Hippocampal neuronal atrophy and cognitive function in delayed poststroke and aging-related dementias.

BACKGROUND AND PURPOSE: We have previously shown delayed poststroke dementia in elderly (≥75 years old) stroke survivors is associated with medial temporal lobe atrophy; however, the basis of the structural and functional changes is unknown. METHODS: Using 3-dimensional stereological methods, we quantified hippocampal pyramidal neuronal volumes and densities in a total of 95 postmortem samples from demented and nondemented poststroke survivors within our prospective Cognitive Function after Stroke study and subjects pathologically diagnosed with vascular dementia, Alzheimer disease, and mixed

Cognitive aging, executive function, and fractional anisotropy: a diffusion tensor MR imaging study.

BACKGROUND AND PURPOSE: Fractional anisotropy (FA) is a useful measure of connectivity in the brain that can be derived from the diffusion tensor imaging (DTI) dataset. This study investigated the relationship between FA and selected measures of cognition across a broad age group to explore a possible structural basis for cognitive changes with age. METHODS: FA images were generated from DTI data acquired at 1.5T in 87 healthy subjects (age range, 20-73 years). Relationships between a range of cognitive measures and FA were explored using regional and voxel-based analysis. RESULTS: Age and r

Effects of motor-cognitive interaction based on dual-task gait analysis recognition in middle age to aging people with normal cognition and mild cognitive impairment.

BACKGROUND: Mild cognitive impairment (MCI) is considered a transitional stage between cognitive normality and dementia among the elderly, and its associated risk of developing Alzheimer's disease (AD) is 10-15 times higher than that of the general population. MCI is an important threshold for the prevention and control of AD, and intervention in the MCI stage may be the most effective strategy to delay the occurrence of AD. MATERIALS AND METHODS: In this study, 68 subjects who met the inclusion criteria were divided into an MCI group (38 subjects) and normal elderly (NE) group (30 subjects).

[Immune dysfunction and cognitive deficit in stress and physiological aging. Part II: New approaches to cognitive disorder prevention and treatment ].

Long-term stress as well as physiological aging result in similar immunological and hormonal disturbances including hypothalamic-pituitary-adrenal) axis depletion, aberrant immune response (regulatory T-cells, Tregs, and T(h17)-lymphocyte accumulation) and decreased dehydroepian-drosterone synthesis both in the brain and in the adrenal glands. Since the main mechanisms of inflammation control, "prompt" (stress hormones) and "delayed" (Tregs), are broken, serum cytokine levels increase and become sufficient for blood-brain-barrier disruption. As a result peripheral cytokines penetrate into the

Cognitive health on the fingertips: Fine motor deficiencies for assessing the risk of mild cognitive impairment in the aging adults.

OBJECTIVE: Mild cognitive impairment (MCI) represents a critical prodromal stage associated with Alzheimer's disease and other age-related neurocognitive conditions in middle-old aging adults, yet research exploring its association with fine motor skill deficits remains relatively limited. This study explores the specific characteristics of fine motor function in middle-old aging adults with MCI using the latest wearable inertial motion capture technology. METHOD: A computerized system with wearable inertial motion capture technology captured and assessed fine motions. The least absolute shri

Neurovascular mechanisms of cognitive aging: Sex-related differences in the average progression of arteriosclerosis, white matter atrophy, and cognitive decline.

Arterial stiffness (arteriosclerosis) has been linked to heightened risks for cognitive decline, and ultimately for Alzheimer's disease and other forms of dementia. Importantly, neurovascular outcomes generally vary according to one's biological sex. Here, capitalizing on a large sample of participants with neuroimaging and behavioral data (N = 203, age range = 18-87 years), we aimed to provide support for a hierarchical model of neurocognitive aging, which links age-related declines in cerebrovascular health to the rate of cognitive decline via a series of intervening variables, such as white

Greater cognitive reserve is related to lower cortical excitability in healthy cognitive aging, but not in early clinical Alzheimer's disease.

OBJECTIVE: To investigate the relationship between cortico-motor excitability and cognitive reserve (CR) in cognitively unimpaired older adults (CU) and in older adults with mild cognitive impairment or mild dementia due to Alzheimer's disease (AD). METHODS: Data were collected and analyzed from 15 CU and 24 amyloid-positive AD participants aged 50-90 years. A cognitive reserve questionnaire score (CRQ) assessed education, occupation, leisure activities, physical activities, and social engagement. Cortical excitability was quantified as the average amplitude of motor evoked potentials (MEP am

Aging, Cognition, and Depression: Unraveling the Psychological Pathways of Self-Perceptions of Aging.

BACKGROUND: Negative self-perceptions of aging (SPA) have been identified as an important risk factor for depression among older adults. However, the mechanisms underlying this association remain unclear. This study examined whether cognitive function mediates the relationship between negative SPA and depression and whether socioeconomic status (SES) moderates this pathway. METHODS: Data were obtained from 2583 Chinese older adults (Mage = 81.45 years, SD = 9.14) who participated in the 2011, 2014, and 2018 waves of the Chinese Longitudinal Healthy Longevity Survey (CLHLS). Brain age gap, dementia risk factors and cognition in middle age.

