Mutational analysis of inherited epilepsies.

Single-gene mutations in mice initiate specific heritable neuronal diseases featuring different patterns of epilepsy. Thirteen chromosomal loci for convulsive seizures and four loci for spike-wave seizures have been assigned. Within this genetic framework, a few general principles are emerging. Mutations located on different chromosomes may result in identical seizure patterns. Interactions between specific genes, demonstrated by the synthesis of compound heterozygotes, can express intermediate syndromes. Interactions with unknown genes in the background can mask neurological disease expressio

[Effect of cerebral hypoxia and hyperventilation hypocapnia on the epileptiform activity of the cerebral cortex of the cat].

Influence of cerebral hypoxia and hyperventilatory hypocapnia on the ECoG and focal epileptiform activity of the cerebral cortex induced with local application of strychnine was studied in cats with transection of spinal cord at C1. Although both hypoxia and hypocapnia produced synchronization of the cerebral cortex electrical activity, i.e. exerted the same effects on the ECoG, their influence on cortical excitability was quite different: hypoxia suppressed the epileptiform activity whereas hypocapnia facilitated it.

How we fall asleep: regional and temporal differences in electroencephalographic synchronization at sleep onset.

OBJECTIVES: We hypothesized that the brain shows specific and predictable patterns of spatial and temporal differences during sleep onset (SO) reflecting a temporal uncoupling of electrical activity between different cortical regions and a dissociated wakelike and sleeplike electrocortical activity in different cortical areas. METHODS: We analyzed full-scalp electroencephalographic (EEG) recordings of 40 healthy subjects to investigate spatial and temporal changes of EEG activity across the wake-sleep transition. We quantified EEG sleep recordings by a fast Fourier transform (FFT) algorithm a

Interpersonal brain synchronization during sensorimotor synchronization in people with different aerobic fitness levels: A fNIRS-based hyperscanning study.

Sensorimotor synchronization (SMS) refers to the temporal coordination of individual actions with perceptible external events and rhythms. Previous research has revealed a potential relationship between SMS abilities and physical activity, with proposed links to underlying inner-brain cognitive processes. However, it needs to be explored that whether good aerobic fitness will have a strong SMS ability, its internal mechanism, and the inter-brain mechanism. In the present study, we recruited 23 dyads of long-distance runners as the experimental group and 22 dyads of non-physical education major

Joint analysis of frontal theta synchrony and white matter following mild traumatic brain injury.

Some of the most disabling aspects of mild traumatic brain injury (mTBI) include lingering deficits in executive functioning. It is known that mTBI can damage white matter tracts, but it remains unknown how this structural brain damage translates into cognitive deficits. This experiment utilized theta band phase synchrony to identify the dysfunctional neural operations that contribute to cognitive problems following mTBI. Sub-acute stage (< 2 weeks) mTBI patients (N = 52) and healthy matched controls (N = 32) completed a control-demanding task with concurrent EEG. Structural MRI was also colle

Reduced movement-related β desynchronisation in juvenile myoclonic epilepsy: a MEG study of task specific cortical modulation.

OBJECTIVE: We investigated differences in task induced responses in occipital and sensorimotor cortex between patients with juvenile myclonic epilepsy (JME) and healthy controls . METHODS: Twelve patients with JME and 12 age-matched non-epilepsy volunteers performed visual and motor tasks during MEG. We used synthetic aperture magnetometry to localise areas of task-related oscillatory modulations, performed time-frequency analyses on the locations of peak task related power changes and compared power and frequency modulation at these locations between patients and controls. RESULTS: Patients

Sodium lactate differently alters relative EEG power and functional connectivity in Alzheimer's disease patients' brain regions.

Bilateral temporo-parietal hypoperfusion and decreased glucose metabolism are characteristic in vivo findings in Alzheimer's disease (AD). Lactate is a metabolic vasodilator and is known to induce increased cerebral blood flow in healthy adults. The present study addresses the issue whether sodium lactate infusion affects functional state and resulting electroencephalographic patterns of AD patients. Twelve late-onset sporadic AD probands participated in this self-control study. The relative power and synchronization likelihood (SL) values of the electroencephalographic samples were calculated

Robustness of Beta Desynchronization from Chronically Implanted Cortical Electrodes on Multiple Time Scales.

Adaptive deep brain stimulation (aDBS) promises a significant improvement in patient outcomes, compared to existing deep brain stimulation devices. Fully implanted systems represent the next step to the clinical adoption of aDBS. We take advantage of a unique longitudinal data set formed as part of an effort to investigate aDBS for essential tremor to verify the long term reliability of electrocorticography strips over the motor cortex as a source of bio-markers for control of adaptive stimulation. We show that beta band event related de-synchronization, a promising bio-marker for movement, is

Atypical somatosensory-motor cortical response during vowel vocalization in spasmodic dysphonia.

OBJECTIVE: Spasmodic dysphonia (SD) is a debilitating voice/speech disorder without an effective cure. To obtain a better understanding of the underlying cortical neural mechanism of the disease we analyzed electroencephalographic (EEG) signals of people with SD during voice production. METHOD: Ten SD individuals and 10 healthy volunteers produced 50 vowel vocalization epochs of 2500 ms duration. Two EEG features were derived: (1) event-related change in spectral power during vocalization relative to rest, (2) inter-regional spectral coherence. RESULTS: During early vocalization (500-1000 ms

Disturbed temporal dynamics of brain synchronization in vision loss.

Damage along the visual pathway prevents bottom-up visual input from reaching further processing stages and consequently leads to loss of vision. But perception is not a simple bottom-up process - rather it emerges from activity of widespread cortical networks which coordinate visual processing in space and time. Here we set out to study how vision loss affects activity of brain visual networks and how networks' activity is related to perception. Specifically, we focused on studying temporal patterns of brain activity. To this end, resting-state eyes-closed EEG was recorded from partially blin

Spike-wave complexes and fast components of cortically generated seizures. IV. Paroxysmal fast runs in cortical and thalamic neurons.

