A Neurotensin Brake on Exploratory Drive under Persistent Threat.

Avoidance behavior is an adaptive response that delays exploration to promote survival. Avoidance is enhanced by psychological stress and is a hallmark of many neuropsychiatric disorders. Neural circuits that control avoidance by integrating stressful stimuli and modulating exploratory behavior remain underexplored. Elucidating the functional dynamics of this highly conserved phenomenon and the underlying neural mechanism of avoidance is an important open-ended question, with relevance to understanding both innate behaviors and neuropsychiatric disorders. Using predator odor as an innate, chro

SIRT1 deacetylates GAPDH to drive microglial glycolysis and neuroinflammation.

INTRODUCTION: Microglial activation drives neuroinflammation through a metabolic switch from oxidative phosphorylation to aerobic glycolysis; however, the molecular mechanisms governing this transition remain poorly defined. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH), sirtuin 1 (SIRT1), lipopolysaccharide (LPS), and interferon-gamma (IFN-γ) are central to this study; GAPDH plays plays a key regulatory role in this switch, and its activity is modulated by reversible acetylation at lysine 254 (K254). It remains unclear whether sirtuin deacetylases regulate this modification in microglia.

Progenitor T cells drive chronic pulmonary type 2 inflammation.

Type 2 inflammation is coordinated by remarkably durable CD4+ T helper 2 (TH2) cell responses, yet the cellular architecture that drives this chronic inflammation and prevents exhaustion remains poorly understood. To define the TH2 landscape in chronic type 2 inflammation, we established a mouse model of long-term pulmonary allergen exposure, finding that type 2 inflammation was broadly sustained over time and included an expanded T cell factor 1-expressing progenitor-like population. In vivo, lung TH2 progenitors were sufficient to both initiate and sustain type 2 inflammation, coupling self-

Clathrin adaptors drive phase separation in endocytosis and trafficking.

Liquid-liquid phase separation (LLPS) underlies the formation of biomolecular condensates that organize processes, including endocytosis and membrane trafficking. Although LLPS has been implicated in clathrin-mediated pathways, the contribution of adaptor proteins to condensate formation and function remains unclear. Here we show that two yeast adaptors, Ent5 and Sla2, undergo LLPS in vitro and this property correlates with their roles in membrane trafficking and condensate recruitment in vivo. Ent5 condensation is driven by a dynamic helix within its disordered region that acts as a molecular

How nanophotonics can drive optical computing toward practical applications.

The rapid advancement of artificial intelligence (AI) imposes unprecedented speed and energy requirements on large-scale computation. Owing to its intrinsic high bandwidth, massive parallelism capacity and low energy consumption, optical computing is widely regarded as one of the most promising technologies to address AI computation demands. Here we offer a synthesis of photonic platforms that have already shown programmability, high integration density and stability, and that can support large-scale AI models, edge computing, and logic and scientific computing. We summarize the requirements t

Eosinophils drive intestinal remodelling and innate defence in reproduction.

Mammalian reproduction requires substantial immune adaptations to safeguard reproductive success and to ultimately shape the evolutionary trajectories of a species. Systemic and placental immunity shift towards tolerance during pregnancy1,2; however, how maternal immunity adapts in barrier tissues-which are sites of frequent infection and inflammation-from pregnancy until the postpartum lactation period remains poorly understood. Here we report a previously unrecognized role for eosinophils, a type of granulocyte typically associated with allergies and helminth infections3,4, in remodelling th

Dominant clones leverage developmental epigenomic states to drive ependymoma.

ZFTA-RELA is the most recurrent genetic alteration seen in paediatric supratentorial ependymoma (EPN) and is sufficient to initiate tumours in mice1. Despite its oncogenic potential, ZFTA-RELA (ZR) is observed nearly exclusively in childhood EPN, with tumours located distinctly in the supratentorial brain of the central nervous system1. We proposed that specific chromatin modules accessible during brain development would render distinct cell lineage programs at direct risk of transformation by ZR. To test this hypothesis, we performed combined single-nucleus assay for transposase-accessible ch

Afferent Renal Nerves Drive Sympathoexcitation and Hypertensive Heart Failure.

BACKGROUND: Sympathetic activation plays a role in heart failure (HF) development. Afferent renal nerve input may induce sympathoexcitation via the hypothalamic paraventricular nucleus (PVN), which projects to the rostral ventrolateral medulla, a center for sympathetic regulation. Central dendritic release of vasopressin from PVN neurons reportedly stimulates neighboring presympathetic neurons, causing sympathoexcitation. This study investigated whether afferent renal nerves contribute to hypertensive cardiac dysfunction and whether the afferent renal nerve-PVN axis mediates sympathoexcitation

ZDHHC18-YTHDF2 axis suppresses USP13 to drive cardiac hypertrophy.

Cardiac hypertrophy, a significant pathological response to various stressors, often culminates in heart failure and necessitates the identification of novel regulatory targets for developing effective therapeutic strategies. In our study, an integrated transcriptomic and proteomic analysis revealed that deubiquitinating enzyme 13 (USP13) was significantly downregulated during cardiac hypertrophy. Subsequent functional experiments confirmed that USP13 overexpression markedly ameliorated transverse aortic constriction-induced cardiac dysfunction and attenuated cardiomyocyte hypertrophy, indicat

Capillary interactions drive the self-organization of bacterial colonies.

