Bladder cancer poses significant clinical challenges due to its high metastatic potential and poor prognosis, especially when it progresses to muscle-invasive stages. Here, we show that the m6A reader YTHDC1 is downregulated in muscle-invasive bladder cancer and is negatively correlated with the expression of epithelial‒mesenchymal transition genes. The functional inhibition or depletion of YTHDC1 increased the migration and invasion of urothelial cells. Integrative analysis of multimodal sequencing datasets provided detailed insights into the molecular mechanisms mediating YTHDC1-dependent ph

Cardiovascular diseases (CVDs) remain the leading cause of death worldwide. Their regulation involves not only classical genetic mechanisms but also dynamic epitranscriptomic control. The fat mass and obesity-associated protein (FTO), an N6-methyladenosine (m6A) RNA demethylase, has been implicated in cardiovascular disease. Evidence shows that the role of FTO in CVDs is strongly context dependent, with both protective and harmful effects reported in different settings. This review summarizes the genetic, molecular, and epitranscriptomic features of FTO and presents a framework in which FTO ac

The dynamic and reversible N6-methyladenosine (m6A) RNA modification has emerged as a pivotal layer of epitranscriptomic regulation in cancer, governing RNA metabolism and function. Concurrently, deubiquitylating enzymes (DUBs), key components of the post-translational modifications (PTMs) system, orchestrate protein stability and signaling pathways crucial for tumorigenesis. Recent evidence reveals extensive and intricate crosstalk between these two regulatory systems, forming a complex network that can influence cancer progression. This review systematically explores the bidirectional interp

N6-methyladenosine (m6A) is the most abundant internal epitranscriptomic modification in eukaryotic mRNA. It dynamically regulates RNA splicing, transport, stability, and translation efficiency through the "Writers-Erasers-Readers" system, thereby playing a broad role in gene expression regulation. Skeletal muscle, a key metabolic and locomotor organ, undergoes precise m6A-mediated regulation during development, regeneration, homeostasis, and aging. In this review, we systematically summarize the composition and function of the m6A modification system, with a focus on its critical roles in ske

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by persistent synovial inflammation and progressive joint destruction. Although N6-methyladenosine (m⁶A) modification is increasingly recognized as an important epitranscriptomic regulatory mechanism, its role in RA remains incompletely understood. This study integrated transcriptomic analysis, m⁶A methylome profiling, and functional experiments to investigate the potential role of m⁶A-associated growth arrest-specific 6 (GAS6) in RA. Synovial tissue samples from patients with RA and healthy controls (HC) were subjected to

RNA modifications represent a dynamic layer of gene expression regulation, RNA stability, and translation with profound implications for cellular function and disease. However, the critical regulation and functions of RNA-modifying proteins (RMPs) remain poorly understood. Here, we present a large-scale characterization of RMPs through 378 multiomics datasets encompassing genomics, bulk and single-cell transcriptomics, epitranscriptomics, proteomics, and posttranslational modifications (PTMs) across 63 human tissues. Our analysis of experimental perturbations of RMPs revealed dynamic different

Systemic sclerosis (SSc) is characterized by progressive fibrosis driven by persistent activation of dermal fibroblasts, yet the epigenetic and epitranscriptomic mechanisms sustaining fibroblast pathogenicity remain incompletely defined. Here, we investigated the role of N-acetyltransferase 10 (NAT10)-mediated N4-acetylcytidine (ac4C) RNA modification in fibroblast activation and fibrotic remodeling. Integrative analyses of single-cell transcriptomics, human SSc skin, in vitro assays, in vivo models, and fibroblast-specific knockout mice revealed that NAT10 is upregulated in pathogenic fibrobl

Pancreatic α-cells are central regulators of glucose and amino acid homeostasis, yet the mechanisms that preserve α-cell identity and function remain incompletely understood. N6-methyladenosine (m6A) is a widespread mRNA modification that is essential for β-cell biology and pancreatic endocrine differentiation. Here we show that m6A is a key regulator of α-cell function and plasticity. In α-cells, metabolic cues that stimulate glucagon secretion such as L-arginine increase METTL3, METTL14 and m6A levels. Loss of m6A impairs amino acid-stimulated glucagon secretion, disrupts α-cell identity pro

N6-methyladenosine (m6A) RNA modification has emerged as a pivotal epitranscriptomic mark shaping RNA metabolism and cellular programs. Among the diverse m6A reader proteins, YTHDF2 has garnered significant attention as a key regulator of RNA stability. Recent studies highlight the multifaceted roles of YTHDF2 in orchestrating hematopoietic stem cell self-renewal and differentiation, fine-tuning immune cell development and function, and modulating tumor-immune interactions within the tumor microenvironment. YTHDF2 has also been shown to function as a reader for RNA 5-methylcytosine (m5C) modif

