Increased dendritic inhibition of dentate gyrus granule cells in a mouse model of Down syndrome.

Down syndrome is the most common genetic neurodevelopmental disorder associated with mild-to-moderate intellectual disability. A disturbed excitation-inhibition balance is thought to be a major cause for the intellectual deficits in DS. In this study, we used patch-clamp electrophysiology, optogenetic stimulation and immunohistochemistry to investigate synaptic inhibition from specific interneuron subpopulations onto granule cells of the dentate gyrus in Ts65Dn mice. Optogenetically evoked inhibitory postsynaptic currents (IPSCs) from somatostatin (SOM) interneurons onto dendrites of granule c

A novel approach for in vivo screening of toxins using the Drosophila Giant Fiber circuit.

Finding compounds that affect neuronal or muscular function is of great interest as potential therapeutic agents for a variety of neurological disorders. Alternative applications for these compounds include their use as molecular probes as well as insecticides. We have developed a bioassay that requires small amounts of compounds and allows for unbiased screening of biological activity in vivo. For this, we paired administering compounds in a non-invasive manner with simultaneous electrophysiological recordings from a well-characterized neuronal circuit, the Giant Fiber System of Drosophila me

Aberrant protein S-nitrosylation contributes to hyperexcitability-induced synaptic damage in Alzheimer's disease: Mechanistic insights and potential therapies.

Alzheimer's disease (AD) is arguably the most common cause of dementia in the elderly and is marked by progressive synaptic degeneration, which in turn leads to cognitive decline. Studies in patients and in various AD models have shown that one of the early signatures of AD is neuronal hyperactivity. This excessive electrical activity contributes to dysregulated neural network function and synaptic damage. Mechanistically, evidence suggests that hyperexcitability accelerates production of reactive oxygen species (ROS) and reactive nitrogen species (RNS) that contribute to neural network impair

Injured adult retinal axons with Pten and Socs3 co-deletion reform active synapses with suprachiasmatic neurons.

Despite advances in promoting axonal regeneration after adult central nervous system injury, elicitation of a large number of lesion-passing axons reform active synaptic connections with natural target neurons remains limited. By deleting both Pten and Socs3 in retinal ganglion cells, we report that optic nerve axons after prechiasm lesion robustly reinnervate the hypothalamus, form new synapses with neurons in the suprachiasmatic nucleus (SCN), and re-integrate with the existing circuitry. Photic or electric stimulation of the retinal axons induces neuronal response in SCN. However both the i

[Two symptoms of elective paresis of individual hand muscles].

The author describes 2 symptoms of elective lesions of the hand muscles: one of the central genesis, the other--peripheral, vegetative. The first symptom is characterized by a weakening of volar and dorsal interossal muscles of the hand in an increased electrical activity, which is beyond the weakened muscles. It is due to a lesion of filogenetically new cortico-spinal path, adjacent to the pyramidal and participating in the innervation of the muscles of the distal parts of extremities. The second symptom is "the symptom of the fifth finger", which is characterized by a selective weakening and

Synaptic vesicle undocking induces low-frequency depression.

Synaptic depression is often interpreted as reflecting depletion of the readily releasable pool (RRP) following exocytosis. Such a mechanism predicts little or no depression at low stimulation frequency, as RRP replenishment should then offset the loss of vesicles by exocytosis. Nevertheless, in several types of mammalian central synapses, repetitive presynaptic stimulation at low frequency (<5 Hz) elicits synaptic depression (low-frequency depression or LFD). In the present work, we count the number of synaptic vesicles released at individual active zones to study the RRP and its replenishmen

Surface charge manipulation and electrostatic immobilization of synaptosomes for super-resolution imaging: a study on tau compartmentalization.

Synaptosomes are subcellular fractions prepared from brain tissues that are enriched in synaptic terminals, widely used for the study of neural transmission and synaptic dysfunction. Immunofluorescence imaging is increasingly applied to synaptosomes to investigate protein localization. However, conventional methods for imaging synaptosomes over glass coverslips suffer from formaldehyde-induced aggregation. Here, we developed a facile strategy to capture and image synaptosomes without aggregation artefacts. First, ethylene glycol bis(succinimidyl succinate) (EGS) is chosen as the chemical fixat

Potentials evoked by alvear tract in hippocampal CA1 region of rats. I. Topographical projection, component analysis, and correlation with unit activities.

1. The field potentials and unit activities evoked by the alvear tract (AT) in CA1 region of the dorsal hippocampus of rats were studied under sodium pentobarbital anesthesia. 2. The localized activity evoked anterior to an AT stimulus began as a compound action potential, followed by a slower negative wave, and ended in a long-lasting, slow positive wave. Observed with a 64-electrode recording array, topographical projections of the AT in CA1 were seen as parallel strips inclined at an angle of 5-30 degrees medially from the sagittal plane. 3. Three overlapping components in the averaged evok

Stack-Engineered Mode Selection in PtMn/(Co/Pd)n Multilayers Enables Deterministic Analog Spin-Orbit Torque Synapses.

Spin-orbit-torque (SOT) devices that support both binary and analog switching can bridge spintronic memory and neuromorphic computing, provided the switching mode can be deliberately assigned. Here, we demonstrate that in PtMn/(Co/Pd)n multilayers, the Co/Pd repeat number, nCo/Pd, serves as a material parameter that determines the reversal mechanism and switching mode. For nCo/Pd ≤ 7, magnetization reversal is governed by nucleation followed by domain-wall propagation, resulting in binary switching that can undergo a transition to analog behavior through electrical conditioning. For nCo/Pd ≥ 8

Differential synaptic depression mediates the therapeutic effect of deep brain stimulation.

