Giardia--from genome to proteome.

In this review, the current status of genomic and proteomic research on Giardia is examined in terms of evolutionary biology, phylogenetic relationships and taxonomy. The review also describes how characterising genetic variation in Giardia from numerous hosts and endemic areas has provided a better understanding of life cycle patterns, transmission and the epidemiology of Giardia infections in humans, domestic animals and wildlife. Some progress has been made in relating genomic information to the phenotype of Giardia, and as a consequence, new information has been obtained on aspects of deve

Human pluripotent stem cells for the modelling of diseases of the retina and optic nerve: toward a retina in a dish.

Human pluripotent stem cells can be differentiated into specific, relevant cell types of interest including the cells of the retina and optic nerve. These cells can then be used to study fundamental biology as well as disease modelling and subsequent screening of potential treatments. Many models of differentiation and modelling have relied on two-dimensional monocultures of specific cell types, which are not representative of the complexity of the human retina and optic nerve. Hence, more complex models of the human retina and optic nerve are required. Three-dimensional organoids and emerging

Retinoic acid induced myelomeningocele in fetal rats: characterization by histopathological analysis and magnetic resonance imaging.

The prevention of human neural tube defects by folic acid administration and the potential for fetal surgical intervention for myelomeningocele (MMC) have renewed interest in the molecular pathways and pathophysiology of spina bifida. Animal models for assessment of the early developmental biology and pathophysiology of this lesion are needed. The goal of this study was to develop and characterize a non-surgical rat model of MMC. Time-dated Sprague-Dawley rats were gavage fed different doses of retinoic acid (RA) dissolved in olive oil at E10 (maternal n = 55, fetal n = 505). Control animals r

Conserved roles of engrailed: patterning tissues and specifying cell types.

More than 40 years ago, studies of the Drosophila engrailed and Hox genes led to major discoveries that shaped the history of developmental biology. We learned that these genes define the state of determination of cells that populate particular spatially defined regions: the identity of segmental domains by Hox genes, and the identity of posterior developmental compartments by engrailed. Hence, the boundaries that delimit spatial domains depend on engrailed. Here, we review the engrailed field, which now includes orthologs in Drosophila and mouse, as well as many other animals. We focus on fly

Advances in tools and strategies for spatiotemporal regulation of gene expression in Caenorhabditis elegans.

Spatiotemporal manipulation of gene expression is essential for dissecting gene function, elucidating developmental processes, and uncovering complex physiological mechanisms. As a model organism, Caenorhabditis elegans has played an important role in basic biology research and provides an ideal experimental system for achieving such precise control. With the development of single-copy transgenesis, CRISPR/Cas9-mediated genome editing, and conditional recombination and inducible systems, the genetic toolkits for spatiotemporal regulation of gene expression, and even at the single-cell-resoluti

Bibliometric analysis of congenital diaphragmatic hernia research models: evolution and insights from the past three decades.

BACKGROUND: Research models of congenital diaphragmatic hernia (CDH) have evolved over time. We aimed to evaluate global research activities in CDH models using bibliometric analysis over the past three decades. METHODS: Studies were retrieved from the Web of Science Core Collection (WoSCC) database between 1995 and 2024. Microsoft Excel, GraphPad Prism, and VOSviewer were applied to collect and analyze the bibliometric variables of this field. RESULTS: A total of 302 original publications from 21 countries were identified. The nitrofen rat model (n = 187) was used in most studies, followed

The Chronological Trigger: The Orchestra Between Homeobox Genes and the Circadian Clock During Development.

As master regulators of embryonic development, regulating homeobox genes is fundamental for developmental biology. Despite the growth of multiple topics regarding fine-tuning homeobox gene expression, the discussion on how the circadian rhythm affects their control and vice-versa still needs to be improved. Due to the intrinsic importance of the circadian clock and its impact on several molecular mechanisms, including development and pregnancy, the interplay between this mechanism and homeobox genes becomes a meaningful discussion. This work aims to review and critically discuss the crosstalk

Signal transduction during C. elegans vulval development: a NeverEnding story.

The Caenorhabditis elegans hermaphrodite vulva is one of the best studied models for signal transduction and cell fate determination during organogenesis. Systematic forward genetic screens have identified a complex and highly interconnected signaling network formed by the conserved EGFR, NOTCH, and WNT signaling pathways that specifies an invariant pattern of cell fates among the six vulval precursor cells (VPCs). Multiple inhibitory interactions between the EGFR and NOTCH pathways ensure the selection of a single 1° VPC that is always flanked by two 2° VPCs thanks to lateral NOTCH signaling.

The eyes of the olms.

