Methylation of the telomerase gene promoter region in umbilical cord blood of patients with gestational diabetes mellitus is associated with decreased telomerase expression levels and shortened telomere length.

OBJECTIVE: This study speculates that gestational diabetes mellitus (GDM) may reduce fetal telomere length (TL),which may be related to modification of methylation in the promoter region of the telomerase (TE) gene promoter region. METHODS: In this study, umbilical cord blood samples from patients with and without GDM (N = 100 each) were analyzed by prospective case-control. The TL, TE expression levels, and methylation levels of TERT and TERC gene promoter regions in two groups were measured. The significance of the methylation level of each CpG locus employed logistic regression analysis of

Long-term exposure to PM2.5 exacerbates dopaminergic neuronal loss through CpG hypermethylation induced down-regulation of PINK1 and DJ-1 genes.

To investigate whether airborne particle (PM2.5) aggravates Parkinson's disease (PD) and alter expression of key PD-related genes by DNA methylation. Two groups of rats were exposed to either clean air or polluted air for 3, 6, and 12 months. The neurotoxin rotenone was injected intraperitoneally to induce a Parkinson's-like disorder. Immunostaining was used to measure the number of dopaminergic neurons in substantia nigra (SN). Real-time PCR was used to measure mRNA levels of PD-related genes PINK1 and DJ-1 in SN. Bisulfate sequencing (BSP) was used to measure DNA methylation levels in gene p

Regulated transcription of human matrix metalloproteinase 13 (MMP13) and interleukin-1β (IL1B) genes in chondrocytes depends on methylation of specific proximal promoter CpG sites.

The role of DNA methylation in the regulation of catabolic genes such as MMP13 and IL1B, which have sparse CpG islands, is poorly understood in the context of musculoskeletal diseases. We report that demethylation of specific CpG sites at -110 bp and -299 bp of the proximal MMP13 and IL1B promoters, respectively, detected by in situ methylation analysis of chondrocytes obtained directly from human cartilage, strongly correlated with higher levels of gene expression. The methylation status of these sites had a significant impact on promoter activities in chondrocytes, as revealed in transfectio

Blood-Based Epigenetic Markers of FKBP5 Gene Methylation in Patients With Dilated Cardiomyopathy.

Background Blood-based DNA methylation patterns are linked to types of diseases. FKBP prolyl isomerase 5 (FKBP5), a protein cochaperone, is known to be associated with the inflammatory response, but the regulatory mechanisms by leukocyte FKBP5 DNA methylation in patients with dilated cardiomyopathy (DCM) remain unclear. Methods and Results The present study enrolled patients with DCM (n=31) and age-matched and sex-matched control participants (n=43). We assessed FKBP5 CpG (cytosine-phosphate-guanine) methylation of CpG islands at the 5' side as well as putative promoter regions by methylation-

miR-342-3p promotes osteogenic differentiation via targeting ATF3.

Through their multiple targets, microRNAs (miRNAs) are involved in numerous physiological and pathological processes. In this study, miR-342-3p was found to be deregulated with ossification of ligament or osteoporosis. We demonstrate that silencing miR-342-3p impairs osteoblast activity and matrix mineralization, while over expression of miR-342-3p promotes osteoblast differentiation significantly. Moreover, miR-342-3p directly targets activating transcription factor 3 (ATF3), which inhibits transcription of pro-osteogenic differentiation-associated genes. In addition, there exists a higher fr

Degree of methylation burden is determined by the exposure period to carcinogenic factors.

Aberrant DNA methylation accumulated in normal tissues, namely methylation burden, is associated with risk of carcinogenesis. The levels of methylation burden are known to be influenced by multiple factors, such as genetic factors and strengths of carcinogenic factors. However, the impact of the degree of exposure to a carcinogenic factor is still unclear. Here, using a Mongolian gerbil model of Helicobacter pylori (H. pylori)-induced gastritis, we aimed to clarify the impact of the degree of exposure on methylation burden in normal gastric tissues. DNA methylation levels of four CpG islands,

Postnatal epigenetic differences in calves following transient fetal infection with bovine viral diarrhea virus.