Deep brain stimulation (DBS) effectively treats drug-resistant neurological and psychiatric disorders, yet its mechanisms remain unclear. Here we show that high-frequency DBS of the subthalamic nucleus (STN), a common target for Parkinson's disease (PD), activates afferent axons while inhibiting STN neurons. These contrasting presynaptic and postsynaptic effects arise from a decrease in local neurotransmitter release with a larger decrease in glutamate than GABA, shifting the excitation/inhibition balance toward inhibition. Chemogenetic inhibition, but not excitation, of STN neurons mimics the

Actions of general anaesthetics on synaptic transmission in the CNS.

This article reviews the actions of general anaesthetics on synapses in the mammalian central nervous system. It is shown that during general anaesthesia, anaesthetics act primarily on the chemical transmission process itself and do not affect the conduction of impulses in nerve axons or change the electrical excitability of neurones. Virtually all general anaesthetics depress excitatory synaptic transmission at concentrations required for surgical anaesthesia but some, notably the barbiturates, also increase the intensity of inhibitory synaptic transmission. Detailed analysis has found that s

Uncoupling of electrotonic synapses by calcium.

The degree of axo-axonal synaptic coupling between nerve cells in the buccal ganglion of Navanax was investigated in relation to intracellular ionic calcium. Increasing intracellular Ca2+ by injection of Ca2+, injection of Na+, or application of ionophore X537A produced uncoupling after at least 90 min, if metabolic inhibitor was present in the medium. Subsequent removal of the metabolic inhibitor reestablished the coupling in less than 30 min. Injected Sr2+ also mimicked the uncoupling action of Ca2+. The presence of a metabolic inhibitor alone had no effect on the coupling. These results lea

Magnetic Domain-Wall Racetrack Grating as a Multichannel Analog Optoelectronic Synapse.

Electrical manipulation of magnetic domain wall motion and its applications constitute a long-standing central topic of spintronics. Recent enthusiasm for magnetically tunable diffractive optics calls for a fast and efficient method of device reconfiguration. Here we develop a chiral domain-wall racetrack grating device whose periodic magnetization structures can be electrically reconfigured by spin-orbit torques without the assistance of external magnetic fields, demonstrating a fast, efficient, and continuous tuning of optical diffraction. We further show that these unique features can be ut

In vitro studies on GABA release.

1 Recent studies have demonstrated growing evidence for a primary action of the benzodiazepines on gabaminergic neurones which induces a facilitation of gamma-aminobutyric acid (GABA)-mediated neurotransmission. As enhancement of GABA release has been suggested to account for their activation of GABA mechanisms, the effect of diazepam and clobazam, and of several other psychotropic drugs, on stimulated GABA release have been studied. 2 Using rat brain cortex slices saturated with [3H]-GABA, the electrically stimulated overflow of GABA is reduced in a concentration-dependent manner in the prese

Hippocampal Commissural Circuitry Shows Asymmetric cAMP-Dependent Synaptic Plasticity.

Hemispheric asymmetries in NMDAR-dependent synaptic plasticity have been described in hippocampal area CA1, but it remains unclear whether similar lateralized mechanisms exist for cyclic adenosine monophosphate (cAMP)-dependent plasticity. Here, we investigated whether cAMP-mediated potentiation of synaptic transmission in mouse CA1 exhibits hemisphere-specific properties. In recordings with electrical stimulation of CA1 inputs, a subset of recordings in the left, but not in the right hemisphere CA1, exhibited a pronounced cAMP-induced potentiation of field excitatory postsynaptic potentials (

Ultrastructural localization of connexins (Cx36, Cx43, Cx45), glutamate receptors and aquaporin-4 in rodent olfactory mucosa, olfactory nerve and olfactory bulb.

Odorant/receptor binding and initial olfactory information processing occurs in olfactory receptor neurons (ORNs) within the olfactory epithelium. Subsequent information coding involves high-frequency spike synchronization of paired mitral/tufted cell dendrites within olfactory bulb (OB) glomeruli via positive feedback between glutamate receptors and closely-associated gap junctions. With mRNA for connexins Cx36, Cx43 and Cx45 detected within ORN somata and Cx36 and Cx43 proteins reported in ORN somata and axons, abundant gap junctions were proposed to couple ORNs. We used freeze-fracture repl

Effects of reduced vesicular filling on synaptic transmission in rat hippocampal neurones.

The consequence of reduced uptake of neurotransmitters into synaptic vesicles on synaptic transmission was examined in rat hippocampal slices and culture using bafilomycin A1 (Baf), a potent and specific blocker of the vacuolar-type (V-type) ATPase, which eliminates the driving force for the uptake of both glutamate and GABA into synaptic vesicles. After incubation with Baf, both the amplitude and frequency of GABAergic miniature inhibitory postsynaptic currents (mIPSCs) were reduced in the slice preparation. Similar effects were seen with glutamatergic miniature excitatory postsynaptic curren

The olivocochlear reflex strength and cochlear sensitivity are independently modulated by auditory cortex microstimulation.

In mammals, efferent projections to the cochlear receptor are constituted by olivocochlear (OC) fibers that originate in the superior olivary complex. Medial and lateral OC neurons make synapses with outer hair cells and with auditory nerve fibers, respectively. In addition to the OC system, there are also descending projections from the auditory cortex that are directed towards the thalamus, inferior colliculus, cochlear nucleus, and superior olivary complex. Olivocochlear function can be assessed by measuring a brainstem reflex mediated by auditory nerve fibers, cochlear nucleus neurons, and
