Microsatellite loci cross-species transferability in Aedes fluviatilis (Diptera:Culicidae): a cost-effective approach for population genetics studies.

BACKGROUND: Aedes fluviatilis is a neotropical mosquito species thought to be a potential vector of Yellow Fever viruses and can be infected with Plasmodium gallinaceum in laboratory. A better understanding of its genetic structure is very important to understand its epidemiologic potential and how it is responding to urbanization. The objective of this study was to survey the transferability of microsatellites loci developed for other Aedes to Ae. fluviatilis. FINDINGS: We tested in Ae. fluviatilis 40 pairs of primers known to flank microsatellite regions in Aedes aegypti, Aedes albopictus a

Low genetic barrier to large increases in HIV-1 cross-resistance to protease inhibitors during salvage therapy.

HIV-1 resistance to protease inhibitors (PIs) is characterized by extensive cross-resistance within this drug class. Some PIs, however, appear less affected by cross-resistance and are often prescribed in salvage therapy regimens for patients who have failed previous PI treatment. To examine the capacity of HIV-1 to adapt to these treatment changes, we have followed the evolution of HIV-1 protease genotypes and phenotypes in 21 protease-inhibitor-experienced patients in whom 26 weeks of an aggressive salvage regimen associating lopinavir, amprenavir and ritonavir failed to suppress viral repli

Genetic Literacy and Communication of Genetic Information in Families Concerned with Hereditary Breast and Ovarian Cancer: A Cross-Study Comparison in Two Countries and within a Timeframe of More Than 10 Years.

Examining genetic literacy in families concerned with hereditary breast and ovarian cancer (HBOC) helps understand how genetic information is passed on from individuals who had genetic counseling to their at-risk relatives. This cross-study comparison explored genetic literacy both at the individual and the family level using data collected from three sequential studies conducted in the U.S. and Switzerland over ≥10 years. Participants were primarily females, at-risk or confirmed carriers of HBOC-associated pathogenic variants, who had genetic counselling, and ≥1 of their relatives who did not

Cytokine and genetic crosstalk in patients with coexistent familial Mediterranean fever and celiac disease: a cross-sectional study from Türkiye.

BACKGROUND: Familial Mediterranean Fever (FMF) and Celiac Disease (CD) are two distinct inflammatory disorders that arise through different immunological pathways, yet they show a notable tendency to coexist, particularly in Mediterranean populations. The immunological consequences of this coexistence have not been adequately characterized. OBJECTIVE: We set out to compare serum cytokine profiles and explore potential genetic interactions in patients with both FMF and CD versus those with FMF alone or CD alone. One hundred adult patients were enrolled and divided into three groups: 40 with co

Dissecting shared genetic architecture between pan-cancer and aging-related traits: a genome-wide cross-trait analysis.

The association between aging and cancer has been extensively documented in observational studies, but their shared genetic basis remains unclear. Leveraging genome-wide association studies summary statistics of aging and pan-cancer (87,531 cases and 314,193 controls) within the European population, genetic correlation and Mendelian randomization analyses were used to estimate genetic correlations, and infer causal relationships between seven aging-related traits and pan-cancer. We further conducted cross-trait and colocalization analyses to identify shared causal variants and then mapped them

Genetic predisposition for depression, psychosocial and behavioural factors and premenstrual symptoms: a cross-sectional study among young women in China.

BACKGROUND: Premenstrual disorders (PMDs) affect approximately one in three women of reproductive age and have a substantial impact on daily functioning and mental health. Given the challenges observed in diagnosis and clinical management, understanding whether genetic predisposition and readily assessable factors jointly mark greater symptom burden may help inform future risk-stratified monitoring and assessment. METHODS: After excluding 424 participants with missing data, we conducted a cross-sectional study of 1528 female college students from the Care of Premenstrual Emotion (COPE) cohort

Genetic architecture shared between body shape phenotypes and preeclampsia-related diseases: a genome-wide cross-trait analysis.

BACKGROUND: Observational studies have established a strong epidemiological connection between obesity and an increased risk of pre-eclampsia (PE). However, the genetic mechanisms underlying this comorbidity remain largely unexplored. In this study, we conducted a comprehensive cross-trait analysis to elucidate the shared genetic architecture between various adiposity-related traits and PE. METHODS: Using the genome-wide association study (GWAS) statistics, we first evaluated the overall genetic overlap between adiposity-related traits and PE through multiple approaches, including linkage dis

GFPT1 as a cross-ancestry validated target for degenerative spinal disease: genetic association in a Chinese cohort and functional characterization in zebrafish.

Degenerative spinal disease (DSD), including spinal stenosis and spondylosis, lacks effective pharmacological treatment. To identify druggable targets and assess cross-ancestry applicability, we integrate multi-omics analyses using Summary-data-based Mendelian Randomization (SMR), colocalization, and two-sample Mendelian randomization with European whole-blood, peripheral-blood, and CSF eQTL/pQTL datasets, followed by whole-genome sequencing (WGS) validation in a Chinese cohort. We identify 7 genes/proteins associated with spinal stenosis and 5 with spondylosis, with GFPT1, GPX1, and SERPINA1

TyG index is associated with colorectal adenomatous polyps and genetically linked to colon polyps: a cross-sectional and Mendelian randomization study.