BACKGROUND: Bovine viral diarrhea virus (BVDV) is the most detrimental pestivirus within the cattle industry. Infection with vertically transmissible BVDV prior to 125 days of gestation results in the generation of a persistently infected (PI) calf. These PI calves are unable to clear the virus in utero, due to an incomplete immune response. However, when infection with BVDV occurs after 150 days of gestation, the fetus clears the transient infection (TI) in utero and is born with antibodies specific to the infecting strain of BVDV. Variations in DNA methylation have been identified in white b

Bacterial DNA confers neuroprotection after optic nerve injury by suppressing CD4+CD25+ regulatory T-cell activity.

PURPOSE: Protective autoimmunity attenuates secondary degeneration after central nervous system (CNS) injury. Such neuroprotection is achieved via activation of autoimmune CD4(+)CD25(-) effector T cells (Teffs) or suppression of naturally occurring CD4(+)CD25(+) regulatory T cells (Tregs). In this study the ability of bacterial DNA, characterized by unmethylated CpG islands, to downregulate Treg activity and therefore, to confer neuroprotection was investigated. METHODS: The effects of CpG on suppressive activity of mouse Tregs were studied by coculturing Tregs with Teffs and measuring prolif

A smoothing method for DNA methylome analysis to enhance epigenomic signature detection in epigenome-wide association studies.

Epigenome-wide association studies (EWAS) are instrumental for mapping DNA methylation changes in human traits and diseases but often suffer from low statistical power and false positives, especially in small cohorts. We developed an EWAS smoothing method that exploits co-methylation of adjacent CpG probes within CpG islands via a sliding-window average and generalized it using Savitzky-Golay filtering. We applied the smoothing approach-with window widths of 1-3 CpGs and, for generalization, Savitzky-Golay filters of varying polynomial orders and window sizes-across five distinct EWAS settings

Exploring the Impact of DNA Methylation on Gene Expression in CRC: A Computational Approach for Identifying Epigenetically Regulated Genes in Multi-Omic Datasets.

BACKGROUND/OBJECTIVES: DNA methylation is a key epigenetic process that regulates gene expression and is often disrupted in colorectal cancer (CRC). Aberrant methylation of promoter CpG islands can silence tumor suppressor genes and drive tumorigenesis. A subset of CRCs exhibits the CpG Island Methylator Phenotype (CIMP), characterized by widespread hypermethylation and distinct clinical outcomes. Identifying genes whose expression is epigenetically regulated by methylation is important for prioritizing candidate biomarkers and therapeutic targets in CRC. METHODS: We developed and compared a

DNA methylation and exposure to ambient air pollution in two prospective cohorts.

Long-term exposure to air pollution has been associated with several adverse health effects including cardiovascular, respiratory diseases and cancers. However, underlying molecular alterations remain to be further investigated. The aim of this study is to investigate the effects of long-term exposure to air pollutants on (a) average DNA methylation at functional regions and, (b) individual differentially methylated CpG sites. An assumption is that omic measurements, including the methylome, are more sensitive to low doses than hard health outcomes. This study included blood-derived DNA methyl

The role of Zearalenone in epigenetic modifications of candidate genes in Nellore heifers.

Mycotoxins are toxic secondary metabolites produced by fungi that proliferate in stored feeds such as grains and silage. Zearalenone (ZEN) is a frequently detected mycotoxin with well-known estrogenic activity. This study aimed to evaluate the effects of ingesting ZEN-contaminated feed on the methylation and hydroxymethylation of CpG islands located in the candidate genes CYP1A1, CYP21A2, and CYP1B1 in Nellore heifers. Twenty heifers were confined for 12 weeks during the finishing phase and assigned to two groups: a control group (non-contaminated feed) and a ZEN-treated group (approximately 3
