Primary nephrotic syndrome (PNS) is common in children, affecting the soft and hard tissues of the oral cavity. This study aimed to investigate the percentage of dental caries, gingivitis, hypertrophic gingivitis, and developmental defects of enamel (DDE) in children with PNS. The association of PNS with these diseases and oral care behavior was also assessed. A total of 407 children with PNS and 407 age- and gender-matched controls were recruited. PNS was diagnosed based on blood and urinary tests. The Simplified Oral Hygiene Index (OHI-S), the Gingival Index (GI), the Gingival Overgrowth Ind

An 11-old boy with Prader-Willi syndrome and partial epilepsy was reported. Muscular hypotonia in early infancy was extreme and developmental milestones were retarded, especially walk and speech. He achieved these landmarks within three years. The first seizure disorder was seen in the 9th year. The patient was characterized by hypotonic musculature, severe mental retardation, obesity (gynaecomasty, excess of fat on the thighs, the abdomen and the trunk), hypogonadism (a minute penis, hypoplastic scrotum and cryptorchidism). Apart from these characteristics, the patient presented some minor mo

Permanent tooth structures can respond to environmental disruptions even after the initiation of hard tissue deposition at the cusp tips, affecting dental morphology. In bioarcheological studies, linear enamel hypoplasia (LEH) provides evidence of both the presence and timing of developmental stressors that can occur during first molar odontogenesis. Yet, previous studies used LEH from the entirety of the anterior tooth surface to relate disruptions to changes in permanent tooth morphology, which could conflate causality by misrepresenting the timing of events. This study assessed associations

INTRODUCTION: Molar-incisor-hypomineralization (MIH), a developmental defect that alters normal mineralization, causing permanent damage to the enamel. The affected enamel shows a reduction in mineral composition, calcium: phosphorus (Ca:P) ratio, and relatively increased carbon content. Among the numerous factors that affect the enamel integrity in MIH, the properties such as salivary and plaque pH might have an impact, but recent studies suggest only minimal data regarding the various factors, making it still obscure to the clinician.
AIM: To determine the salivary and plaque pH in 8-15-yea

Children's developing teeth may be sensitive to environmental dioxins, and in animal studies developing teeth are one of the most sensitive targets of toxicity of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). Twenty-five years after the dioxin accident in Seveso, Italy, 48 subjects from the contaminated areas (zones A and B) and in patches lightly contaminated (zone R) were recruited for the examination of dental and oral aberrations. Subjects were randomly invited from those exposed in their childhood and for whom frozen serum samples were available. The subjects were frequency matched with 65

Molar incisor hypomineralization (MIH) is a developmental defect of dental enamel that shares features with hypomineralized second primary molars (HSPM). Prior to permanent tooth eruption, second primary molars could have predictive value for permanent molar and incisor hypomineralization. To assess this possible relationship, a cross-sectional study was conducted in a sample of 414 children aged 8 and 9 years from the INMA cohort in Valencia (Spain). A calibrated examiner (linear-weighted Kappa 0.83) performed the intraoral examinations at the University of Valencia between November 2013 and

Short Root Defects defined by a reduced ratio of root to crown, may culminate in root resorption and subsequent tooth loss, in spite of the absence of apparent symptoms. Such defects present considerable impediments to orthodontic treatment and restoration. Recent identification of Fam20a, an emergent pseudokinase, has been associated with enamel development and tooth eruption, yet its definitive role in root formation and eruption remains ambiguous. In this research, we initially ascertained that the targeted knockout of Fam20a within the epithelium led to truncated tooth roots, irregular bre

Molar incisor hypomineralization (MIH) is a qualitative developmental enamel defect that affects one to four permanent first molars, with or without involvement of permanent incisors. Its etiology is of systemic origin, but is not well understood. Therefore, we conducted this cross-sectional study to examine pre-, peri-, and postnatal risk factors for MIH among children, 7-8 yr of age, in urban areas of Khon Kaen, Thailand. Molar incisor hypomineralization defects were diagnosed using the European Academy of Pediatric Dentistry criteria. Mothers or primary caregivers were interviewed on matern

AIM: This case report illustrates the multidisciplinary treatment of a 12-year-old boy with esthetic challenges and endodontic problems in his maxillary incisors after severe dental injury at the age of 2½ years.
BACKGROUND: The close anatomic relationship of the primary tooth to the permanent tooth germ explains why traumatic dental injuries in primary dentition may affect the development of permanent teeth especially in the maxillary anterior region. Developmental defects of enamel (DDE) as well as crown/root dilacerations are often seen after displacement injuries such as intrusion or avul

BACKGROUND/PURPOSE: Regional odontodysplasia (RO) is a rare developmental anomaly affecting dental tissue formation, typically confined to a single quadrant. It manifests through hypoplastic enamel, irregular dentin, short roots with open apices, and a characteristic "ghost-like" radiographic appearance. Its etiology remains unclear, with vascular, genetic and inflammatory factors proposed.
MATERIALS AND METHODS: In this study, we report the case of a 15-year-old female with mandibular RO affecting canine, premolars, and molars, focusing on the clinical, radiographic, and microscopic analysis

Inositol 1,4,5-trisphosphate receptors (IP3R) mediate Ca2+ release from intracellular stores, contributing to complex regulation of numerous physiological responses. The involvement of the three IP3R genes (ITPR1, ITPR2 and ITPR3) in inherited human diseases has started to shed light on the essential roles of each receptor in different human tissues and cell types. Variants in the ITPR3 gene, which encodes IP3R3, have recently been found to cause demyelinating sensorimotor Charcot-Marie-Tooth neuropathy type 1J (CMT1J). In addition to peripheral neuropathy, immunodeficiency and tooth abnormali

Hypotonia, ataxia, developmental delay, and tooth enamel defects syndrome (HADDTS) is a recently identified disorder linked to a heterozygous mutation in the C-terminal Binding Protein 1 (CTBP1) transcriptional corepressor. The predominant mutation (p.R342W) is located within the major protein binding cleft (PXDLS), crucial for CtBP1's interaction with transcriptional regulatory proteins. To investigate the mutation's functional consequences, we generated isogenic induced pluripotent cell lines (iPSCs) carrying the CTBP1 mutation in heterozygous and homozygous conditions using the CRISPR/Cas9

MicroRNAs (miRNAs) are a class of small, non-coding RNAs that provide an efficient pathway for regulation of gene expression at a post-transcriptional level. Tooth development is regulated by a complex network of cell-cell signaling during all steps of organogenesis. Most of the congenital dental defects in humans are caused by mutations in genes involved in developmental regulatory networks. Whereas the developmental morphological stages of the tooth development already are thoroughly documented, the implicated genetic network is still under investigation. The involvement of miRNAs in the reg
