Vasculopathic and cardiomyopathic changes induced by low-protein high-carbohydrate tapioca based diet in bonnet monkey. Vasculopathic and cardiomyopathic changes in induced malnutrition.

Mucoid vasculopathy is an entity characterized by a generalized mucopolysaccharidosis of blood vessels. It has been observed in patients from our Institute. Bonnet monkeys (Macaca radiata) were given low-protein normal-carbohydrate and low-protein high-carbohydrate tapioca based diets for experimental periods of 3 or 5 months. The latter diet is similar to that consumed by people from this part of India (Kerala) where mucoid vasculopathy is observed. Vascular lesions were seen in all test animals as a generalized mucopolysaccharidosis and as hyperplastic changes in intimal and medial smooth mu

Comparative effects of bile salts and free bile acids on intestinal health of grouper (Epinephelus spp.) fed a high cottonseed protein concentrate diet.

The physiological impacts of bile acids (BA) conjugation states remain underexplored in aquatic species. This study investigated the effects of supplementing conjugated BA (bile salts [BS]) or non-conjugated BA (free bile acids [FBA]) on the intestinal health of pearl gentian grouper (Epinephelus fuscoguttatus ♂ × Epinephelus lanceolatus ♀) fed a high plant-protein diet for 8 weeks. A total of 300 healthy juveniles (approximately 90-d-old; initial body weight 15.21 ± 0.04 g) were randomly distributed into four dietary groups in triplicate (25 fish per tank): CD group (a fishmeal-based control

The role and mechanism of fatty acid-binding protein 7 in spinal reactive astrocytes in prolonged postoperative pain induced by high-fat diet.

Obesity markedly exacerbates nociceptive sensitivity and substantially compromises the quality of life of affected people. Astrocytes orchestrate metabolic regulation and homeostatic maintenance in the central nervous system. Notably, fatty acid binding protein 7 (FABP7) is highly expressed in astrocytes that governs intracellular fatty acid uptake and transport. While systemic hyperlipidemia is pathognomonic of obesity, the mechanistic contribution of FABP7 in astrocytes to obesity-associated pain pathophysiology remains poorly characterized. The present study established a model of high-fat

Dynamin-Related Protein 1 Binding Partners MiD49 and MiD51 Increased Mitochondrial Fission In Vitro and Atherosclerosis in High-Fat-Diet-Fed ApoE-/- Mice.

Novel components of the mitochondrial fission machinery, mitochondrial dynamics proteins of 49 kDa (MiD49) and 51 kDa (MiD51), have been recently described, and their potential therapeutic targets for treating cardiovascular disease have been shown, including acute myocardial infarction (AMI), anthracycline cardiomyopathy and pulmonary arterial hypertension (PAH). Here, we examined the role of MiD49 and MiD51 in atherosclerosis. MiD49/51 expression was increased in the aortic valve endothelial cells (ECs) of high-fat diet-induced atherosclerosis in ApoE-/-mice and IL-8-induced human umbilical

Deletion of Thioredoxin-Interacting Protein (TXNIP) Abrogates High Fat Diet-induced Retinal Leukostasis, Barrier Dysfunction and Microvascular Degeneration in a Mouse Obesity Model.

We have shown that a high fat diet (HFD) induces the activation of retinal NOD-like receptor protein (NLRP3)-inflammasome that is associated with enhanced expression and interaction with thioredoxin-interacting protein (TXNIP). Here, the specific contribution of TXNIP and the impact of HFD on retinal leukostasis, barrier dysfunction and microvascular degeneration were investigated. Wild-type (WT) and TXNIP knockout (TKO) mice were fed with normal diet or 60% HFD for 8-18 weeks. TXNIP was overexpressed or silenced in human retinal endothelial cells (REC). At 8 weeks, HFD significantly induced r

Role of 14-3-3η protein on cardiac fatty acid metabolism and macrophage polarization after high fat diet induced type 2 diabetes mellitus.

