Opioid-induced respiratory depression (OIRD) causes death following an opioid overdose, yet the neurobiological mechanisms of this process are not well understood. Here, we show that neurons within the lateral parabrachial nucleus that express the µ-opioid receptor (PBL Oprm1 neurons) are involved in OIRD pathogenesis. PBL Oprm1 neuronal activity is tightly correlated with respiratory rate, and this correlation is abolished following morphine injection. Chemogenetic inactivation of PBL Oprm1 neurons mimics OIRD in mice, whereas their chemogenetic activation following morphine injection rescues

Previous work has demonstrated that fusion of a luciferase to an opsin, to create a luminescent opsin or luminopsin, provides a genetically encoded means of manipulating neuronal activity via both chemogenetic and optogenetic approaches. Here we have expanded and refined the versatility of luminopsin tools by fusing an alternative luciferase variant with high light emission, Gaussia luciferase mutant GLucM23, to depolarizing and hyperpolarizing channelrhodopsins with increased light sensitivity. The combination of GLucM23 with Volvox channelrhodopsin-1 produced LMO4, while combining GLucM23 wi

Cataplexy is a hallmark of narcolepsy characterized by the sudden uncontrollable onset of muscle weakness or paralysis during wakefulness. It can occur spontaneously, but is typically triggered by positive emotions such as laughter. Although cataplexy was identified >130 years ago, its neural mechanism remains unclear. Here, we show that a newly identified GABA circuit within the central nucleus of the amygdala (CeA) promotes cataplexy. We used behavioral, electrophysiological, immunohistochemical, and chemogenetic strategies to target and manipulate CeA activity selectively in narcoleptic (or

Temporal lobe epilepsy (TLE) is a common form of epilepsy in adults. The anterior nucleus of the thalamus (ANT) is a potential therapeutic target due to its role in epileptic networks. Using calcium imaging, we show that glutamatergic neurons in the anterodorsal thalamic nucleus (AD), a subarea of the ANT, are activated during hippocampal seizures. Chemogenetic inhibition of AD glutamatergic neurons suppresses seizure onset and generalization in TLE models, whereas activation promotes seizures. Trans-monosynaptic tracing identifies the postsubiculum (PoSub) as an important downstream target of

The anterior cingulate cortex (ACC) is one of the critical brain areas for processing noxious information. Previous studies showed that peripheral nerve injury induced broad changes in the ACC, contributing to pain hypersensitivity. The neurons in layer 3 (L3) of the ACC receive the inputs from the mediodorsal thalamus (MD) and form the feedforward inhibition (FFI) microcircuits. The effects of peripheral nerve injury on the MD-driven FFI in L3 of ACC are unknown. In our study, we record the enhanced excitatory synaptic transmissions from the MD to L3 of the ACC in mice with common peroneal ne

Loss-of-function mutations of the gene Cul3 have been identified as a risk factor for autism-spectrum disorder (ASD), but the pathogenic mechanisms are not well understood. Conditional Cul3 ablation in cholinergic neurons of mice (ChatCRECul3F/+) recapitulated ASD-like social and sensory gating phenotypes and caused significant cognitive impairments, with diminished activity of cholinergic neurons in the basal forebrain (BF). Chemogenetic inhibition of BF cholinergic neurons in healthy mice induced similar social and cognitive deficits. Conversely, chemogenetic stimulation of BF cholinergic ne

BACKGROUND: Neural tissue has limited regenerative ability. To cope with that, in recent years a diverse set of novel tools has been used to tailor neurostimulation therapies and promote functional regeneration after axonal injuries.
METHOD: In this report, we explore cell-specific methods to modulate neuronal activity, including opto- and chemogenetics to assess the effect of specific neuronal stimulation in the promotion of axonal regeneration after injury.
RESULTS: Opto- and chemogenetic stimulations of neuronal activity elicited increased in vitro neurite outgrowth in both sensory and co

Dementia is characterized by loss of cognitive function, social deficits, and emotional impairment and is prevalent in tau-mediated disorders. However, the mechanisms underlying this cognitive and behavioral dysfunction remain uncertain, as phenotypes do not necessarily correlate well with the presence of tau pathology. Here, we identify critical roles for the cerebellum in regulating cognition and behavior in a tauopathy mouse model. We find that social and fear memory deficits emerge much later than tau pathology and that cerebellar Purkinje cell (PC) dysfunction coincides with the onset of

Pain of unknown etiology is frequent in individuals with the tumor predisposition syndrome neurofibromatosis 1 (NF1), even when tumors are absent. Nerve Schwann cells (SCs) were recently shown to play roles in nociceptive processing, and we find that chemogenetic activation of SCs is sufficient to induce afferent and behavioral mechanical hypersensitivity in wild-type mice. In mouse models, animals showed afferent and behavioral hypersensitivity when SCs, but not neurons, lacked Nf1. Importantly, hypersensitivity corresponded with SC-specific upregulation of mRNA encoding glial cell line-deriv

The hypothalamus is key in the control of energy balance. However, strategies targeting hypothalamic neurons have failed to provide viable options to treat most metabolic diseases. Conversely, the role of astrocytes in systemic metabolic control has remained largely unexplored. Here, we show that obesity promotes anatomically restricted remodeling of hypothalamic astrocyte activity. In the paraventricular nucleus (PVN) of the hypothalamus, chemogenetic manipulation of astrocytes results in bidirectional control of neighboring neuron activity, autonomic outflow, glucose metabolism, and energy b

