Plants from the genus Pterocaulon are popularly used as antifungal and wound-healing agents. Such activities have been related to coumarins, which are abundant in those plants. Coumarins are soluble in organic solvents, such as hexane and dichloromethane, and some of them are also soluble in hot water. Considering that infusion and decoctions of these plants are used in traditional medicine, the aim of this study was to identify and quantify the coumarins in the aqueous extract of Pterocaulon balansae. The aqueous extract was obtained by dynamic maceration and the compounds were characterized

A series of coumarins and benzocoumarins incorporating methyl and hydroxyl moieties in the heterocyclic ring were investigated for the inhibition of the zinc enzyme carbonic anhydrase (CA, EC 4.2.1.1). These coumarins were very weak or ineffective as inhibitors of the house-keeping, offtarget isoforms CA I and II, but showed effective, submicromolar inhibition of the transmembrane, tumor-associated isoforms CA IX and to a slightly less extent, CA XII. The nature and position of the groups substituting the coumarin ring influenced CA inhibitory properties. 4-Methyl-5,7-dihydroydroxycoumarin sho

Eight coumarins, which carry a terminal alkene tethered by a CH(2)XCH(2) group to their 4-position (X = CH(2), CMe(2), O, S, NBoc, NZ, NTs, NBn), were synthesized in overall yields of 51-80 %. Starting materials for the syntheses were either commercially available 4-hydroxycoumarin or 4-formylcoumarin. The intramolecular [2+2] photocycloaddition of these coumarins gave diastereoselectively products with a tetracyclic 3,3a,4,4a-tetrahydro-1H-cyclopenta[2,3]cyclobuta[1,2-c]chromen-5(2H)-one skeleton. Direct irradiation at λ = 300 nm in dichloromethane (c = 10 mM) led to product formation in good

Hypoxic tumor regions remain challenging to treat in breast cancer therapy due to extracellular acidification, chemoresistance, and other adaptive mechanisms promoted by the coordinated actions of tumor-associated carbonic anhydrase IX (hCA IX), heat shock protein 90 alpha (HSP90α), and other proteins. Inspired by the privileged role of coumarins as selective hCA IX inhibitors and as HSP90α modulators, together with the ATP-mimetic and anticancer properties of phenyl quinazolinones, a series of alkyl coumarin-tethered phenyl quinazolinones (4a-l) were rationally designed and synthesized as hyp

Although fig leaves are widely used for dermatological treatment in traditional medicine, research on the bioactivity of their components is limited. In this study, an ultraviolet-guided approach was employed to isolate 2 novel benzofurans (1-2), 12 known benzofurans (3-14), and 13 known coumarins (15-26a/26b). Compounds 4-10, 13-14, 17, 20, 24, and 26a/26b were detected in fig leaves for the first time. Their structures were elucidated through comprehensive spectral analysis, 1-phenyl-3-methyl-5-pyrazolone (PMP) derivatization, and literature comparison. The absolute configurations of compoun

Different 2,4-thiazolidinedione-tethered coumarins 5a-b, 10a-n and 11a-d were synthesised and evaluated for their inhibitory action against the cancer-associated hCAs IX and XII, as well as the physiologically dominant hCAs I and II to explore their selectivity. Un-substituted phenyl-bearing coumarins 10a, 10 h, and 2-thienyl/furyl-bearing coumarins 11a-c exhibited the best hCA IX (KIs between 0.48 and 0.93 µM) and hCA XII (KIs between 0.44 and 1.1 µM) inhibitory actions. Interestingly, none of the coumarins had any inhibitory effect on the off-target hCA I and II isoforms. The sub-micromolar

(1) Background: carbonic anhydrases (CAs) are attractive targets for the development of new anticancer therapies; in particular, CAs IX and XII isoforms are overexpressed in numerous tumors. (2) Methods: following the tail approach, we have appended a hydrophobic aromatic tail to a pharmacophore responsible for the CA inhibition (aryl sulfonamide, coumarin). As a linker, we have used squaramides, featured with strong hydrogen bond acceptor and donor capacities. (3) Results: Starting from easily accessible dimethyl squarate, the title compounds were successfully obtained as crystalline solids,