The experiments of the Austrian biologist Paul Kammerer to breed eyes in blind olms is probably one of the most notable manifesations of Lamarckian thinking and research at the beginning of the 20th century. If living in the environment of the dark caves in the Slovenian Kraijna for thousands of years has reduced the eyes of the olms until they nearly disappeared, then is it possible to influence the development in the other direction and speed it up? Will a transformed milieux or media (in a Lamarckian sense) conduct olms to vision, to the mysteries of light? Kammerer's legendary skill in tak

On the trail of the 'new head' in Les Treilles.

The vertebrate brain develops in association with neighboring tissues: neural crest, placodes, mesoderm and endoderm. The molecular and evolutionary relationships between the forming nervous system and the other craniofacial structures were at the focus of a recent meeting at the Fondation des Treilles in France. Entitled 'Relationships between Craniofacial and Neural Development', the meeting brought together researchers working on diverse species, the findings of whom provide clues as to the origin and diversity of the brain and facial regions that are involved in forming the 'new head' of v

Meckel on developmental pathology.

Before Schleiden and Schwann, Darwin and Mendel there passed briefly a towering giant, Johann Friedrich Meckel the Younger (1781-1833), now glimpsed only fleetingly and obscurely through the mist of time and former controversies, who can nowadays easily and clearly be identified as the father of a "pre-modern" developmental biology. At his beginning this prodigiously gifted physician-scholar had, as one would say nowadays, an unfair advantage, his cradle having been rocked, as it were, by the preparators in his father's and grandfather's huge collection of normal and abnormal anatomical "speci

Integrated Genetic Networks and Epigenetic Regulation inTooth Development and Maturation.

Tooth development or odontogenesis is a complex morphogenetic process that requires tightly regulated interactions between the oral epithelium and mesenchyme of neural crest origin. In this narrative review, we compile existing knowledge regarding gene regulatory networks and epigenetic factors throughout tooth development from initiation to eruption. Signaling between the epithelium and mesenchyme is mediated by four conserved pathways-Wnt/β-catenin, bone morphogenetic protein (BMP), fibroblast growth factor (FGF), and Sonic hedgehog (Shh)-which operate iteratively and interact through extens

Primary tooth emergence in young children is associated with a stress biomarker.

Early tooth emergence may reflect developmental stress exposures and compose part of a broader set of growth measures that may be accelerated in response to early stress exposure. We test the hypothesis that timing of primary tooth emergence is associated with stress exposure and stress biology in young children. Tooth emergence was tracked prospectively in a diverse sample of initially caries-free 1-3 year-olds; socio-economic and psychosocial determinants of oral health, oral microbiology, and salivary stress markers were assessed at baseline and at 6-monthly intervals for 2 years. Experimental and Computational Approaches to Identify Noncoding Pathogenic Variation in Rare Disease.

Noncoding variants occur within noncoding genes as well as within the regulatory nontranslated regions of protein-coding genes. It is important to be aware that these variants have been increasingly implicated in developmental disease through a variety of mechanisms. However, they remain difficult to interpret clinically due to their unclear effect on transcript or protein abundance compared with coding variants. Here, we review methods to identify pathogenic noncoding variants in rare disease, which can present challenges due to the inaccessibility of disease-relevant tissue for many conditio

Antibody staining of Parhyale hawaiensis embryos.

The great diversity of arthropod body plans, together with our detailed understanding of fruit fly development, makes arthropods a premier taxon for examining the evolutionary diversification of developmental patterns and hence the diversity of extant life. Crustaceans, in particular, show a remarkable range of morphologies and provide a useful outgroup to the insects. The amphipod crustacean Parhyale hawaiensis is becoming established as a model organism for developmental studies within the arthropods. This protocol provides a simplified protocol for antibody staining of P. hawaiensis embryos

Relative parameter sensitivity in prenatal toxicity studies with substances classified as developmental toxicants.

Developmental toxicity testing according to the globally standardized OECD 414 protocol is an important basis for decisions on classification and labeling of developmental toxicants in the European Union (EU). This test requires relatively large animal numbers, given that parental and offspring generations are involved. In vitro assay designs and systems biology paradigms are being developed to reduce animal use and to improve prediction of human hazard. Such approaches could benefit from the long-term experience with animal protocols and more specifically from information on the relevance of

Quantitative lipidomic analysis of Ascaris suum.

Ascaris is a soil-transmitted nematode that causes ascariasis, a neglected tropical disease affecting predominantly children and adolescents in the tropics and subtropics. Approximately 0.8 billion people are affected worldwide, equating to 0.86 million disability-adjusted life-years (DALYs). Exploring the molecular biology of Ascaris is important to gain a better understanding of the host-parasite interactions and disease processes, and supports the development of novel interventions. Although advances have been made in the genomics, transcriptomics and proteomics of Ascaris, its lipidome has