Maintaining body temperature within a narrow range is vital for warm-blooded animals. In rodents, the preoptic area (POA) of the hypothalamus detects and regulates core body temperature. However, knowledge about the thermal regulation center in primates remains limited. Here, we show that activating a subpopulation of POA neurons by a chemogenetic strategy reliably induces hypothermia in anesthetized and freely moving macaques. Comprehensive monitoring of physiological parameters reveals that such hypothermia is accompanied by autonomic changes including a rise in heart rate, skeletal muscle a

Understanding the neural mechanisms underlying sleep state transitions is a fundamental goal of neurobiology and important for the development of new treatments for insomnia and other sleep disorders. Yet, brain circuits controlling this process remain poorly understood. Here we identify a population of sleep-active glutamatergic neurons in the ventrolateral medulla (VLM) that project to the preoptic area (POA), a prominent sleep-promoting region, in mice. Microendoscopic calcium imaging demonstrate that these VLM glutamatergic neurons display increased activity during the transitions from wak

Chronic neuropathic pain is frequently comorbid with anxiety disorders, yet the neural circuits underlying this interaction remain poorly defined. The parafascicular nucleus (PF) of the thalamus integrates nociceptive and affective signals, but its specific regulatory mechanisms in pain-anxiety comorbidity are not well known. Using spared nerve injury (SNI) model mice, we combined viral neural tracing, chemogenetics, pharmacology, and electrophysiology to dissect the locus coeruleus (LC)-PF neural pathway. Viral tracing revealed monosynaptic projections from norepinephrinergic (NEergic) neuron

BACKGROUND: Vascular cognitive impairment (VCI) is a prevalent and heterogeneous condition, both clinically and pathophysiologically, that still lacks approved treatment. Peripheral electrical nerve stimulation (PENS) shows promise for VCI management, yet its underlying neurobiological mechanisms are not well understood.
METHODS: VCI was induced in rats via permanent bilateral common carotid artery occlusion. Cognitive function was assessed using the Morris water maze, Y-maze, and novel object recognition. Synaptic plasticity was evaluated through long-term potentiation recordings and Golgi s

The medial prefrontal cortex (mPFC), which abundantly expresses corticotropin-releasing hormone (CRH) and its receptors, is a key region for integrating somatosensory, cognitive, and emotional information. Although CRH has been implicated in itch modulation, the cellular and local-circuit roles of CRH-positive neurons in the mPFC remain poorly defined. Here, we combined behavioral assays, in vivo fiber photometry, chemogenetics, optogenetics, immunofluorescence, and ex vivo whole-cell patch-clamp recordings to determine how mPFC CRH neurons regulate itch-related scratching in mice. mPFC CRH ne

BACKGROUND: The mechanisms underlying volatile anaesthetic-induced unconsciousness remain unclear. Glutamatergic pyramidal neurones and fast-spiking interneurones in cerebral cortex circuits play distinct roles in cortical network dynamics under volatile anaesthesia. We investigated the roles of medial prefrontal cortex (mPFC) pyramidal and fast-spiking interneurones in volatile anaesthetic-induced hypnosis.
METHODS: The electrophysiological properties of pyramidal and fast-spiking parvalbumin (PV+) neurones were explored by in vivo multichannel recordings and in vitro patch-clamp electrophys

Chronic itch, a devastating dermatological disorder, lacks targeted therapies due to incomplete understanding of its neural circuitry. Building on seminal studies that identified neuropeptide Y (NPY) inhibitory interneurons and their downstream urocortin 3-positive (Ucn3+)/Y1R-expressing neurons, calcitonin receptor-like receptor-positive (Calcrl+) neurons, identified as spinal projection neurons, have been proposed to contribute to mechanical itch signaling, though their underlying mechanistic role remains undefined. In the present study, using chemogenetic manipulation, behavioral tests, mor

BACKGROUND: Impulsive action and risk-related decision-making (RDM) are associated with various psychiatric disorders, including drug abuse. Both behavioral traits have also been linked to reduced frontocortical activity and alterations in dopamine function in the ventral tegmental area (VTA). However, despite direct projections from the medial prefrontal cortex (mPFC) to the VTA, the specific role of the mPFC-to-VTA pathway in controlling impulsive action and RDM remains unexplored.

BACKGROUND: To investigate specific brain regions and neural circuits that are responsible for migraine chronification.
METHODS: We established a mouse model of chronic migraine with intermittent injections of clinically-relevant dose of nitroglycerin (0.1 mg/kg for 9 days) and validated the model with cephalic and extracephalic mechanical sensitivity, calcitonin gene-related peptide (CGRP) expression in trigeminal ganglion, and responsiveness to sumatriptan or central CGRP blockade. We explored the neurons that were sensitized along with migraine chronification and investigated their roles o

BACKGROUND: Intrinsic and extrinsic trauma to the temporomandibular joint (TMJ) increases the incidence of painful temporomandibular disorders. TMJ trauma also causes increased degeneration of the TMJ structure, leading to TMJ osteoarthritis (TMJOA). Clinically, it is important to identify risk factors of post-traumatic TMJOA, such as sex, injury severity, and persistent post-traumatic pain.
OBJECTIVES: We determined the effects of potential risk factors on post-traumatic TMJ degeneration using a mouse model.
METHODS: A repeated forced-mouth opening (FMO) model was used to determine the effe
