Parallel adaptations enabling the use of plant cells as the primary food source have occurred multiple times in distinct nematode clades. The hallmark of all extant obligate and facultative plant-feeding nematodes is the presence of an oral stylet, which is required for penetration of plant cell walls, delivery of pharyngeal gland secretions into host cells and selective uptake of plant assimilates. Plant parasites from different clades, and even within a single clade, display a large diversity in feeding behaviours ranging from short feeding cycles on single cells to prolonged feeding on high

BACKGROUND: Beneficial plant-associated microorganisms are key drivers of sustainable agriculture. However, their discovery and practical application remain insufficiently explored. Here, we present the first comprehensive comparative genomic analysis of a newly identified Sinomonas strain together with all publicly available genomes to assess its agronomic potential. RESULTS: Our results demonstrate that Sinomonas has an open pangenome and harbors multiple pathways for IAA biosynthesis, including a rare pathway found in only a limited number of bacterial taxa. The genus also contains genes in

Thidiazuron (TDZ) is a synthetic growth regulator with cytokinin-like activity, widely used in biotechnology, but its physiological effects during the early stages of plant development are not yet fully understood. In this study, the impact of increasing concentrations of TDZ on the germination and early development of Arabidopsis thaliana was evaluated, with particular attention to physiological responses and their dose-dependent patterns. Seeds of the Col-0 ecotype were germinated in the presence of TDZ and analyzed using integrated approaches including morphological observations, photosynth

Fruit shape diversity in melon is governed by complex genetic networks, with ethylene biosynthesis playing a pivotal yet poorly characterized role. In this study, we identified a rare CmACS7A57V/frameshift double mutant through fine mapping of the fsq2 locus. Ethylene-mediated ovary growth regulation has been completely lost in the CmACS7A57V/frameshift double mutant, driving a transition from elongated to spherical fruit. Transcriptome analysis was performed to clarify the core role of CmACS7 in the ethylene signaling pathway. The loss of CmACS7 function regulates key genes in the ethylene re

Nitric oxide (NO) is a multiregulatory signal molecule that integrates development and stress responses. To elucidate the molecular mechanisms of NO phytoeffects and to identify NO-associated genes, both genetic screens and genome-wide transcriptome analysis have been employed in numerous studies. Forward genetic screens have linked NO signalling to key biological processes, such as photosynthesis, cytokinin metabolism, stress adaptation, and cell cycle regulation. Reverse genetics has further characterized the role of NO-related genes involved in NO biosynthesis (e.g., NIA1/NIA2, NOA1), signa

Plants are frequently exposed to high salinity, negatively affecting their development and productivity. This review examined the complex roles of membrane transporters (MTs) and phytohormones in mediating salt stress. MTs are crucial in capturing sodium ions (Na+) and maintaining a delicate balance between sodium (Na+) and potassium (K+), essential for supporting cellular homeostasis and enhancing overall plant health. These MTs were instrumental in regulating ion balance and promoting the absorption and segregation of vital nutrients, thereby enhancing salt stress tolerance. Various plant ho

The miR156 gene is known to play an important role in regulating growth and development in plants. This gene is involved in the transition from juvenile to adult stages, leaf morphology, and root development, among other processes. While the function of miR156 is similar in many plants, there are also differences in the function of this gene between herbaceous and native species. We obtained BpmiR156 overexpression transgenic lines in Betula platyphylla, and the transgenic lines exhibited traits such as delayed development, dwarfism, increased leaf epidermal hairs, larger leaf basal angle and

N,N-dimethylhexadecylamine (DMHDA) is a bacterial volatile organic compound that affects plant growth and morphogenesis and is considered a cross-kingdom signal molecule. Its bioactivity involves crosstalk with the cytokinin and jasmonic acid (JA) pathways to control stem cell niches and induce iron deficiency adaptation and plant defense. In this study, through genetic analysis, we show that the DMHDA-JA-Ethylene (ET) relations determine the magnitude of the defensive response mounted during the infestation of Arabidopsis plants by the pathogenic fungus Botrytis cinerea. Plants can be regenerated from various explants/tissues via de novo shoot meristem formation. Most of these regeneration pathways are indirect and involve callus formation. Besides plant hormones, the role of polyamines (PAs) has been implicated in these processes. Interestingly, the lateral root primordia (LRPs) of Arabidopsis can be directly converted to shoot meristems by exogenous cytokinin application. In this system, no callus formation takes place. We report that the level of PAs, especially that of spermidine (Spd), increased during meristem conversion and the application of exogenous

Kinetin (N6-furfuryladenine), a plant growth substance of the cytokinin family, has been shown to modulate aging and various age-related conditions in animal models. Here we report the synthesis of kinetin isosteres with the purine ring replaced by other bicyclic heterocycles, and the biological evaluation of their activity in several in vitro models related to neurodegenerative diseases. Our findings indicate that kinetin isosteres protect Friedreich́s ataxia patient-derived fibroblasts against glutathione depletion, protect neuron-like SH-SY5Y cells from glutamate-induced oxidative damage, a

