Doxorubicin-encapsulated thermosensitive liposomes modified with poly(N-isopropylacrylamide-co-acrylamide): drug release behavior and stability in the presence of serum.

In the field of the temperature sensitive drug delivery systems, we studied on the surface modification of liposomes by using poly(N-isopropylacrylamide-co-acrylamide) (PNIPAM-AAM) and polyethyleneglycol (PEG) to increase the release of doxorubicin (DOX) from liposomes and prolong the stability of liposomes in the presence of serum. The release of DOX from the PNIPAM-AAM/PEG modified liposomes is enhanced around the transition temperature of the polymer. In addition, the stability of the PNIPAM-AAM/PEG modified liposomes in serum shows a high level comparing with polymer unmodified liposomes.

Eco-friendly stability-indicating HPLC method development, validation, and LC-MS-based characterization of degradation products of the polar drug imeglimin.

Imeglimin (IMG) is a recently developed cyclic biguanide compound used for the management of hyperglycaemia. To comprehend the stability behavior of the polar drug IMG under various degradation environments, an HPLC method was developed, and the major degradation products (DPs) were characterized using mass-spectrometric studies. The mobile phase comprising a 20 mM potassium dihydrogen phosphate (KH2PO4) buffer (pH 3.1) along with ethanol in a simple isocratic mode (98:02, v/v) was employed for optimum resolution of the drug and its DPs using a reverse-phase Luna C18(2) (250 × 4.6 mm, 5 µm) co

Liposomes as versatile agents for the management of traumatic and nontraumatic central nervous system disorders: drug stability, targeting efficiency, and safety.

Various nanoparticle-based drug delivery systems for the treatment of neurological disorders have been widely studied. However, their inability to cross the blood-brain barrier hampers the clinical translation of these therapeutic strategies. Liposomes are nanoparticles composed of lipid bilayers, which can effectively encapsulate drugs and improve drug delivery across the blood-brain barrier and into brain tissue through their targeting and permeability. Therefore, they can potentially treat traumatic and nontraumatic central nervous system diseases. In this review, we outlined the common pro

Stability-indicating high-performance thin-layer chromatographic determination of levonorgestrel and ethinyloestradiol in bulk drug and in low-dosage oral contraceptives.

A stability-indicating high-performance thin-layer chromatography (HPTLC) method was developed and validated for simultaneous determination of steroidal hormones levonorgestrel and ethinyloestradiol both in bulk drug and in low-dosage oral contraceptives. Optimization of conditions for the spectrodensitometric procedure was reached by eluting HPTLC silica gel plates in a 10 cm x 10 cm horizontal chamber. The solvent system consisted of hexane-chloroform-methanol (1.0:3.0:0.25, v/v/v). This system was found to give compact, dense and typical peaks for both levonorgestrel (R(f)=0.65+/-0.03) and

Influence of pH on hydrolytic decomposition of diethylpropion hydrochloride: stability studies on drug substance and tablets using high-performance liquid chromatography.

The influence of pH on the hydrolysis of diethylpropion hydrochloride was investigated. Hydrolytic decomposition of the drug in solution at 45 degrees occurred at a very slow and constant rate at pH 3.5 and below but increased rapidly as the pH was raised above 3.5. Results of stability studies performed on six commercial tablet products from five manufacturers showed a similar relationship between the decomposition rate and the formulation pH. A reaction pathway based on the formation of an enamine structure is proposed for the pH-dependent hydrolysis. A high-performance liquid chromatographi

Physicochemical compatibility and stability of ketamine co-administered with selected supportive care drugs and parenteral nutrition in pediatric oncology.

PURPOSE: Ketamine has been previously administered as an anesthetic but recently gained importance as an analgesic in pediatric oncology. Despite its efficacy, continuous intravenous administration to children with cancer is interrupted when other essential drugs require infusion. This occurs because its compatibility with many drugs and lipid emulsions is unknown, causing inadequate pain management and patient discomfort. This study has assessed the physicochemical compatibility and stability of ketamine with selected drugs, based on usage frequency data, and lipid emulsion formulations admin

Surfactant-rescued gelation: Stabilizing P123-chitosan hydrogels in blood-isotonic aqueous solutions for controlled drug delivery.

The loss of gelation in Chitosan-Pluronic composite hydrogels under physiological saline conditions restricts their potential for drug delivery applications. To this effect, we present a dynamic and composite hydrogel system based on Pluronic P123 and chitosan, which restores gelation upon the addition of sodium dodecyl sulfate (SDS), even in the presence of NaCl at blood-isotonic concentrations. The entry of SDS induces electrostatic interactions and hydrophobic associations, enabling robust gel formation under physiological conditions. The gelation behavior can be desirably modified by varyi

Linc00347 suppresses ferroptosis in drug-resistant gastric cancer cells through stabilizing FoxO6 mRNA mediated by YBX1.

Chemoresistance remains a major therapeutic challenge in gastric cancer (GC). We report that the long non-coding RNA (lncRNA) Linc00347 is markedly upregulated in oxaliplatin (Oxa)-resistant GC tissues relative to Oxa-sensitive counterparts, and its elevated expression correlates with poor patient prognosis. Functionally, Linc00347 conferred Oxa resistance and accelerated tumor growth in vivo by repressing ferroptosis. Mechanistically, Linc00347 directly interacted with the RNA-binding protein YBX1 and prevented its proteasomal degradation by recruiting the deubiquitinase USP10. The stabilized

LMI-Enabled Absolutely Stabilizing PID Control of Pharmacological Systems for Closed-Loop Automated Intravenous Drug Administration.

OBJECTIVE: We developed a linear matrix inequality-enabled absolutely stabilizing proportional-integral-derivative control design approach for pharmacological systems applicable to intravenous drug administration. METHODS: We developed a proportional-integral- derivative control design approach that does not require detailed knowledge of the dose-response relationship other than its sector bound. It repetitively solves a set of linear matrix inequalities, which encapsulate the Lyapunov stability conditions against unknown dose-response relationship, over a broad proportional-integral-derivati

Mast cell stabilizer, an anti-allergic drug, reduces ventricular arrhythmia risk via modulation of neuroimmune interaction.

Mast cells (MCs) are important intermediates between the nervous and immune systems. The cardiac autonomic nervous system (CANS) crucially modulates cardiac electrophysiology and arrhythmogenesis, but whether and how MC-CANS neuroimmune interaction influences arrhythmia remain unclear. Our clinical data showed a close relationship between serum levels of MC markers and CANS activity, and then we use mast cell stabilizers (MCSs) to alter this MC-CANS communication. MCSs, which are well-known anti-allergic agents, could reduce the risk of ventricular arrhythmia (VA) after myocardial infarction (

Thermo-responsive Diels-Alder stabilized hydrogels for ocular drug delivery of a corticosteroid and an anti-VEGF fab fragment.

In the present study, a novel in situ forming thermosensitive hydrogel system was investigated as a versatile drug delivery system for ocular therapy. For this purpose, two thermosensitive ABA triblock copolymers bearing either furan or maleimide moieties were synthesized, named respectively poly(NIPAM-co-HEA/Furan)-PEG6K-P(NIPAM-co-HEA/Furan) (PNF) and poly(NIPAM-co-HEA/Maleimide)-PEG6K-P(NIPAM-co-HEA/-Maleimide) (PNM). Hydrogels were obtained upon mixing aqueous PNF and PNM solutions followed by incubation at 37 °C. The hydrogel undergoes an immediate (<1 min) sol-gel transition at 37 °C. In
