New small-molecule drug design strategies for fighting resistant influenza A.

Influenza A virus is the major cause of seasonal or pandemic flu worldwide. Two main treatment strategies-vaccination and small molecule anti-influenza drugs are currently available. As an effective vaccine usually takes at least 6 months to develop, anti-influenza small molecule drugs are more effective for the first line of protection against the virus during an epidemic outbreak, especially in the early stage. Two major classes of anti-influenza drugs currently available are admantane-based M2 protein blockers (amantadine and rimantadine) and neuraminidase (NA) inhibitors (oseltamivir, zana

Structural analysis of trypanosomal sirtuin: an insight for selective drug design.

The infectious disease burden imposed by trypanosomatidae family continues to create burden in countries that are least equipped to bring new medicines to the clinic. For sickness caused by this family of parasites (African trypanosomiasis, Chagas disease, and leishmaniasis) no vaccines are available, and currently available drugs suffer from insufficient efficacy, excessive toxicity, and steady loss of effectiveness due to resistance. Availability of the genome sequence of pathogens of this family offers a unique avenue for the identification of novel common drug targets for all three pathoge

Structure and functionality in flavivirus NS-proteins: perspectives for drug design.

Flaviviridae are small enveloped viruses hosting a positive-sense single-stranded RNA genome. Besides yellow fever virus, a landmark case in the history of virology, members of the Flavivirus genus, such as West Nile virus and dengue virus, are increasingly gaining attention due to their re-emergence and incidence in different areas of the world. Additional environmental and demographic considerations suggest that novel or known flaviviruses will continue to emerge in the future. Nevertheless, up to few years ago flaviviruses were considered low interest candidates for drug design. At the star

Biological characteristics of dengue virus and potential targets for drug design.

Dengue infection is a major cause of morbidity in tropical and subtropical regions, bringing nearly 40% of the world population at risk and causing more than 20,000 deaths per year. But there is neither a vaccine for dengue disease nor antiviral drugs to treat the infection. In recent years, dengue infection has been particularly prevalent in India, Southeast Asia, Brazil, and Guangdong Province, China. In this article, we present a brief summary of the biological characteristics of dengue virus and associated flaviviruses, and outline the progress on studies of vaccines and drugs based on pot

Drug design targeting the main protease, the Achilles' heel of coronaviruses.

Coronaviruses (CoVs), a genus containing about 26 known species to date, cause highly prevalent diseases and are often severe or fatal in humans and animals. In 2003, a previously unknown coronavirus was identified to be the etiological agent of a global outbreak of a form of life-threatening pneumonia called severe acute respiratory syndrome (SARS). No efficacious therapy is currently available, and vaccines and drugs are under development to prevent SARS-CoV infection in many countries. The CoV main protease (M(pro)), which plays a pivotal role in viral gene expression and replication throug

The bacterial ribosome, a promising focus for structure-based drug design.

Recent crystal structures of the bacterial ribosome have identified the complex molecular interactions involved in antibiotic-ribosome recognition. Insights into the binding of aminoglycosides, macrolides, tetracyclines and other antibiotics provide opportunities for computational, structure-based approaches to be used in the design of appropriate modifications to existing antibiotics as well as in the discovery of completely new drug classes.

Nature-inspired novel drug design paradigm using nanosilver: efficacy on multi-drug-resistant clinical isolates of tuberculosis.

Despite discovery of the pathogen more than 100 years ago, tuberculosis (TB) continues to be a major killer disease worldwide. Currently a third of world population is infected and multiple-drug-resistant (mdr) TB registers maximum mortality by a single pathogen. Nanomedicine provides enormous opportunity for developing novel drugs. We have recently demonstrated surface-modified-lipophilic-nanosilica as drug to combat malaria and 100% lethal virus, BmNPV. Nanosilver possesses inherent antibacterial properties, but toxicity is a major concern. We hypothesized that capping with nature-inspired b

Exploring optimal drug targets through subtractive proteomics analysis and pangenomic insights for tailored drug design in tuberculosis.