Blue light photoreceptors, cryptochromes (CRYs), regulate multiple aspects of plant growth and development. However, our knowledge of CRYs is predominantly based on model plant Arabidopsis at early growth stage. In this study, we elucidated functions of CRY1a gene in mature tomato (Solanum lycopersicum) plants by using cry1a mutants and CRY1a-overexpressing lines (OE-CRY1a-1 and OE-CRY1a-2). In comparison with wild-type plants, cry1a mutants are relatively tall, accumulate low biomass, and bear more fruits, whereas OE-CRY1a plants are short stature, and they not only flower lately but also bea

An effective system for isolating and regenerating protoplasts is crucial for research in genome engineering. This study focused on refining a protocol for the isolation and regeneration of mesophyll protoplasts from the leaves of Solanum tuberosum cv. Kufri Jyoti. Key factors influencing protoplast yield and viability, such as dark pretreatment, pre-plasmolysis, enzyme concentrations, and osmoticum levels, were thoroughly assessed and optimized. The highest protoplast yield and viability were achieved with an enzyme mixture of 1.0% cellulase R-10 and 0.5% macerozyme R-10 after 16 h of incubat

Salt stress seriously restricts crop yield and quality, leading to an urgent need to understand its effects on plants and the mechanism of plant responses. Although phytohormones are crucial for plant responses to salt stress, the role of phytohormone signal transduction in the salt stress responses of stress-resistant species such as Sophora alopecuroides has not been reported. Herein, we combined transcriptome and metabolome analyses to evaluate expression changes of key genes and metabolites associated with plant hormone signal transduction in S. alopecuroides roots under salt stress for 0

Magnesium (Mg) deficiency is becoming a widespread limiting factor for crop production. How crops adapt to Mg limitation remains largely unclear at the molecular level. Using hydroponic-cultured tomato seedlings, we found that total Mg2+ content significantly decreased by ∼80% under Mg limitation while K+ and Ca2+ concentrations increased. Phylogenetic analysis suggested that Mg transporters (MRS2/MGTs) constitute a previously uncharacterized 3-clade tree in planta with two rounds of asymmetric duplications, providing evolutionary evidence for further molecular investigation. In adaptation to

During their lifetime, plants encounter numerous biotic and abiotic stresses with diverse modes of attack. Phytohormones, including salicylic acid (SA), ethylene (ET), jasmonate (JA), abscisic acid (ABA), auxin (AUX), brassinosteroid (BR), gibberellic acid (GA), cytokinin (CK) and the recently identified strigolactones (SLs), orchestrate effective defense responses by activating defense gene expression. Genetic analysis of the model plant Arabidopsis thaliana has advanced our understanding of the function of these hormones. The SA- and ET/JA-mediated signaling pathways were thought to be the b

Mutations that affect the splicing of pre-mRNA are a major cause of human disease. Familial dysautonomia (FD) is a recessive neurodegenerative disease caused by a T to C transition at base pair 6 of IKBKAP intron 20. This mutation results in variable tissue-specific skipping of exon 20. Previously, we reported that the plant cytokinin kinetin dramatically increases exon 20 inclusion in RNA isolated from cultured FD cells. The goal of the current study was to investigate the nature of the FD splicing defect and the mechanism by which kinetin improves exon inclusion, as such knowledge will facil

The increasing challenges, posed by plant biotic and abiotic stress, significantly impact agroecosystems, particularly in the context of rapid global climate change. Phytohormones function as critical chemical messengers, enabling plants to endure these stresses through a complex regulatory framework, thereby playing an essential role in plant survival. Recent research has increasingly underscored the significance of abscisic acid (ABA), in facilitating a prominent role under adverse conditions within plants. ABA has been shown to engage with major phytohormones, such as gibberellins (GAs), et

Hydrogen sulfide (H2S) has been postulated to be the third gasotransmitter in both animals and plants after nitric oxide (NO) and carbon monoxide (CO). In this review, the physiological roles of H2S in plant growth, development and responses to biotic, and abiotic stresses are summarized. The enzymes which generate H2S are subjected to tight regulation to produce H2S when needed, contributing to delicate responses of H2S to environmental stimuli. H2S occupies a central position in plant sulfur metabolism as it is the link of inorganic sulfur to the first organic sulfur-containing compound cyst

Coconut, Cocos nucifera L., is a tree that is cultivated to provide a large number of products, although it is mainly grown for its nutritional and medicinal values. Coconut oil, derived from the coconut fruit, has been recognised historically as containing high levels of saturated fat; however, closer scrutiny suggests that coconut should be regarded more favourably. Unlike most other dietary fats that are high in long-chain fatty acids, coconut oil comprises medium-chain fatty acids (MCFA). MCFA are unique in that they are easily absorbed and metabolised by the liver, and can be converted to
