA protein as a therapeutic approach against multidrug resistant Acinetobacter baumannii.

Acinetobacter baumannii is a notorious pathogen associated with life-threatening infections, with its outer membrane protein A (OmpA) being a key contributor to its pathogenicity by targeting epithelial cell apoptosis. The study presents an in silico analysis of chalcone derivatives as potential therapeutic agents against the outer membrane protein A (OmpA) of Acinetobacter baumannii. We performed molecular docking to evaluate the binding interactions, revealing that isobavachalcone exhibited the highest binding affinity. Further fragment optimization (FOI) of isobavachalcone improved its bind

Biological evaluation and molecular Docking studies of the prepared chalcone derivatives as potential anti-Alzheimer agents.

Alzheimer's disease (AD) is a neurological disorder characterized by memory loss, language difficulties, and the loss of planning and coordination skills. Research done by neurologists suggest that the decline in acetylcholine and the accumulation of pathological forms of amyloid beta plaques produced by both the β- and γ-secretase enzymes in the brain as some of the primary causes of AD. Due to the multifactorial pathway associated with the progression of the disease, the inhibition of cholinesterase, β-secretase and other anti-inflammatory related enzymes have been reported as one of the maj

A hydroxyl coumarin-chalcone-based fluorescent probe for sensing copper ions in plant and living cells.

Copper ions are essential trace elements for living organisms. However, its excessive levels in the liver and kidneys may lead to severe health issues. Therefore, developing effective fluorescent probes for detecting copper ions is crucial. A hydroxyl coumarin chalcone derivative featuring a hydroxyl substituent has been developed as a fluorescent probe for monitoring Cu2+. The probe can selectively detect Cu2+ in aqueous solutions, exhibiting a visible color change from red to light yellow to the naked eye. This is accompanied by a quenching of the red fluorescence, with a limit of detection

Morpholinodiazenyl chalcone blocks influenza A virus capsid uncoating by perturbing the clathrin-mediated vesicular trafficking pathway.

Influenza A virus (IAV) is a highly contagious respiratory pathogen that significantly threatens global health by causing seasonal epidemics and occasional, unpredictable pandemics. To identify new compounds with therapeutic potential against IAV, we designed and synthesized a series of 4'-morpholinodiazenyl chalcones using the molecular hybridization method, performed a high-content screen against IAV, and found that (E)-1-{4-[(E)-morpholinodiazenyl]phenyl}-3-(3,4,5-trimethoxyphenyl)prop-2-en-1-one (MC-22) completely neutralized IAV infection. While MC-22 allowed IAV to successfully internali

In Vivo Safety and Efficacy of Chalcone-Loaded Microparticles with Modified Polymeric Matrix against Cutaneous Leishmaniasis.

Current chemotherapy of cutaneous leishmaniasis (CL) is based on repeated systemic or intralesional administration of drugs that often cause severe toxicity. Previously, we demonstrated the therapeutic potential of biodegradable poly(lactic-co-glycolic acid) (PLGA) microparticles (MPs) loaded with 8% of the nitrochalcone CH8 (CH8/PLGA) prepared by a conventional bench method. Aiming at an industrially scalable process and increased drug loading, new MPs were prepared by spray drying: CH8/PDE with PLGA matrix and CH8/PVDE with PLGA + polyvinylpyrrolidone (PVP) matrix, both with narrower size di
