The Role of Hydrogen Sulfide in the Dilatation of Mesenteric Lymphatic Vessels in Bulls.

We studied the effects and mechanisms of action of NaHS, an H2S donor, on bovine isolated mesenteric efferent lymphatic vessels pre-contracted with phenylephrine. NaHS induced concentration-dependent relaxation of lymphatic vessels. Removal of the endothelium reduced, but did not completely abolish the relaxing effect of NaHS. Application of NO synthase inhibitor L-NAME, soluble guanylyl cyclase inhibitor ODQ, blocker ATP-sensitive K+ channels glibenclamide, and a combination of blockers of Ca-activated K+ channels of small and intermediate conductance charybdotoxin and apamin attenuated relax

Contribution of activation of K+ channels to glyceryl trinitrate-induced relaxation of rabbit aorta.

1. Possible contribution of K+ channel opening to the relaxation by glyceryl trinitrate (GTN) was examined using isolated rabbit aorta. 2. While glibenclamide and apamin failed to affect relaxation by GTN, both charybdotoxin (ChTx) and iberiotoxin (IbTx) effectively attenuated GTN-induced relaxation. 3. The increase in cGMP produced by GTN was not attenuated by ChTx and IbTx. 4. The inhibitory effect of ChTx on GTN-induced relaxation was not reduced in the presence of zaprinast, indicating that cGMP but not GMP was responsible for activation of the K+ channel. 5. Pb2+ activates potassium currents in bovine adrenal chromaffin cells.

Inorganic lead (Pb2+) is a potent environmental toxin which adversely affects several aspects of neuronal and secretory cell function. In this report, we provide evidence that at subnanomolar concentrations, Pb2+ activates the outward K+ currents in bovine adrenal chromaffin cells. Whole-cell patch clamp combined with intracellular perfusion was employed to monitor outward K+ currents in bovine chromaffin cells before and after intracellular application of EGTA-Pb buffers. Intracellular Pb2+ > or = 10(-10) M enhanced the K(+)-currents in a concentration dependent manner, with apparent K0.5 app

Functional modification of a Ca2+-activated K+ channel by trimethyloxonium.

Single Ca2+-activated K+ channels from rat skeletal muscle plasma membranes were studied in neutral phospholipid bilayers. Channels were chemically modified by briefly exposing the external side to the carboxyl group modifying reagent trimethyloxonium (TMO). TMO modification, in a "multi-hit" fashion, reduces the single-channel conductance without affecting ion selectivity. Modification also shifts the voltage activation curve toward more depolarized voltages and reduces the affinity of the channel blocker charybdotoxin (CTX). CTX, bound to the channel during the TMO exposure, prevents the TMO

Properties of ShB A-type potassium channels expressed in Shaker mutant Drosophila by germline transformation.

We have used P element-mediated germline transformation to express ShB channels in Shaker mutant Drosophila and have examined their properties by patch-clamp of embryonic myotubes. The transformed ShB cDNA was placed under the transcriptional control of a heat shock promoter (hsp70). Northern blots revealed that transformed DNA is efficiently transcribed in response to heat shock. Cultured myotubes from the transformants produced large A-type potassium currents in response to heat shock. Although qualitatively similar to native Shaker A-currents in wild-type myotubes, transformant A-current in

[The rational evolution of scorpion toxins].

A theoretical method for the rational design of a "universal" scorpion toxin with a wider spectrum of specificity for K+ channels and a more stable alpha/beta-folding than in its natural homologues is described. On the basis of the analysis of molecular hydrophobic potentials (MHP) of the protein spatial structures, structural features for a family of five short scorpion toxins were revealed. The analysis of the maps of two-dimensional intramolecular MHP contacts allowed the identification of amino acid residues responsible for the folding of the protein and/or for the manifestation of its spe

High-level expression and functional reconstitution of Shaker K+ channels.

