Intra-Articular Injection of Chitin Nanofiber Attenuates Osteoarthritis: An Experimental Study in a Rat Model of Osteoarthritis.

BACKGROUND: This study aimed to evaluate the effect of chitin nanofibers (CNF) produced from crab shells as a medical material for the knee in an osteoarthritic rat model. METHODS: The effect of intra-articular CNF injection was evaluated histologically among three groups: saline, hyaluronic acid (HA), and CNF injection groups. The Osteoarthritis Research Society International (OARSI) cartilage, subchondral bone, synovial, and meniscus scores were used for scoring. RESULTS: At 4 weeks, the CNF group had significantly lower scores than the saline group. The Synovial score was lower in HA and

A septin-based hierarchy of proteins required for localized deposition of chitin in the Saccharomyces cerevisiae cell wall.

Just before bud emergence, a Saccharomyces cerevisiae cell forms a ring of chitin in its cell wall; this ring remains at the base of the bud as the bud grows and ultimately forms part of the bud scar marking the division site on the mother cell. The chitin ring seems to be formed largely or entirely by chitin synthase III, one of the three known chitin synthases in S. cerevisiae. The chitin ring does not form normally in temperature-sensitive mutants defective in any of four septins, a family of proteins that are constituents of the "neck filaments" that lie immediately subjacent to the plasma

Tuning the micropore/mesopore ratio of chitin-derived hierarchically porous carbon for high-performance lithium-selenium batteries.

Lithium-selenium (Li-Se) batteries are considered a promising energy storage alternative, mainly because of the high theoretical volumetric capacity of selenium cathodes. However, the polyselenide shuttle effect, large volume expansion and slow diffusion kinetics during cycling continue to impede their practical application. Herein, we report an enhancement of the Li+ diffusion kinetics via tuning the micropore/mesopore ratio of the chitin-derived hierarchically porous carbon for Li-Se batteries. In this structure, confining Se in the hierarchically porous carbon matrix leads to the direct for

Ion Desolvation and Electric Double-Layer Formation in Confined Ultramicropores of Activation-Free Chitin-Derived Porous Carbons.

Understanding ion transport and charge storage in confined carbon nanopores is important for the design of electric double-layer capacitors (EDLCs), but the relationship between ultramicropores, ion desolvation, and practical rate performance remains difficult to clarify. Here, chitin-derived porous carbons (CDPCs) were prepared by activation-free carbonization at 800-900 °C and evaluated as model nanoporous carbon electrodes in aqueous 6 M KOH. The CDPCs possessed moderate BET surface areas of 314-328 m2 g-1 determined from N2 adsorption isotherms, together with ultramicropores identified by

Designing cryogels through cryostructuring of β-chitin nanofibers to reinforce polyelectrolyte complexes for non-compressible hemorrhage control.

Non-compressible hemorrhage remains a major clinical challenge, requiring hemostatic materials with rapid blood absorption and sufficient mechanical robustness. Conventional carboxymethyl chitosan (CMCS)/sodium alginate (SA) polyelectrolyte complex (PEC) sponges exhibit high swelling capacity but limited uptake kinetics and mechanical strength. Here, β-chitin nanofibers (β-ChNFs) were incorporated into the CMCS/SA system to establish a freeze-thaw reinforcement strategy depended on β-ChNFs. Repeated freeze-thaw cycles had little effect on the storage modulus (G') of CMCS/SA alone, whereas G' p

Reactive Oxygen Species Drive the Aberrant Immune Response to a C. neoformans Chitin Synthase 3 (chs3Δ) Mutant.

Cryptococcus neoformans is a globally distributed fungal pathogen that causes severe opportunistic infections, particularly in individuals with impaired cell-mediated immunity, such as AIDS patients and organ transplant recipients. Chitosan, a deacetylated derivative of chitin, plays a key role in modulating immune responses during Cryptococcus neoformans infection. We previously showed that mice inoculated with a heat-killed (HK) chitosan-deficient C. neoformans strain lacking chitin synthase 3 (chs3Δ) undergo rapid, neutrophil-dependent mortality within 36 hours, despite the absence of viabl

Acute hypoxia stress impairs Litopenaeus vannamei resistance to white spot syndrome virus by downregulating chitin-binding proteins.

