Gene transfer using micellar nanovectors inhibits corneal neovascularization in vivo.

PURPOSE: The efficacy of gene therapy using nonviral vector based on polyplex micelle has been studied against corneal neovascularization in mice. METHODS: A block copolymer, poly(ethylene glycol) (PEG)-block-polycation carrying ethylenediamine units in the side chain (PEG-b-P[Asp(DET)]), was prepared. PEG-b-P[Asp(DET)] formed a polyplex micelle through the polyion complex formation with plasmid DNA. To evaluate in vivo gene transfer efficiency, PEG-b-P[Asp(DET)] micelle was injected into the subconjunctival space of mice, and the expression of the reporter gene was assessed. Furthermore, mou

Deletion of the FHL2 gene attenuating neovascularization after corneal injury.

PURPOSE: The four-and-one-half LIM domain-containing protein2 (FHL2) is a member of the four-and-a-half LIM domain-only (FHL) gene family. Although FHL2 is expressed in the cornea, its role in angiogenesis is unclear. The aim of this study was to investigate the role of the FHL2 gene in corneal angiogenesis after chemical injury. METHODS: FHL2-LacZ knock-in mice were used to trace FHL2 gene expression before and after corneal injury. Corneal angiogenesis between FHL2-null mice and wild-type mice that underwent chemical and mechanical denudation of corneal and limbal epithelium were compared.

Topical and subcutaneous alpha-interferon fails to suppress corneal neovascularization.

Corneal neovascularization is a potentially blinding complication of a variety of corneal disorders. alpha-Interferon has been shown to inhibit endothelial cell migration and proliferation. It has been used successfully in the treatment of pediatric pulmonary hemangioma and hairy cell leukemia. This study was undertaken to evaluate the effect of topical and subcutaneous (s.c.) alpha-interferon on corneal neovascularization. Corneal neovascularization was induced in 40 male New Zealand white rabbits by placing silk sutures (7.0) bilaterally in each rabbit eye at the 3 and 9 o'clock positions of

[Morphological study of experimental corneal neovascularization after anterior uveal ischemia].

We morphologically investigated what kinds of inflammatory cells infiltrate the corneoscleral limbus by light and electron microscopy and what manner of keratocytes and vascular endothelial cells appear at the corneal limbus, when the corneal edema was aggravated severely and clinical corneal new vessels were found initially, i.e. until three days after anterior segmental ischemia in rabbit eyes. The majority of infiltrating inflammatory cells were polymorphonuclear leukocytes and the others were lymphocytes and histiocytes etc. Peripheral keratocytes around limbal vessels were stimulated and

[Corneal neovascularization following short-term wear of soft contact lenses].

Between 1980 and 1982 the author treated 9 patients suffering from severe, partly irreversible bilateral damage to the cornea resulting from the use of soft contact lenses. After three months to two years the patients complained of a foreign body sensation, dryness and of a substantial reduction in visual acuity. The high degree of comfort experienced initially encourages patients to wear soft lenses for excessively long periods of time and this can lead to severe complications: while at first there was only slight capillary proliferation at the limbus, superficial and deep vascular proliferat

Rapamycin inhibits corneal inflammatory response and neovascularization in a mouse model of corneal alkali burn.

Alkali burn-induced corneal injury often causes inflammation and neovascularization and leads to compromised vision. We previously reported that rapamycin ameliorated corneal injury after alkali burns by methylation modification. In this study, we aimed to investigate the rapamycin-medicated mechanism against corneal inflammation and neovascularization. Our data showed that alkali burn could induce a range of different inflammatory response, including a stark upregulation of pro-inflammatory factor expression and an increase in the infiltration of myeloperoxidase- and F4/80-positive cells from

Biomicroscopic and histopathologic observations after corneal laser photocoagulation in a rabbit model of corneal neovascularization.

Corneal neovascularization complicates many anterior segment diseases. Corneal laser photocoagulation using yellow light (577 nm) has been shown to reduce corneal neovascularization. No histopathologic studies of the effects of this treatment on the eye have been reported, however. Target (cornea) and nontarget (iris, lens, retina, and choroid) ocular tissue were studied 1, 24, and 48 h and 5 days after yellow dye corneal laser photocoagulation in a rabbit model of corneal neovascularization. Biomicroscopic examination of the corneas revealed intracorneal hemorrhage in five of 24 (21%) eyes of

Development of Miniprotein Binders to Inhibit VEGF-Induced Corneal Neovascularization.

PURPOSE: Corneal neovascularization (CNV) disrupts corneal transparency and impairs vision and is driven primarily by vascular endothelial growth factor (VEGF-A165). Current therapies rely on off-label anti-VEGF agents, which are limited by poor corneal penetration and the need for repeated dosing. Herein, we aimed to rationally design and validate novel hyperstable VEGF-A165 miniprotein inhibitors against VEGF-A165 for preventing CNV. METHODS: VEGF-A165 miniprotein binders were generated using a de novo diffusion-based protein design protocol and expressed in Escherichia coli (E. coli). Sele

PROTAC based targeted degradation of LRG1 for mitigating corneal neovascularization.

Leucine-rich alpha-2-glycoprotein 1 (LRG1), a secretory glycoprotein associated with angiogenesis, inflammation, fibrosis, and other pivotal pathophysiological processes, is significantly upregulated in corneal neovascularization (CNV), where it drives neovascularization via the TGF-β-Smad signaling pathway, making it a potential therapeutic target for CNV. This study employs proteolysis-targeting chimera (PROTAC) technology, utilizing our newly developed PROTAC agent, ETTAC-2, to selectively degrade LRG1 in a mouse model of alkali burn-induced CNV. The cellular study revealed that ETTAC-2 eff