Voltage-gated K+ channels were expressed in COS cells transiently transfected with a plasmid carrying a cDNA for an inactivation-removed Shaker K+ channel driven by an adenovirus promoter. Channel expression was followed by immunological detection, binding of radioactive charybdotoxin (CTX), and functional reconstitution into planar lipid bilayers. About 10(7) channels per transfected cell are expressed on the plasma membrane. The expressed channels are glycosylated and competent to bind CTX with the expected characteristics. Channels observed after insertion into planar lipid bilayers display

Noxiustoxin and leiurutoxin III, two homologous peptide toxins with binding properties to synaptosomal membrane K+ channels.

Noxiustoxin (NTX), a short-chain peptide toxin from the scorpion C. noxius, binds to a single class of non-interacting binding sites in brain synaptosomal membranes with a KD = 160-300 nM and a Bmax = 2.2 pmols.mg-1 protein. The K+ channel blocking agents tetraethylammonium, 4-aminopyridine and Charybdotoxin partially inhibit the binding of [125I]NTX, while a variety of toxins targeted against Ca2+ and Na+ channels have no effect, suggesting that NTX interacts with a bona fide K+ channel protein. NTX totally displaces the binding of [125I]Leiurutoxin III, a novel peptide from L. quinquestriatu

Effects of K+ channel agonists cromakalim and pinacidil on rat basilar artery smooth muscle cells are mediated by Ca(++)-activated K+ channels.

Whole-cell and cell-free inside-out patch-clamp recording techniques were used to examine the actions of potassium channel openers pinacidil and cromakalim in enzymatically isolated smooth muscle cells of rat basilar artery. Delayed rectifier and calcium-dependent potassium currents were identified from the whole-cell recordings. Only the calcium-dependent potassium current was increased by cromakalim and pinacidil. Recordings from inside-out membrane patches revealed a large conductance voltage- and calcium-dependent potassium channel, which was blocked by charybdotoxin but unaffected by ATP

ATP-sensitive K+ channels from aortic smooth muscle incorporated into planar lipid bilayers.

Glibenclamide binding sites were identified in a membrane preparation from canine aortic smooth muscle. The dissociation constant for [3H]glibenclamide binding was 10 +/- 2 nM, with a density of 420 +/- 108 fmol/mg protein. The properties of ATP-sensitive potassium (KATP) channels from the same membrane preparation incorporated into planar lipid bilayers were investigated. ATP was a potent inhibitor of the channels with half-maximal inhibition of channel activity by 41 microM ATP. Glibenclamide inhibited channel activity, and cromakalim activated the channel in the presence of ATP. Blockers of

Evidence for two calcium-dependent potassium conductances in lizard motor nerve terminals.

Action potentials and afterpotentials were recorded with a microelectrode inserted into lizard motor axons within a few millimeters of their motor terminals. In the presence of 1 mM 4-aminopyridine (4-AP), the duration of the action potential recorded near motor terminals was Ca-sensitive: repolarization was more rapid when bath [Ca] was elevated, and became slower when bath [Ca] was removed or when 0.1-1 mM Mn was added. Repolarization was also slowed following addition of 3-10 nM charybdotoxin or 100-300 microM tetraethylammonium (TEA) to the bath, and following intra-axonal injection of the

Modulation of chloride, potassium and bicarbonate transport by muscarinic receptors in a human adenocarcinoma cell line.

1. Short-circuit current (I(SC)) responses to carbachol (CCh) were investigated in Colony 1 epithelia, a subpopulation of the HCA-7 adenocarcinoma cell line. In Krebs-Henseleit (KH) buffer, CCh responses consisted of three I(SC) components: an unusual rapid decrease (the 10 s spike) followed by an upward spike at 30 s and a slower transient increase (the 2 min peak). This response was not potentiated by forskolin; rather, CCh inhibited cyclic AMP-stimulated I(SC). 2. In HCO3- free buffer, the decrease in forskolin-elevated I(SC) after CCh was reduced, although the interactions between CCh and

Electrophysiological recordings and calcium measurements in striatal large aspiny interneurons in response to combined O2/glucose deprivation.