Tuberculosis (TB), caused by Mycobacterium tuberculosis, ranks among the top causes of global human mortality, as reported by the World Health Organization's 2022 TB report. The prevalence of M. tuberculosis strains that are multiple and extensive-drug resistant represents a significant barrier to TB eradication. Fortunately, having many completely sequenced M. tuberculosis genomes available has made it possible to investigate the species pangenome, conduct a pan-phylogenetic investigation, and find potential new drug targets. The 442 complete genome dataset was used to estimate the pangenome

Fusion Protein Targeted Antiviral Peptides: Fragment-Based Drug Design (FBDD) Guided Rational Design of Dipeptides Against SARS-CoV-2.

Infectious diseases caused by viruses have become a serious public health issue in the recent past, including the current pandemic situation of COVID-19. Enveloped viruses are most commonly known to cause emerging and recurring infectious diseases. Viral and cell membrane fusion is the major key event in the case of enveloped viruses that is required for their entry into the cell. Viral fusion proteins play an important role in the fusion process and in infection establishment. Because of this, the fusion process targeting antivirals become an interest to fight against viral diseases caused by

DEG2MOL: Conditional Latent Flow Matching for Transcriptome-Guided de novo Drug Design.

Transcriptomic profiles capture cellular responses to perturbations, offering a principled foundation for biologically grounded de novo drug design. However, existing transcriptome-guided generative approaches remain limited in biological specificity, transcriptomic coverage, and evaluation across diverse perturbation contexts. Here, we propose DEG2MOL, a biologically grounded conditioning framework that encodes differentially expressed gene profiles through a Gene Ontology (GO)-informed encoder and maps this representation into the latent space of a frozen, pre-trained scaffold-aware VAE via

Drug repurposing-inspired design of miconazole-based 1,2,3-triazoles as antichagasic agents.

Drug-repurposing strategies have been widely explored to accelerate the development of new treatments for Chagas disease, leveraging compounds with previously established safety profiles. Among these, azole antifungals have attracted attention due to their inhibitory activity against 14-α-lanosterol demethylase (CYP51), an enzyme also present in Trypanosoma cruzi and essential for its ergosterol biosynthesis. Inhibition of this pathway compromises parasite membrane integrity and function and several azoles, including posaconazole, ravuconazole, and miconazole, have demonstrated significant in

Barrier-Oriented Design of Next-Generation Polymeric Nanocarriers for Targeted Drug Delivery.

Targeted drug delivery remains difficult because multiple biological barriers interfere with the stable transport of therapeutics to the site of action. Polymeric nanocarriers have gained broad attention as delivery platforms since their composition and surface properties can be adjusted to improve circulation behavior and cellular delivery. This review discusses the major biological barriers involved in targeted drug delivery and describes how polymeric nanocarriers are engineered to overcome them. Major carrier types, including polymeric nanoparticles and micelles, are considered with emphas

Computer-aided Drug Design for Alzheimer's Disease: Recent Advancements and Future Perspectives.

Alzheimer's disease (AD) is a neurodegenerative disorder marked by a decline in cognitive function and memory loss, primarily resulting from cholinergic dysfunction, the accumulation of amyloid plaques, the formation of tau tangles, and the progressive degeneration of neurons. While existing treatments offer limited symptomatic relief, they do not effectively halt or reverse the underlying progression of the disease, presenting a major global challenge in Alzheimer's research. Developing therapeutic strategies for AD remains complex, largely due to the inability of current medications to signi

Exposure Robustness by Design: a translational framework for oral drug product development.

Oral drug product development has traditionally been optimized around maximizing mean bioavailability in healthy volunteers, yet clinically relevant exposure liabilities including food effects, pH sensitivity, transit dependence and drug-drug interactions often emerge only after first-in-human (FIH) studies, driving label restrictions or reformulation. We introduce Exposure Robustness by Design (ERbD) - a prospective framework that reframes formulation development from maximizing mean bioavailability toward achieving exposure robustness across the physiological variability of the intended pati
