Japanese quail acute exposure to methamidophos: experimental design, lethal, sub-lethal effects and cholinesterase biochemical and histochemical expression.

We exposed the Japanese quail (Coturnix coturnix japonica) to the organophosphate methamidophos using acute oral test. Mortality and sub-lethal effects were recorded in accordance to internationally accepted protocols. In addition cholinesterases were biochemically estimated in tissues of the quail: brain, liver and plasma. Furthermore, brain, liver and duodenum cryostat sections were processed for cholinesterase histochemistry using various substrates and inhibitors. Mortalities occurred mainly in the first 1-2h following application. Sub-lethal effects, such as ataxia, ruffled feathers, trem

Day and nighttime concentrations of 5-methoxytryptophol and melatonin in the retina and pineal gland from different classes of vertebrates.

5-Methoxytryptophol (ML) and melatonin concentrations were determined by radioimmunoassay in the retinae and pineal glands of frogs (Rana esculenta), chickens (Gallus domesticus), quails (Coturnix coturnix), and European hamsters (Cricetus cricetus), killed in the afternoon (1500-1700 hr) and around midnight (2400-0200 hr). ML was only detectable in significant amounts in the frog retina. In the pineal gland ML levels showed the opposite profile to retinal ML, with measurable levels in the chicken, quail, and hamster and undetectable levels in the frog (less than 2.0 pg/gland). A significant d

Effect of photoperiod and estrogen on expression of arginine vasotocin and its oxytocic-like receptor in the shell gland of the Japanese quail.

The avian neurohypophysial hormone arginine vasotocin (AVT) is an important regulatory hormone involved in many physiological processes including fluid balance, blood pressure regulation, stress responses and reproductive events including oviposition. The mechanisms by which AVT stimulates myometrial contraction during oviposition are not well established in birds. In the present study, immunohistochemistry and in situ hybridization were used to localize AVT and the oxytocin-like VT3 receptor in the shell gland of Japanese quail (Coturnix coturnix japonica). Using an AVT-specific antibody, imm

Ultrastructural study of post-hatching development in the pineal gland of the Japanese quail.

The pineal gland of the Japanese quail (Coturnix coturnix japonica) retained numerous follicular structures, each of which was composed mostly of pinealocytes and supporting cells radially arranged around the follicular lumen during the first 6 weeks of age. In the pinealocytes, paraboloids and regular stacks of lamellar disks, which were demonstrated at 1 day of age in a previous study [14], rapidly decreased in number after 1 week of age. From 1 to 6 weeks of age, the pinealocytes had more developed lamellar whorls in the follicular lumen with the increased numbers of mitochondria in their a

Anatomical and histological insights into gastrointestinal adaptations of adult Egyptian quail.

Coturnix coturnix is widely used as a laboratory model due to its small size, ease of care, and rapid maturation. This study examined the relationship between the structure of its alimentary tract and feeding habits using anatomical, morphometric, histological, histochemical, and ultrastructural analyses. The stomach comprised a glandular proventriculus and a muscular gizzard, while the ileum was the longest intestinal segment. Paired, well-developed caeca were present at the ileocecal junction. Histologically, the alimentary canal showed the typical four layers: mucosa, submucosa, muscularis,

Treatment with an inhibitor of matrix metalloproteinase 9 or cathepsin K lengthens embryonic lower jaw bone.

OBJECTIVES: Skeletal malocclusions are common, and severe malocclusions are treated by invasive surgeries. Recently, jaw bone length has been shown to be developmentally controlled by osteoclasts. Our objective was to determine the effect of inhibiting osteoclast-secreted proteolytic enzymes on lower jaw bone length of avian embryos by pharmacologically inhibiting matrix metalloproteinase-9 (MMP9) or cathepsin K (CTSK). METHODS: Quail (Coturnix coturnix japonica) embryos were given a single dose of an inhibitor of MMP9 (iMMP9), an inhibitor CTSK (iCTSK), or vehicle at a developmental stage wh

Migratory and opportunistic wild and domestic birds, as Toxoplasma gondii carriers.

Toxoplasmosis is among the most prevalent parasitic diseases worldwide. Birds are important intermediate hosts and are regarded as good carriers or reservoirs for Toxoplasma gondii, because of their high dispersal capabilities and moreover they can act as sentinels of environmental contamination with oocysts. Birds can become infected by ingesting sporulated oocysts from contaminated feed, water, or soil, or for carnivorous birds, by ingesting tissue cysts of infected prey. On the other hand, infected birds, as frequent prey for felines, can transmit T. gondii to the definitive hosts. To the b

[Rhabdomyolysis: have you considered food poisoning from quails?].

Rhabdomyolysis (R) is a complex condition involving the rapid dissolution of damaged or injured skeletal muscle. This leads to the direct release of intracellular components, including myoglobin, creatine kinase, aldolase, and lactate dehydrogenase, as well as electrolytes, into the bloodstream and extracellular space. Clinically, R shows a triad of symptoms: myalgia, limb weakness, and myoglobinuria without hematuria, while myoglobin has been recognized as playing a part in the development of acute kidney injury. Coturnism is a relatively rare disease, mostly found in the European countries b

Experimental studies of St. Louis encephalitis virus in vertebrates.