RNA modifications, particularly in the realm of epitranscriptomics, have emerged as crucial regulatory factors in cancer progression, influencing cancer cell behavior by reprogramming metabolic pathways. These modifications, such as N⁶-methyladenosine (m⁶A), 5-methylcytosine (m⁵C), N¹-methyladenosine (m¹A), N⁷-methylguanosine (m⁷G), N⁴-acetylcytidine (ac⁴C) and pseudouridine (Ψ), modulate mRNA stability, splicing, translation efficiency, and non-coding RNA function, thereby fine-tuning the expression of metabolic enzymes and transporters. Importantly, these marks converge on shared metabolic e

Lung cancer is the leading cause of cancer-related mortality worldwide, and its tumorigenesis involves the accumulation of genetic and epigenetic events in the respiratory epithelium. Epigenetic modifications, such as DNA methylation, RNA modification, and histone modifications, have been widely reported to play an important role in lung cancer development and in other pulmonary diseases. Whereas the functionality of DNA and chromatin modifications referred to as epigenetics is widely characterized, various modifications of RNA nucleotides have recently come into prominence as functionally imp

RNA 5-methylcytosine (m5C) is an emerging epitranscriptomic modification implicated in the progression of multiple cancers, yet the landscape of m5C in bladder cancer (BCa) and underlying regulatory mechanisms remains largely elusive. In this study, we identified NOP2, an RNA methyltransferase, as a key driver of BCa progression. NOP2 was significantly upregulated in BCa tumors and associated with poor prognosis. Functionally, NOP2 promoted cell proliferation and invasion, enhancing tumor growth in xenograft models. Mechanistically, NOP2-mediated m5C deposition facilitates the recruitment of t

Plasmodium falciparum artemisinin (ART) resistance is driven by mutations in kelch-like protein 13 (PfK13). Quiescence, a key aspect of resistance, may also be regulated by a yet unidentified epigenetic pathway. Transfer RNA modification reprogramming and codon bias translation is a conserved epitranscriptomic translational control mechanism that allows cells to rapidly respond to stress. We report a role for this mechanism in ART-resistant parasites by combining tRNA modification, proteomic and codon usage analyses in ring-stage ART-sensitive and ART-resistant parasites in response to drug. Different organs undergo distinct transcriptional, epigenetic and physiological alterations that guarantee their functional maturation after birth. However, the roles of epitranscriptomic machineries in these processes have remained elusive. Here we demonstrate that expression of RNA methyltransferase enzymes Mettl3 and Mettl14 gradually declines during postnatal liver development in male mice. Liver-specific Mettl3 deficiency causes hepatocyte hypertrophy, liver injury and growth retardation. Transcriptomic and N6-methyl-adenosine (m6A) profiling identify the neutral sphingomyelinase, Smpd3,

RNA methylation, notably m6A modification, is a predominant epitranscriptomic alteration in mRNA, yet its evolutionary properties and the selective constraints acting on it across mammals remain poorly understood. Here, we generated a single-base-resolution m6A modification atlas in liver, kidney, and brain tissues across 21 non-model mammals using Nanopore direct RNA sequencing. We found that 25.54-35.70% of orthologous transcripts across examined species harbor m6A modifications, with m6A-modified sites exhibiting significantly greater conservation than nearby unmodified regions, probably un

Male infertility accounts for approximately 50% of infertility cases, with nearly 30% remaining unexplained after standard evaluations. This highlights the need for a better understanding of sperm function to advance diagnostic and therapeutic strategies. Sperm were long considered translationally quiescent; however, emerging evidence suggests that sperm translate messenger RNAs (mRNAs) to synthesize proteins crucial for sperm functions. However, mRNA translation during capacitation remains poorly understood, despite its potential importance for fertility diagnostics. RNA-binding proteins (RBP

BACKGROUND: N6-methyladenosine (m6A), the most prevalent and reversible post-transcriptional RNA modification, is involved in the progression of various diseases. Nonetheless, the role of m6A modification in Tuberculosis (TB) pathogenesis remains unknown. Here, we investigated the general expression patterns and potential functions of m6A regulators in TB.
METHODS: The differentially expressed m6A genes between the healthy and TB groups were evaluated using the public Gene Expression Omnibus (GEO) database, and quantitative real-time PCR (qRT-PCR) was used to test the expression of key m6A re

Several strategies, including chemotherapy and radiotherapy, have improved therapeutic outcomes among cancer patients in clinical practice. However, due to their heterogeneity, cancer cells frequently display primary or acquired therapeutic resistance, thereby resulting in treatment failure. The mechanisms underlying cancer therapeutic resistance are complex and varied. Among them, N6-methyladenosine (m6A) RNA modification has gained increasing attention as a potential determinant of therapy resistance within various cancers. In this review, we primarily describe evidence for the effect of the

Hepatocellular carcinoma (HCC) is a highly lethal malignant tumor with substantial heterogeneity, posing formidable challenges to overcoming drug resistance. Accumulating evidence highlights the pivotal role of epitranscriptomic regulation, particularly N6-methyladenosine (m6A) modification, in reshaping tumor drug resistance. This review elaborates on the dual functions and mechanisms of m6A erasers FTO and ALKBH5 in HCC drug resistance. In cancer cells, these erasers post-transcriptionally regulate key transcripts to orchestrate metabolic reprogramming, enhance anti-apoptotic signaling, and