Many bacteria inhabit thin water layers on solid surfaces. These thin films occur both naturally - in soils, on hosts, on textiles - and in the lab on agar hydrogels. In these environments, cells the water menisci formed around bacteria lead to capillary attraction between cells while still allowing them to slide past one another. We develop an experimental apparatus that allows us to control bacterial collective experience capillary forces but it is unclear how these forces shape bacterial collective behavior. Here, we show that behavior by varying the strength and range of capillary forces.

Alpha synuclein overexpression can drive microbiome dysbiosis in mice.

Growing evidence indicates that persons with Parkinson disease (PD), have a unique composition of indigenous gut microbes. Given the long prodromal or pre-diagnosed period, longitudinal studies of the human and rodent gut microbiome before symptomatic onset and for the duration of the disease are currently lacking. PD is partially characterized by the accumulation of the protein α-synuclein (α-syn) into insoluble aggregates, in both the central and enteric nervous systems. As such, several experimental rodent and non-human primate models of α-syn overexpression recapitulate some of the hallmar

Microglia-drive IRF8 upregulates complement pathway in Parkinson's disease.

Parkinson's disease (PD) is a widespread degenerative disorder of the central nervous system. The gradual degeneration of dopaminergic neurons in the substantia nigra region is one of the primary pathological features of PD. Glial cells in SN are also linked to the pathological PD alterations. To discern the role of neurons and glial cells as well as their corresponding genetic modifications in PD, we utilized diverse bioinformatics techniques and performed biological experiments on cell and animal models. Several transcriptome datasets of the substantia nigra region were collected from the Ge

Nuclear Import Defects Drive Cell Cycle Dysregulation in Neurodegeneration.

Neurodegenerative diseases (NDDs) and other age-related disorders have been classically defined by a set of key pathological hallmarks. Two of these hallmarks, cell cycle dysregulation (CCD) and nucleocytoplasmic transport (NCT) defects, have long been debated as being either causal or consequential in the pathology of accelerated aging. Specifically, aberrant cell cycle activation in post-mitotic neurons has been shown to trigger neuronal cell death pathways and cellular senescence. Additionally, NCT has been observed to be progressively dysregulated during aging and in neurodegeneration, whe

Extracellular matrix anchored neutrophils drive pulmonary fibrosis in mice.

Pulmonary fibrosis (PF) is a progressive lung disease characterized by chronic immune dysregulation and excessive extracellular matrix (ECM) remodeling. Neutrophils, traditionally linked to acute inflammation, are increasingly recognized for their role in fibrosis. In this study, we identified a unique subset of neutrophils undergoing reverse transendothelial migration (rTEM) in the lungs of a silicosis mouse model induced by intratracheal SiO₂ instillation. Using single-cell RNA sequencing (scRNA-seq), spatial transcriptomics (ST), and ECM proteomics, we found that rTEM neutrophils were retai

How pathogens drive adipose tissue loss in the host.

Weight loss is a hallmark of many infections, including those caused by bacteria, fungi and parasites. This loss is often attributed to infection-induced anorexia and the need to mobilise energy from internal sources to cope with the pathogens. Weight loss during infection results from a significant reduction of muscle and fat mass, two organs that together account for approximately 60% of body mass in the healthy state. While muscle wasting is a well-documented aspect of infection-related weight loss, adipose tissue loss via lipolysis also plays a critical role and can determine disease outco

Using perioperative patient safety guidelines to drive systems change.

A European consensus study in this issue addresses perioperative patient safety recommendations by synthesising multiple patient safety guidelines covering the entire patient journey to provide a single point of reference. In this editorial we address what is considered as evidence and seek inclusion of broader ways of knowing and understanding the complexity of safety in healthcare. We suggest prioritising feasibility could limit improvements to specific clinical practices rather than systems-wide change. Safety is more than a collection of isolated recommendations. Safety interventions shoul

Arterial pulsations and transmantle pressure synergetically drive glymphatic flow.

Clearance of waste material from the brain by the glymphatic system results from net flow of cerebrospinal fluid (CSF) through perivascular spaces surrounding veins and arteries. In periarterial spaces, this bulk flow is directed from the cranial subarachnoid space towards the brain's interior. The precise pumping mechanism explaining this net inflow remains unclear. While in vivo experiments have shown that the pulsatile motion in periarterial spaces is synchronized with arterial pulsations, peristalsis alone has been deemed insufficient to explain bulk flow. In this study we examine an alter

Entamoeba histolytica extracellular vesicles drive pro-inflammatory monocyte signaling.

The parasitic protozoan Entamoeba histolytica secretes extracellular vesicles (EVs), but so far little is known about their function in the interaction with the host immune system. Infection with E. histolytica trophozoites can lead to formation of amebic liver abscesses (ALAs), in which pro-inflammatory immune responses of Ly6Chi monocytes contribute to liver damage. Men exhibit a more severe pathology as the result of higher monocyte recruitment and a stronger immune response. To investigate the role of EVs and pathogenicity in the host immune response, we studied the effect of EVs secreted