Diabetic cardiomyopathy (DCM), a metabolic disorder, is one of the leading causes of mortality around the world and its pathogenesis involves cardiac inflammation and altered metabolic profile. Altered fatty acid metabolism during DCM can cause macrophage polarization in which inflammatory M1 phenotype dominates over the anti-inflammatory M2 phenotype. Hence, it is essential to identify a specific target, which could revert the metabolic profile and thereby reducing the M1 macrophage polarization. 14-3-3η protein has several cellular protective functions especially in the heart as plenty of re

Cynanchum wilfordii Radix attenuates liver fat accumulation and damage by suppressing hepatic cyclooxygenase-2 and mitogen-activated protein kinase in mice fed with a high-fat and high-fructose diet.

Excessive consumption of fat and fructose augments the pathological progression of nonalcoholic fatty liver disease through hepatic fibrosis, inflammation, and hepatic de novo lipogenesis. We hypothesized that supplementation with Cynanchum wilfordii extract (CWE) decreases fat accumulation in the liver by suppressing cyclooxygenase-2 (COX-2), the nuclear translocation of nuclear factor κB (NF-κB), and p38 mitogen-activated protein kinase (MAPK). The beneficial effect of CWE was evaluated in a murine model of nonalcoholic fatty liver disease. Mice were fed either a normal diet or an atherogeni

The impact of high fat diet on global protein abundance and fractional synthetic rate in liver and mammary gland of peak lactation ICR mice.

Maternal metabolic environment creates the developmental environment for offspring. Previous studies demonstrated high fat (HF) diet increased neonate growth rate during lactation, which related to increased milk lactose content and increased fatty acyl chain length and unsaturation of milk lipids. To understand how HF diet alters maternal metabolism, we measured liver and mammary gland global protein abundance and fractional synthetic rate (FSR) of peak lactation mice after enriching body water with deuterium oxide for 24 h. In both tissues, HF altered abundance of proteins that reflected les

Bioavailability of lead in contaminated soils elevated with high carbohydrate, protein, and fat diets via influences on gut microbiota and intestinal barrier function.

Accidental ingestion of lead (Pb) contaminated soil will interact with different types of foods in the gastrointestinal tract. To determine the effects of food composition on soil Pb oral bioavailability, a mouse bioassay was conducted to evaluate the relative bioavailability (RBA) of Pb with high-fat, high-protein, high-carbohydrate, and high-dietary fiber diets. Results indicated that high-fat diet most effectively enhanced Pb-RBA (91.4% for soil A and 89.7% for soil B, respectively), while high-dietary fiber reduced it. Compared with the basal diet group, high-carbohydrate, high-fat, and hi

Mulberry leaf and konjac compound powder improves the metabolic capacity of old mice on a high-protein diet by regulating the structure of the intestinal microbiota.

BACKGROUND: The health of elderly individuals is closely linked to their protein intake and the abundance of intestinal microbiota. To investigate the impact of a compound powder made from mulberry leaf and konjac (hereinafter referred to as 'compound powder') on regulating the structure of intestinal microbiota in 15-month-old BALB/c mice that were fed a high-beef-protein diet, 16S rRNA high-throughput sequencing, reverse transcription quantitative polymerase reaction, western blot, and other biochemical methods were used to analyze the differences in intestinal microbiota, protein metabolism

High fat diet incorporated with meat proteins changes biomarkers of lipid metabolism, antioxidant activities, and the serum metabolomic profile in Glrx1-/- mice.

Red and processed meat consumption has been associated with oxidative stress, diabetes and non-alcoholic fatty liver disease (NAFLD). This study was aimed at exploring the effects of high-fat meat protein diets on potential metabolite biomarkers in Glrx1-/- mice, a well-documented mouse model to study NAFLD. Male Glrx1-/- mice were fed a control diet with 12% energy (kcal) from fat, a high-fat diet supplemented with casein (HFC) with 60% energy (kcal) from fat, and a high-fat diet supplemented with fish (HFF) or mutton proteins (HFM) for 12 weeks. Impact of high-grain diet feeding on mucosa-associated bacterial community and gene expression of tight junction proteins in the small intestine of goats.

The objective of this study was to investigate the impact of a high-grain (HG) diet on the microbial fermentation, the composition of the mucosa-associated bacterial microbiota, and the gene expression of tight junction proteins in the small intestine of goats. In the present study, we randomly assigned 10 male goats to either a hay diet (n = 5) or a HG diet (56.5% grain; n = 5) and then examined changes in the bacterial community using Illumina MiSeq sequencing and the expression of tight junction proteins using qRT-PCR in the mucosa of the small intestine. The results showed that HG diet dec

New frontiers in gut nutrient sensor research: monosodium L-glutamate added to a high-energy, high-protein liquid diet promotes gastric emptying: a possible therapy for patients with functional dyspepsia.