Brain Age Gap has been associated with dementia in old age. Less is known relating brain age gap to dementia risk-factors or cognitive performance in middle-age. Cognitively healthy, middle-aged subjects from PREVENT-Dementia had comprehensive neuropsychological, neuroimaging and genetic assessments. Brain Ages were predicted from T1-weighted 3T MRI scans. Cognition was assessed using the COGNITO computerized test battery. 552 middle-aged participants (median [interquartile range] age 52.8 [8.7] years, 60.0% female) had baseline data, of whom 95 had amyloid PET data. Brain age gap in middle-ag

Structural brain indicators of cognitive performance in middle and late adulthood: The Human Connectome Project in Aging/Aging Adult Brain Connectome cohort.

Although cortical thinning and atrophy are well documented in aging and neurodegenerative disease, the cortical structural features associated with preserved or superior cognition across typical adulthood remain incompletely characterized. We examined associations between cortical thickness and cognitive performance in a large, typically aging cohort and tested whether they vary by age and cognitive domain. We analyzed cross-sectional data from cognitively healthy adults in the Human Connectome Project-Aging/Aging Adult Brain Connectome, spanning middle (36-59 years), early older (60-79), and

Bilingualism and cognitive aging in ethnically diverse older adults: cross-sectional and longitudinal evidence from Health and Aging Brain Study-Health Disparities.

OBJECTIVES: Identifying modifiable factors that shape cognitive aging trajectories is a central goal in aging research. This study examined whether bilingualism is associated with dementia status and longitudinal cognitive decline, and whether associations vary by ethnicity or gender. METHODS: We analyzed Wave 7 data from the Health and Aging Brain Study-Health Disparities (HABS-HD), a longitudinal U.S. cohort of adults aged ≥50 years. Bilingualism was defined by self-reported use of a second language. Cross-sectional analyses included 4,140 participants (1,013 bilinguals); longitudinal analy

Cognitive Aging in Same- and Different-Sex Relationships: Comparing Age of Diagnosis and Rate of Cognitive Decline in the Health and Retirement Study.

INTRODUCTION: The ongoing marginalization of lesbian, gay, and bisexual people has been hypothesized to produce poorer late-in-life cognitive outcomes, according to mechanisms posited by minority stress and allostatic load theories. Yet the existence of those outcomes remains understudied, and results of existing studies have been contradictory. Using a population-based longitudinal aging study, this paper will compare age at diagnosis of Alzheimer's disease (AD) or a related dementia and rates of cognitive decline between participants in same-sex relationships (SSRs) and different-sex relatio

Combining Cognitive and Sensorimotor Assessments to Facilitate Detection of Age-Related Cognitive Decline.

Early detection of cognitive decline is critical for reducing progression of neurodegenerative diseases. Bioimaging- and biofluids- based approaches are invasive and expensive; clinical screening tools have difficulties in discerning subtle cognitive changes. Eye tracking measures have been used to detect cognitive deficits effectively and reduction in sensorimotor performance may precede cognitive declines by several years. Therefore, we developed a combined cognitive and sensorimotor evaluation system by integrating an arm-hand exoskeleton and an eye-tracking device to facilitate the early d

Sex Differences in Dementia, Cognition, and Health in the Cognitive Function and Ageing Studies (CFAS).

BACKGROUND: There is renewed interest in whether sex differences in dementia risk exist, and what influence social and biological factors have. OBJECTIVE: To review evidence from the Cognitive Function and Ageing Studies (CFAS), a multi-center population-representative cohort study in the UK; focusing on dementia and cognition, incorporating findings on participants' health and social circumstances. METHODS: After identifying all CFAS publications, the results of all sex-stratified primary analyses of CFAS data were narratively reviewed. RESULTS: Of 337 publications, 94 report results by se

The interplay of life satisfaction and cognitive reserve: implications for cognitive changes in old age.

BACKGROUND: Life satisfaction (LS), a widely used measure of psychological well-being, has been previously linked with cognitive decline, although sex-differences, as well as the interplay of LS and other established correlates of cognitive health – particularly cognitive reserve (CR) – remain unknown. Thus, we explored the sex-specific interaction between LS an CR for cognitive change over 15 years during aging. METHOD: For this population-based longitudinal cohort study, data from the Swedish National Study on Aging and Care in Kungsholmen (SNAC-K) were used. The sample included 1871 cogniti

Cognitive trajectory in mild cognitive impairment due to primary age-related tauopathy.

Primary age-related tauopathy is increasingly recognized as a separate neuropathological entity different from Alzheimer's disease. Both share the neuropathological features of tau aggregates and neuronal loss in the temporal lobe, but primary age-related tauopathy lacks the requisite amyloid plaques central to Alzheimer's disease. While both have similar clinical presentations, individuals with symptomatic primary age-related tauopathy are commonly of more advanced ages with milder cognitive dysfunction. Direct comparison of the neuropsychological trajectories of primary age-related tauopathy

Understanding cognitive ageing: individuals, institutions, and inequalities.

Population ageing has drawn growing attention to cognitive ageing as a major public health and social policy issue. However, cognitive ageing is marked by substantial heterogeneity, reflecting not only biological and genetic processes but also the cumulative influence of social, economic, and institutional conditions across the life course. This editorial introduces a Special Issue that advances a social determinants perspective on cognitive ageing by integrating life-course, inequality, and contextual approaches across diverse geographical settings. Drawing on longitudinal surveys, administra