BACKGROUND: The triglyceride-glucose (TyG) index, a simple and validated surrogate marker of insulin resistance, may be clinically relevant to colorectal polyp development because insulin resistance can promote colorectal neoplasia through hyperinsulinemia, chronic inflammation, oxidative stress, and dysregulated insulin-like growth factor signaling. However, the relationship between TyG index and colorectal polyps remains incompletely understood. This study aimed to investigate the association between TyG index and colorectal polyp risk using both observational and genetic approaches. METHOD

Genetic diversity of collaborative cross mice enables the establishment of a novel Chlamydia muridarum female genital tract infection model.

Chlamydial infection in women displays wide variation in bacterial burden, persistence, and risk of upper genital tract pathology, yet the host genetic factors underlying this heterogeneity remain poorly defined. We evaluated genital tract infection with Chlamydia muridarum across 20 Collaborative Cross (CC) strains, a recombinant inbred mouse panel that captures broad genetic diversity with high within-strain reproducibility. CC strains exhibited striking differences in early bacterial burden, time to clearance, and oviduct pathology, including prolonged low-inflammatory infections and burden

Shared genetics between metabolic traits and sensorineural hearing loss: a large-scale genome-wide cross-trait analysis.

Sensorineural hearing loss (SNHL) has been epidemiologically linked to metabolic traits, yet their shared genetic basis remains unclear. We performed the largest genome-wide association study (GWAS) meta-analysis for SNHL and analyzed GWAS data from 14 metabolic traits to assess genetic correlations and infer causal relationships. Significant genetic correlations were observed between SNHL and high triglyceride (TG) levels and low high-density lipoprotein (HDL) levels. Cross-trait meta-analyses identified dozens of shared loci and pleiotropic genes, especially between SNHL and HDL or TG. Funct

Genetic evidence of the causal relationship between serum micronutrients and Graves' disease: A Mendelian randomization and cross-sectional observational study.

BACKGROUND AND OBJECTIVES: Exploring the effects of circulating micronutrients on Graves' disease (GD) through observational research or randomized controlled trials has drawn more attention. In order to investigate the putative causal inference, we provide an illustrative estimate of two-sample Mendelian randomization (MR) study. METHODS AND STUDY DESIGN: Inverse-variance weighted (IVW) method was employed as the primary approach to determine the causal relationships between micronutrients level and GD. Several complementary sensitivity analyses were also undertaken to evaluate the impact of

Genetic pleiotropy underlying obesity and autoimmune disorders: a large-scale cross-trait gwas analysis in European ancestry populations.

BACKGROUND: Obesity and autoimmune disorders represent a significant comorbidity burden, yet their shared genetic architecture is not fully understood. Elucidating the pleiotropic genetic basis underlying both conditions is crucial for unraveling the mechanisms driving their co-occurrence and advancing therapeutic strategies. METHODS: We conducted a large-scale cross-trait analysis integrating genome-wide association study (GWAS) summary data for obesity and 17 autoimmune diseases. Genetic correlations were assessed using LD score regression and high-definition likelihood. Cross-trait pleiotro

Shared genetic basis connects smoking behaviors and bone health: insights from a genome-wide cross-trait analysis.

Although the negative association of tobacco smoking with osteoporosis is well-documented, little is known regarding the shared genetic basis underlying these conditions. In this study, we aim to investigate a shared genetic architecture between smoking and heel estimated bone mineral density (eBMD), a reliable proxy for osteoporosis. We conducted a comprehensive genome-wide cross-trait analysis to identify genetic correlation, pleiotropic loci and causal relationship of smoking with eBMD, leveraging summary statistics of the hitherto largest genome-wide association studies conducted in Europe

Genetic Risk of Primary Aldosteronism and Its Contribution to Hypertension: A Cross-Ancestry Meta-Analysis of Genome-Wide Association Studies.

BACKGROUND: Hypertension imposes substantial health and economic burden worldwide. Primary aldosteronism (PA) is one of the most common causes of secondary hypertension, causing cardiovascular events at higher risk compared with essential hypertension. However, the germline genetic contribution to the susceptibility of PA has not been well elucidated. METHOD: We conducted a genome-wide association analysis of PA in the Japanese population and a cross-ancestry meta-analysis combined with UK Biobank and FinnGen cohorts (816 PA cases and 425 239 controls) to identify genetic variants that contri

PheWAS and cross-disorder analysis reveal genetic architecture, pleiotropic loci and phenotypic correlations across 11 autoimmune disorders.

INTRODUCTION: Autoimmune disorders (ADs) are a group of about 80 disorders that occur when self-attacking autoantibodies are produced due to failure in the self-tolerance mechanisms. ADs are polygenic disorders and associations with genes both in the human leukocyte antigen (HLA) region and outside of it have been described. Previous studies have shown that they are highly comorbid with shared genetic risk factors, while epidemiological studies revealed associations between various lifestyle and health-related phenotypes and ADs. METHODS: Here, for the first time, we performed a comparative p

Clinical-Genetic Features Influencing Disability in Spastic Paraplegia Type 4: A Cross-sectional Study by the Italian DAISY Network.

BACKGROUND AND OBJECTIVES: Hereditary spastic paraplegias (HSPs) are a group of inherited rare neurologic disorders characterized by length-dependent degeneration of the corticospinal tracts and dorsal columns, whose prominent clinical feature is represented by spastic gait. Spastic paraplegia type 4 (SPG4, SPAST-HSP) is the most common form. We present both clinical and molecular findings of a large cohort of patients, with the aim of (1) defining the clinical spectrum of SPAST-HSP in Italy; (2) describing their molecular features; and (3) assessing genotype-phenotype correlations to identify