In the preceding papers of this series, we have analyzed the cellular patterns and synchronization of neocortical seizures occurring spontaneously or induced by electrical stimulation or cortical infusion of bicuculline under a variety of experimental conditions, including natural states of vigilance in behaving animals and acute preparations under different anesthetics. The seizures consisted of two distinct components: spike-wave (SW) or polyspike-wave (PSW) at 2-3 Hz and fast runs at 10-15 Hz. Because the thalamus is an input source and target of cortical neurons, we investigated here the s

Uniting functional network topology and oscillations in the fronto-parietal single unit network of behaving primates.

The functional communication of neurons in cortical networks underlies higher cognitive processes. Yet, little is known about the organization of the single neuron network or its relationship to the synchronization processes that are essential for its formation. Here, we show that the functional single neuron network of three fronto-parietal areas during active behavior of macaque monkeys is highly complex. The network was closely connected (small-world) and consisted of functional modules spanning these areas. Surprisingly, the importance of different neurons to the network was highly heterog

Effects of external body vibrations on cognitive performance through brain-cardio-respiratory dynamics.

External body vibrations (EBV) affect the human physiological and cognitive states. Here we construct a neurophysiological model integrating a cortical model and the cardiovascular system, exploring impacts of EBV on physiological and cognitive function. We adopted a reservoir computing model for cortical dynamics to investigate cognitive effects, focusing on binocular rivalry. This model successfully replicated changes in human perceptual alternation observed during voluntary respiratory control and cycling exercise. Our simulation indicates that EBV exerts frequency-dependent effects on perc

EEG oscillations: how are they modulated during different phases of repetitive movements?

Voluntary movements are planned through the relative timing between submovements of movement sequences as part of the motor program. Different movement phases are characterized by specific amplitude modulation of cortical oscillations. The latter represent neurophysiological correlates of specific synchronization or desynchronization of different neuronal groups. In this Neuro Forum, we review recent evidence regarding the temporal relation between neurophysiological correlates of different phases of a repetitive motor task using electroencephalography and source localization using individuali

Cutaneous Sensory Stimulation Intensity Modulates Beta-Band Event-Related Desynchronization and Synchronization Amplitudes.

Beta-band event-related desynchronization and synchronization elicited by somatosensory stimulation could be potential neurophysiological markers of motor function, specifically in neurological disorders; however, their characteristics in healthy individuals remain unclear. This study aimed to examine whether the amount of afferent input modulates beta-band oscillatory responses and if they are associated with motor performance in healthy adults. Magnetoencephalographic data were recorded for 24 healthy participants during cutaneous electrical stimulation of the right index finger at three int

Transcranial Magnetic Stimulation in Autism Spectrum Disorders: Neuropathological Underpinnings and Clinical Correlations.

Despite growing knowledge about autism spectrum disorder (ASD), research findings have not been translated into curative treatment. At present, most therapeutic interventions provide for symptomatic treatment. Outcomes of interventions are judged by subjective endpoints (eg, behavioral assessments) which alongside the highly heterogeneous nature of ASD account for wide variability in the effectiveness of treatments. Transcranial magnetic stimulation (TMS) is one of the first treatments that targets a putative core pathologic feature of autism, specifically the cortical inhibitory imbalance tha

Prestimulation phase predicts the TMS-evoked response.

Prestimulation oscillatory phase and power in particular frequency bands predict perception of at-threshold visual stimuli and of transcranial magnetic stimulation (TMS)-induced phosphenes. These effects may be due to changes in cortical excitability, such that certain ranges of power and/or phase values result in a state in which a particular brain area is more receptive to input, thereby biasing behavior. However, the effects of trial-by-trial fluctuations in phase and power of ongoing oscillations on the brain's electrical response to TMS itself have thus far not been addressed. The present

Rhythm patterns interaction--synchronization behavior for human-robot joint action.

Interactive behavior among humans is governed by the dynamics of movement synchronization in a variety of repetitive tasks. This requires the interaction partners to perform for example rhythmic limb swinging or even goal-directed arm movements. Inspired by that essential feature of human interaction, we present a novel concept and design methodology to synthesize goal-directed synchronization behavior for robotic agents in repetitive joint action tasks. The agents' tasks are described by closed movement trajectories and interpreted as limit cycles, for which instantaneous phase variables are

Altered pattern of motor cortical activation-inhibition during voluntary movements in Tourette syndrome.

In patients with Gilles de la Tourette syndrome (GTS) alterations of motor cortex (M1) excitability at rest have been evidenced. In contrast, there has so far been little research into changes of motor cortical reactivity during the time course of voluntary movements in GTS patients. The present study investigates neuromagnetic event-related desynchronization (ERD) and event-related synchronization (ERS) of bilateral M1 in 11 GTS patients and 11 healthy control subjects. ERD represents motor cortical activation, whereas ERS most likely indicates its inhibition. Subjects performed a self-paced

Effects of naloxone on ethanol and acetaldehyde-induced electroencephalographic changes in rabbits.

Rabbits treated with a single IV dose of ethanol, 0.6 g/kg, exhibited rapid EEG synchronization and behavioral changes that were reversed and, in part, prevented by the opiate antagonist, naloxone, 40 micrograms/kg. The continuous intracerebroventricular infusion of acetaldehyde, 10 or 120 micrograms/min. caused biphasic changes with EEG activation and severe bradycardia followed by overt synchrony during the post-infusion period. The results of the present study provide further evidence for the ability of naloxone to counteract some acute effects of ethanol. Moreover, they do not support a ro
