PpAPRR2 regulates chlorophyll biosynthesis and chloroplast development during early fruit development in peach.

PpAPRR2 promotes chlorophyll accumulation in peach fruit, and its overexpression enhances fruitgreening in tomato. Fruit peel color represents an important quality trait in peach (Prunus persica L.), determined by the dynamic balance between chlorophyll and anthocyanin metabolism. While the regulatory networks controlling anthocyanin regulation have been well documented, the molecular mechanisms governing chlorophyll accumulation and peel greening remain less understood. Here, we identified nine ARR-B family transcription factors in peach and found that PpAPRR2 was predominantly expressed duri

The complete chloroplast genome of Lamium purpureum L. var. purpureum (Lamiaceae) and phylogenetic analysis.

Lamium purpureum var. purpureum, the autonym of L. purpureum, is one of five infraspecific taxa currently recognized within the species. In this study, we report the complete chloroplast genome of Lamium purpureum var. purpureum (150,474 bp) with a typical quadripartite structure. A total of 116 unique genes were annotated, comprising 79 protein-coding genes, 33 transfer RNA genes, and four ribosomal RNA genes. Phylogenetic analysis recovered the subfamily Lamioideae and the genus Lamium as monophyletic with strong bootstrap support (100%). Within Lamium, L. purpureum var. purpureum was resolv

Chloroplast Protein Degradation Pathways During Leaf Senescence: Coordination and Roles in Nitrogen Remobilization.

Leaf senescence is a highly programmed developmental process that enables nutrient remobilization from source to sink organs in plants. Chloroplasts serve as the primary nitrogen reservoir, and the timely degradation of their constituent proteins is essential for sustaining seed fill and crop yield. For efficient nitrogen recycling, the chloroplast proteome must be dismantled in a controlled manner through coordinated proteolytic pathways that function both inside and outside the organelle. Protein abundances are progressively reduced during senescence through the balanced action of degradatio

Cross talk between plant photoreceptor signaling and chloroplast signaling: implications for physiological regulation.

This review focuses on the bidirectional integrated network formed by light signaling and photosynthesis, providing insights for research aimed at optimizing plant energy balance, resource allocation, and stress resilience to facilitate crop improvement. Photoreceptor-mediated light signaling constitutes the core pathway through which plants perceive light quality, intensity, and duration to precisely modulate growth and development. Crucially, this signaling network extensively interacts with and engages in cross talk with the photosynthetic system, forming a sophisticated regulatory circuit.

Chloroplast-mitochondria synergy modulates responses to iron limitation in two Thalassiosira diatom species.

Iron is naturally present at low levels in modern oceans, where it is essential for life. Ocean-dwelling organisms such as oceanic phytoplankton must therefore adapt to the available levels. Among phytoplankton, diatoms are a highly diverse and successful taxon that includes the Thalassiosira genus. As a group, diatoms contribute around 20% of global primary photosynthetic productivity; they have also developed specific resources allowing them to thrive in low-iron regions. However, the major biological factors underlying their success in these ocean environments remain unknown. Here, we compa

How Did Thylakoids Emerge in Cyanobacteria, and How Were the Primary Chloroplast and Chromatophore Acquired?

The emergence of thylakoid membranes in cyanobacteria is a key event in the evolution of all oxygenic photosynthetic cells, from prokaryotes to eukaryotes. Recent analyses show that they could originate from a unique lipid phase transition rather than from a supposed vesicular budding mechanism. Emergence of thylakoids coincided with the great oxygenation event, more than two billion years ago. The acquisition of semi-autonomous organelles, such as the mitochondrion, the chloroplast, and, more recently, the chromatophore, is a critical step in the evolution of eukaryotes. They resulted from pr

Cucumber NUCLEAR FACTOR-YC2/-YC9 target translocon component CsTIC21 in chloroplast photomorphogenesis.