Functional dyspepsia is a clinical syndrome that features abdominal symptoms centered in the upper abdomen without an organic basis. Three possible mechanisms of gastric dysfunction could be related to functional dyspepsia: 1) delayed gastric emptying, 2) impaired gastric accommodation to food intake, and 3) hypersensitivity to gastric distention. Delayed gastric emptying has been suggested to lead to prolonged antral distension that causes dyspeptic symptoms. Delayed gastric emptying is therefore a focal point of debate about anorexia caused by dyspepsia, and prokinetic agents are often admin

High-protein diets increase microbiota associated p-cresol production in the colon and reduce gut barrier function in a sex-dependent manner.

Protein is an essential nutrient, but the detrimental effects of excess dietary protein on gut health are often overlooked. Protein fermentation by colonic microbiota may impair barrier function by increasing toxic metabolite production. We previously identified sex-by-protein interactions affecting the microbiota and its metabolites in vitro. Do sex-by-protein interactions in colonic protein fermentation lead to a sexually dimorphic response in gut barrier function in vivo? We hypothesised that high-protein diets would elicit sex-specific effects on microbiota and barrier function. Twenty sib

Evaluation of Effects of a High-Protein Hypocaloric Diet on Body Composition and Cardio-Metabolic Factors in Women with Polycystic Ovary Syndrome and Overweight or Obesity.

BACKGROUND: The optimal composition of a hypocaloric diet for women with polycystic ovary syndrome (PCOS) remains uncertain. The aim of this study was to evaluate and compare the impacts of a hypocaloric high-protein diet (HPD) versus an isocaloric conventional calorie-restricted diet (CRD) with normal protein intake on the body composition and biochemical profiles of women diagnosed with PCOS combined with overweight or obesity. METHODS: This was a dietary intervention study evaluating the effects of two types of diets on women with PCOS who initiated weight loss independently at the Clinica

Interaction of Programmed Obesity and Postnatal High Fat Diet but Not (-)-Epicatechin Treatment Modifies Muscle Atrophy Proteins Levels in Male Wistar Rats.

We determine whether the offspring of obese mothers and a postnatal high-fat diet (HFD) modify protein levels related to muscle synthesis (p70S6K-alpha) or atrophy (Murf and MAFbx), and if the administration of (-)-epicatechin (Epi) can modify these alterations. We hypothesized that the ubiquitin ligases Murf and MAFbx would be increased in the obesogenic context, either by in utero obesogenic environment or by a postnatal high-fat diet, while the p70S6K-alpha kinase and its activation might be decreased. Eight groups of six male Wistar offspring formed eight experimental groups: control (C),

Effect of low-carbohydrate diets high in either fat or protein on thyroid function, plasma insulin, glucose, and triglycerides in healthy young adults.

A low-carbohydrate diet, frequently used for treatment of reactive hypoglycemia, hypertriglyceridemia, and obesity may affect thyroid function. We studied the effects of replacing the deleted carbohydrate with either fat or protein in seven healthy young adults. Subjects were randomly assigned to receive seven days of each of two isocaloric liquid-formula, low-carbohydrate diets consecutively. One diet was high in polyunsaturated fat (HF), with 10%, 55%, and 35% of total calories derived from protein, fat, and carbohydrate, respectively. The other was high in protein (HP) with 35%, 30%, and 35

Modelling maternal obesity: the effects of a chronic high-fat, high-cholesterol diet on uterine expression of contractile-associated proteins and ex vivo contractile activity during labour in the rat.

Maternal obesity is associated with prolonged and dysfunctional labour and emergency caesarean section, but the mechanisms are unknown. The present study investigated the effects of an adiposity-inducing high-fat, high-cholesterol (HFHC) diet on uterine contractile-associated protein (CAP) expression and ex vivo uterine contractility in term non-labouring (TNL) and term labouring (TL) rats. Female rats were fed either control chow (CON n=20) or HFHC (n=20) diet 6 weeks before conception and during pregnancy. On gestational day 21 (TNL) or day 22 (TL) CON and HFHC (n=10) rats were killed to det