This paper reports on dihydroxycoumarins as fluorescent probes suitable for the detection and determination of the nitroxide radical, namely 4-amino-TEMPO. Since 4-amino-TEMPO is used as a spin label for the detection of various radicals and damage caused by these species, its determination under physiological conditions might help us to understand the mechanism of the oxidative stress. Among different coumarins studied, only dihydroxy-substituted derivatives show high sensitivity, specificity, and selectivity for the nitroxide radical. In this assay, dihydroxy-substituted coumarins under the

Free ortho-hydroxy cinnamate ester derivatives are evaluated in the synthesis of structurally diverse 4-aryl-coumarins via a tandem Heck-Matsuda cyclization reaction. Free phenolic groups were considered incompatible with such a reaction, which usually provide the corresponding diazo dyes. A concise and scalable route employing a ligand-free, Pd-catalyzed Heck-Matsuda arylation under aerobic conditions for the preparation of (R)-Tolterodine in high overall yield and ee is also presented.

The SARS-CoV-2 main protease (Mpro) plays a crucial role in viral replication, making it an attractive target for antiviral drug development. This study evaluates the inhibitory potential of nineteen 6-substituted coumarin derivatives (1a-p and 2a-c) against Mpro using a FRET-based enzymatic assay. Among them, the chlorinated derivative 1f (AKE-9306) exhibited the highest potency with an IC50 value of 61.77 µM. Enzyme kinetics analysis through Michaelis-Menten and Lineweaver-Burk plots revealed that compound 1f may act through a mixed-type inhibition mechanism with a preference for the free en

Nine previously undescribed coumarin congeners, including ailancoumaquinone A (1) and ailancoumarins M-T (2-9), were isolated from the root bark of Ailanthus altissima (Mill.) Swingle. Their structures were confirmed through spectral analyses, computational approaches, and the Mosher's method. Among these compounds, ailancoumaquinone A (1) was identified as a rare hybrid of a coumarin and a naphthoquinone, featuring an unprecedented carbon skeleton in its naphthoquinone moiety. Furthermore, 1 demonstrated significant anti-inflammatory potential, as it not only inhibits pro-inflammatory factors

A series of carboxamide derivatives of 6- and 7-substituted coumarins have been prepared by an original procedure starting from the corresponding 6- or 7-hydroxycoumarins which were alkylated with ethyl iodoacetate, and the obtained ester was converted to the corresponding carboxylic acids which were thereafter reacted with a series of aromatic/aliphatic/heterocyclic amines leading to the desired amides. The new derivatives were investigated as inhibitors of two enzymes, human carbonic anhydrases (hCAs) and soy bean lipoxygenase (LOX). Compounds 4a and 4b were potent LOX inhibitors, whereas ma

The major threat to public health due to the outbreak of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) infection has been recognised as a global issue. The increase in morbidity is primarily due to the lack of SARS-CoV-2 specific drugs. One of the major strategies to combat this threat is to deactivate the enzymes responsible for the replication of corona virus. To this end, 3-arylidene/3-hydroxycoumarin induced deactivation of 3-chymotrypsin like protease (3CLpro) enzyme, which takes the pivotal role in the replication and maturation, was investigated. For ready availability of

Coumarins are generally considered to be produced by natural plants. Fungi have been reported to produce coumarins, but their biosynthetic pathways are still unknown. In this study, Fusarium oxysporum GU-7 and GU-60 were isolated from Glycyrrhiza uralensis, and their antioxidant activities were determined to be significantly different. Abundant dipeptide, phenolic acids, and the plant-derived coumarins fraxetin and scopoletin were identified in GU-7 by untargeted metabolomics, and these compounds may account for its stronger antioxidant activity compared to GU-60. Combined with metabolome and