Light signals promote photomorphogenesis and photosynthesis, allowing plants to establish photoautotrophic growth. Chloroplasts are organelles responsible for photosynthesis in which light energy is converted into chemical energy and stored as organic matter. However, how light regulates chloroplast photomorphogenesis remains unclear. Here, we isolated a cucumber (Cucumis sativus L.) mutant albino seedling (as) from an ethyl methane sulfonate mutagenesis library with an albino phenotype. Map-based cloning revealed that the mutation occurred in a component of cucumber translocon at the inner me

The complete chloroplast genome of Bupleurum longicaule var. strictum, an annual herb endemic to China.

Bupleurum species are well-known for their antipyretic, analgesic, gallbladder and other functions so that they are very popular in traditional Chinese medicine. However, to our knowledge, there is no completed chloroplast genome of Bupleurum genus in China. In the present study, we determined the complete chloroplast genome sequences for Bupleurum longicaule var. strictum using IIumina sequencing. Bupleurum longicaule var. strictum is 155,578 bp which is composed of two inverted repeats (IR: 26,257bp), a large single copy region (LSC: 86,977 bp), and a small single-copy region (SSC: 16,087 bp

Cold Stress Activates the Expression of Genes of the Chloroplast Transcription Apparatus in Arabidopsis thaliana Plants.

The physiological and molecular responses of Arabidopsis thaliana plants to cold stress were studied. Exposure to a low non-freezing temperature (4°C, 5 days) caused a decrease in the physiological functions and activity of a number of photosynthetic genes and elevation in expression of the cold stress gene COR15a, the product of which protects chloroplasts. It was shown for the first time that in parallel to a general inhibition of physiological functions under hypothermia, an increase in the expression of most genes for the chloroplast transcription apparatus was observed. This is obviously

Development of species-specific chloroplast markers for the authentication of Gynostemma pentaphyllum and their distribution in the Korean peninsula.

Gynostemma pentaphyllum is a traditional oriental medicinal herb used as tea since ancient time. Among Gynostemma species, G. pentaphyllum has more active chemical components and better therapeutic effect. It is used to cure depression, diabetes, anxiety, hyperlipidemia, fatigue, immunity, cancer, and oxidative stress. Overexploitation of G. pentaphyllum for its medicinal benefits has been on a rise, due to which they are adulterated or mistakenly identified with other members of Gynostemma species. Hence, we used chloroplast universal regions such as ycf3, accD, petD, psbB and their polymorph

A variation on chloroplast development: the bizonoplast and photosynthetic efficiency in the deep-shade plant Selaginella erythropus.

UNLABELLED: • PREMISE OF THE STUDY: Chloroplast development and structure are highly conserved in vascular plants, but the bizonoplast of Selaginella is a notable exception. In the shade plant S. erythropus, each dorsal epidermal cell contains one bizonoplast, while other cells have normal chloroplasts. Our quest was to (1) determine the origin of bizonoplasts, (2) explore developmental plasticity, and (3) correlate developmental changes with photosynthetic activity to provide insights unavailable in other green plants with more constrained development.• METHODS: Bizonoplast development was

Algal chloroplast produced camelid VH H antitoxins are capable of neutralizing botulinum neurotoxin.

We have produced three antitoxins consisting of the variable domains of camelid heavy chain-only antibodies (VH H) by expressing the genes in the chloroplast of green algae. These antitoxins accumulate as soluble proteins capable of binding and neutralizing botulinum neurotoxin. Furthermore, they accumulate at up to 5% total soluble protein, sufficient expression to easily produce these antitoxins at scale from algae. The genes for the three different antitoxins were transformed into Chlamydomonas reinhardtii chloroplasts and their products purified from algae lysates and assayed for in vitro

Biochemical characterization of the chloroplastic β-carbonic anhydrase from Flaveria bidentis (L.) "Kuntze".

C3 and C4 plant carbonic anhydrases (CAs) are zinc-enzymes that catalyze the reversible hydration of CO2. They are sub-divided in three classes: α, β and γ, being distributed between both photosynthetic subtypes. The C4 dicotyledon species Flaveria bidentis (L.) "Kuntze" contains a small gene family encoding three distinct β-CAs, named FbiCA1, FbiCA2 and FbiCA3. We have expressed and purified recombinant FbiCA1, which is localized in the chloroplast where it is thought to play a role in lipid biosynthesis and antioxidant activity, and biochemically characterized it by spectroscopic and inhibit

The complete chloroplast genome of banana (Musa acuminata, Zingiberales): insight into plastid monocotyledon evolution.