Hypoxia stress is a key environmental stressor that can induce white spot syndrome virus (WSSV) outbreaks, but underlying mechanisms remain unclear. Here, transcriptome analysis and RT-PCR were conducted to investigate the impact of acute hypoxia stress on gene expression profiles in shrimp. We revealed that acute hypoxia stress significantly altered the expression profile of genes in shrimp gill, particularly downregulating genes encoding chitin-binding proteins (CBPs), including Mucin-19, GAAP, CP1114, SgAbd-1, pro-Resilin, Obstructor-E, CP, GP1, and AMP1A. Downregulation of Mucin-19, GAAP,

Molecular architecture of chitin and chitosan-dominated cell walls in zygomycetous fungal pathogens by solid-state NMR.

Zygomycetous fungal infections pose an emerging medical threat among individuals with compromised immunity and metabolic abnormalities. Our pathophysiological understanding of these infections, particularly the role of fungal cell walls in growth and immune response, remains limited. Here we conducted multidimensional solid-state NMR analysis to examine cell walls in five Mucorales species, including key mucormycosis causative agents like Rhizopus and Mucor species. We show that the rigid core of the cell wall primarily comprises highly polymorphic chitin and chitosan, with minimal quantities

Design, synthesis and biological evaluation of novel spiro-quinazolinone derivatives as chitin synthase inhibitors and antifungal agents.

A series of spiro-quinazolinone scaffolds were constructed based on the bioactivity of quinazolinone and the inherent feature of spirocycle to design novel chitin synthase inhibitors that possess mode of action different from that of the currently used antifungal agents. Among them, the spiro[thiophen-quinazolin]-one derivatives containing α, β-unsaturated carbonyl fragments had shown inhibitory activities against chitin synthase and antifungal activities. The enzymatic experiments showed that among the sixteen compounds, compounds 12d, 12g, 12j, 12l and 12m exhibited inhibitions against chiti

Morphological changes induced by class III chitin synthase gene silencing could enhance penicillin production of Penicillium chrysogenum.

Chitin synthases catalyze the formation of β-(1,4)-glycosidic bonds between N-acetylglucosamine residues to form the unbranched polysaccharide chitin, which is the major component of cell walls in most filamentous fungi. Several studies have shown that chitin synthases are structurally and functionally divergent and play crucial roles in the growth and morphogenesis of the genus Aspergillus although little research on this topic has been done in Penicillium chrysogenum. We used BLAST to find the genes encoding chitin synthases in P. chrysogenum related to chitin synthase genes in Aspergillus n

Acute high carbonate alkalinity alters ion regulatory functions, chitin turnover, and intestinal microbial communities in Penaeus monodon.

Carbonate alkalinity is an important environmental stressor in saline-alkali aquaculture. However, its comprehensive effects on the physiological regulation, transcriptional responses, and intestinal microbial homeostasis of Penaeus monodon remain unclear. In this study, shrimp were exposed to high carbonate alkalinity stress for 96 h. Key physiological indicators were evaluated, and gill transcriptome analysis and intestinal 16S rRNA gene sequencing were performed. The results showed that high alkalinity reduced the survival rate of P. monodon, increased hemolymph osmotic pressure and carboni

An anthocyanin-grafted chitin film with enhanced stability prepared via Mannich multicomponent reaction for food freshness monitoring.

Anthocyanins are widely used as natural indicators for food freshness monitoring. However, their practical applications are limited by insufficient stability. In this study, a sustainable chitin-based intelligent indicator film was developed by covalently grafting anthocyanins onto chitin matrix via an in situ Mannich multicomponent reaction. The grafting behavior and structural properties of the resulting chitin-anthocyanin (Ch-A7) film were systematically investigated. The results demonstrated that anthocyanins were successfully immobilized onto the chitin matrix, and the apparent grafting d

Porous chitin nanofibers combined with adipose-derived stem cells promote diabetic wound healing via PPAR-γ activation.