Serologically negative birds and mammals of species, known from other studies to be exposed naturally to St. Louis encephalitis (SLE) virus in Memphis, Tennessee, and other selected species were inoculated experimentally with strains of SLE virus to determine their potential as natural hosts. Mosquitoes (Culex sp.) were allowed to feed on some of the inoculated vertebrate species, held for 14 days, and tested for SLE infection. The cardinals (Richmondena cardinalis), robins (Turdus migratorius), and baby chicks (Gallus gallus) all became viremic; 97% of the bobwhites (Colinus virginianus) and

Effect of di(n-butyl) phthalate on the blood-testis barrier during puberty onset.

Di(n-butyl) phthalate (DBP) is considered a substance of serious concern because of its reproductive toxicity and endocrine-disrupting properties. Exposure to DBP causes morphological and functional changes in the male reproductive system of birds and mammals. However, there are no detailed reports on the effects of DBP on the Sertoli cell and junctional complexes of the blood-testis barrier (BTB) in birds. The present study investigated dose-related ultrastructural changes in Sertoli cells and junctional complexes of the BTB in adult Japanese quail (Coturnix coturnix japonica) exposed to DBP

Fine mapping of complex traits in non-model species: using next generation sequencing and advanced intercross lines in Japanese quail.

BACKGROUND: As for other non-model species, genetic analyses in quail will benefit greatly from a higher marker density, now attainable thanks to the evolution of sequencing and genotyping technologies. Our objective was to obtain the first genome wide panel of Japanese quail SNP (Single Nucleotide Polymorphism) and to use it for the fine mapping of a QTL for a fear-related behaviour, namely tonic immobility, previously localized on Coturnix japonica chromosome 1. To this aim, two reduced representations of the genome were analysed through high-throughput 454 sequencing: AFLP (Amplified Fragme

Combined exposure of Japanese quails to cyanotoxins, Newcastle virus and lead: oxidative stress responses.

Wild birds are continually exposed to many anthropogenic and natural stressors in their habitats. Over the last decades, mass mortalities of wild birds constitute a serious problem and may possibly have more causations such as natural toxins including cyanotoxins, parasitic diseases, industrial chemicals and other anthropogenic contaminants. This study brings new knowledge on the effects of controlled exposure to multiple stressors in birds. The aim was to test the hypothesis that influence of cyanobacterial biomass, lead and antigenic load may combine to enhance the effects on birds, includin

Developmental toxicity of glyceryl trinitrate in quail embryos.

BACKGROUND: Although glyceryl trinitrate (GTN) is used extensively to treat angina and heart failure, little is known about its effects on the conceptus during organogenesis. The goal of these studies was to investigate the effects of GTN in a model organism, the quail (Coturnix coturnix japonica) embryo. METHODS: To identify the effects of GTN on quail embryo development, fertilized quail eggs (n = 10-12 eggs/group) were injected with GTN (0, 4.4, 44, or 440 μM) at Hamburger-Hamilton (HH) stage 0, 9, or 19 and examined 7 days later. Next, HH 9 embryos were injected with GTN (0, 0.88, 4.4, 8.

Functional disorder of the retina in manganese-deficient Japanese quail revealed by electroretinography using a contact lens electrode with built-in light source.

Manganese deficiency results in neurological and skeletal defects, together with ultrastructural disarrangement of the retina in rats. Wild birds show a range of Mn concentrations in their tissues, including the liver, raising the possibility of Mn-related disorders in the wild. Electroretinography (ERG) provides a useful noninvasive approach to evaluate visual function. This method is especially useful in birds, as objective analysis of them is very difficult, while they have well-developed vision. In this study, we carried out a convenient and reliable ERG recording using a contact lens elec

Circadian rhythms of corneal mitotic rate, retinal melatonin and immunoreactive visual pigments, and the effects of melatonin on the rhythms in the Japanese quail.

We investigated circadian ocular rhythms in the Japanese quail, Coturnix coturnix japonica. The birds were placed under light-dark cycles (LD 12:12), constant light (LL) and constant darkness (DD), and the retinas were dissected out at four-hour intervals throughout 24 h. Following measurements were performed. (1) Melatonin content in the retina was measured by radioimmunoassay. It was low in light and several folds higher in darkness under LD 12:12. The rhythm continued in DD, but disappeared in LL. (2) Mitotic figures in the corneal epithelium were counted. Similar rhythms to the melatonin c

The neuroendocrine control of gonadotrophin release in the Japanese quail. III. The role of the tuberal and anterior hypothalamus in the control of ovarian development and ovulation.

Reproduction in the Japanese quail (Coturnix coturnix japonica), as in many other species, is controlled by the length of the daily photoperiod. The present experiments have investigated the neuroendocrine regulation of both the photoperiodically induced 'tonic' gonadotrophin secretion, which causes the development and maintenance of the ovary, and the 'cyclic' surge release of luteinizing hormone (LH), which stimulates ovulation. Areas within the anterior and tuberal hypothalamus were destroyed by means of electrolytic lesions while a small knife was used to isolate, to various degrees, the t