BACKGROUND: Banana (genus Musa) is a crop of major economic importance worldwide. It is a monocotyledonous member of the Zingiberales, a sister group of the widely studied Poales. Most cultivated bananas are natural Musa inter-(sub-)specific triploid hybrids. A Musa acuminata reference nuclear genome sequence was recently produced based on sequencing of genomic DNA enriched in nucleus. METHODOLOGY/PRINCIPAL FINDINGS: The Musa acuminata chloroplast genome was assembled with chloroplast reads extracted from whole-genome-shotgun sequence data. The Musa chloroplast genome is a circular molecule o

Role of a disulfide bond in the gamma subunit in activation of the ATPase of chloroplast coupling factor 1.

The relationship between activation of the latent ATPase activity of isolated chloroplast coupling factor 1 (CF1) and reduction of a disulfide in the gamma subunit has been assessed. The sulfhydryl residues involved in the disulfide bond are distinct from residues normally accessible to maleimide modification during incubation of thylakoids in the dark or the light. Dithiothreitol-induced activation is time dependent, and correlates with reduction of the disulfide. Sulfhydryl residues exposed during activation can be reoxidized to disulfide by incubation with iodosobenzoate , with a concomitan

Leaf senescence in a non-yellowing mutant of Festuca pratensis : I. Chloroplast membrane polypeptides.

Soluble and thylakoid membrane polypeptides from senescing leaf tissue of Rossa, a normal yellowing Festuca pratensis genotype, were fractionated by sodium dodecyl sulphate polyacrylamide gel electrophoresis and compared with those of the non-yellowing mutant Bf 993. Subunits of ribulose-1,5-bisphosphate carboxylase were the major soluble polypeptides and declined to low levels in senescing leaves of both genotypes. The major thylakoid polypeptides were those associated with the chlorophyllprotein complexes CPI and CPII. The levels of all thylakoid polypeptide species fell during senescence of

[Purification and characterization of chloroplast carbonate dehydratase from leaves of Lactuca sativa (author's transl)].

Two isoenzymes of carbonate dehydratase were identified in green leaf tissue of Lactuca sativa. Their molecular weights were found to be 195 000 and 250 000. The lighter isoenzyme (I) was further characterized. It is localized in the chloroplast fraction. With polyacrylamide gel electrophoresis in the presence of dodecyl sulphate, subunits with a molecular weight of 34 000 and higher aggregates of this size could be detected. This is interpreted as an indication of an hexameric enzyme structure. The 900-fold purified polymer contained 5 - 6 atoms of zinc. Amino acid composition and inhibition

Cold chain and virus-free oral polio booster vaccine made in lettuce chloroplasts confers protection against all three poliovirus serotypes.

To prevent vaccine-associated paralytic poliomyelitis, WHO recommended withdrawal of Oral Polio Vaccine (Serotype-2) and a single dose of Inactivated Poliovirus Vaccine (IPV). IPV however is expensive, requires cold chain, injections and offers limited intestinal mucosal immunity, essential to prevent polio reinfection in countries with open sewer system. To date, there is no virus-free and cold chain-free polio vaccine capable of inducing robust mucosal immunity. We report here a novel low-cost, cold chain/poliovirus-free, booster vaccine using poliovirus capsid protein (VP1, conserved in all

Chlorophyll luminescence as an indicator of stress-induced damage to the photosynthetic apparatus. Effects of heat-stress in isolated chloroplasts.

A brief review is given of investigations on stres-induced alterations of ms-to s-luminescence yield of chlorophyll in plants. Three different approaches are considered: phytoluminography, luminescence-temperature curves, and luminescence induction curves. The remainder of this article presents new results of the effect of heat stress on luminescence induction curves of isolated chloroplasts. Three parameters with widely different heat resistances were resolved from induction curves. A fast valinomycin sensitive transient, L'i, with a 50% inhibition temperature of 33 to 34°C was correlated wit