Electrophysiological recordings and calcium measurements in striatal large aspiny interneurons in response to combined O2/glucose deprivation. The effects of combined O2/glucose deprivation were investigated on large aspiny (LA) interneurons recorded from a striatal slice preparation by means of simultaneous electrophysiological and optical recordings. LA interneurons were visually identified and impaled with sharp microelectrodes loaded with the calcium (Ca2+)-sensitive dye bis-fura-2. These cells showed the morphological, electrophysiological, and pharmacological features of large striatal c

Oxytocin-induced changes in single cell K+ currents and smooth muscle contraction of guinea-pig gastric antrum.

To study the effects of oxytocin on both spontaneous phasic contractions and K+ outward currents (IK) of the so-called 'non-target' smooth muscle cells, physiological concentrations of oxytocin ranging between 10(-12) mol/l and 10(-8) mol/l were applied to smooth muscle preparations and single voltage-clamped cells isolated from the circular layer of the guinea-pig gastric antrum. Oxytocin (10(-12) mol/l to 10(-8) mol/l) suppressed, in a dose-dependent manner, the tetrodotoxin- and atropine-resistant spontaneous phasic contractions and shifted rightward the dose-response curves of 10(-7) mol/l

Atomic structure and plasticity of the CTX-MthK complex investigated by cryo-EM, NMR, and MD simulations.

Scorpion toxins block potassium channels, disrupting cellular excitability and causing symptoms such as pain, muscle spasms, or paralysis. Here, we use an integrated structural biology approach to uncover the binding mode of the scorpion toxin charybdotoxin (CTX) to the MthK channel, a model system for human large-conductance potassium (BK) channels. Cryo-EM defines the overall architecture of the MthK-CTX complex, while complementary solution- and solid-state NMR experiments identify key binding residues and show that toxin engagement alters the selectivity filter (SF) ion configuration witho

Distinct soluble astrocytic factors induce expression of outward K+ currents and ramification of brain macrophages.

Isolated cultured murine brain macrophages (BM) were treated with supernatants of enriched astrocytic cultures. The astrocyte-conditioned medium (ACM) induced ramification of BM. In parallel, BM expressed voltage-gated outward K+ currents (I(K)) during the first 2 days after the application of ACM. However, in ramified BM which were treated once with ACM, I(K) disappeared 5 days after that treatment. In contrast, BM expressed I(K) over a period of more than 5 days when cells were treated daily with ACM. A blockade of I(K) by charybdotoxin or by kaliotoxin did not inhibit ramification of the ce

Determination of the subunit stoichiometry of a voltage-activated potassium channel.

The voltage-activated K+, Na+ and Ca2+ channels are responsible for the generation and propagation of electrical signals in cell membranes. The K+ channels are multimeric membrane proteins formed by the aggregation of an unknown number of independent subunits. By studying the interaction of a scorpion toxin with coexpressed wild-type and toxin-insensitive mutant Shaker K+ channels, the subunit stoichiometry can be determined. The Shaker K+ channel is found to have a tetrameric structure. This is consistent with the sequence relationship between a K+ channel and each of the four internally homo

Solution structure of BmBKTx1, a new BKCa1 channel blocker from the Chinese scorpion Buthus martensi Karsch.

BmBKTx1 is a 31-amino acid peptide identified from the venom of the Chinese scorpion Buthus martensi Karsch, blocking high-conductance calcium-activated potassium channels. Sequence homology analysis indicates that BmBKTx1 is a new subfamily of short-chain alpha-KTx toxins of the potassium channel, which we term alpha-KTx19. Synthetic BmBKTx1 was prepared by using solid-phase peptide synthesis. Two-dimensional NMR spectroscopy techniques were used to determine the solution structure of BmBKTx1. The results show that the BmBKTx1 forms a typical cysteine-stabilized alpha/beta scaffold adopted by