Chronic wounds of diabetes are one of the serious complications that cause disability or death in patients with diabetes, posing a significant clinical challenge. Therefore, porous chitin nanofibers (CR) were prepared from silk and squid cartilage, and combined with adipose-derived stem cells (ADSCs) transplantation to promote wound healing in type 2 diabetes mellitus (T2DM) in this study. The results of scanning electron microscopy (SEM), fourier transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD) indicated that the CR had an obvious three-dimensional porous structure. Rnai-mediated knockdown of nuclear receptors impairs cuticle formation by disrupting chitin metabolic pathway in Holotrichia oblita.

Nuclear receptors (NRs) are ligand-regulated transcription factors that play crucial roles in insect growth and development. However, their functions in Holotrichia oblita Faldermann remain uncharacterized. In this study, we identified and characterized three HoNRs from H. oblita. Spatiotemporal expression analysis revealed that all three HoNRs were predominantly expressed in eggs and first-instar larvae. Tissue localization showed high expression in the epidermis, fat body, and rectum. RNA interference (RNAi) experiments demonstrated that silencing these HoNRs reduced larval feeding, impaired

Effect of chitin-architected spatiotemporal three-dimensional culture microenvironments on human umbilical cord-derived mesenchymal stem cells.

Mesenchymal stem cell (MSC) transplantation has been explored for the clinical treatment of various diseases. However, the current two-dimensional (2D) culture method lacks a natural spatial microenvironment in vitro. This limitation restricts the stable establishment and adaptive maintenance of MSC stemness. Using natural polymers with biocompatibility for constructing stereoscopic MSC microenvironments may have significant application potential. This study used chitin-based nanoscaffolds to establish a novel MSC three-dimensional (3D) culture. We compared 2D and 3D cultured human umbilical c

Rational design of silver NPs-incorporated quaternized chitin nanomicelle with combinational antibacterial capability for infected wound healing.

Bacterial and biofilm infections are prevalent, photothermal antibacterial therapy exploiting Ag NPs was an alternative. However, various matrix materials including polysaccharides used to stabilize Ag NPs are not efficiently utilized. In this study, catechol functionalized quaternized chitin (DQC) is first synthesized, then Ag+ is in situ reduced to small Ag NPs stabilized and well-dispersed by DQC to form Ag NPs-incorporated quaternized chitin (DQCA) nanomicelle in a green and simple way. The photothermal conversion efficiency of the DQCA was up to be 65 %, which was much higher than that of

RNAi for chitin synthase 1 rather than 2 causes growth delay and molting defect in Henosepilachna vigintioctopunctata.

Chitin synthase (CHS) plays a critical role in chitin synthesis and excretion. In most insects, CHSs have been segregated into 1 and 2 classes. CHS1 is responsible for chitin production in the ectodermally-derived epidermal cells. CHS2 is dedicated to chitin biosynthesis in the midgut peritrophic matrix (PM). Henosepilachna vigintioctopunctata is a serious pest of Solanaceae and Cucurbitaceae plants. In this study, we identified HvCHS1 and HvCHS2. We found that HvCHS1 was abundantly transcribed in the larval tracheae and epidermis, whereas HvCHS2 was mainly expressed in the guts. Escherichia c

Extraction of Chitin From Shrimp Shell by Successive Two-Step Fermentation of Exiguobacterium profundum and Lactobacillus acidophilus.

As an environmentally friendly and efficient method, successive two-step fermentation has been applied for extracting chitin from shrimp shells. To screen out the microorganisms for fermentation, a protease-producing strain, Exiguobacterium profundum, and a lactic acid-producing strain, Lactobacillus acidophilus, were isolated from the traditional fermented shrimp paste. Chitin was extracted by successive two-step fermentation with these two strains, and 85.9 ± 1.2% of protein and 95 ± 3% of minerals were removed. The recovery and yield of chitin were 47.82 and 16.32%, respectively. Fourier tr
